        Intron 4 VNTR (4a/b) Polymorphism of the Endothelial Nitric Oxide Synthase Gene Is Associated with Breast Cancer in Mexican Women

        The endothelial nitric oxide synthase (eNOS) gene plays an important role in several biological functions. Polymorphisms of the eNOS gene have been associated with cancer. It has been suggested that the VNTR 4 a/b polymorphism may affect the expression of eNOS and contributes to tumor promotion in the mammary gland. We examined the role of the eNOS4 a/b polymorphism by comparing the genotypes of 281 healthy Mexican women with the genotypes of 429 Mexican women with breast cancer (BC). The observed genotype frequencies for control and BC patients were 0.6% and 0.7% for a/a (polymorphic); 87% and 77% for a/a (wild type); and 12% and 22% for a/b respectively. We found that the odds ratio (OR) was 1.9, with a 95% confidence interval (95%CI) of 1.29-2.95, P=0.001 for genotypes a/a-a/b, b/c. The association was also evident when comparing the distribution of the a/a-a/b genotypes in patients with high levels of glutamate-oxaloacetate transaminase (SGOT) (OR, 1.93; 95% CI, 1.14-3.28; P=0.015); undergoing menopause with high levels of SGOT (OR, 2.0; 95% CI, 1.1-3.84); and with high levels of glutamic-pyruvic transaminase (SGPT) (OR, 3.5; 95% CI, 1.56-8.22). The genotypes a/a-a/b are associated with BC susceptibility in the analyzed samples from the Mexican population.

        User Preferences Related to Multimedia Elements of a Mobile Application to Prevent Diabetes

        Objectives: To understand user preferences related to the characteristics of an application that promotes and provides educationon healthy habits to correctly design multimedia elements. Methods: We conducted a comprehensive qualitative studywith a hermeneutical strategy, which gathered information using well-researched questions that were posed to focus groupsconsisting of 32 participants. These participants were asked for opinions related to multimedia elements to display educationalmessages about physical activity and healthy eating in a mobile application. There were three analysis categories ofmultimedia elements: text, visual elements, and audio elements. Results: The majority of the participants, 93.75%, were in thelow socioeconomic stratum; 68.75% are in a civil union with their partner; 53.12% completed or failed to complete secondaryschool, and 68.75% were housewives. Based on the qualitative results, we found that mobile applications become mediatingtools that support the adoption of actions that tend to improve lifestyles and increase knowledge about proper nutrition andphysical activity. Text messages used in mobile applications should promote healthy habits and remind users of their benefits. Images and videos should be accompanied by text and audio to provide greater clarity regarding recommendations of healthyhabits. Conclusions: Technology must provide accessibility and coverage opportunities, while meeting the needs and expectationsof users. It should facilitate primary health intervention through education to transform unhealthy behaviors andgenerate lifestyles that improve the health of the user and their family context.

        Transcription Factors Tec1 and Tec2 Play Key Roles in the Hyphal Growth and Virulence of Mucor lusitanicus Through Increased Mitochondrial Oxidative Metabolism

        Mucormycosis is a lethal and difficult-to-treat fungal infection caused by fungi of the order Mucorales. Mucor lusitanicus, a member of Mucorales, is commonly used as a model to understand disease pathogenesis. However, transcriptional control of hyphal growth and virulence in Mucorales is poorly understood. This study aimed to investigate the role of Tec proteins, which belong to the TEA/ATTS transcription factor family, in the hyphal development and virulence of M. lusitanicus. Unlike in the genome of Ascomycetes and Basidiomycetes, which have a single Tec homologue, in the genome of Mucorales, two Tec homologues, Tec1 and Tec2, were found, except in that of Phycomyces blakesleeanus, with only one Tec homologue. tec1 and tec2 overexpression in M. lusitanicus increased mycelial growth, mitochondrial content and activity, expression of the rhizoferrin synthetase-encoding gene rfs, and virulence in nematodes and wax moth larvae but decreased cAMP levels and protein kinase A (PKA) activity. Furthermore, tec1- and tec2-overexpressing strains required adequate mitochondrial metabolism to promote the virulent phenotype. The heterotrimeric G beta subunit 1-encoding gene deletant strain (Δgpb1) increased cAMP-PKA activity, downregulation of both tec genes, decreased both virulence and hyphal development, but tec1 and tec2 overexpression restored these defects. Overexpression of allele-mutated variants of Tec1(S332A) and Tec2(S168A) in the putative phosphorylation sites for PKA increased both virulence and hyphal growth of Δgpb1. These findings suggest that Tec homologues promote mycelial development and virulence by enhancing mitochondrial metabolism and rhizoferrin accumulation, providing new information for the rational control of the virulent phenotype of M. lusitanicus.

        Prophylaxis for invasive fungal infection in pediatric patients with allogeneic hematopoietic stem cell transplantation

        Background Antifungal prophylaxis is recommended for hematopoietic stem cell transplantation (HSCT) to decrease the incidence of invasive fungal infections (IFI). This study aimed to compare the two groups of antifungal prophylaxis in pediatric patients undergoing allogeneic HSCT. Methods This observational, analytic, retrospective cohort study compared the incidence of IFI with antifungal prophylaxis with voriconazole vs. other antifungals in the first 100 days after allogeneic HSCT in patients aged <18 years between 2012 and 2018. The statistical analysis included univariate and multivariate analyses and determination of the cumulative incidence of invasive fungal infection by the Kaplan‒Meier method using STATA 14 statistical software. Results A total of 139 allogeneic HSCT were performed. The principal diagnosis was acute leukemia (63%). The 75% had haploidentical donors, and 50% used an antifungal in the month before transplantation. Voriconazole (69%) was the most frequently administered antifungal prophylaxis. The cumulative incidence of IFI was 5% (7 cases). Of the patients with IFIs, four began prophylaxis with voriconazole, one with caspofungin, and one with fluconazole. Additionally, six were possible cases, one was proven (Candida parapsilosis), and 1/7 died. Conclusion There were no differences in the incidence of IFI between patients who received prophylaxis with voriconazole and other antifungal agents.

        Chemical bath deposition of CdS films on different substrate orientations

        Cadmium sulfide (CdS) thin films were prepared by the chemical bath deposition (CBD) technique on glass substrates under different substrate orientation and two agitation modes of the bath. Groups of CdS films were grown onto horizontal, vertical, and 45° inclined substrates, meanwhile the chemical bath was affected by magnetic or oscillating agitation. Optical homogeneity was studied by means of the band gap energy measured on different sites of the film. Morphology, thickness, stoichiometry and crystalline structure of films were characterized by atomic force microscopy, profilemetry, energy dispersive spectroscopy, and X-ray diffraction techniques, respectively. Relations between measured properties,substrate orientation and agitation types are discussed. In all cases, a cubic phase with (111) as main peak diffraction was obtained in films. Oscillating agitation in bath produces large grain size with low roughness, constant deposition rate and better crystal orientation as compared with magnetic agitation. CdS films grown on vertical substrates result with low roughness, high Cd/S ratio, band gap energy values near to the typical value (2.42 eV) and better crystalline structure than other substrate orientation.

        Sources, Characteristics, and Therapeutic Applications of Mesenchymal Cells in Tissue Engineering

        Tissue engineering (TE) is a therapeutic option within regenerative medicine that allows to mimic the original cell environment and functional organization of the cell types necessary for the recovery or regeneration of damaged tissue using cell sources, scaffolds, and bioreactors. Among the cell sources, the utilization of mesenchymal cells (MSCs) has gained great interest because these multipotent cells are capable of differentiating into diverse tissues, in addition to their self-renewal capacity to maintain their cell population, thus representing a therapeutic alternative for those diseases that can only be controlled with palliative treatments. This review aimed to summarize the state of the art of the main sources of MSCs as well as particular characteristics of each subtype and applications of MSCs in TE in seven different areas (neural, osseous, epithelial, cartilage, osteochondral, muscle, and cardiac) with a systemic revision of advances made in the last 10 years. It was observed that bone marrow-derived MSCs are the principal type of MSCs used in TE, and the most commonly employed techniques for MSCs characterization are immunodetection techniques. Moreover, the utilization of natural biomaterials is higher (41.96%) than that of synthetic biomaterials (18.75%) for the construction of the scaffolds in which cells are seeded. Further, this review shows alternatives of MSCs derived from other tissues and diverse strategies that can improve this area of regenerative medicine.

        Management of large class II lesions in molars: how to restore and when to perform surgical crown lengthening?

        The restoration of endodontic tooth is always a challenge for the clinician, not only due to excessive loss of tooth structure but also invasion of the biological width due to large decayed lesions. In this paper, the 7 most common clinical scenarios in molars with class II lesions ever deeper were examined. This includes both the type of restoration (direct or indirect) and the management of the cavity margin, such as the need for deep margin elevation (DME) or crown lengthening. It is necessary to have the DME when the healthy tooth remnant is in the sulcus or at the epithelium level. For caries that reaches the connective tissue or the bone crest, crown lengthening is required. Endocrowns are a good treatment option in the endodontically treated tooth when the loss of structure is advanced.

        Correlation Between the Sinew Channels with the Myofascial System, Pathology, and Treatment

        The sinew channels are a tendon and muscle network, and their description is based on the observation presented on the Huangdi Neijing Ling Shu. However, the myofascial system is an uninterrupted series of connective tissue that is comprised of layers that run in different directions. The similarities on these pathways are compared, such as a brief description on the myofascial pain syndrome and its similitude with the Impediment disorder from the Traditional Chinese Medicine (TCM). Furthermore, we discuss the treatment of these conditions from a Traditional Chinese Medicine perspective.

        Mechanosynthesis of graphene nanoribbons from waste zinc–carbon batteries

        The mechanosynthesis route is a physical top–down strategy to produce different nanomaterials. Here, we report the formation of graphene nanoribbons (GNRs) through this route using carbon bars recovered from discarded alkaline batteries as raw material. The mechanosynthesis time (milling time) is shown to have an influence on different features of the GNRs such as their width and edges features. TEM revealed the presence of GNRs with widths of 15.26, 8.8, and 23.55 nm for the milling times of 6, 12, and 18 h, respectively. Additionally, the carbon bars evolved from poorly shaped GNRs for the shortest milling time (6 h) to well-shaped GNRs of oriented sheets forming for the longest milling time. Besides GNRs, graphene sheets (GNS) of different sizes were also observed. The Raman analysis of the 2D bands identified the GNS signal and confirmed the GNRs nature. ID/IG values of 0.21, 0.32, and 0.40 revealed the degree of disorder for each sample. The in-plane sp2 crystallite sizes (La) of graphite decreased to 91, 60, and 48 nm with increasing peeling time. The RBLM band at 288 cm−1 confirmed the formation of the GNRs. Mechanosynthesis is a complex process and the formation of the GNRs is discussed in terms of a mechanical exfoliation, formation of graphene sheets and its fragmentation to reach GNR-like shapes. It is shown that the synthesis of GNRs through the mechanosynthesis route, besides the use of recycled materials, is an alternative for obtaining self-sustaining materials.

        Ultrathin Polymer Fibers Coated with an Amorphous SiO2–CaO–P2O5 Bioactive Powders for Biomedical Applications

        The development of new biomaterials with improved properties is a trend in tissue regeneration. In this way, an innovative approach is employed in this work for obtaining polymer fibers coated with nanoparticles resulting from the simultaneous application of poly (lactic acid) (PLA)/polycaprolactone (PCL) electrospinning and bioactive particles from an amorphous multicomponent silica–calcium–phosphorus system (AMS) electrospraying. The osteogenesis was evaluated in vitro and in vivo using male rats, in which total protein, alkaline phosphatase, and biological performance through histological and histomorphometric analysis were discussed. The morphological results assessed by scanning electron microscopy showed a mesh of PLA/PCL fibers associated with AMS. The spraying of 17.44% of AMS particles in the PLA/PCL electrospun fibers decreased the Young’s modulus and tensile strength. However, the amount of AMS particles sprayed was enough to promote a reduction of 17.8% in the measured contact angle values. Phosphatase alkaline higher mean value was also observed in the fibers when in contact with the AMS, but nonstatistical difference was observed (p > 0.05). It was possible to observe the presence of mineralized nodules deposited on the bottom of the plate and between the fibers. The newly formed bone into defect filled with PLA/PCL-AMS fibers was higher than that observed in the control group. These findings suggest PLA/PCL-AMS fibers as a multifunctional composite system that may be attractive for both bone and dental tissue engineering applications.
