        Recognition of the Importance of Imidazolidinone Motif for Cytotoxicity of 4-Phenyl-1-arylsulfonylimidazolidinones Using Thiadiazolidine-1,1-Dioxide Analogs

        For probing the importance of planarity of imidazolidinone motif of 4-phenyl-1-(N-acylindoline- 5-sulfonyl)imidazolidinones 1 for their cytotoxicity, 4-phenyl-1-(N-acylindoline-5-sulfonyl)[ 1,2,5]thiadiazolidine-1,1-dioxides 2 were prepared and their cytotoxicity were measured against human lung carcinoma (A549), human colon carcinoma (COLO205), human ovarian cancer (SK-OV-3), human leukemic cancer (K562), and murine colon adenocarcinoma (Colon26) cell lines in vitro. Although only carbonyl moiety of imidazolidinone ring was replaced with sulfonyl group, compounds 2 do not show any activity against all five cancer cell lines unlike 1. Therefore the planarity of imidazolidinone ring of 1 should be an important factor for their cytotoxic activity.

        Anti-Inflammatory Mode of Isoflavone Glycoside Sophoricoside by Inhibition of Interleukin-6 and Cyclooxygenase02 in Inflammatory Response

        Soy, high dietary intake for the oriental population, is a main source of isoflavonoids. Sophoricoside (SOP) an isoflavone glycoside was isolated from immature fruits of Sophora japonica (Leguminosae family) and its inhibitory effect on chemical mediators involved in inflammatory response was investigated in this study. SOP inhibited the interleukin (IL)-6 bioactivity with an IC50 value of 6.1 mM whereas it had no effects on IL-1b and TNF-a bioactivities. SOP was identified as a selective inhibitor of cyclooxygenase (COX)-2 activity with an IC50 value of 4.4 mM, but did not show inhibitory effect on the synthesis of COX-2. However, SOP had no effect on the production of reactive oxygen species including superoxide anions and nitric oxide. These results revealed that in vitro anti-inflammatory action of SOP is significantly different from that of genistein known as a phytoestrogen of soy products. This experimental study has documented an importance of dietary soy isoflavonoids as multifunctional agents beneficial to human health, and will help to clarify protective mechanisms of SOP against inflammatory conditions.

        Importance of Sulfonylimldazolidinone Motif of 4-Phemy-1-arylsulfonylimidazolidinones for Their Cytotoxicity: Synthesis of 2-Benzoyl-4-phenyl[1,2,5]thiazolidine-1,1-dioxides and Their Cytotoxcity

        For probing the importance of planarity of imidazolidinone motif of 4-phenyl-1-(benzenesulfonyl) imidazolidinones 1 for their cytotoxicity, 4-phenyl-2-(benzoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2a), 4-phenyl-2-( p-toluoyl)[1,2,5]thiadiazolidine-1,1-dioxide (2b), 4-phenyl-2-(phenylcarbamoyl) [1,2,5]thiadiazolidine-1,1-dioxide (3a), and 4-phenyl-2-( p-tolylcarbamoyl)[1,2,5]thiadiazolidine- 1,1-dioxide (3b) were prepared along with their regioisomers (5a, 5b, 9a, 9b) and their cytotoxicity were measured against human lung carcinoma (A549), human colon carcinoma (COLO205), human ovarian cancer (SK-OV-3), human leukemic cancer (K562), and murine colon adenocarcinoma (Colon26) cell lines in vitro. All compounds prepared do not show any activity against all five cancer cell lines unlike 1. Compounds 1 possess planarity of imidazolidinone, especially in sulfonylurea moiety (-SO2NHCONH-). However compounds 2 and 3 have nonplanar 5-membered ring, [1,2,5]thiadiazolidine-1,1-dioxides. Such structural differentiation might result in the loss of activity. Therefore the inactivity of 2 and 3 could also be an indication for the necessity of planarity of imidazolidinone ring of 1 for their cytotoxic activity.

        Development of Tobacco Mosaic Virus (TMV) resistant Flue-cured F1 Hybrid, KF118

        Tobacco mosaic virus (TMV), TMV-common strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. Controlling TMV outbreak with cultivating practices and chemicals are limited, thus, the best way would be developing TMV resistant variety. A new flue-cured hybrid, KF118, was developed from a cross of maternal parent MSTC175 and TC853 at the Agro-tech. Research Group, KT&G Central Research Institute. KF118 exhibited more vigorous growth characteristics in early stage than that of NC82, standard variety in Korea, while other characters were favorable to those of NC82, It is highly resistant to TMV and bacterial wilt(Ral-stonia solanacerum), and black shank(phytophthora parasitica var. nicotianae) is comparable to that of NC82. It should adapt well to the flue-cured production area, and can reduce premature flowering under unfavorable weather conditions. Yield of KF118 is 1 to 2% higher, and leaf quality is 4% higher than NC82. Fl hybrid, KF118, met acceptable standards for chemical and physical characteristics of cured leaf, and ranked high in good smoking taste by panel members in KT&G.
