        Phosphoketolase assay was used for the quantification of bifidobacteria in kimchi or fermented milk. The viable bifidobacterial change during fermentation in kimchi with of Bifidobacterium longum was measured by phosphoketolase assay and plate count method. Phosphoketolase activity was influenced by treatment of sonication or Triton X-100 to the microorganims to disrupt the cells. The result of phosphoketolase assay was similar to that of plate count method and showed different enzyme activities among different strains. This assay method took about one hour for the quantification of Bifidobacterium longum in food.

      • 청각 자극용 전기자극기 개발

        This paper introduces a development of an electrical stimulator for auditory stimulation. The electrical stimulator is useful in neurotological diagnosis, audiological evaluation, candidate selection for cochlear implantation, optimal device selection and decision making of MAP strategy for severe-to-profound hearing impaired persons. The development was based on sound parameters of auditory brainstem responses and auditory electrophysiological characteristic such as effective firing of auditory nerve and recording evoked potentials during refractory period of neuron. Besides pulse parameter could adjustable by programming for more varied electrical stimulation evoked response audiometry. Using the electrical stimulator, electrical square pulse was applied to promontory, and electrically evoked auditory brainstem response and electrically middle latency response were successfully recorded in cats.

      • Agrobacterium tumefaciens C58에 依하여 形質轉換된 人蔘組織에서의 saponin 生産

        The present study was conducted to obtain some basic information on the transformation of ginseng tissue, identification of opine compound and protein, and saponin production from ginseng tissue transformed with pTiC58. Ginseng tumor callus induced by pTiC58 could be continuously cultured on the phytohormone-free basic medium. The transformation was reconfirmed by the detection and identification of opine compounds from the tumor callus. The transformed ginseng calli contained higher amount of protein than normal calli and the protein pattern of transformed calli was quite different from that of normal callus. Higher saponin content was found in the transformed callus than the normal callus, and ginsenoside composition of the transformed callus was similar to that of cultivated ginseng root, but quite different from that of normal callus.

      • 와우 갑각 전기자극 뇌간유발반응용 원추형 전극의 개발

        This paper introduces a new zinc coated copper wire electrode with coiled cone shape which has low surface resistance and tolerance to the motion artifact for promontory stimulation electrically auditory brainstem responses (PSEABR). Auditory brainstem responses (ABR) can be used to predict hearing threshold level with a great deal of accuracy particularly for a young child who cannot cooperate mechanically and some hearing impaired who are exaggerating a hearing loss for economic compensation. While severe profound sensorineural hearing losses may not be implemented by auditory potentials, PSEABR is proven as a useful tool even for some sensorineural related hearing impaired. It was shown that PSEABR gives the electrical stimuli to promontory of the cochlear instead of giving acoustic stimuli. For this reason, PSEABR can be used as an alternative for cochlear implantation, and can also be used as an optimal device selection and neural information for MAP. It was found that the role of electrode is very important in PSEABR. Even though this cone-shaped electrode was applied in animal experiments, waveforms are well produced by PSEABR. Thus, it was concluded that cone-shaped electrode turned out to be a useful preoperative audiological evaluation tool in deciding time for cochlear implantation surgery.

      • Ti-plasmid에 依하여 形質轉換된 煙草組織의 特性과 再分化 樣相

        Studies were conducted to examine the characteristics of teratoma shoots obtained from diploid and haploid tobacco crown gall tumor tissues induced by the inoculation of Agrobacterium tumefaciens and the effects of plant hormones and active carbon in the cultural media on the growth and regeneration patterns of the teratoma shoots. Results can be summarized as follows. 1. Teratoma shoots could be obtained when the crown gall tumor, induced by the inoculation of A. tumefuciens strain C-58 to the stem, was successively grown on hormone-free medium. 2. The growth of tumor callus induced by C-58 strain was inhibited when several plant hormones were added to the cultural media. Plant regeneration could not be obtained from the tumor callus or the teratomas. 3. Addition of active carbon and benzyl adenine to cultural media was effective for shoot elongation and reduction in multiple shoot formation. 4. The growth of tumor callus obtained from the haploid plant was promoted by the addition of plant hormones to the media. Whole plants were also regenerated from the teratoma shoots formed from tumor callus. 5. Normal or transformed plants were regenerated from the tumor callus formed by the inoculation of a strain of A. tumefaciens collected from field soil. The transformed plants are characterized by having twisted dark-green leaves, shorter internodes, and abundant axillary shoots.

        궤적 정보를 이용한 냉동 컨테이너 모니터링 시스템

        위성 통신 기술의 발달로 국부적으로 수행리던 위치 추적의 범위가 선 세계로 3~4간되었다 하지만 기존의 물류 관제 시스템들은 관제 서버와 물류간의 통신을 위하여 휴대폰 통신망을 이용하기 때문에 일부 지역 내에서 이동하는 물류에 대해서만 관제가 가능하다. 이 논문에서는 위성 통신을 이용하여 전 세계를 이동하는 물류 매체인 냉동 컨테이너의 위치 및 상태 정보를 관리하기 위한시스템을 제안하였다. 제안 시스템은 냉동 컨테이너의 상태를 직절 관제하는 데이터 수집부, 관제서버와의 통신을 담당하는 위성 통신부, 냉동 컨테이너 정보를 판리하는 관제부로 구성된다. 시스템의 관제부에서는 냉동 컨테이너의 위치 주적 및 관제를 효과적으로 수행하기 위하여 기존의 TB 트리를 기반으로 하는 다중 버전 이동 객체 색인을 사용하였다. 제안된 냉동 컨테이너 관제시스템은 화주, 운송회사 등 육해상 물류관련 기관에서 냉동컨테이너 및 선박 위치추적, 컨테이너 통제, 컨테이너 통계 등 다양한 용도로 사용될 수 있어 화주에게는 컨테이너 운송에 대한 서비스를 제공하며, 선사에게는 선박안전경보시스템, 선박 관리, 냉동컨테이너를 효율적인 관리 기능을 제공하게 한다. As developing satellite communication, the tracking range of the moving objects which move in local area is expanded to the whole world. However previous logistics management system is able to monitor freight which transporting in local area using mobile communication system. In this paper, we propose the reefer container management system that manages the location information and other related information such as temperature, humidity of container using the satellite system. The proposed system consists of three parts; data collector, satellite communication manager, reefer container information manager. And the proposed system uses the moving object index for managing the trajectory of container and tracing the location of container or vessel that is transporting the container, and supports various services such as reefer container and vessel tracking, container control and container statistics to logistic companies like shipper and forwarding agent. And the system can be increasing the quality of container transportation service to the shipper, and it makes the efficient management of reefer container to the shipping company.

      • Helicobacter pylori와 대장균의 Shuttle Vector 개발

        In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.

      • 이명과 청각민감증을 동반한 편측 고음 급추형 감각신경성 난청의 청각 재활

        In case of the hearing impairment with tinnitus and increased sound sensitivity, it is known that the patients tend to appeal the psychologically oriented social handicap rather than communication disability. The audiologist who is responsible for such patients in aural rehabilitation should pay special attention to the counseling techniques inducting tinnitus retain therapy (TRT), ear protector, noise generator, or specific acoustic training based on dose cooperation and rapport. And then the audiologist should try to lessen their reaction to the tinnitus by using a hearing aid. This therapies tries to focus not a total approach but a treatment to lessen the severity of tinnitus. This paper as a case report that a unilateral sharply slopped sensorineural hearing impaired person with tinnitus and increased sound sensitivity by using four channel digital signal processing (DSP) hearing aid with programming increment at low level (PILL).

      • 예비 초등교사들의 세포에 대한 개념

        교육대학교에 재학 중인 1학년에서4학년까지 총 127명의 남녀 학생들을 대상으로 하여 면담에 의해 세포에 대한 개념이 어느 정도 형성되어 있으며, 어떤 유형의 오개념을 가지고 있는가를 조사하였다. 조사한 모든 예비 초등교사들은 생물체를 구성하는 기본단위가 세포라는 과학지식을 가지고 있었으나 감자나 고기와 같이 우리가 음식물로 이용하는 생물체의 일부분은 단순히 양분 덩어리인 것으로 인식하는 등, 학습에서 얻은과학 개념과 실제 세계에 대한 또 다른 개념 체계를 병행하여 가지고 있는 것으로 나타났다. 예비 초등교사들은 핵, 엽록체 및 미토콘드리아 등과 관련된 세포의 기능에 대한 개념이 세포에 대한 다른 개념들보다는 낮게 형성되어 있으나 기 보고(장명덕과 송남희, 1978)된 초등교사들의 개념 형성 정도보다는 10%정도 높았다. 그러나 개체의 성장을 세포의 수준에서 바르게 인식하고 있는 예비 초등교사는 조사 대상자의 57% 정도였으며 초등교사들보다 17% 정도 낮았다. 본 연구와 기 보고된 초등교사들을 대상으로 하며 나타난 오개념들의 유형이 서로 유사한 것으로 미루어 보아 이러한 오개념들은 교정되지 않고 장기간 지속되는 것이었다. 그러므로 예비 교사의 양성과정이나 현직교사들의 연수과정에서 이러한 기본 개념들이 바르게 확립될 수 있도록 교육 프로그램을 개발, 적용할 필요가 있다. The purpose of this study is to investigate degree of the concept formation and to clarify the patterns of misconception on cell in preservice elementary school teachers. Data were collected by interviews with 120 presservice elementary school teachers, ranged from freshman to senior, studying at Taegu National University of Education, Korea. The instrument used in this study was developed by Jang and Song(1998). The results were discussed with those on elementary school teachers' cell conceptions(Jandg and Song, 1998).
