Prevalence of tumor BRCA1 and BRCA2 dysfunction in unselected patients with ovarian cancer

Objective The therapeutic benefits of poly(ADP-ribose) polymerase inhibitors highlight the need to evaluate BRCA1/2 defects in tubal/ovarian cancer (OC). We sought to determine the pattern and disease characteristics associated with tumor BRCA1/2 mutations and BRCA1 methylation in women with OC. Methods We obtained 111 OC specimens from 2 university hospitals and assessed BRCA1/2 mutations and BRCA1 methylation in tumor DNA. The frequency and pattern of BRCA1/2 defects were examined. Associations between patient/disease characteristics and BRCA1/2 defects were ascertained (Fisher’s exact test). Platinum-free interval (PFI), progressionfree survival (PFS), and overall survival (OS) based on the underlying BRCA1/2 defect were determined (Kaplan-Meier analysis [log-rank test]). Results We observed a BRCA1/2 dysfunction rate of 40% (28/70) in high-grade serous tubal/ovarian cancer (HGSC), including 14.3% BRCA1 methylation (n=10), 7.1% BRCA1 mutation (n=5), and 18.6% BRCA2 mutation (n=13). Defects in BRCA1/2 genes were associated with stage III/IV HGSC (BRCA1 methylation: P=0.005 [stage III/IV] and P=0.004 [HGSC]; BRCA1/2 mutation: P=0.03 [stage III/IV] and P<0.001 [HGSC]). Patients with BRCA1/2-mutated cancers showed improved OS (hazard ratio [HR], 0.65; 95% confidence interval [CI], 0.43-0.99; P=0.045) and a trend toward improved PFI (HR, 0.48; 95% CI, 0.22-1.06; P=0.07) and PFS (HR, 0.72; 95% CI, 0.51-1.03; P=0.07). No survival differences were observed between BRCA1-methylated and BRCA1/2 wild-type non-BRCA1-methylated cancers. Conclusion We observed a high tumor BRCA1/2 dysfunction rate in HGSC with a unique predominance of BRCA2 over BRCA1 mutations. While BRCA1/2 mutations conferred survival benefits in OC, no such association was observed with BRCA1 methylation.
Pain Assessment in Children Using a Modified Wong Baker Faces Pain Rating Scale

Objective: Pain assessment in children is an important measure in treatment planning and to establish effectiveness of treatment. To evaluate the efficiency of a Modified Wong–Baker Faces Scale (MWBFS) and evaluate its comfortability with Wong–Baker Faces Scale (WBFS). Methods: Study consists of 50 children aged 3 to 12 years old. Following dental treatment like restoration, extraction, pulp therapy etc, patients were asked to relate the pain experienced during treatment by selecting a face from the MWBFS template and WBFS template. Patients’ comfortability with both the scales recorded. Results: All the 50 children preferred the MWBFS (100% preference) over the WBFS. A statistical significance of <0.001 indicates true reliability of MWBFS. Conclusion: MWBFS is a reliable scale. Hence, it can be used successfully in pediatric dentistry by clinicians and researchers.
Gastric Adenocarcinoma with Prostatic Metastasis

Metastasis of gastric adenocarcinoma to the prostate gland is extremely rare. Herein, we report a case of gastric adenocarcinoma in a 56-year-old man with prostatic metastasis diagnosed through the analysis of biopsy specimens from representative lesions in the stomach and prostate gland. Immunohistochemistry of the prostatic tissue showed positive staining for cytokeratin 7 and negative staining for prostate-specific antigen (PSA), whereas the serum PSA level was normal, confirming the diagnosis of prostatic metastasis from carcinoma of the stomach.

Sulfonated poly(fluorenyl ether sulfone) (PFES) with a terminally-crosslinked network structure was prepared by heat-induced crosslinking of the allyl-terminated telechelic sulfone polymers using a bisazide. The crosslinked polymer membrane with a sulfofluorenyl moiety of 70% (PFES-70) showed excellent hydrolytic,dimensional, and mechanical stability. The crosslinked PFES-70 membrane revealed a proton conductivity of 0.04S/cm at 20 oC, which increased significantly with increasing temperature. The conductivity of 0.38 S/cm at 100 oC for PFES-70 was higher than that of both non-crosslinked (0.33 S/cm) and Nafion® (0.17 S/cm). In addition, the crosslinked PFES-70 showed a methanol permeability of only 1.4% (3.9×10-8 cm2 /s) compared to Nafion®.

The synthesis and properties of a solvent-free gel polymer electrolyte based on an imidazolium-poly(ethyleneoxide) (PEO)-based ionene electrolyte, a poly(vinylidene fluoride-hexafluoropropylene) (PVDF(HFP)) support, andthe lithium bis(trifluoromethane sulfonyl) imide (LiTFSI) salt are reported. A ternary gel polymer electrolyte film,produced from mixtures of the imidazolium-PEO-based ionene ([IPI][Br]), PVDF(HFP), and LiTFSI, displayedgood thermal and mechanical stability. A very high room temperature conductivity of 2.6×10-3 S/cm and a broadelectrochemical stability window were obtained, demonstrating the utility of the electrolyte for practical Li-ion battery applications.

Public service motivation (PSM) has been found to predict job satisfaction (JS) of government and nonprofit organization employees in many varied industry, job grade, country, and culture contexts. However, the role that culture plays in this link, vis-à-vis the politico-economic context of countries, is not well understood. This meta-analysis looks at a combined sample size of 45,812 individuals, from 10 countries and 32 studies; and attempts to disambiguate the influence of these two moderators. Subgroup analysis and meta-regression techniques are employed to test hypotheses and contribute toward theory building. The study finds Asian countries to have a stronger correlation between PSM and JS, compared to Western countries; while two cultural traits, performance orientation and gender egalitarianism, demonstrate significantly higher strengthening impact in correlation effect sizes in Asia vis-a-vis the West. Future research directions and limitations are discussed.

Higher prediction efficacy is a very challenging task in any field of engineering. Due to global warming, there is a considerable increase in the global sea level. Through this work, an attempt has been made to find the sea level variability due to climate change impact at Haldia Port, India. Different statistical downscaling techniques are available and through this paper authors are intending to compare and illustrate the performances of three regression models. The models: Wavelet Neural Network (WNN), Minimax Probability Machine Regression (MPMR), Feed-Forward Neural Network (FFNN) are used for projecting the sea level variability due to climate change at Haldia Port, India. Model performance indices like PI, RMSE, NSE, MAPE, RSR etc were evaluated to get a clear picture on the model accuracy. All the indices are pointing towards the outperformance of WNN in projecting the sea level variability. The findings suggest a strong recommendation for ensembled models especially wavelet decomposed neural network to improve projecting efficiency in any time series modeling.
Modelling land surface temperature using gamma test coupled wavelet neural network

The climate change has made adverse effects on land surface temperature for many regions of the world. Several climatic studies focused on different downscaling techniques for climatological parameters of different regions. For statistical downscaling of any hydrological parameters, conventional Neural Network Models were used in common. However, it seems that in any modeling study, uncertainty is a vital aspect when making any predictions about the performance. In this paper, Gamma Test is performed to determine the data length selection for training to minimize the uncertainty in model development. Another measure to improve the data quality and model development are wavelet transforms. Hence, Gamma Test with Wavelet decomposed Feedforward Neural Network (GT-WNN) model is developed and tested for downscaled land surface temperature of Patna Urban, Bihar. The results of GT-WNN model are compared with GT-FFNN and conventional Feedforward Neural Network (FFNN) model. The effectiveness of the developed models is illustrated by Root Mean Square Error and Coefficient of Correlation. Results showed that GT-WNN outperformed the GT-FFNN and conventional FFNN in downscaling the land surface temperature. The land surface temperature is forecasted for a period of 2015-2044 with GT-WNN model for Patna Urban in Bihar. In addition, the significance of the probable changes in the land surface temperature is also found through Mann-Kendall (M-K) Test for Summer, Winter, Monsoon and Post Monsoon seasons. Results showed an increasing surface temperature trend for summer and winter seasons and no significant trend for monsoon and post monsoon season over the study area for the period between 2015 and 2044. Overall, the M-K test analysis for the annual data shows an increasing trend in the land surface temperature of Patna Urban.
