Proceedings of International Symposium on East-West Medicine, Seoul. 212-230, 1999. -Hepatitis B is caused by hepadnavirus. Hepatitis B virus replicates through 3.5kb pregenomic RNA intermediate which is regulated by core promoter. Pathogenesis of hepatitis B virus has been bilieved the result of host immune response. But recently many studies have reported that high level of viral replication caused by mutation in core promoter might result in severs hepatitis. But these studies were performed in vitro, not in vivo. So there is yet debate about which factor, viral of host factor, is more important in pathogenesis of hepatitis B virus. So we measured real viral replication level in 204 chronic hepatitis B patients by quantifying HBV DNA from sera by our novel PCR-based more sensitive method, and compared these results with ALT level measured from same sera, which indicates liver cell damage. Surprisingly there are no significant correlation between HBV DNA quantity and ALT level. Then we cloned core promoter region. In SSCP, we found that many viral mutants coexist in one patient. Base on SSCP result, we chose main viral core promoter type in each patients, which is thought to determine overall viral replication level in this patient. Main type of core promoter region of each 41 patients were directly sequenced. And with these we measured promoter activity by luciferase assay system and compared promoter activity with on another. We found tha there were some differences in promoter activity according to core promoter sequences. And we constructed replication-competent viral constructs with core promoter from 41 patients and Transfected these into HepG2 cell and measured HBV DNA by southern blot. There were also differences in HBV DNA quantity according to core promoter sequences. On these all results we investigated correlation between the effect of HBV core promoter on viral replication in vitro and HBN DNA quantity, ALT level from sera of each patients. We found there is no significant correlation among them. As a result, we concluded that in determining severity chronic hepatitis B patients, host factors of each patient is more important rather than replicative activity of virus itself.

        Disruption of rsmA Gene of Pectobacterium carotovorum subsp. carotovorum LY34 and Effect on Pathogenicity

        The rsmA gene was cloned from soft-rot bacterium Pectobacterium carotovorum subsp. carotovorum LY34 (Pcc LY34), and its role in pathogenicity was investigated by marker exchange mutagenesis. From a cosmid library of Pcc LY34 genomic DNA, a positive clone carrying the rsmA gene was selected, and the gene was cloned by polymerase chain reaction (PCR) amplification. The gene is 186 bp in size and encodes a protein of 62 amino acids with a predicted molecular mass of 6,839 Da. The calculated pI of the RsmA is 8.16. The phylogenetic tree showed that the RsmA of Pcc LY34 appeared genetically identical to the CsrA of Pectobacterium atrosepticum SCRI1043 (100% identity) and similar to the CsrA of Yersinia pestis KIM10+(98.3%). The gene was disrupted by the $Km^r$ gene, and the cells became mutated (i.e., $RsmA^-$ mutant). The pathogenicity test revealed that the disease rating of the $RsmA^-$ mutant only differed slightly from that of the wild type on a slice of potato tuber and a Chinese cabbage stalk. These results suggest that RsmA is not an essential factor for the pathogenicity of Pcc LY34 and that the rsmA gene of Pcc LY34 is not completely derepressed in the $RsmA^-$ mutant for virulence-related genes, contrary to the results of Erwinia carotovora subsp. carotovora $RsmA^-$ mutant, which proved hypervirulent for celery petioles. These results showed that the microenvironmental conditions of the host and/or strain of pathogen are important for the coordination of virulence gene expression.

        Evaluation of Hot Deformation and Dynamic Recrystallization Behaviors of Advanced Reduced-Activated Alloy (ARAA)

        The hot deformation behavior of advanced reduced-activation alloy (ARAA) was investigated using hot torsion tests. Thefl ow stress decreased as deformation temperature increased and as strain rate decreased. The fl ow behavior demonstratedthe typical dynamic recrystallization (DRX). Based on the constitutive analysis of peak stress, the activation energy for hotdeformation was found to be 330.3 kJ mol −1 . Peak stress was analyzed as a function of the Zener–Hollomon parameter, andcalculated and experimental values were in good agreement. A DRX kinetic model for ARAA was derived with deformationconditions based on the Avrami-type model. It was confi rmed that the volume fraction of dynamically recrystallized grainsincreased as deformation temperature increased and as strain rate decreased. The necklace structure and grain boundarybulging were observed in the deformed microstructure of ARAA. The suggested DRX mechanism for ARAA during hotworking is discontinuous DRX.

      • Regulation of paclitaxel-induced programmed cell death by autophagic induction: A model for cervical cancer

        Objective: Autophagy plays a vital role in homeostasis by combining organelles and cellular proteins with lysosome under starvation conditions. In addition, autophagy provides tumor cells with a source of energy. Continued autophagy will induce cells death. Here we aim to see if autophagic induction has an effect on conventional chemotherapeutic agents. Methods: Rapamycin, or mammalian target of rapamycin and paclitaxel, apoptosis-inducing agents were used autophagy in HeLa cervical cancer cells. Results: Growth inhibition of cells was not observed after the application of 0, 10, 20 nM of paclitaxel with or without rapamycin. Using a 5 nM concentration of paclitaxel, rapamycin administration inhibited cell growth significantly compared to no treatment. This implies the synergic antitumor effect of paclitaxel and rapamycin. Paclitaxel itself did not show any autophagic effect on cells but did show cell apoptosis by flow cytometry. Light chain 3, a microtubule-associated protein, which reflect autophagy, was increased with 5 nM of paclitaxel after pretreatment with 10 nM of rapamycin. Conclusion: These findings suggest that the autophagic inducer, rapamycin, can potentiate autophagic cell death when added as an apoptosis-inducing chemotherapeutic agent. In conclusion, the control of autophagy may be a future target for chemotherapy.

        K-식(式) 침전관(沈澱管)에 의한 간질충란(肝蛭蟲卵) 간이검사법(簡易檢査法)에 관한 연구(硏究)

        피내반응법(皮內反應法)에 의(依)해 간질(肝蛭) 양성반응(陽性反應)으로 판정(判定)된 유우(乳牛)를 대상(對像)으로 분편검사(糞便檢査)를 실시(實施)하여 충란(蟲卵)을 확인(確認)한 후(後), 현재(現在) 사용(使用)되고 있는 간질(肝蛭) 충란검사법(蟲卵儉査法)들의 단점(短點)을 개선(改善)하기 위하여 잔재물(殘滓物)을 성력적(省力的)으로 제거(除去)하는 사(篩)의 조합법(組合法)과 K-식(式) 침전관(沈澱管)을 고안(考案)하여, 인위적(人爲的)으로 분즙(糞汁)에 충란(蟲卵)을 혼입(混入)하여 K-식(式) 침전관법(沈澱管法)에 따라 검사(檢査)한 결과(結果) 현재(現在)까지 보고(報告)된 타방법(他方法)들에 비(比)하여 조작(操作)이 간편(簡便)하고 검출율(檢出率)도 높아 이상적(理想的)인 방법(方法)으로 사료(思料)되었기에 그 개요(槪要)와 결과(結果)를 보고(報告)하는 바이다. 1. 간질충란(肝蛭蟲卵)을 여과(慮過)하고 잔재물(殘滓物)을 제거(除去)하기 사(篩)의 조합(組合)을 3단계(段階)(100mesh, 150mesh, 250mesh)로 조합(組合)하여 성력적(省力的)인 방법(方法)으로 개선(改善)하였다. 2. 3종(種)의 침전관중(沈澱管中) 제(第)III형(型)의 침전관(沈澱管)에 의(依)한 간질충란(肝蛭蟲卵) 회수율(回收率)이 가장 높았으므로 제(第)III형(型)을 K-식(式) 침전관(沈澱管)으로 지칭(指稱) 채택(採擇)하였다. 3. K-식(式) 침전관법(沈澱管法)에서 A-II, A-III, B-III, C-III형(型)의 1구(回) 경검성적(鏡檢成績)은 99~100%였으며 A-I, B-II, C-I 및 C-II형(型)은 71~91%였다. 4. A형(型)($5m{\ell}$, 5-10침전(沈澱)), B형(型)($10m{\ell}$, 10-10침전(沈澱)) 및 C형(型)($15m{\ell}$, 15-15침전(沈澱))은 각기(各其) 특징(特徵)을 가진 방법(方法)들로서 판정시간(判定時間)을 단축(短縮)하는데는 A형(型)이 가장 유용(有用)한 방법(方法)이었다. 5. K-식(式) 침전관법(沈澱管法)은 다른 검출법(檢出法)에 비(比)하여 가장 검출율(檢出率)이 높은 간편(簡便)한 방법(方法)이었으며 E. P. G가(價)도 동시(同時)에 얻을 수 있는 이상적(理想的)인 방법(方法)으로서 임상응용가치(臨床應用價値)가 매우 높은 것으로 사료(思料)되었다. These studies were carried out to investigate of detection of Fasciola egg from positive cattle faces in the intradermal reaction method by laborsaving composition of sieves and Kim's sedimental tube method. The results obtained are summerized as follows: 1. Detection method of Fasciola eggs was improved by 3 meshes(100mesh, 150mesh, 250mesh) for filtration of fecal fluid contained Fasciola eggs. 2. III type of tube in the 3 kinds of sedimental tube had the highest recovery rate of Fasciola egg. Thus, this type of tube was selected and called as Kim's sedimental tube. 3. A-II, A-III, B-III and C-III type by Kim's sedimental tube method had the recovery rates of eggs in the 1st dropping as ranged 99 to 100% and A-I, B-II, C-I and C-II as 71 to 91%. 4. All types as themselves contained the special aspects. In the these type was useful to determine the results in the urgent cases add to shorten the time of the diagnostic procedures. 5. Kim's sedimental tube method high recovery rate and convenient procedures as compared with other detection methods reported. In addition, through this method can also obatained E. P. C. value. Therefore, it is desirable that Kim's sedimental tube method be recommended by clinicians.

The results obtained are summarized as follows: 1. The detection method of Fasciola eggs was improved by 3 meshes (100mesh, 150mesh, 250mesh) for filteration of fecal fluid contained Fasciola eggs. 2. Third type of tube in the 3 kinds of sedimental tube had the highest recovers rate of Fasciola eggs. Thus, this type of tube was selected and called as Kim's sedimental tube. 3.         Meteorogical Mechanisms Associated with Long-range Transport of Asian Dust Observed at the West Coast of North America in April 2001

        Meteorological mechanisms in association with long-range transport of Asian dust in April 2001 have been investigated using weather maps, satellite images, TOMS and surface PM_10 data, backward trajectories, plus modeling output results (geopotential heights, horizontal wind vectors, potential temperatures, and streamlines). The results indicated that long-range transport of Asian dust to the west coast of North America was associated with strong westerlies between the Aleutian low and the Pacific high acting as a conveyor belt. Accelerating westerly flows due to cyclogenesis at the source regions over East Asia transported pollution from the continent to the central Pacific. When the system reached the Aleutian Islands, the intensity of troughs and the westerlies were amplified in the North Pacific. Thereafter the winds between the Aleutian Islands and the Pacific Ocean were more intensified from the air flow transport of the conveyor belt. Consequently, the strong wind in the conveyor belt enhanced the dust transport from the Pacific Ocean to the west coast of North America. This was evidenced by PM_10 concentration (maximum of about 100㎍m­³) observed in California. Further evidence of the dust transport was found through the observation of satellite images, the distribution of TOMS aerosol index, and the analyses of streamlines and backward trajectories.

        Physical and chemical Effects on the sonication Treatment of chitosan solution

        물리적 방법을 이용한 키토산 올리고당의 제조에 관한 기초 자료를 제시하기 위하여 키토산용액을 20kHz의 초음파로 처리하였으며, 초음파 처리 효과에 미치는 처리 조건의 영향을 조사하였다. 키토산 용액의 고유점도는 초음파 처리 5분까지는 급속하게 감소하였고 그 이후로는 서서히 감소하였다. 키토산 용액의 부피가 작을수록 초음파 처리효과는 컸으며, 키토산 용액의 온도는 초음파 처리에 별다른 효과가 없는 것으로 나타났다. 5분간 초음파 처리 후의 점도는 용매의 종류에 따라 큰 차이가 없었으나, acetate buffer의 경우 초음파 처리효과가 가장 컸다. pH가 높을수록 초음파 처리효과는 크게 나타났으며, 이온강도와 염의 종류에 따른 효과는 나타나지 않았다. 이상의 결과에서, 용액의 부피는 10~20ml, 온도는 20~30℃, 용액의 pH는 4.5, 그리고 용매의 종류는 acetate buffer로 하는 것이 최적 초음파 처리조건으로 추정된다. As the first step of studies related to production of chitooligosaccharides by physical methods, chitosan solution were sonicated with 20kHz and various treatment effects were examined to present fundamental data of sonicated chitosan solution. Intrinsic viscosity of chitosan solution sharply decreased from 3.76dl/g to 2.90dl/g until 5 minutes of sonication and then slowly decreased. With low volume of chitosan solution, sonication was very effective and temperature of chitosan solution slightly affected the efficiency of sonication. In case of changing the solvent, no significant differences were observed on the effect of sonication, however, acetate buffer had highest sonication effect among various solvents. The sonication effect was increased as the increasement of the value of pH, on the contrary, ionic strength and type of counterions showed no effect on sonication, With these results, we assumed that optimal sonication treatment would be as follows, solution volume was 10 ~20ml, temperature range was 20~30℃ , pH value of solution was 4.5 and type of solvent was acetate buffer
