Proceedings of International Symposium on East-West Medicine, Seoul. 201-211, 1999. -Nephroprotective effects of a crude drug-prearation (E-kong-san) were determined from cisplatin on rabbit kidney proximal tubule and human renal cortical cells by MTT assays and sustained glucose consumption on ciplatin-induced human renal cortical tissue. Levels of creatinine and blood ure nitrogen(BUN) in serum after administration of cisplatin(0.75mg/kg,i.p.) to E-kong-san(0.75g/kg/day,p.o.)-pretreated rats were markedly lower compared to those of cisplatin-treated rats. Moreover, the administration of E-kong-san significantly inhibited the loss of body weight of cisplatin injected rts. These findings suggest that E-kong-san on the cisplatin induced nephrotoxicity led us to investigate whether the effect of this water extract was a result of anti-oxidation. E-kong-san showed strong free radical scavenger activities on 1,1-dipheny1-2picrylhydrazil(DPPH) radical and xanthine/xanthine oxidase(XOD) generated superoxide anion radical(O2-). We further studied the effects of E-kong-san on lipid peroxidation in rat liver microsomes induced by enzymatic and nonenzymatic methods. E-kong-san exhibited significant inhibition on both ascorbic acid/Fe2+and ADP/NADPH/Fe3+ induced lipid peroxidation in rat liver microsomes. Based on these results, we suggest that-E-kong-san attenuate the cisplatin induced cytotoxicity and its mechanism can be eplained by antioxidant.

        판상형 충전제의 함량과 배향에 따른 PP복합체의 영률 변화 연구

        이 논문은 판상형 충전제의 함량과 배향에 따라 폴리프로필렌 복합체의 영률 변화에 미치는 영향을 연구 하였다. 이론적인 예측은 Lee와 그의 연구자들에 의해 제안된 두 개의 종횡비, ρα=a1/a3과 ρβ=a1/a2 를 이용하여 3차원 타원체의 형상을 계산하였고 실험값과 비교 분석하였다. 판상형 충전제로 운모와 탈크를 사용하였다. 충전제의 형태는 SEM을 이용해 관찰하였고, 종횡비는 고분자의 흐름을 기준으로 좌표축을 설정하여 통계적 방법으로 계 산하였다. 충전제가 운모인 경우에 ρα=13.5와 ρβ=1.8이었고, 탈크의 경우는 ρα=3.8과 ρβ=1.4이었다. 또한 충전제 의 2개의 종횡비와 함량이 기계적 물성에 미치는 영향을 실험적으로 연구하였다. 운모의 경우 30 wt% 함량일 때 E11이 약 2.7배 향상되었다. 탈크의 경우는 E11이 약 2.3배 향상하였다. This paper studied the effects of the content and the orientation of the disk shape fillers on the modulus of PP composites. The experimental results were compared with the theoretical calculations which included the three dimensional ellipsoids and analyzed by two aspect ratios, ρα=a1/a3 and ρβ=a1/a2 proposed by Lee and his researchers. Mica and talc were used as disk shape fillers in the composites. The shapes of mica and talc were observed by SEM and aspect ratios were statistically calculated. For the case of mica, the average aspect ratios were ρα=13.5 and ρβ=1.8, and for the case of talc, they were ρα=3.8 and ρβ=1.4. Also, the effects of two aspect ratios and the content of filler on the mechanical properties were studied: For 30 wt% of mica, E11 increased up to about 2.7 times, and for the other case of talc, E11 increased up to about 2.3 times, respectively.

        Background: The outcome of hepatitis B virus (HBV) infection after liver transplantation (LT) was improved by hepatitis B immunoglobulin (HBIG) and nucles(t)ide analogue (NUA). However, HBV recurrence after LT is critical because the recurrence is occasionally accompanied by a progressive destruction of graft and poor survival. The aims of this study were to investigate the significance HBV recurrence and identity factors associated with HBV recurrence. Methods: From October 1999 to February 2011, a total of 2684 consecutive LT recipients who underwent HBV-associated LT were retrospectively enrolled from 7 transplantation centers in Korea. Results: Prophylaxis regimens were HBIG monotherapy (67.7%) or a combination of HBIG with NUA (22.3%). The recurrence rate of HBV was 6.1% (164 recipients) during mean follow-up duration of 10.9 years. The median time from transplantation to recurrence was 2.1 years (0.1-7.9 years). Of the 1,071 patients with hepatocellular carcinoma (HCC) prior to LT, 155 patients (14.5%) had HCC recurrence after transplantation, and 48 patients (31.0%) had HBV recurrence. Of the 48 patients with recurrence of both HBV and HCC, 25 patients (52.1%) experienced HBV recurrence after HCC recurrence. In the multivariate analysis, pretransplant HCC, pretransplant HBV DNA above 5.5 log copies/mL was independent clinical factors influencing HBV recurrence after LT. The mortality rate among the recipients with HBV recurrence was 34.1% (56 recipients). Mean overall survival was 6.4 years in the HBV-recurrence group and 9.9 years in the HBV-nonrecurrence group (p<0.001). HBV recurrence was not an independent prognostic factor for overall survival. HCC recurrence was the most important factor for overall survival. Conclusions: The overall outcome of LT in HBV-related liver disease was excellent with the current prophylaxis regimen Choon Hyuck David Kwon,8 Suk-Koo Lee8 Pretransplant HBV DNA and HCC were important factors for HBV recurrence. HBV recurrence after LT did not significantly influence on the overall survival without combining of HCC recurrence.

      • The Changes of Steroid Receptors by Irradiation in rat Mammary Tumors

        To centralize the determining estrogen and progesterone receptor concentrations in metropolitan area which are essential to decide the therapeutic method in managing the breast cancer patients and to realize the changes of hormonal receptors after irradiation authors induced mammary tumors in female rats by single intragastric feeding of 20㎎ 7, 12-dimethylbenzanthracene(DMBA). The estrogen and progesterone receptor concentrations were estimated before and four weeks after irradiation using the dextran-coated charcol procedures and the tumor size was checked before and weekly after irradiation for 4 weeks. 1. Only one of the nine positives was changed into negative state in estrogen receptor concentration after irradiation without statistical significance. 2. The positive seven cases were changed into negative state in progesterone receptor concentration except one and statistically significant reduction was observed after irradiation. 3. The average tumor size of single irradiation group before irradiation was 11.45±4.49㎝^2 and changed to 5.55±2.88㎝^2 at irradiated 2 weeks, 0.89±0.36㎝^2 at irradiated 4 weeks. 4. The average size in divided dose group was 16.17±7.22㎝^2 and changed to 11.85±7.81㎝^2 firstly irradiated 2 weeks, 7.85±5.07㎝^2 at secondarily irradiated 2 weeks.

        Generation of disease-specific induced pluripotent stem cells from patients with rheumatoid arthritis and osteoarthritis

        <P><B>Introduction</B></P><P>Since the concept of reprogramming mature somatic cells to generate induced pluripotent stem cells (iPSCs) was demonstrated in 2006, iPSCs have become a potential substitute for embryonic stem cells (ESCs) given their pluripotency and “stemness” characteristics, which resemble those of ESCs. We investigated to reprogram fibroblast-like synoviocytes (FLSs) from patients with rheumatoid arthritis (RA) and osteoarthritis (OA) to generate iPSCs using a 4-in-1 lentiviral vector system.</P><P><B>Methods</B></P><P>A 4-in-1 lentiviral vector containing Oct4, Sox2, Klf4, and c-Myc was transduced into RA and OA FLSs isolated from the synovia of two RA patients and two OA patients. Immunohistochemical staining and real-time PCR studies were performed to demonstrate the pluripotency of iPSCs. Chromosomal abnormalities were determined based on the karyotype. SCID-beige mice were injected with iPSCs and sacrificed to test for teratoma formation.</P><P><B>Results</B></P><P>After 14 days of transduction using the 4-in-1 lentiviral vector, RA FLSs and OA FLSs were transformed into spherical shapes that resembled embryonic stem cell colonies. Colonies were picked and cultivated on matrigel plates to produce iPSC lines. Real-time PCR of RA and OA iPSCs detected positive markers of pluripotency. Immunohistochemical staining tests with Nanog, Oct4, Sox2, Tra-1-80, Tra-1-60, and SSEA-4 were also positive. Teratomas that comprised three compartments of ectoderm, mesoderm, and endoderm were formed at the injection sites of iPSCs. Established iPSCs were shown to be compatible by karyotyping. Finally, we confirmed that the patient-derived iPSCs were able to differentiate into osteoblast, which was shown by an osteoimage mineralization assay.</P><P><B>Conclusion</B></P><P>FLSs derived from RA and OA could be cell resources for iPSC reprogramming. Disease- and patient-specific iPSCs have the potential to be applied in clinical settings as source materials for molecular diagnosis and regenerative therapy.</P>

      • Constructing compact and effective graphs for recommender systems via node and edge aggregations

        <P><B>Abstract</B></P> <P>Exploiting graphs for recommender systems has great potential to flexibly incorporate heterogeneous information for producing better recommendation results. As our baseline approach, we first introduce a naïve graph-based recommendation method, which operates with a heterogeneous <I>log-metadata</I> graph constructed from user log and content metadata databases. Although the naïve graph-based recommendation method is simple, it allows us to take advantages of heterogeneous information and shows promising flexibility and recommendation accuracy. However, it often leads to extensive processing time due to the sheer size of the graphs constructed from entire user log and content metadata databases. In this paper, we propose node and edge aggregation approaches to constructing compact and effective graphs called ‘<I>Factor</I>-<I>Item</I> bipartite graphs’ by aggregating nodes and edges of a <I>log-metadata</I> graph. Experimental results using real world datasets indicate that our approach can significantly reduce the size of graphs exploited for recommender systems without sacrificing the recommendation quality.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We present an aggregation technique for building compact graphs for recommendation. </LI> <LI> We present an algorithm for ranking nodes for recommendation. </LI> <LI> Node and edge aggregation can significantly reduce the graph size. </LI> <LI> Using a compact graph for recommendation does not sacrifice much accuracy. </LI> </UL> </P>

        Characterization of Thermostable Tyrosine Phenol-Lyase from an Obligatory Symbiotic Thermophile, Symbiobacterium sp. SC-1

        Tyrosine phenol-lyase of thermophilic Symbiobacterium sp. SC-1, which is obligately and symbiotically dependent on thermophilic Bacillus sp. SK-1, was purified and characterized. The enzyme is composed of four identical subunits and contains approximately 1 mol of pyridoxal 5'-phosphate (PLP) per mol subunit as a cofactor. The enzyme showed absorption maxima at 330 and 420nm, and lost this absorption profile by treatment with phenylhydrazine. The apparent dissociation constsnt, K'??, for PLP was determined with the apoenzyme to be about 1.2 μM. The isoelectric point was 4.9. The optimal temperature and pH for the α,β-elimination of L-tyrosine were found to be 80℃ and pH 8.0, respectively. The substrate specificity of the enzyme was very broad: L-amino acids including L-tyrosine, 3,4-dihydroxyphenyl-L-alanine (L-DOPA), L-cysteine, L-serine, S-methyl-L-cysteine, β-chloro-S-alanine, and S-(o-nitrophenyl)-L-cysteine all served as substrates. D-Tyrosine and D-serine were also decomposed into pyruvic acid and ammonia at rates of 7% and 31% relative to their corresponding L-enantiomers, respectively. D-Alanine, which was inert as a substrate in α,β-elimination, was the only D-amino acid racemized by the enzyme. The K?? values for L-tyrosine, L-DOPA, S-(o-nitrophenyl)-L-cysteine, β-chloro-L-alanine, and S-methyl-L-cycsteine were 0.19, 9.9, 0.36, 12, and 5.5 mM, respectively.

      • Detection of Minimal Lesion and Identification of Clonality in Malignant Lymphoma

        The bone marrow biopsy is an integral of the staging process in patients with malignant lymphomas. Marrow involvement indicates stage Ⅳ disease, but there are always some cases in which clear distinction is not possible based on morphology alone. Additional difficulties are caused by morphologic discordance between the bone marrow and the primary lymphoma. Immunohistochemical stain, mRNA in situ hybridization and polymerase chain reaction were performed to find out the minimal lesion and the clonality in formalin fixed paraffin embedded tissues of 39 primary lymphomas and corresponding bone marrow biopsy specimens. As a result, nine morphologically negative bone marrows of 18 lymphomas were positive by PCR (Group Ⅰ). Among the 6 lymphomas with morphologically suspicious bone marrow involvement (Group Ⅱ), one was confirmed to be positive for marrow involvement by both mRNA ISH and PCR and other four cases by PCR alone. The positive bone marrows of Group Ⅰ and Ⅱ revealed gene rearrangement at the same time as the primary lesion, suggesting the same clonality. Thirteen of 15 lymphomas with morphologically positive bone marrow(Group Ⅲ) had the same clonality in the primary lymphomas and the bone marrow lesion. Three cases among the Group Ⅲ with morphologic discordance also revealed the same clonality by PCR. This study shows that combination of mRNA ISH and PCR in addition to immunohistochemical stain improve the diagnostic sensitivity in the detection of bone marrow involvement and identification of clonality, and among the three methods used, PCR is the most sensitive in detecting minimal lesion.

        Clinical research on the efficacy and safety of Bosinji for low back pain with radiculopathy caused by herniated intervertebral disc of the lumbar spine : A protocol for a multicenter, randomized, controlled equivalence trial

        <P><B>Abstract</B></P><P><B>Background:</B></P><P>A lumbar herniated intervertebral disc (LHIVD) is a common problem that usually causes low back pain and radiating pain. The effectiveness of Bosinji, one of the herbal medicines used for low back pain and radiating pain in patient with LHIVD, has been reported in several studies; however, little clinical evidence is available owing to the methodological limitations in previous studies. Hence, the present study aims to establish the clinical evidence regarding the efficacy and safety of Bosinji in improving pain, function, and quality of life in LHIVD patients.</P><P><B>Method/design:</B></P><P>This is a multicenter, open-label, randomized, controlled, and equivalence trial with 2 parallel arms. A total of 74 patients who have low back pain and radiating pain due to LHIVD will be recruited and randomly allocated to the experimental group and control group. The patients in the experimental group and control group will take 2.5 g of Bosinji granule (1.523 g of Bosinji extract) or Loxonin tablet (60 mg of loxoprofen) 3 times a day for 6 weeks. Additionally, both groups will receive the same acupuncture treatment once a week for 6 weeks as a concurrent treatment. Changes in the 100-mm visual analogue scale (VAS) for low back pain after 6 weeks from baseline will be assessed as the primary outcome. Furthermore, the 100-mm VAS for radiating pain, Oswestry disability index (ODI), Roland–Morris disability questionnaire (RMDQ), EuroQol 5 Dimensions 5 Levels (EQ-5D-5L), global perceived effect (GPE), and deficiency syndrome of kidney index (DSKI) will be used to evaluate secondary outcomes. Outcomes will be assessed at baseline and at 3, 6, and 10 weeks after screening. For the safety evaluation, laboratory examinations including complete blood count, liver function test, renal function test, blood coagulation test, inflammation test, and urine analysis will be conducted before and after taking the medications.</P><P><B>Discussion:</B></P><P>The results of this trial will be used to establish clinical evidence regarding the use of Bosinji with acupuncture treatment in the treatment of patients with LHIVD.</P><P><B>Trial registration number:</B></P><P>NCT03386149 (clinicaltrials.gov) and KCT0002848 (Clinical Research Information Service of the Republic of Korea).</P>
