Signal transduction pathways in erythrocyte nitric oxide metabolism under high fibrinogen levels

Previous studies show that the fibrinogen molecule modulates the metabolism of nitric oxide (NO) in erythrocyte. The in vitro induced hiperfibrinogenemia interferes in the metabolism of the NO in the erythrocyte in dependence of the phosphorylation degree of the band 3. The soluble form of fibrinogen binds into CD47 protein present in the erythrocyte membrane. The soluble thrombomodulin is an inflammatory marker that binds to the erythrocyte CD47 in a site with a sequence peptide known as 4N1K. A study done in vitro shows that when hiperfibrinogenemia was induced in the presence of the peptide 4N1K agonist of CD47 it were observed variations in the efflux of NO from erythrocyte and an increase in the concentrations of GSNO, peroxinitrite, nitrite and nitrate of the erythrocytes. The aim of this work was to study the influence of the peptide 4N1K, on the metabolism of NO in the erythrocyte under high fibrinogen concentration and in the presence of inhibitors of the status of phosphorylation of protein band 3. In this in vitro study, whole blood samples were harvested from healthy subjects and NO, peroxynitrite, nitrite, nitrate and S-nitroglutathione (GSNO) were determined in presence of 4N1K, calpeptine, Syk inhibitor and under high fibrinogen concentrations. The results obtained in erythrocytes under high fibrinogen levels when 4N1K is present with the Syk inhibitor or with calpeptine, showed in relation to the control samples increased significant concentrations of efflux of NO and of peroxynitrite, nitrite, nitrate and GSNO. In conclusion it was verified that in the in vitro model of hiperfibrinogenemia the peptide 4N1K, agonist of CD47, induces mobilization of NO in the erythrocyte in dependence of the status of phosphorylation of protein band 3.

tions such as Brahma Rasayan and Chyawanprash. However, this ethnomedicinal use has not been investigated scientifically. This study evaluated the effect of the ethanolic extract of Mesua ferrea flowers (MFE) on memory in scopolamine-induced models of cognitive dysfunction. MFE was administered to rats (100, 200 and 400 mg/kg, p.o) for a period of 14 days, after which amnesia was induced by giving scopolamine (1 mg/kg, s.c) on the 14th day. Piracetam (200 mg/kg, p.o) was given as a positive control. The models employed to assess memory in the rats were the T-maze continuous alternation task (T-CAT) and novel object recognition test (NORT). Pretreatment with MFE ameliorated the memory deficit caused by scopolamine; which was evidenced by a significantly greater relative proportion of spontaneous alternation percentage in the T-CAT, and a significant increase of discrimination index in the NORT. Further, MFE significantly inhibited anticholinesterase activity in the brain, elevated the levels of reduced glutathione and catalase, and decreased malondialdehyde and nitrite levels in the brain. The results of this study show that MFE exhibited significant anticholinesterase and antioxidant activities in scopolamine treated rats, which could be the possible underlying mechanism of its memory-enhancing activity and of its ethnomedicinal use as a brain tonic.
Pacing Strategy of Single Scullers during Rowing World Championships

This study aimed to describe and compare the pacing strategy and performance of world level under 19 (U19), under 23(U23) and senior single scullers during world championship races. Data are from 8 years of Rowing World Championships A and B finals of single scull races for U19, U23 and senior age scullers. Pacing was determined for velocity, stroke rate and, distance per stroke (SD). Scullers presented a fast start strategy for all races investigated. There was no significant difference in pacing between age groups. Velocity peaked at the 100m (U19) and 150m (U23 and senior) mark. The scullers in the older age groups showed superior performance scores of the studied variables. U19, Under 23 and senior single scullers present similar pacing regardless of their age group. Performance of single sculler improve from U19, to U23 to senior for both lightweight and heavyweight classes. The results of this study may serve as a reference for athletes, coaches, and sports scientists to be implemented in training.

Antimicrobial activity of Amazonian oils Andiroba and Copaiba against Paenibacillus larvae has been recently determined, indicating their potential use for the control of American Foulbrood Disease (AFB), but the use of essential oils in the environment still represents a challenge. The oils present several volatile elements in its composition, such high volatility being the cause of a sharp decline in antimicrobial activity. In this context the nanostructuration of these amazon oils may decrease the volatile characteristic of such products. The following research aimed to evaluate the activity of nanoemulsions prepared with Andiroba and Copaiba oils against Paenibacillus species. The toxicity of nanoemulsions has also been investigated with larvae and adult worker bees. Nanoemulsions (NE1, 10% Andiroba oil; NE2, 10% Copaiba oil; and NE3, 10% medium-chain triglyceride as negative control) were prepared in a high pressure homogenizer. The particle sizes were determined as 192, 211, and 178 nm for NE1, NE2, and NE3, respectively. The z potential values were −56.4, −47.1, and −27.2, respectively. NE1 and NE2 showed minimal inhibitory concentration (MIC) values lower than 0.39% for most Paenibacillus species tested. None of the strains were inhibited by negative control NE3. The timeresponse effect of the nanoemulsions has been tested on P. larvae ATCC9545, resulting in a decrease in the number of viable cells to less than 1 log CFU/ml for NE1. The nanoemulsion NE1 showed a significant toxic effect for the larvae (26% mortality) when compared with NE2 (13%) and NE3 (7%). The toxic effect of nanoemulsions has also been evaluated for 72 h in adult worker bees and low mortality rate was only observed for the NE1 treatment (8.3%). This study shows for the first time that nanoemulsions of Copaiba oil can be a potential candidate for the treatment or prevention of AFB.

The present report describes the endodontic treatment of an Oehlers type II dens invaginatus in a maxillary lateral incisor with 5 root canals, an extremely rare condition. Apical periodontitis and related symptoms were noted. Cone-beam computed tomography was used to aid the diagnosis, reveal tooth morphology, and assist in canal location. The pulp chamber was carefully accessed, and the root canals were explored under magnification. All root canals were prepared with an R25 Reciproc Blue system and sodium hypochlorite (NaOCl) irrigation. After initial preparation, a self-adjusting file (SAF) with NaOCl and ethylenediaminetetraacetic acid was used to complement the disinfection. Additionally, calcium hydroxide medication was applied. Vertical compaction was used to fill the canals with a calcium silicate-based endodontic sealer and gutta-percha. After 12 months, the patient exhibited healing of the periapical region, absence of symptoms, and normal dental function. In conclusion, this nonsurgical treatment protocol was successful in promoting the cure of apical periodontitis. Both complementary disinfection with an SAF and use of calcium hydroxide medication should be considered when choosing the best treatment approach for dens invaginatus with very complex anatomy.
HIV/AIDS-related lymphoma: perspective from a regional cancer center in India

BackgroundIndia has the third largest population of people living with HIV/AIDS (PLHA). Lymphoma is the second most common malignancy among PLHA. However, data are lacking regard-ing HIV/AIDS-related lymphoma (ARL) in India. This study evaluated the epidemiology and clinical outcomes of ARL from a regional cancer center in India.MethodsThis retrospective analysis included cases of ARL between March 2011 and September 2017. Data were obtained from patient record files for the assessment of epidemiology and clinical outcomes. Statistical analysis was performed using GraphPad Prism 6. Comparisons of subtype-specific survivals were performed using log-rank tests.ResultsOf 1,226 lymphoma cases, 80 (6.5%) were ARL. Details were available for 70 patients. The median age at diagnosis was 40.5 (9‒74) years with a male:female ratio of 2:1. AIDS-defining lymphomas (ADL) constituted 78.6% of cases, while 21.4% had non-AIDS defining lymphoma (NADL). The mean CD4 counts were 193.15±92.85 and 301.93±107.95 cells/L, respectively (t-test; P=0.0002). Extranodal involvement was present in 55.7%, B symptoms were reported in 60%, and lactate dehydrogenase (LDH) was elevated in 64.3% of patients. The median overall survival times were 6 months for plasmablastic lymphoma (PBL), 23 months for diffuse large B-cell lymphoma (DLBCL), and was not reached for Hodgkin’s lymphoma (log-rank test; P=0.0011). Other histo-logical subtype cases were too few to draw meaningful survival outcomes.ConclusionARL is a heterogeneous disease. Histologic subtype is a major determinant of the clinical outcome. ADL has significantly lower CD4 counts than those of NADL. There is an urgent and unmet need for uniform management guidelines for improving outcomes in this un-der-represented patient population.
