        한국인에서 5,10-Methylenetetrahydrofolate Reductase 유전자 다형성 분석 및 임신의 결과에 미치는 영향

        목적: 본 연구는 한국인 임신 여성에서 MTHFR 유전자형 변이와 혈청내 호모시스테인 및 엽산의 농도와 이들이 임신에 미치는 영향을 알아보고자 하였다. 방법: 임신부 600명에게서 혈액을 채취하여 얻어진 DNA로 MTHFR C677T와 A1298C 유전자의 다형성을 PCR-RELP로 분석하였고 혈청내 호모시스테인은 HPLC 방법으로, 엽산은 RIA 방법으로 측정하였다. 결과: 혈청내 호모시스테인은 MTHFR C677T 유전자형이 C/C형이나 C/T인 군에 비해 T/T인 군에서 유의하게 높았으며 MTHFR A1298C 유전자형이 A/C형이나 C/C인 군에 비해 A/A인 군에서 역시 유의하게 높았다( p<0.05). 혈청 호모시스테인은 모든 MTHFR 유전자형에서 혈청 엽산농도와 역상관관계를 보였으나, 변이형 유전자형(T/T와 A/A유전자형)에서 더 큰 역 상관관계를 보였다. 15?mol/L 이상의 고호모시스테인혈증의 임신부에서 임신 주수와 출생체중이 36.1주, 3053.8 g으로 15 ?mol/L미만인 군의 38.3 주, 3215.3 g 보다 유의하게 낮았다( p<0.05). 결론: 혈청 호모시스테인의 농도는 MTHFR C677T와 A1298T 유전자의 다형성에 의해 영향을 받으며, MTHFR C677T와 A1298T 유전자의 다형성과 혈청 호모시스테인은 모두 임신주수와 신생아의 체중에 영향을 미친다. Objective: The purpose of this study was to analyze MTHFR polymorphism among the Korean population and to evaluate the relationship between serum levels of homocysteine and MTHFR polymorphism and also to investigate the effect on pregnancy outcomes. Methods: DNA was extracted from whole blood of 600 pregnant women. All samples were genotyped for the C677T and A1298C polymorphisms in MTHFR gene by PCR-RELP assay. Serum levels of homocysteine and folate were measured by high performance liquid chromatography for homocysteine and radioassay for folate. Pregnancy outcomes were estimated by gestational weeks and birth weights of newborns. Results: Serum homocysteine was higher in women with the T/T genotype than those with the C/T or C/C genotype of the MTHFR C677T polymorphism (p<0.05). And also serum homocysteine was higher in women with the A/A genotype than those with the A/C or C/C genotype of the MTHFR A1298C polymorphism (p<0.05). Serum homocysteine was negatively correlated with serum folate in all MTHFR genotypes, especially prominent in T/T genotype of MTHFR C677T polymorphism and A/A genotype of MTHFR A1298C polymorphism. Gestational age and the birth weight of infant from hyperhomocysteinemic mothers whose homocysteine levels higher than 15 μmol/L were 36.1 weeks, 3053.8 g, respectively, which were significant lower than those from normohomocysteinemic mothers (38.3 weeks, 3,215.3 g) (p<0.05). Conclusion: Serum homocysteine was influenced significantly by MTHFR C677T polymorphism and MTHFR A1298C polymorphism. MTHFR C677T and A1298C polymorphism and serum homocysteine levels affect pregnancy outcomes, although not mainly by serum folate level.

        소화기 ; 대전 지역에서 급성 A형 간염의 유전자형에 따른 임상 특성 고찰

        목적: B형 간염이나 C형 간염 등은 유전자형에 따른 차이가 알려져 있지만, 국내에서 증가하는 IIIA 유전자형과 기존의 IA 유전자형에 의한 간염의 차이가 거의 알려진 바 없다. 따라서 대전 지역에서 발생한 급성 A형 간염의 유전자형에 따른 차이를 알고자 한다. 방법: 2009년 11월부터 2010년 6월까지 급성 A형 간염으로 진단되면서 유전자형이 확인된 81명의 환자를 대상으로 임상적 소견들과 검사실 소견들을 전향적으로 연구하였다. 결과: 평균 나이는 32.6±7.4세이며, 평균 입원 기간은 7.7±2.4일이었다. 직업은 서비스직, 사무직 그리고 교육직이 가장 많았다. 유전자형 간의 급성 간염에 따른 증상들의 종류는 별다른 차이가 없었다. 유전자형에 따른 증상의 빈도는 IIIA 유전자형에서 구토와 설사가, IA 유전자형에서는 복통과 피부 반점이 상대적으로 많았다. 검사실 소견에서는 IIIA 유전자형에서 헤모글로빈의 평균, CRP 평균, AST 최대 수치 평균은 상대적으로 유의하게 높았다. 그리고 AST와 ALT 및 총 빌리루빈 최대 수치들과 INR의 수치가 IIIA 유전자형에서 상대적으로 높게 분포하는 경향을 보였다. 결론: 대전 지역에서 유행하는 급성 A형 간염에서 IIIA 유전자형에 따른 급성 간염은 기존의 IA 유전자형과 차이가 있을 것으로 보이며, 다기관 연구를 통한 심도 깊은 연구가 필요하겠다. Background/Aims: Acute viral hepatitis A is a major health problem in Korea and the influx of genotype IIIA is thought to be one reason. We examined the differences in the clinical characteristics and laboratory findings of genotypes IA and IIIA in Daejeon. Methods: From November 2009 to June 2010, 81 patients positive for IgM anti-HAV were enrolled prospectively. The hepatitis A was genotyped using real-time polymerase chain reaction. The clinical characteristics and laboratory results were compared on the Results: The mean patient age was 32.6±7.4 years. The mean hospitalization was 7.7±2.4 days. The patient occupation varied. Clinically, vomiting and diarrhea were relatively more prevalent in genotype IIIA than in IA. Abdominal pain and skin spots were relatively more prevalent in genotype IA than in IIIA. The hemoglobin, peak aspartate aminotransferase (AST) level, and C-reactive protein were statistically higher in genotype IIIA than in IA. The distributions of the peak AST, alanine aminotransferase (ALT) and total bilirubin values tended to be perched in genotype IIIA than in IA. The international normalized ratio (INR) tended to be slightly prolonged in genotype IIIA than in IA. Conclusions: Recently, genotype IIIA of acute viral hepatitis A has become prevalent in Daejeon. Hepatitis A genotype IIIA probably causes worse laboratory abnormalities than genotype IA.

        교반 및 정치배양에 따른 사과식초의 품질특성

        국내산 사과를 원료로 일체의 첨가물을 사용하지 않고 교반 및 정치배양법으로 각각의 사과식초를 제조하여 품질을 비교하였다. 사과식초의 pH는 초산발효기간이 지남에 따라 낮아졌으며, 총산은 증가하였다. 교반배양 사과식초(A)는 발효 8일째에 6.08%, 정치배양 사과식초(B)는 발효 60일째에 5.20%로 나타나 교반배양법(A)으로 제조한 사과식초가 총산은 높게 나타났으며, pH와 당도는 식초 간에 큰 차이는 없었다. L값은 정치배양 사과식초(B)가 교반배양 사과식초 (A)에 비해 더 낮게 나타났으나 a값, b값은 높게 나타났다. 유기산 함량은 교반배양 사과식초(A)에서는 acetic acid만 높았으나 정치배양 사과식초(B)에서는 malic acid, citric acid 및 succinic acid도 높게 나타났다. 유리당은 교반배양 사과식초(A), 정치배양 사과식초(B) 모두에서 fructose, glucose가 검출되었다. 사과식초의 관능검사 결과, 맛과 전반적 기호도에서 정치배양 사과식초(B)가 가장 높은 평점을 얻었다. 이상의 결과 전통적인 정치배양법으로 고품질의 사과식초 제조가 가능할 것으로 기대된다. Quality characteristics of apple vinegar by using agitated and static cultures without any additive were compared. pH was reduced with passage of acetic acid fermentation time. Total acidity of the agitated culture vinegar (A) was 6.08% at the 8 day according to the progress of fermentation and that of the static culture vinegar (B) recorded 5.20% at the 60 day of fermentation. There was no significant difference in sugar content of (A) and (B). L value was lower in (B) than (A) but a and b values were higher in (B) than (A). Organic acid content of (A) was high only in acetic acid but malic, citric and succinic acids appeared high in (B). As free sugar, fructose and glucose were observed in both of them. In sensory examination results, (B) showed the highest taste and overall preferences. From all of these results traditional static culture are expected to be used to make high-quality vinegar.

        <P><B>Abstract</B></P><P>Unregulated activation of microglia is a key risk factor contributes to neurodegenerative diseases and suppression of this phenomenon is considered as a potential therapeutic target. The compound isolated from sea horse <I>Hippocampus kuda</I> Bleeler; 1-(5-bromo-2-hydroxy-4-methoxyphenyl)ethanone [SE1] was characterized for its ability in suppressing LPS mediated activation of murine BV-2 cells. Despite the presence of various active molecular groups in the structure, SE1 has not well explored for its biological activities. The outcome of this study clearly indicated that SE1 inhibited the production of inflammatory mediators; nitric oxide, prostaglandin E<SUB>2</SUB> and pro-inflammatory cytokines. Furthermore, it inhibited the protein and gene expression levels of inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β and interleukin-6. The responsible signaling mechanisms leading to these inhibitions were identified as SE1 mediated blocking of phosphorylation of mitogen activate protein kinase (MAPK) molecules; C-jun-N-terminal kinase (JNK), p38 and nuclear translocation of nuclear factor-κB (NF-κB) p65 and p50 subunits. These results suggest that SE1 has the potential to be further developed as therapeutic against neuro-inflammation.</P> <P><B>Graphical abstract</B></P><P><ce:figure id='f0045'></ce:figure></P>

        The whole wheat effect and refined with E-selectin polymorphism on breast cancer

        Wheat is rich in sources of fiber, oligosaccharides, and resistant starch, simple carbohydrates which may have a protective role against carcinoma. Additionally, Whole wheat/bran as well includes contains phytochemicals such as flavonoids, lignans, folate, phytosterols, phenolic acids, and tocols. The above phytochemicals suitable forms antioxidant and cholesterol-reducing activities. Phytoestrogens are regarded as especially essential in the preventative measures of hormonally dependent malignancies such as breast cancer (BC). In this study lowered BC risk has been associated with whole grain/bran consumption with an odds ratio (OR=0.24 and 95 %CI=0.10-0.56). Wheat/bran appears to have a reliable protective impact against BC. While intake of white bread has been associated with a high risk of BC (OR=2.63 and CI 95 %=1.07-6.48). Also, the E-Selectin (SELE) Single nucleotide polymorphisms (SNPs) rs5353 A/G and rs932307 C/T were investigated using the sanger sequences approach. There was a positive association between genotypes (rs5353 GG+AG) and (rs932307 TT+CT) with rate consumption of wheat-white/bread and these genotypes were more frequent in patients had BC in comparison with a significant difference (P=0.03) (P=0.01), respectively. The genotypes (GG+AG) frequency of the rs5353 polymorphism and (TT+CT) genotypes of the rs932307 polymorphism in the present study had a high risk of cancer with (OR=3.05), (OR=4.17) respectively. While these genotypes showed no significant association with the rate of whole grain consumption in patients and control. Therefore, the type of wheat may associate with increased incidence of disease-related with type of Polymorphism because some present genotypes of SNPs showed high (OR) which may refer to their positive associated with disease, the white wheat consumption may active the risky association between SNPs and BC.

        The ubiquitin conjugating enzyme E2 (UBC E2) mediates selective ubiquitination, acting with E1 and E3 enzymes to designate specific proteins for subsequent degradation. In the present study, we characterized the function of the mung bean VrUBC1 gene (Vigna radiata UBC 1). RNA gel-blot analysis showed that VrUBC1 mRNA expression was induced by either dehydration, high salinity or by the exogenous abscisic acid (ABA), but not by low temperature or wounding. Biochemical studies of VrUBC1 recombinant protein and complementation of yeast ubc4/5 by VrUBC1 revealed that VrUBC1 encodes a functional UBC E2. To understand the function of this gene in development and plant responses to osmotic stresses, we overexpressed VrUBC1 in Arabidopsis (Arabidopsis thaliana). The VrUBC1-overexpressing plants displayed highly sensitive responses to ABA and osmotic stress during germination, enhanced ABA- or salt-induced stomatal closing, and increased drought stress tolerance. The expression levels of a number of key ABA signaling genes were increased in VrUBC1-overexpressing plants compared to the wild-type plants. Yeast two-hybrid and bimolecular fluorescence complementation demonstrated that VrUBC1 interacts with AtVBP1 (A. thaliana VrUBC1 Binding Partner 1), a C3HC4-type RING E3 ligase. Overall, these results demonstrate that VrUBC1 plays a positive role in osmotic stress tolerance through transcriptional regulation of ABA-related genes and possibly through interaction with a novel RING E3 ligase.

        녹용의 단백질가수분해 및 추출조건에 따른 특성 변화

        녹용을 식품소재로 활용하고자 단백질 가수분해조건 설정 및 추출조건에 따른 품질특성을 조사하였다. 효소제 종류에 따른 영향을 조사한 결과 효소제 종류에 따른 pH는 차이는 없었으나, °Brix, 고형분 및 조단백질 함량은 ProteAX(A)에서 높게 나타났다. 효소제 A 농도에서는 0.5%(w/w)까지 °Brix, 고형분 및 조단백질 함량은 증가하였으며, 1.0%(w/w 이상에서는 큰 변화가 없었다. 가수분해 시간은 5시간까지 °Brix, 고형분 및 조단백질 함량은 증가하였으나 그 후 큰 변화가 없어 가수분해 시간을 5시간으로 설정하였다. 녹용에 효소 A와 C를 각각 0.5%(w/w)씩 혼합첨가 하여 60℃에서 5시간 가수분해할 때 고형분 및 단백질 함량이 가장 높게 나타났다. 상기 설정된 조건으로 가수분해 시킨 녹용가수분해물(DAH)과 대조구(DA)를 95℃ 상압조건(AP, 6~18시간) 및 120℃ 가압조건(UP, 15~60분)으로 각각 추출한 결과 pH는 효소처리 및 추출조건에 따른 큰 차이는 없었다. °Brix는 DA 1.5°Brix, DA-UP 및 DAH-AP에서는 2.2°Brix로 나타났으며, DAH-UP에서 60분 추출하였을 때 2.7°Brix로 가장 높게 나타났다. 또한 DAH-UP에서는 가용성 고형분 1.97%, 조단백질 함량 742.7 ㎎/100 g, 콜라겐 함량 498.8 ㎎/100 g으로 가장 높게 나타났다. 이상의 결과 가압추출조건이 녹용의 품질특성 향상에 더 효과적인 것으로 나타났으며, 향후 다양한 소재로 활용이 기대되었다. This study was conducted to investigate the proteolysis and extraction conditions of deer antler for application of food materials. ProteAX (A) was the most effective enzyme for proteolysis of deer antler and the proteolysis condition was 0.5% (w/w) for enzyme concentration and 5 hr for proteolysis time. The effect of mixing enzyme ProteAX (A)+KFEN 2 (C) treatment in 60℃, 5 hr was investigated; soluble solid and protein content were the highest with A 0.5% (w/w) and B 0.5% (w/w) concentration. Result for DAH (deer antler hydrolysate) and DA (deer antler) prepared with extraction in 95℃ atmospheric pressure (AP, 6~18 hr) and extraction under 120℃ pressure condition (UP, 15~60 min) after hydrolysis on preceding established condition descriptions indicated that difference in pH according to enzyme treatment and extraction conditions was not significant. Sugar content of DA was 1.5oBrix, DA-UP (under pressure) and DAH-AP (atmospheric pressure) were 2.2°Brix; the highest sugar content of 2.7°Brix was observed in DAH-UP for 60 min extraction. Also total free sugar, crude protein and collagen content were the highest in DAH-UP for 60 min recording at 1.97%, 742.7 ㎎/100 g and 498.8 ㎎/100 g, respectively. From these results, deer antler hydrolysate prepared with extraction under pressure was the most effective for functional characteristics enhancement. Hereafter, various practical uses of materials with enhanced characteristics of antler is expected.

        Suppressive Effect of Fermented Angelica tenuissima Root Extract against Photoaging: Possible Involvement of Hemeoxygenase-1

        Angelica tenuissima root has historically been used as a traditional medicine in Korea. Previous studies have identified the anti-melanogenic effects of the extract of A. tenuissima root fermented by Aspergillus oryzae (FAT). This study investigated the protective effects of FAT against ultraviolet light B exposure (UVB; 30 mJ/㎠) in HaCaT (human keratinocyte) or Hs68 (human foreskin fibroblast) skin cells. FAT treatment was able to stimulate wound healing rate at the basal condition. FAT also favored the maintenance and/or improvement of extracellular matrix impairment caused by UVB irradiation through: 1) upregulation of procollagen Type-1 synthesis and secretion; 2) suppression of MMP-1 and elastase expression. FAT was able to play a role in the attenuation of inflammatory responses caused by UVB irradiation via upregulation of photo-protective hemeoxygease-1 and suppression of proinflammatory cyclooxygenase-2 expression. After further verification of the anti-photoaging potential of FAT, it could be utilized as an effective ingredient in anti-aging and anti-wrinkle cosmetics.
