      • Curcumol Induces Apoptosis in SPC-A-1 Human Lung Adenocarcinoma Cells and Displays Anti-neoplastic Effects in Tumor Bearing Mice

        Curcumol is a sesquiterpene originally isolated from curcuma rhizomes, a component of herbal remedies commonly used in oriental medicine. Its beneficial pharmacological activities have attract significant interest recently. In this study, anti-cancer activity of curcumol was examined with both in vitro and in vivo models. It was found that curcumol exhibited time- and concentration-dependent anti-proliferative effects in SPC-A-1 human lung adenocarcinoma cells with cell cycle arrest in the G0/G1 phase while apoptosis-induction was also confirmed with flow cytometry and morphological analyses. Interestingly, curcumol did not display growth inhibition in MRC-5 human embryonic lung fibroblasts, suggesting the anti-proliferative effects of curcumol were specific to cancer cells. Anti-neoplastic effects of curcumol were also confirmed in tumor bearing mice. Curcumol (60 mg/ kg daily) significantly reduced tumor size without causing notable toxicity. In conclusion, curcumol appears a favorable anti-cancer candidate for further development.

      • Decreased Expression of LKB1 Correlates with Poor Prognosis in Hepatocellular Carcinoma Patients Undergoing Hepatectomy

        Aim: To study any correlation of LKB1 expression with prognosis in hepatocellular carcinoma (HCC) cases. Methods: A total of 70 HCC patients and 20 primary intrahepatic stone patients in the first affiliated hospital of Wenzhou Medical College were enrolled in this study. LKB1 expression was detected by immunohistochemistry. Patients were followed-up and prognostic factors were evaluated. Result: LKB1 expression was decreased in the HCC samples. Loss of LKB1 expression in HCC was significantly related to histologic grade (P=0.010), vascular invasion (P=0.025) and TMN stage (P=0.011). Patients showing negative LKB1 expression had a significantly shorter disease-free and overall survival than those with positive expression (P = 0.001, P=0.000, respectively). Multivariate Cox regression analysis indicated that LKB1 expression level was an independent factor of survival (P = 0.033). Conclusion: HCC patients with decreased expression LKB1 have a poor prognosis. The loss of LKB1 expression is correlated with a lower survival rate.

        Influences of heating processes on properties and microstructure of porous CeO2 beads as a surrogate for nuclear fuels fabricated by a microfluidic sol-gel process

        The control of microstructure is critical for the porous fuel particles used for infiltrating actinide nuclides. This study concerns the effect of heating processes on properties and microstructure of the fuel particles. The uniform gel precursor beads were synthesized by a microfluidic sol-gel process and then the porousCeO2 microspheres, as a surrogate for the ceramic nuclear fuel particles, were obtained by heatingtreatment of the gel precursors. The fabricated CeO2 microspheres have a narrow size distribution andgood sphericity due to the feature of microfluidics. The effects of heating processes parameters, such asheating mode and peak temperatures on the properties of microspheres were studied in detail. Anoptimized heating mode and the peak temperature of 650 C were selected to produce porous CeO2microspheres. The optimized heating mode can avoid the appearance of broken or crack microspheres inthe heating process, and as-prepared porous microspheres were of suitable pore size distribution andpore volume for loading minor actinide (MA) solution by an infiltration method that is used for fabricationof MA-bearing nuclear fuel beads. After the infiltration process, 1000 C was selected as the finaltemperature to improve the compressive strength of microspheres.

        Power Cable Ampacity and Influential Factors Analysis under Operation

        With the increasing of urban electricity demand, making the most use of the power cable carrying capacity has become an important task in power grid system. Contrary to the rated ampacity obtained under extremely conservative conditions, this paper presents the various steady value of cable ampacity by using the changing surrounding parameters under operation, which is based on cable ampacity calculation equation under the IEC-60287 standard. To some degree, the cable ampacity analysis of actual surroundings improves the transmission capacity of cables. This paper reveals the factors that influence cable ampacity such as insulating layer thickness, allowable long-term conductor temperature, the ambient temperature, soil thermal resistance coefficient, and so on, then gives the class of the influence of these parameters on the ampacity, which plays a great role in accurately calculating the real-time ampacity and improving the utilization rate of cable in the complex external environment condition. Furthermore, the transient thermal rating of the cable is analyzed in this paper, and temperature variation of the conductor under different overload conditions is discussed, which provides effective information for the operation and control of the system.

        Migraine Susceptibility Genes in Han Chinese of Fujian Province

        Background and Purpose Five single-nucleotide polymorphisms (SNPs) (rs4379368, rs10504861, rs10915437, rs12134493 and rs13208321) were recently identified in a Western population with migraine. These migraine-associated SNPs have not been evaluated in a Han Chinese population. This study investigated the associations of specific SNPs with migraine in a Han population. Methods This was a case-control study of Han Chinese residing in Fujian Province. Polymerase chain reaction—restriction-fragment-length polymorphism analysis and direct sequencing were used to characterize the relationships of SNPs in a control group of 200 subjects and in a migraine group of 201 patients. Results The frequencies of the five SNPs did not differ between patients with migraine and healthy non migraine controls. However, subgroup analysis indicated certain SNPs were more strongly associated with migraine with aura or migraine without aura than with controls. The CT genotype of rs4379368 was more common in migraine patients with aura (75%) than in migraine patients without aura (47.9%) and controls (48.5%) (p<0.05), and the TT genotype of rs10504861 was more common in migraine patients with aura than in controls (8.3% vs. 0.5%) (p<0.05). Meanwhile, the CC genotype of rs12134493 was less common in migraine patients without aura than in controls (80.6% vs. 88%) (p<0.05). Conclusions Our findings suggest that the rs4379368 and rs10504861 SNPs are markers for susceptibility to migraine with aura and that rs12134493 is a marker for the risk of migraine without aura in this Han population. Future studies should further explore if these associations vary by ethnicity.

        Numerical and experimental investigation on electromechanical aileron actuation system with joint clearance

        A closed-loop coupling model at the system-level is developed to analyze the effects of joint clearance on the dynamic responses of electromechanical aileron actuation system. The proposed model considers the coupling effects between the electromechanical actuator (EMA) control performance and dynamic characteristics of linkage mechanism with joint clearance. Besides, the experiments are conducted in a test rig, which verifies the effectiveness of the proposed closed-loop coupling model. The nonlinear contact force model and modified Coulomb friction model are adopted in the joint clearance of the linkage mechanism, and the influences of clearance size on the dynamic behaviors of electromechanical aileron actuation system are studied. The numerical and experimental results indicate that the novel closed-loop coupling model, considering the EMA control performance and dynamics of linkage mechanism with joint clearance at the same time, is an effective model to predict the dynamic characteristics of electromechanical aileron actuation with joint clearance, which provides a practical method to analyze the dynamic performance of electromechanical coupling multibody systems with joint clearance.

        Study on the dynamic interaction of multiple clearance joints for flap actuation system with a modified contact force model

        To analyze the effects of clearance joint number and dynamic interaction on responses of the flap actuation system, a dynamic analysis model is proposed based on a modified contact force model. The modified contact force model can take contact properties with small clearance, heavy load, large contact area and variable contact stiffness coefficient into consideration for the flap actuation system. Numerical results show that the actuation system presents violent fluctuation due to the dynamic interaction between multiple clearance joints, and the clearance joint closest to the input part suffers more serious contact effects. Furthermore, the combination motion modes between the multiple clearance joints are helpful to judge and analyze the motion state and dynamic behaviors of actuation systems. These simulation findings can provide a theoretical foundation for the optimization design, control strategy and engineering experiments of the actuation systems with multiple clearances.

        Polymorphisms in TYMS for Prediction of Capecitabine-Induced Hand-Foot Syndrome in Chinese Patients with Colorectal Cancer

        Purpose Capecitabine is an extensively used oral prodrug of 5-fluorouracil in treatment of colon cancer and is known to cause hand-foot syndrome (HFS). As the target enzyme for capecitabine, thymidylate synthase (TYMS) plays a key role for 5-fluorouracil metabolism and has been associated with some side effects caused by capecitabine. The aim of our study is to identify the possible genetic predictors of capecitabine-induced HFS (CAP-HFS) in Chinese colorectal cancer patients.Materials and Methods Whole exons of TYMS were sequenced for 288 extreme phenotype HFS patients, including 144 severe or early-onset (first 2 cycles) moderate HFS extreme cases and 144 extreme controls with no reported HFS. The associations between polymorphisms and CAP-HFS were analyzed using logistic regression under an additive model.Results We identified a novel risk mutation (c.1A>G, chr18:657743), was associated with severe HFS in an extreme case who was affected during the first cycle of treatment. Moreover, we identified three new variants, rs3786362, rs699517, rs2790, and two previously reported variants, 5’VNTR 2R/3R and 3′-untranslated region 6-bp ins-del, which were significantly associated with CAP-HFS (p < 0.05). In silico analysis revealed that the effect of these polymorphisms in the TYMS region on the development of HFS might not be restricted solely to the regulation of TYMS expression, but also the TYMS catalytic activity through the indirect effect on ENOSF1 expression.Conclusion This study identified new polymorphisms in TYMS gene significantly associated with CAP-HFS, which may serve as useful genetic predictors for CAP-HFS and help to elucidate the underlying mechanism of HFS.

      • Reduced Telomere Length in Colorectal Carcinomas

        Purpose: Telomeres play a key role in the maintenance of chromosome integrity and stability, and telomere shortening is involved in initiation and progression of malignancies. The aim of this study was to determine whether telomere length is associated with the colorectal carcinoma. Patients and methods: A total of 148 colorectal cancer (CRC) samples and corresponding adjacent non-cancerous tissues were evaluated for telomere length, P53 mutation, and cyclooxygenase-2 (COX-2) mutation detected by fluorescent immunohistochemistry. Telomere length was estimated by real-time PCR. Samples with a T/S>1.0 have an average telomere length greater than that of the standard DNA; samples with a T/S<1.0 have an average telomere length shorter than that of the standard DNA. Results: Telomeres were shorter in CRCs than in adjacent tissues, regardless of tumor stage and grade, site, or genetic alterations (P=0.004). Telomere length in CRCs also had differences with COX-2 status (P=0.004), but did not differ with P53 status (P=0.101), tumor progression (P=0.244), gender (P=0.542), and metastasis (P=0.488). There was no clear trend between T/S optimal cut-off values (<1 or > 1) and colorectal tumor progression, metastasis, gender, P53 and COX-2 status. Conclusion: These findings suggesting that telomere shortening is associated with colorectal carcinogenesis but does not differ with tumor progression, gender, and metastasis.
