국내 재배 흰 민들레 부위별 에탄올 추출물의 항산화 활성 및 세포독성

In the present study, the total content of polyphenols and flavonoids, the antioxidant activities, and cytotoxic effects of the ethanol extracts from different parts of Taraxacum coreanum Nakai were investigated for their use as functional foods. The extract yields of the flower, leaf, and root were $32.15{\pm}3.21%$, $31.63{\pm}0.63%$, and $27.48{\pm}2.47%$, respectively. Total polyphenol and flavonoid content of the flower extract were $61.29{\pm}2.11mg/g$ and $46.11{\pm}1.88mg/g$, respectively, which were much higher than those of any other plant parts. The antioxidant activities of the flower, leaf, and root extracts were $89.99{\pm}2.83%$, $85.29{\pm}2.22%$, and $37.88{\pm}2.34%$, respectively, at a concentration of 1.0 mg/mL. Cell cytotoxicity effects of AGS (human gastric carcinoma), HCT116 (human colon carcinoma), and A549 (human pulmonary carcinoma) cells were the highest in the flower extract, with values of $62.85{\pm}4.63%$, $69.89{\pm}3.44%$, and $85.72{\pm}4.17%$, respectively, at a concentration of 400 mg/kg. Both the antioxidant activities and cytotoxic effects of the ethanol extracts from all the parts of the T. coreanum Nakai increased dose-dependently. These results provide preliminary data for the development of T. coreanum Nakai as an edible functional food material.

Epimedium koreanum Nakai is a wild medicinal plant commonly consumed in South Korea due to its beneficial health effects. In this study, the antimutagenic and immunological activities of E. koreanum Nakai extracts were investigated for their use in food. In the immunomodulating activity, the effects of E. koreanum Nakai on the B cell (Rhamos) and T cell (Molt-4) were investigated. The results showed that the growth and viability of the B and T cells were increased and activated more in the ethylacetate (1.35 and 1.48 times) and water fraction (1.30 and 1.40 times), respectively. In the Ames test, none of the fractions produced a mutagenic effect on Salmonella. typhimurium TA98 and TA 100. The ethylacetate fraction showed a strong antimutagenic effect (98%) on and a high butanol fraction (84%) of B(α)P in S. typhimurium TA98 and TA100, respectively. In 4-nitroquinoline-1-oxide (4NQO), all the solvent fractions showed an over 70% antimutagenic effect, except for the chloroform extract. Especially, ethylacetate and butanol showed strong inhibition of the mutagenic effects (80 and 90%) on 4NQO in S. typhimurium TA98 and TA100, respectively. These results provide preliminary data for the development of E. koreanum Nakai as an edible food material.
국내 재배 흰민들레 부위별 열수 추출물의 항산화 및 항암 활성

본 연구는 흰민들레 꽃, 잎, 뿌리의 부위별 성분 및 생리활성을 탐색하고자 열수 추출물에서 총 폴리페놀, 총 플라보노이드, 항산화 및 항암 활성을 분석하였다. 흰민들레 부위별 시료의 열수 추출조건에 따른 추출수율은 꽃, 잎, 뿌리가 각각 30.25%, 34.53%, 66.25%로 뿌리> 잎> 꽃의 순으로 나타났다. Folin-Denis 방법에 따른 총 폴리페놀 함량은 꽃 추출물에서 50.54 mg/g으로 가장 높게 나타났으며, 그다음이 잎(41.11 mg/g), 뿌리(10.27 mg/g) 순으로 나타났다. 흰민들레 부위별 열수 추출물에서의 총 플라보노이드 함량은 총 폴리페놀 함량과 같은 경향으로 꽃(35.26 mg/g), 잎(28.91 mg/g), 뿌리(8.28 mg/g)의 순으로 각각 나타났다. DPPH 라디칼 소거 활성으로 분석한 흰민들레 부위별 열수 추출물의 전자공여능은 추출물 농도 의존적으로 나타났으며, 꽃, 잎 및 뿌리 열수 추출물 1.0 mg/mL의 농도에서 각각 91.04 %, 88.22%, 38.58%로 꽃 열수 추출물이 가장 높게 나타났다. 이는 폴리페놀 및 플라보노이드 함량이 DPPH 라디칼 소거능과 관련이 있음을 보여주었다. 흰민들레 부위별 열수추출물 400 mg/kg 첨가 시 위암 세포주(AGS), 폐암 세포주(A-549) 및 대장암 세포주(HCT-116)에 대한 암세포 생존율을 확인한 결과 위암 세포주(AGS)에서 꽃(40.34%), 잎(44.56%), 뿌리(95.12%)의 순으로, 대장암 세포주(HCT-116)에서 꽃(39.56%), 잎(48.55%), 뿌리(95.11%)의 순으로, 폐암 세포주(A-549)에서 꽃(17.52%), 잎(23.57%), 뿌리(88.15%)의 순으로 모든 암세포주들에 대하여 꽃 열수추출물이 가장 높았고 뿌리에서 가장 낮은 것으로 나타났으며, 모두 농도 의존적으로 항암 활성이 증가하는 것으로 나타났다. 이와 같은 결과 향후 국내 흰민들레 재배면적 확대 뿐만 아니라 기능성 식의약품 소재로서의 이용에 있어 유용한 기초 자료로 활용될 수 있을 것으로 기대된다. Extract yields of flower, leaf, and root were 30.25%, 34.53%, and 66.25%, respectively. Total polyphenols and total flavonoids contents of flower extract were 50.54 mg/g and 35.26 mg/g, respectively, which were much higher than any other parts. The electron donating abilities of flower, leaf, and root extracts were 91.04%, 88.22%, and 38.58%, respectively, at a concentration of 1.0 mg/mL. Cell viability of AGS for human gastric carcinoma, HCT-116 for human colon carcinoma, and A-549 for human pulmonary carcinoma showed the lowest values in flower extracts (40.34%, 39.56%, and 17.52%, respectively), indicating the highest cytotoxicity at a concentration of 400 mg/kg. Both antioxidative and anticancer activities of water extracts from all T. coreanum Nakai parts dose-dependently increased. These results provide preliminary data for the development of T. coreanum Nakai as an edible functional food material.

Hypsizigus marmoreus is a wild mushroom commonly consumed in South Korea due to its beneficial effects on health. In the present study, the general chemical and inorganic-element composition, the total amino acid contents, and the volatile-flavor compounds of H. marmoreus were investigated for food uses. The proximate compositions consisted of 60.1% carbohydrate, 32.0% crude protein, 8.98% moisture, 5.0% ash, and 2.0% crude lipid. The minerals in H. marmoreus were found to be as follows; potassium (429.5 mg), phosphorus (101.9 mg), sodium (20.3 mg), magnesium (54.86 mg), calcium (2.7 mg), zinc (0.8 mg), iron (0.7 mg), manganese (0.2 mg), and copper (0.1 mg), based on 100 g of mushroom dry weight. Seventeen kinds of total amino acids were found in H. marmoreus, with the glutamine acid content being the highest (2,340 mg/100 g), followed by the asparagine, serine, arginine, and leucine contents. The volatile-flavor components of H. marmoreus were collected via simultaneous steam distillation extraction (SDE), and were analyzed via gas chromatography-massspectrophotometry (GC-MS). A total of 17 volatile-flavor compounds were identified, including eight aldehydes, seven alcohols, one acids, and one other compounds. The most abundant compound was 2,3,6-trimethyl pyridine, which accounted for more than 40% of the total volatiles; other important compounds were 1-octen-3-ol, butylhydroxytoluene (BHT), isooctyl phthalate, 3-octanal, 1-undecanol, and 2-amylfuran. These results provide preliminary data for the development of H. marmoreus as an edible food material.
Icariin 함량에 따른 삼지구엽초 추출물의 항돌연변잉 및 면역활성

Epimedium koreanum Nakai is a wild herb commonly consumed in South Korea due to its beneficial health effects. In the present study, the antimutagenic and immunoactivities of extracts from E. koreanum Nakai containing different icariin quantities were investigated for food use. In the Ames test, both the water and ethanol extracts were found not to have a mutagenic effect on Salmonella typhimurium TA98 and TA100, respectively. The E. koreanum Nakai extracts showed over 80 and 90% antimutagenic effects on benzo(α)pyrene (B(a)P) in S. typhimurium TA98 and TA100, respectively. Moreover, all the extracts showed over 70% antimutagenicity on S. typhimurium TA98 and TA100 against 4-nitroquinoline-1-oxide (4NQO). The E. koreanum Nakai extract with ethanol showed strong antimutagenic activity, higher than that of the water extract and the sequenced KE9412, KE9408, and KE9405. In the immunomodulating activity test, the effect of E. koreanum Nakai on the B (Rhamos) and T (Jukat) cells were investigated. The immunoactivity results showed that the growth and viability of the B and T cells increased and were activated more in KE9405 (1.8 times), KE9408 (1.6 times), and KE9412 (1.32 times) in the water extracts, and least in KE9412 (1.74 times), KE9408 (1.52 times), and KE9405 (1.4 times) in the ethanol extracts. In the case of both the water and ethanol extracts (500 μg/mL) from E. koreanum Nakai, the highest cell number of the human B (Rhamos) and T (Jukat) cells was observed on day 4 in KE9405 and KE9412, and on day 5 in KE9408. Based on the obtained results, the development of E. koreanum Nakai as a food material is recommended.
