        Dynamic Changes of Transcriptome and Metabolites During Ripening of Alpinia Oxyphylla Fruit (AOF)

        Alpinia oxyphylla (Yizhi) is an economically valuable plant, not only used as edible fruit, but also as an important traditional medicinal material. However, there are few studies on fruit development and metabolic pathways. To elucidate the dynamic changes of metabolites and transcription levels during fruit development of A. oxyphylla, the four parts of A. oxyphylla fruit from different periods, including early fruit (EF), middle fruit (MF), late pericarp (LP) and late seed (LS) were collected. KEGG pathway analysis shows that DEGs in EF, MF, LP are involved in amino sugar and nucleotide sugar metabolism, starch and sucrose metabolism, and plant hormone signal transduction. However, at the late part of fruit development (LS), most of the DEGs between LS and LP were enriched in pyruvate metabolism, glycolysis/gluconeogenesis, tryptophan metabolism. We found that in the early and middle parts of fruit development (EF, MF), many differential metabolites were enriched in piperidine tropane and biosynthesis of secondary metabolites, pyridine alkaloid biosynthesis. In addition, important active components, such as nootkatone in A. oxyphylla fruits accumulated in the LS part, but did not show significant differences in the first three parts. Some key transcription factors that are significantly positively correlated with the maturation of AOF have also been identified. Together, our research provides new insights into the accumulation of metabolites and the dynamic changes of the transcriptome during fruit development.

        Establishment of a special pathogen free Chinese Wuzhishan Minipigs Colony

        To meet the increasing demands of specific pathogen free (SPF) minipigs in biomedical researches, 8 pregnant Chinese Wuzhishan minipigs (WZSP) sows with clear background were chosen to obtain SPF WZSP by hysterectomy. At $111{\pm}days2$ of the pregnancy, piglets were aseptically taken out from the sows and artificially suckled for 40 to 45 days in the positive isolators. Then, the piglets defined as F0 were transferred to barrier environment and fed with standard feeds. The original SPF colony was formed for breeding by selected piglets from F0 group of 6-8 months old. Biological characteristics of SPF WZSP were collected and further compared to those of conventional (CV) WZSP, including growth performance, reproductive performance, hematology and blood biochemistry, and major pathogens detection. As a result, 61 F0 piglets were obtained from 8 candidate sows, and 55 out of them survived. After strictly selection, 35 F0 piglets were used to form the original SPF colony, which produced 14 litters of SPF piglets defined as F1. Piglet survival rates, growth performance, and reproductive performance of SPF WZSP were similar to CV WZSP. Some hematology and blood biochemistry parameters showed significant differences between SPF and CV WZSP. Eighteen kinds of pathogens were identified to be free in F0 and F1 SPF colony by repeated pathogen detections. In conclusion, we established a satisfied SPF WZSP colony maintaining original characteristics, free of controlled diseases, and being proved to be a suitable laboratory animal.

      • Clinical Risk Factor Analysis for Breast Cancer: 568,000 Subjects Undergoing Breast Cancer Screening in Beijing, 2009

        Objectives: Although there are many reports about the risk of breast cancer, few have reported clinical factors including history of breast-related or other diseases that affect the prevalence of breast cancer. This study explores these risk factors for breast cancer cases reported in Beijing in 2009. Materials and Methods: Data were derived from a Beijing breast cancer screening performed in 2009, of 568,000 women, from 16 districts of Beijing, all aged between 40 and 60 years. In this study, multilevel statistical modeling was used to identify clinical factors that affect the prevalence of breast cancer and to provide more reliable evidence for clinical diagnostics by using screening data. Results and Conclusion: Those women who had organ transplants, compared with those with none, were associated with breast cancer with an odds ratio (OR)=65.352 [95% confidence interval (CI): 8.488-503.165] and those with solid breast mass compared with none had OR=1.384 (95% CI: 1.022-1.873). Malignant tendency was strongly associated with increased risk of breast cancer, OR=207.999(95% CI: 151.950-284.721). The risk of breast cancer increased with age, $OR_1$=2.759 (95% CI: 1.837-4.144, 56-60 vs. 40-45), $OR_2$=2.047 (95% CI: 1.394-3.077, 51-55 vs. 40-45), $OR_3$=1.668 (95% CI: 1.145-2.431). Normal results of B ultrasonic examination show a lower risk among participants, OR= 0.136 (95% CI: 0.085-0.218). Those women with ductal papilloma compared with none were associated with breast cancer, OR=6.524 (95% CI: 1.871-22.746). Therefore, this study suggests that clinical doctors should pay attention to these high-risk factors.

        A Study on the Customer Experience Design through analyzing Smart Hotels in China

        코로나(COVID-19) 폭발로 인해 비접촉 서비스의 특징과 장점이 갈수록 두드러졌고 스마트호텔의 발전이 가속 화되었다. 본 연구는 서로 다른 접촉점에서고객 체험 과정에 발생하는 스마트 서비스 체험을 식별, 분류 및 정의하는 데에 목적을 두고 있다. 기존 연구와 문헌을 바탕으로 스마트 호텔의 개념과 특징에 대해 연구 토론해 보았다. 스마트 호텔의 스마트 체험 인자와 고객 접촉점을 활용하여 분석 프레임 워크를 디자인하였다. 이로써 중국에서 선별된 스마트 호텔에 대해 현장 조사 및 분석을 수행한다. 사례 분석 결과를 보면 스마트 호텔의 고객 체험 디자인이 교호성, 개별화, 접근 가능성, 정보성, 프라이버시 안전성에 어느 정도 발전하고 있다는 것을 알 수가 있다. 위의 결과를 바탕으로 본 논문에서는 미래의 스마트 호텔 CX 디자인이 개인화된 서비스 관련 체험을 개발하기 위해 데이터를 통합하는 방향으 로 발전하는것을 제안한다. The outbreak of covid-19 has brought the characteristics and advantages of non-contact services to increased prominence, and the development of smart hotels”has accelerated. This study aims to identify, categorize and define the smart service experience at different touch points of the customer experience. The concept and characteristics of the smart hotel were examined based on existing research and literature. An analytical framework was designed using smart experience factors and customer touch points of smart hotels. Selected Chinese smart hotels were then examined under this framework. The case analysis results show that the customer experience design of smart hotels has developed to different degrees, in terms of interactivity, personalization, accessibility, information and privacy security. Based on the above findings, this article suggests that the design of smart hotels should use integrated data to further enhance personalized service experience.

        The Fingerprinting of Huangjinju Powder for Injection on Chinese Patent Medicine by XRD Fourier

        The purpose is to study the identification method of Huangjinju powder for injection and the medicinal materials by the fingerprint off-ray Diffraction Fourier (XRDF). We used the same method on both the studying of Huangjinju and the medicinal materials. Then we selected a few components alignment to compare. We analyzed the data by setting up the deviation $d({\AA})$ as ${\pm}0.05$ to calculate the rate of special mark on the sample (Px) and on the patent (P). The special XRDF of Huangjinju$[d({\AA})/(I/I_0)]$ have 5 peaks that have not expressed in medicinal materials. Therefore Px is 22.73%. Flos Trollii Chinensis has 3 special marks and Px is 17.65%. Flos Chrysanthemi Indici has 1 special mark and Px is 3.57%. Its coincided interplanar spacing with the patent is $2.907{\AA}$. Flos Lonicerae Japonicae has 6 special marks and Px is 23.08%. Its special mark in the patent are 4.95/14 and 4.50/15, respectively. The P is 9.09%. Its coincided interplanar spacing with the patent is $2.910{\AA}\;and\;3.05{\AA}$, respectively. The number of special XRDF mark peaks of baicalin is 9 and Px is 18.37%. Its coincided interplanar spacing with the patent is $2.910{\AA}$. It has visible mark and specificity adopting XRDF fingerprint to identify Huangjinju and medicinal materials. Establishing the quality standard is a synthetic index that depends both on special marks in the medicinal materials of the patent and on the coincidence peak data.

      • A Study of Cultural Tolerance Reference Frame for Culture-Based Design

        Smart Dining Table is an ongoing project in our Interaction Design Lab. It is a table top display designed to facilitate dining process. Smart Dining Table uses touchable interface to interact with users, providing a new, interesting and efficient experience in dining process. In GuangZhou, the south city of China, Yam Cha is a traditional and local way of entertainment in which normally elders gather together with friends or relatives to have tea and snack, and most importantly, to chat. Yam Cha has a long history in China, and Guangzhou has developed its own unique "Yam Cha" culture. In this paper, we discuss about previous research we did on Yam Cha in GuangZhou, and the approach we propose to manage and analyze issues related to dining table to help designing specific version of Smart Dining Table which can fit right into the Yam Cha process and cultural environment.

        Impaired Voluntary Wheel Running Behavior in the Unilateral 6-Hydroxydopamine Rat Model of Parkinson's Disease

        Objective : The aim of this study was to investigate voluntary wheel running behavior in the unilateral 6-hydroxydopamine (6-OHDA) rat model. Methods : Male Sprague-Dawley rats were assigned to 2 groups : 6-OHDA group (n=17) and control group (n=8). The unilateral 6-OHDA rat model was induced by injection of 6-OHDA into unilateral medial forebrain bundle using a stereotaxic instrument. Voluntary wheel running activity was assessed per day in successfully lesioned rats (n=10) and control rats. Each behavioral test lasted an hour. The following parameters were investigated during behavioral tests : the number of running bouts, the distance moved in the wheel, average peak speed in running bouts and average duration from the running start to the peak speed. Results : The number of running bouts and the distance moved in the wheel were significantly decreased in successfully lesioned rats compared with control rats. In addition, average peak speed in running bouts was decreased, and average duration from the running start to the peak speed was increased in lesioned animals, which might indicate motor deficits in these rats. These behavioral changes were still observed 42 days after lesion. Conclusion : Voluntary wheel running behavior is impaired in the unilateral 6-OHDA rat model and may represent a useful tool to quantify motor deficits in this model.

      • Mangiferin Induces Apoptosis by Regulating Bcl-2 and Bax Expression in the CNE2 Nasopharyngeal Carcinoma Cell Line

        To investigate the anti-proliferative mechanism of mangiferin in a human nasopharyngeal carcinoma cell line, CNE2 cells were incubated with different concentrations of mangiferin (12.5, 25, 50, 100, 150 and $200{\mu}M$) or with PBS as a control for 72 hours. Analyses were made of the cell cycle and apoptosis with measurement of mRNA and protein levels of two apoptosis-related genes, Bcl-2 and Bax. Flow cytometry assays showed mangiferin could inhibit CNE2 cell proliferation via G2/M arrest and induction of early apoptosis. Real time PCR and Western blotting showed the mRNA and protein level of Bcl-2 to be down-regulated, while those of Bax were upregulated, when CNE2 cells were treated with mangiferin. This investigation indicated anti-proliferation effects of mangiferin through induction of cell apoptosis regulated by Bcl-2 and Bax expression.

        Survival of tissue-resident memory T cells requires exogenous lipid uptake and metabolism

        Tissue-resident memory T (T<SUB>RM</SUB>) cells persist indefinitely in epithelial barrier tissues and protect the host against pathogens. However, the biological pathways that enable the long-term survival of T<SUB>RM</SUB> cells are obscure. Here we show that mouse CD8<SUP>+</SUP> T<SUB>RM</SUB> cells generated by viral infection of the skin differentially express high levels of several molecules that mediate lipid uptake and intracellular transport, including fatty-acid-binding proteins 4 and 5 (FABP4 and FABP5). We further show that T-cell-specific deficiency of Fabp4 and Fabp5 (Fabp4/Fabp5) impairs exogenous free fatty acid (FFA) uptake by CD8<SUP>+</SUP> T<SUB>RM</SUB> cells and greatly reduces their long-term survival in vivo, while having no effect on the survival of central memory T (T<SUB>CM</SUB>) cells in lymph nodes. In vitro, CD8<SUP>+</SUP> T<SUB>RM</SUB> cells, but not CD8<SUP>+</SUP> T<SUB>CM</SUB> cells, demonstrated increased mitochondrial oxidative metabolism in the presence of exogenous FFAs; this increase was not seen in Fabp4/Fabp5 double-knockout CD8<SUP>+</SUP> T<SUB>RM</SUB> cells. The persistence of CD8<SUP>+</SUP> T<SUB>RM</SUB> cells in the skin was strongly diminished by inhibition of mitochondrial FFA β-oxidation in vivo. Moreover, skin CD8<SUP>+</SUP> T<SUB>RM</SUB> cells that lacked Fabp4/Fabp5 were less effective at protecting mice from cutaneous viral infection, and lung Fabp4/Fabp5 double-knockout CD8<SUP>+</SUP> T<SUB>RM</SUB> cells generated by skin vaccinia virus (VACV) infection were less effective at protecting mice from a lethal pulmonary challenge with VACV. Consistent with the mouse data, increased FABP4 and FABP5 expression and enhanced extracellular FFA uptake were also demonstrated in human CD8<SUP>+</SUP> T<SUB>RM</SUB> cells in normal and psoriatic skin. These results suggest that FABP4 and FABP5 have a critical role in the maintenance, longevity and function of CD8<SUP>+</SUP> T<SUB>RM</SUB> cells, and suggest that CD8<SUP>+</SUP> T<SUB>RM</SUB> cells use exogenous FFAs and their oxidative metabolism to persist in tissue and to mediate protective immunity.
