      • 신경교세포 및 RAW 264.7 세포에서 Protein kinase의 활성에 의한 유도성 Nitric oxide synthase의 발현

        NO(nitric oxide) plays an important role as neurotransmitter or cytokine, and pathologic factor for some diseases by the large amount production with iNOS(inducible NO synthase) expression in macrophages or glial cells. The expression of iNOS is regulated by various cytokines, protein kinases and transcription factors. In this experiment, to investigate the roles of progein kinase and NF-kB for iNOS expression, the effects of PMA(phorbol 12-myristate 13-acetate), cAMP, and various protein kinase inhibitors on LPS(lipopolysaccharide)-induced iNOS mRNAN expression and nuclear NF-kB binding complex were examined in C6 glial cells and RAW 264.7 cells. In C6 glial cells, iNOS mRNA expression by LPS was induced from 1 hour and peak at 3 hour after treatment. In RAW 264.7 cells, the mRNA was observed from 3 hour and peak at 6 hour. PMA enhanced markedly LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but did not much influence on LPS-induced iNOS mRNA expression in RAW 264.7 cells, in spite of increased LPS-induced NF-kB binding complex at 30 min. cAMP(dibutyryl cAMP) did not much influence on LPS-induced iNOS mRNA expression, by increased LPS-induced NF-kB binding complex in C6 glial cells. However, in RAW 264.7 cells, cAMP increased slightly LPS-induced iNOS mRNA expression without change of NF-kB binding complex. Staurosporine did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression and NF-kB binding complex. Ro-31-8220 did not much influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased significantly LPS-induced iNOS mRNA expression in spite of increased LPS-induced NF-kB binding complex for 3hours. G 6976 did not much influence on LPS-induced iNOS mRNA expression with decreased NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased iNOS mRNA expression without influence on LPS-induced NF-kB binding complex. Genistein did not influence on LPS-induced iNOS mRNA expression and NF-kB binding complex in C6 glial cells, but in RAW 264.7 cells, decreased LPS-induced iNOS mRNA expression inspite of increased NF-kB binding complex. These results suggest that LPS-induced regulation of iNOS expression or NF-kB activity in C6 glial cells, might be different from RAW 264.7 cells through various protein kinases or other factors.

      • 층간분리 형상계수(f_(S))가 FRMLs의 층간분리 성장률(dA_(D)/da)에 미치는 영향

        Most previous researches for the hybrid composite materials such as FRMLs (Al/AFRP, Al/GFRP) have evaluated the fatigue delamination behavior using the traditional fracture mechanism. However, most previous researches have not generally been fumed yet. Because delamination growth behavior in hybrid composite should be consider delamination growth rate, dA_(D)/da using the delamination shape factors, f_(S) instead of traditional fracture mechanic parameters. The major purpose of this study was to evaluate the relationship between delamination shape factor, f_(S) s and delamination growth rate, dA_(D)/da. And a propose parameter on the delamination aspect ratio, b/a. The details of the study are as follow : 1) Relationship between crack length, a and delamination width, b. 2) Relationship between delamination aspect ratio, b/a and delamination area rate, (A_(D))_(N)/(A_(D))_(All). 3) Variation of delamination growth rate, dA_(D)/da was attendant on delamination shape factors, f_(S1), f_(S2), f_(S3). The test results indicated the delamination growth rate depends on delamination shape factors.

      • 만성 골수성 백혈병에 대한 Recombinant Interferon-gamma와 Hydroxyurea 병합요법

        To evaluate the therapeutic effect and toxicity, 8 patients with chronic phase chronic myelogenous leukemia were treated with IFN―gamma(5×10 Units/㎡/day i.m. from Monday to Friday every week) and Hydroxyurea (1―4gm/day po till WBC<20,000/cu mm, when the therapy is switched to IFN―gamma plus hydroxyurea 1―4 gm/day po×7 days every 3 weeks). 6 patients (75%) achieved responses (3 complete hematologic response, 3 partial hematologic response). The median duration of responses was 3 mos(1―10 mos). Early toxicities of interferons (fever, chill, fatigue and myalgia) were noted in 8―100% of the patients, but were well controlled with acetaminophen. The long term results of this trial needs to be followed by more patient accural and for the evaluation of whether this combined modality therapy could delay and prevent the progression to blastic phase from chronic phase of CML.

        근관와동 가봉재의 변연누출에 관한 실험적 연구

        ABSTRACTThe purpose of this study was to evaluate the sealing properties of endodontic cavity filling materials according to the time intervals after filling. Access cavities were prepared in extracted human premolar or molar teeth and filled with caviton, zinc oxide eugenol cement, zinc oxide eugenol cement with a base of gutta percha stopping and gutta percha stopping. After filling at the intervals of immediate, 2 days and 2 weeks the teeth were immersed'for 2 weeks in 1% methylene blue solutions. Longitudinal sections were obtained from approximately center of teeth and the depth of dye penetration into the access cavities were observed by 1 Ox macrolens.The following results were obtained. 1. All the materials experimented showed varying depth of dye penetration. 2. Of the material tested, caviton showed the best marginal sealing qualities regardless of the time intervals after filling and the sealing properties of the gutta percha stopping was the worst. 3. Both in zinc oxide eugenol cement and zinc oxide eugenol cement with a base of gutta percha stopping, the fillings allowed to mature for 2 days in normal saline solution showed the best sealing properties and those with no maturing time revealed the worst sealing qualities. 4. The sealing qualities of zinc oxide eugenol cement with a base of gutta percha stopping reveal- ed slightly lower depth of dye penetration than that of zinc oxide eugenol cements.

        Enterococcus faecalis 추출물이 임파구의 IL-2, IL-4, TGF-β1분비에 미치는 영향에 관한 연구

        In order to examine the immunoresponse of host cells to Enterococcus faecalis, this in vitro study monitored the production of Interleukin-2 (IL-2), Interleukin-4 (IL-4) and Transforming growth factor-β1 (TGF-β1) in human lymphocytes. Lymphocytes were activated with PHA in the presence or abscence of sonicated extracts of E. Faecalis (SEF) and further incubated for 72 hours. The level of each cytokine was measured by ELISA. Data were analyzed with Kruskal-Wallis test and Mann-Whitney U test (P < 0.05). PHA-activated group did exhibit higher level of IL-2 and IL-4 than untreated control group. The levels of expression of both cytokines were significantly decreased following the treatment of high (25㎍/㎖) and medium concentration (12.5㎍/㎖) of SEF (P > 0.05) than those of PHA activated group. But low concentration (5㎍/㎖) of SEF showed th similar level of IL-2 and IL-4 production as those of PHA activated group. TGF-β1 was unaffected by SEF treatment. These results suggested that E. faecalis may suppress IL-2 and IL-4 production by lymphocytes and this could be one of possible factors why E. faecalis are found frequently in the teeth with failed endodontic treatment. 근관치료의 실패원인 중 중요한 세균으로 알려진 Enterococcus faecalis는 최근에 중요성이 더해지며 많은 연구들이 진행중이다. 여러가지 기전들이 보고되고 있으나 면역반응에 관한 연구는 거의 알려져 있지 않은 상태이다. 본 연구에서는 Enterococcus faecalis의 초음파 분쇄 추출물을 성인의 말초혈액으로부터 얻은 임파구에 적용시켜서 여기서 분비되는 interleukin-2, interleukin-4, transforming growth fater-β1의 농도를 Enzyme linked immunosorbent assay (ELISA)로 측정하여 비교, 평가하는 것을 목적으로 한다. E. faecalis를 적절한 조건에서 배양한 뒤 초음파 분쇄를 하여 추출물을 얻어냈다. 임파구는 건강한 성인의 말초혈액에서 추출하여 분리하였다. 임파구를 적절한 농도의 mitogen (Phytohemagglutinin: PHA)으로 자극시킨 뒤에 다양한 농도의 E. faecalis 초음파 추출물을 적용시키고 72시간 동안 배양하였다 ELISA를 이용하여 IL-2, IL-4,TGF-β1 의 농도를 측정하였다. 실험결과는 Kruskal-Wallis test, Man-Whitney rank sum test (p < 0.05)를 사용하여 통계처리 하였다. 실험결과 PHA로 처리한 군은 아무것도 처리하지 않은 군에 비해서 IL-2, IL-4의 수치가 유의성 있게 높았다 (p < 0.05). PHA로 처리한 군중에서 고농도와 중농도의 sonic extract of E. faecalis (SEF)로 처리한 군은 그렇지 않은 군에 비해서 IL-2 IL-4의 농도가 유의성 있게 낮았다 (p < 0.05). PHA로 처리한 군중에서 저농도의 SEF로 처리한 군은 그렇지 않은 군과 비교하여 유의할 만한 차이를 보이지 않았다.TGF-β1의 농도는 모든 군에서 유의할 만한 차이를 보이지 않았다 (p > 0.05). 따라서, E. faecalis의 추출물은 임파구의 IL-2, IL-4의 분비능력을 저하시킨다고 할 수 있다.

      • 혈관 내 폐보조장치에서의 압력손실 예측

        The purpose of this study was making a equation to predict relationship between pressure drop and frontal area in 40% glycerol solution at 37 ℃. Before using whole blood, we estimated pressure drop with experimental equation, and compared the results with experimental results. Also, we tried to make module design that contained the maximum number of hollow fiber membranes in 3㎝diameter tube. We have observed pressure drop of 40% glycerol solution flow rate in each modules that have been changed by the number of the hollow fibers, and studied about the equation related between flow rate and pressure drop, friction factor and Reynolds number. The results showed that the pressure drop which was calculated with the various frontal area was similar to the pressure drop of using the equation obtained from the experimental. Also, the maximum number of hollow fiber is 675, which can be inserted in 3㎝diameter tube when the out side of diameter was 380㎛. It satisfied pressure drop(<15㎜Hg).

        진동에 의한 혈관 내 폐 보조장치의 산소전달 특성

        본 연구는 급성호흡부전환자를 치료하기 위하여 사용되고 있는 혈관 내 폐 보조장치의 기체전달을 향상시키기 위하여 중공사막에 진동기법을 사용하여 기체전달을 향상시키고자 시도하였다. 그리고 혈관 내 폐 보조장치를 정맥에 삽입하기 전, 혈관 내 폐 보조장치의 설계조건을 실험적 모델을 통하여 기체전달을 예측할 수 있는 예측식을 만들고자 하였다. 실험결과, 본 연구에서는 진동기법이 기체전달을 향상시키는데 효과적임을 확인할 수 있었다. 뿐만 아니라, 충진율과 가징 주파수의 함수관계를 이용하여 기체전달을 예측 할 수 있었다. 실험에 의하여 얻어진 결과는 예측식에 의하여 얻어진 결과와 유사한 결과를 얻었다. 그러므로 본 연구에서는 충진율과 가진 주파수의 함수를 이용하여 VIVLAD의 기체전달을 예측할 수 있었다. In this paper, we tried to improve gas exchange of the vibrating intravascular lung assist device(VIVLAD) using vibrating method in the hollow fiber membrane, for the patients suffering from acute respiratory distress syndrome(ARDS). And we tried to formularize prediction equations to make a prediction about gas transfer for designing intravenous artificial lung assist device, and designed modules under various conditions were studied through an experimental modeling before inserted the artificial lung assist device into as venous. As a result, we are convinced that vibration method is very useful for the has transfer increasing. Also, we can estimate the gas transfer as a function of the packing density and excited frequencies. The gas transfer obtained from the experiment was similar to that from the prediction equation, confirming the usefulness prediction equation. Therefore, we can estimate the gas transfer of the VIVLAD as a function of the packing density and excited frequencies.

        인천 인현동 호프집 화재 피해자 분석

        Purpose : This study was conducted to develop field triage, transportation, distribution, and prehospital care at a fire disaster by analyzing the victims of the fore that broke out at a bar in Incheon. Method : We analyzed the cases of the victims of a fire in Incheon in Oct. 1999. We determined the primary care hospital, the arrival time, the burn size, the outcome, and the injury type from the medical records, the concerned organ records, and interviews with concerned persons. Result : The total number of victims was 137: 56 prehospital deaths, 1 hospital death, and 80 survivals. The Pearson correlation coefficient between the burn size and the severity was -0.175. There were 121(89.6%) cases of inhalation injury, 59 (43.7%) cases of flame burns, 66 (48.9%) cases of hypoxic brain damage, and 16 (11.9%) cases involving other types of injury. Conclusion : The causes of death of the fire victims were inhalation injury and hypoxic brain damage due to CO poisoning and other toxic inhalants. We propose the use of a simple triage and rapid treatment(START) system and a reassessment the delayed category in fire disasters.

      • 성인 급성 백혈병의 치료에 관한 연구

        19 adult ALL patients were treated with follwing regimens; VP regimen to 5 patients, VPA regimen to 4 patients, multi-drug combined intensive therapy of Hoelzer regimen to 10 patients. The results of therapy are as follows: 1. Complete remission occured in 3 patients on VP regimen(60%), 4 patients on VPA regimen (100%), 8 patients on Hoelzer regimen (80%), respectively, and overall complete remission rate was 78.9%. 2. Mean survival time of total patients was 13.8 months, and in complete remission group, it was 16.2 month, but in non-complete remission group, it was 1.8 month. 3. Factors favorable for complete remission were absence of organomegaly(P<0.05) and initial leukocyte count under 30,000/㎟(P<0.01), factors favorable for the survival duration were initial leukocyte count under 30,000/㎟(P<0.05) and remission within 4 weeks of treatment.(P<0.01). 4. Recurrence rate were 40% in VP regimen, 50% in VPA regimen, and 20% in Hoelzer regimen. 5. During the remission induction period, common side effects of chemotherapy were alopecia, nausea, vomiting, mucositis, and general weakness and it was severe in VPA regimen and Hoelzer reimen than in VP regimen. But no significant difference was noted between VPA regimen and Hoelzer regimen. From the above results, we could confirm the importance of multiple drug combined intensive chemotherapy of remission regimen and periodic reinduction chemotherapy.
