      • Helicobacter pylori와 대장균의 Shuttle Vector 개발

        In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

      • 탐구형 기하 소프트웨어 환경에서 피드백을 활용한 증명학습에 관한 연구

        탐구형 기하 소프트웨어의 환경에서 기하교육이 유용하다는 많은 연구 결과가 있었다. 본 연구에서는 소프트웨어의 환경에서 교사가 적절한 피드백을 주었을 때 작도문제를 통한 증명학습에 어떠한 영향을 주는지에 대하여 분석하였다. 그 결과 교사는 학생들의 수준을 적절히 판단하여 적합한 피드백을 줄 수 있고 학생들의 사고력의 향상과 지속성을 관찰할 수 있었고 학습태도에 긍정적인 영향을 끼치는 것으로 나타났다. The purpose of this study is to investigate, through case study, how the feedback with dynamic geometry software has influence on the improvement of proof learning and student's attitude. Analysis of data including worksheets, computer activities, and interviews revealed that collaborative learning is enhanced and student's attitudes could be changed positively.

      • 형광검출 역상 액체크로마토그래피에 의한 Histamine의 정량

        Histamine을 정확하고 신속하게 정량하기 위해 9.Fiuorenylmethyl chloroformate를 형광유도체화제로 하여 역상 HPLC법으로 정량하였다. 히스타민을 형광유도체화할 때 반응액의 pH, 반응시간, 형광유도체화제의 농도 등 최적 반응 조건을 검토하였다. 이 방법으로 히스타민을 분석한 결과 0.1~0.5ug/ml의 농도범위에서 상관계수가 0.922인 양호한 직선성을 나타내었으며 검출한계는 0.01??/ml였다. A rapid and simple method for the determination of histamine by reverse-phase high performance liquid chromatography with fluorescence detection was established. 9-Fluorenylmethyl chloroformate(FMOC) was used as fluorescent derivative reagent. The optimum conditions for the derivatiation such as pH, reaction time and he concentration of FMOC were investigated. Linearity of calibration curve was obtained between 0.1ug/ml/and 0.5ug/ml(r=0.922) and the limit of detection was 0.01ug/ml.

      • 김치식이가 흰쥐의 항산화시스템에 미치는 영향

        김치는 배추를 주재료로 하여 마늘, 고추, 생강 등의 양념류를 포함하는 여러 재료들로 만들어지며 이들 재료 중에는 항산화효과를 가진 것으로 알려진 성분들이 많이 함유되어 있어 김치도 항산화효과를 나타낼 것으로 여겨진다. In vitro 실험계 및 세포 모델계에서 김치 재료 및 김치의 항산화효과는 확인된바 있으나 In vivo 시스템에서의 김치의 항산화효과에 관한 자료는 부족하다. 따라서 본 실험에서는 김치 섭취의 생체 내 항산화효과를 규명하기 위하여 흰쥐에게 동결건조한 김치 3, 5, 10% 첨가 식이를 6주간 섭취시키면서 간조직의 산화와 항산화효소계의 활성을 측정하였다. 간의 MDA 함량은 대조군에 비해 김치 투여군에서 낮았고 특히 5% 김치섭취군에서 현저히 낮은 값을 나타내었다. 단백질 산화를 나타내는 카르보닐 함량도 대조군에 비해 김치섭취군에서 낮아 생체 내에서 김치식이가 과산화물 생성 억제효과를 나타내었다. 항산화 효소계의 활성을 측정한 결과 SOD 활성은 대조군에 비해 김치섭취군에서 현저하게 높았고 김치첨가 함량이 증가할수록 SOD 활성이 유의적으로 높아져 10% 김치섭취군에서는 대조군에 비해 70% 이상 높은 활성을 나타내었다. Catalase 활성은 각 군에서 유의적인 차이가 나타나지 않았으나 GSH-px의 활성은 김치 섭취가 높을수록 상대적으로 높은 증가율을 나타내어 SOD 활성과 동일한 경향을 나타내었다. 따라서 김치식이는 생체 내에서 치질 및 단백질 과산화를 현저하게 억제하였고 항산화효소계인 SOD와 GSH-px의 활성을 농도의존적으로 증진시켜 김치식이가 체내의 산화적 스트레스를 효과적으로 억제하는데 기여함을 알 수 있었다. Kimchi is a Kruean traditional fermented food containing various ingredients such as Chinese cabbage, garlic, ginger, red pepper and fish sauce. Some of their components were known as its antioxidative activities and antioxidative effects of kimchi have been studied using in vitro and cell model system. To investigate the antioxidative effect of dietary kimchi, 4 groups of male S.D. rats (n=36) were fed 0, 3%, 5%, and 10% freeze-dried kimchi for 6 weeks and hepatic lipid peroxidation, protein oxidation and the activities of antioxidative enzymes were measured. Hepatic MDA content fed kimchi diet was lower than control diet. Protein carbonyl values were decreased in kimchi diet group, but not significant. SOD activities were significantly increased in kimchi added groups at dose-dependent manner. At 10% kimchi added group, SOD activity increased by 70% compared with control diet group. Catalase activities were not different between experimental groups and control group, whereas GSH-px activities were significantly increased in kimchi added group. As a result kimchi diet inhibited lipid and protein oxidation and induced antioxidative enzymes, SOD and GSH-px in rat hepatic tissue. It suggests that dietary kimchi is expected to prevent the oxidative stress in animal and human.

        오리엔탈나리의 球根 收穫時期와 低溫處理 期間이 生育에 미치는 影響

        This study was carried out to determine the effect of the duration of chilling treatment and the lifting time of bulbs on growth of Lilium Oriental hybrids for product of cut flower in the type of forcing or semiforcing culture in Korea. "Casablanca", "Acapulco" and "Le Reve" in lilium Oriental hybrids were used in these experiment and carried out from 1999 to 2000 in Iksan, Korea. The bulbs for experiment were lifted on August 27, September 27 and October 27 and the all of them were chilled at 5±1℃ for 0,8,10,12 weeks. Development of the bulbs lifted on October 27 was superior than that of lifted bulbs on August 27 in the three cultivars. The bulbs lilted lately and chilled for a long time were sprouted more earlier and grown faster. In order to increase bulb sprouting until 100%, the duration of chilling treatment of bulbs was need for 12 weeks on August 27, 10 weeks on September 27 and above 8 weeks on October 27. The flowering according to the duration of chilling treatment and the lifting time of bulbs was same above. Consequently, the bulbs should be chilled at 5±1℃ for chilled for 10 weeks at late September and 8 weeks in producted bulbs at late October in earlyㆍMidseason variety fur product of cut flower hut bulbs of late variety should be chilled for 12 weeks at late September and 10 weeks at late October.

      • 방사선 치료를 받는 암 환자들의 사회적 지지와 삶의 질과의 관계

        Objective : This study was performed to investigate the relationship between social support and quality of life among cancer patients receiving radiation therapy. Matehals and Methods : The data were collected from 98 patients, who were receiving radiation therapy at two university hospitals located in GwangUJu, used by structured questionnaire. For statistical analyses of the association between quality of life and various characteristics, data were analyzed using t-test, ANOVA, Pearson 's correlation, and multiple regression analysis. Results: 1. There were 56(57.1%) males and 42(42.9%) females. Age ranged from 21 to 82 years. The primary sites of cancer were gastrointestinal tract (24.5%), lung (23.5%), breast (21.4%), and head and neck (11.2%) in order. 2. The mean scores of social, family, and medical support were 4.30 0.58, 4.49 0.78, 4.11 0.65, respectively. The score of quality of life was 5.83 1.63 (range: 1.95 ~ 9.05). 3. An analysis of the association between several factors of patients and quality of life showed that the statistically significant factors were age, the presence of distant metastasis, family support, medical support and social support. 4. As a result of the multiple regression analysis, only social support was significant (β=0.932, P=0.02) with quality of life, but age and presence of distant metastasis were not significant. Conclusion : This suggests that quality of life in cancer patients could be improved by strengthening the social support which consists of family and medical support. Further study would be necessary to evaluate separately several aspects of quality of life among cancer patients.

      • 평균대 경기 시 앞 공중돌기 동작에 대한 운동학적 분석

        본 연구는 평균대에서 수행되는 앞 공중돌기 동작의 효율적인 동작을 연기하는데 필요한 정보를 제공하기 위한 운동학적 요인을 살펴보는 데 있다. 연구대상은 부산동아시아 체조경기대회(1997년)평균대 종목별 결승에 참가한 한국선수 1명, 카자흐스탄 2명, 일본선수 1명을 대상으로 3차원 영상 분석을 통하여 운동학적 요인을 분석한 결과 다음과 같은 결론을 얻었다. 1. 각국면별 소요시간 도움닫기 시간은 스텝시간이 대상자별로 다르게 실시함으로서 수행시간간에 차이를 보였다. 도약 시간은 선행연구 결과에 비해 본 연구의 대상자들이 보다 짧은 시간을 나타냈으며, 체공시간은 길게 소요하고 있는 것으로 나타났다. 2. 체공 및 착자시 인체중심 높이는 선행연구된 마루 운동의 수행 결과들 보다는 다소 낮게 나타났다. 3. 도약시 인체중심 속도는 수직속도보다 수평속도가 높게 나타났다. 4. 도약 및 착지시 선정된 각 관절의 각도는 선행연구 결과들 보다 무릎각을 제외한 발목, 고관절 각도에서 적은 각도를 나타내 불안정한 착지를 수행하는 것으로 나타났다. The purpose of this study was to analyze the kinematic variable of a forward somersault motion in balance beam. Two video cameras were used for 3-dimentional analysis. The subjects were four female gymnasts who is participated in the '97 east asian game. The conclusions are follows. 1. Time of each phase The time of run-up was shown a difference between performance time as type of step patterns of each subjects. The time of support was display a more short time in the gymnasts of present study compared to gymnasts of previous study, and it was shown a long time in the flight phase. 3. The velocity of center of gravity in supports phase was shown more high levels in horizontal velocity than vertical velocity. 4. The angle of each joint in support and landing was display a low angles in ankle and hip joint angles except to knee joint angles. Therefore, it was performing a unbalanced landing.

        청소년기 (8세에서 16세) 구순부 성장변화에 관한 누년적 연구

        악안면 부위의 연조직은 기능, 심미성 그리고 치료후의 안정에 크게 영향을 주므로 진단과 치료계획 수립시 경조직 뿐 아니라 연조직을 포함한 평가가 이루어져야 한다. 구순부는 전체적인 안모형태 결정에 상당한 영향을 미치며 교정치료로 인한 치아의 이동에 따라 구순부 외형이 변화하므로 교정치료시 고려해야할 중요한 부분이다. 본 연구에서는 구순부 외형의 연령 증가에 따른 변화 양상을 규명하여 치료계획 수립과 진단 및 치료의 예후판정등, 임상 교정학 분야에 도움이 되는 지침을 얻고자 8세부터 16세까지 (남자 15명, 여자 15명) 촬영된 연속 두부 방사선 규격사진을 이용하여 구순부 외형의 누년적인 변화를 조사하여 다음과 같은 결론을 얻었다. 1. 상하순 고경은 남,녀 모두가 14세에서 가장 많은 성장량을 보였다. 2. 상하순 후경은 대부분의 연령군에서 여자보다 남자에서 큰 값을 보였으며 성장량에 있어서 A point에서는 14세에서 가장 많았고 Ls, Li, B point에서는 10-11세 이후로 성장량의 감소를 보였다. 3. 하순에 의해 가려지는 상악 절치부의 길이는 남자에서 9-11세때 가장 큰 값을 보였고 여자는 연령에따라 점차 증가하였다. 4. Nasolabial angle은 남, 녀 모두에서 연령이 증가함에 따라 약간씩 감소하였다. 5. Mentolabial angle은 변이도(variability)가 매우 높았다. This study was undertaken to assess the effect of growth on the lips. Not only does lip growth influence the stability of such orthodontic treatment; it also directly influence facial profile, in which the lips have an important part An understanding of the growth of lips is thus central to a consideration of profile change in orthodontics. By analyzing the serial lateral cephalograms of 15 male and 15 female of 8 years old to 16 years old who have normal occlusion. The result of this study were summerized as follows; 1. The largest growth increments in the length of the lips was occured age of 14 in both sexes. 2. The thickness of lips showed lager value for the male than that of the female in the most age group. The lagest growth increments at A point was occured age of 14, while Ls, Li, B point decreased after the age of 10-11. 3. The largest increase in the interval between crest of lower lip and edge of upper incisors was occured between ages 9 and 11 in males. The interval decreased slightly from 8 to 16 years in females. 4. The nasolabial angle decreased slightly from 8 to 16 years in both sexes. 5. The mentolabial angle showed large variation.

        가토 대퇴정맥에서 헤파린 국소 적용과 혈관냉동이 미세혈관문합부 혈전 형성에 미치는 영향

        The effect of topical heparin application and vascular freezing on thrombosis was evaluated in the femoral vein of rabbits. The femoral veins were crushed, incised transverersely and treated by 4 different ways; 1) group 1, heparin treated only, 2) group 2, frozen only, 3) group 3, heparin treated and frozen,4) group 4, treated with saline as control. The patency was evaluated by empty-and-refill test and thrombus formation was judged by stereoscopic and SEM observation. The results obtained were as follows : 1.Thirty minutes after suture, the patency was more improved in the three groups than the control, but there was no significant difference among groups. 2.Three days after suture, the patency was more improved in the three groups than the control, and was prominent in group 1 and group 3 (P<0.05). 3.Significantly reduced thrombus could be seen in most case of group 3. These results suggest that topical application of heparin and vascular freezing is effective in
