        알쯔하이머병 환자 혈청에서의 베타 아밀로이드 단백질에 대한 특이 항체량 측정 : 알쯔하이머병의 생화학적 진단지표 개발 Development of Biomarker for Alzheimer's Disease Diagnosis

        Background:Alzheimer's disease (Ad) is a neurodegenerative disorder that is rapidly increasing with the aging society, requiring a need for early diagnosis and prevention. However, diagnosis on AD has only been possible through limited methods such as neuropsychological examination or MRI. AD is characterized by deposition of senile plaques and neurofibrillary tangles in the brain. Aβ peptide in senile plaques seems to play a central role in the neuropathology of AD. Several biochemical markers for AD are available, including reduced Aβ protein, a change in ratio between Aβ40 and 42 and increased level of tau protein in the cerebrospinal fluid (CSF). Methods:This study analyzes anti-Aβ antibody from serums of AD patients using the ELISA. The levels of anti-Aβ antibody from patients with idiopathic Parkinson's disease or stroke and from normal control were compared to that of AD patients. Results:Our results showed a significantly lower anti-Aβ antibody level in AD compared to those with other neurological diseases or control. Conclusions:These data showed that the anti-Aβ antibody level in the serum may be used to diagnose the presence of AD.

      • Mitochondrial dysfunction and calcium deregulation by the RanBP9-cofilin pathway

        <P>Mitochondrial dysfunction and synaptic damage are important features of Alzheimer's disease (AD) associated with amyloid β (Aβ) and tau. We reported previously that the scaffolding protein RanBP9, which is overall increased in brains of patients with AD and in mutant APP transgenic mice, simultaneously promotes Aβ generation and focal adhesion disruption by accelerating the endocytosis of APP and β1-integrin, respectively. Moreover, RanBP9 induces neurodegeneration <I>in vitro</I> and <I>in vivo</I> and mediates Aβ-induced neurotoxicity. Here we show in primary hippocampal neurons that RanBP9 potentiates Aβ-induced reactive oxygen species (ROS) overproduction, apoptosis, and calcium deregulation. Analyses of calcium-handling measures demonstrate that RanBP9 selectively delays the clearance of cytosolic Ca<SUP>2+</SUP> mediated by the mitochondrial calcium uniporter through a process involving the translocation of cofilin into mitochondria and oxidative mechanisms. Further, RanBP9 retards the anterograde axonal transport of mitochondria in primary neurons and decreases synaptic mitochondrial activity in brain. These data indicate that RanBP9, cofilin, and Aβ mimic and potentiate each other to produce mitochondrial dysfunction, ROS overproduction, and calcium deregulation, which leads to neurodegenerative changes reminiscent of those seen in AD.—Roh.         Locoregional recurrence is the most common type of recurrence in surgical operation of gastric adenocarcinoma, and peritoneal dissemination is one of the most difficult problems in advanced gastric adenocarcinoma treatment. Because the peritoneal cavity is the most common site of the first recurrence after gastric cancer resection, intraperitoneal chemotherapy seems a logical choice for cancer chemotherapy. In this study, Mitomycin C (MMC)-activated charcoal (CH)-alginate (ALG) beads were prepared by the mixtures of CH particles adsorbed with MMC as an anti-cancer drug and aqueous alginate solution. The alginate is recognized as biodegradable, nontoxic, and biocompatible. The release of MMC from the beads in 0.1 M Tris buffer was stable and continuous until about 1 week. The MMC-CH-ALG beads can be applied in the peritoneal cavity for intraperitoneal chemotherapy since they provide a good adhesiveness on the tissue and controlled release pattern of the drugs.

        The method of finite element analysis is modified in this study to predict the material nonlinear behavior of reinforced concrete structure more properly. It is difficult to predict the behavior of reinforced concrete structure due to the development of concrete crack, the behavior of compressive failure, bond properties between concrete and steel, and shear resistance. To predict the material nonlinear behavior of reinforced concrete structure more properly, the stress-strain relationship of confined concrete which proposed by Attard, Setunge is considered. Also, shear retention model is considered. To prove the efficiency of the analysis, analytical results are compared to experimental results.

        18세기 이후 유럽의 뮤지엄 건축의 동선체계에 나타난 보편적 경향과 다원적 경향에 관한 연구

        Since the 18th century the museums in Europe have accepted the social demand of the publicity and have played a role of social education and it lasted to the present. Recently, the society began to change it's direction. It is from the Universalism to the Pluralism. The Universalism is due to democratic society after 'the Civil Revolution' and standardized social system after 'the Industrial Revolution' The Pluralism that starts with the decrease of the modern society is about the diversity and the activation of identity which has been disregarded since the Modernism. This study is to analyze and to clarify how the museum's educational environment which had begun since the 18th century has been changed since 1990 and how these are related to the social alteration from the Universalism to the Pluralism. As a result of this study, it was confirmed that there had been a simple circulation system and a unified spatial experience in the museum of Europe since 18th century to 1980's(ph1) while there had been a complex circulation system and a diverse spatial experience in the museum of Europe after 1990's(ph2). These mean that the museum of ph1 gave a unified social education to the visitors and they came to have a similar knowledge and emotion after their seeing the exhibition material while the museums of 'ph2' gave a diverse one and the visitor came to have a different knowledge and emotion to the same exhibition material. The museum which gives a unified social education is to produce a unified thoughts of people and it is because the social direction is the Universalism. In addition to this, the museum of diverse social education is to produce people of diversity and it is related to the Pluralism.

        그들이 현실세계에서 이루고자 했던 정치적 이상이 결국 좌절되면서 대신 인간의 상상력이라 하는 정신 세계의 문제로 내면화 되었을 때 이미 그들의 시는 정치와 괴리될 수밖에 없었던 것이다. 다시 말해서 그들이 잠시나마 보여 주었던 정치에 대한 놀라운 관심과 발언, 즉 그들이 보여 준 “the old connection between literature and politics"는 이미 그들의 시에서부터 깨어질 운명이었던 것이다. 여기에 그들 시세계의 이상과 한계가 공존하는 것이며, 이러한 한계는 바로 현대시의 것이기도 하다.

      • 공기 중 혼합 유기용제 측정실태와 성분 분석

        This study was performed to investigate ingredients of organic solvents and present status of environmental measruements with official records of working environmental measurements of 799 workplaces in 179 workshops used organic solvents. The result were as follows: 1. The mean of total sampling time and actual time measuring organic solvent were 158?80 min and 72?37 min. In 62.5% of 799 sample the air flowrates were above 0.2?/min. 2 Thirty kinds of organic solvent were detected in the working environments. The most common organic solvent was toluene(95.4%) and followed by xylene(47.1%), MEK(36.5%) and n-hexane(35.3%) in descending order. 3. The average number of species detected per a mixed organic solvent sample was 4.4?2.6, and there was no significant difference in the detection number among type of workplaces. 4. The mean exposure concentrations was the highest in acetone(86.6?167.8ppm) and followed by MEK(37.7?59.6ppm) and toluene(28.7?48.4ppm). While considering the TLV, the mean exposure concentrations were 7.9?10.8ppm(TLV:10ppm) in N.N-dimethy1 formamide, 4.9?5.0ppm(TLV:10ppm) in benzene and 2.1?1.5(TLV:5ppm)in methy1 cellosolve acetate in descending order. 5. Among the 799 workplaces, 20% workplaces were evaluated with exceed TLV in case of sample organic solvents. In case of mixed organic solvents, the TLV exceed rate of workplaces was 40.0%. 6. 72.1 of workplaces were received two consecutive environmental measurements a year and 57.3% of them was not exceeded TLV. The average concentration of organic solvents in the 1st half year was 1.16?1.09 and most of them were above 1.0 TLV. The average concentration of the 2nd half year period was 0.81?0.59 which was somewhat lower than those of the 1st half period. 7. The percentage of workplaces with local ventilation was 65.1%. Among them 25.2% local ventilation was evaluated with poor and 34.9% workplaces was not in-stalled and local ventiliator. According to the performance of ventilator, the concentration of the organic solvents was 0.44?0.30 at well-instituted ventilator, that with poorly-instituted was 1.32?0.66 and that with unistitutied was 1.75?1.27. On the basis of about data, ingradient analysis of mixed organic solvents should be perform prior to eveluation of organic solvents and should evaluated with mixed organic solvent TLV.

        In this report, Bal I endonuclease from brevibacterium albium (ATCC 15831) strain was purified characterized. Bal I endonuclease was purified by the following procedures; ammonium sulfate fractionation, DEAE-cellulose column chromatography, phosphocellulose column chromatography. Bal I endonuclease has a specific site for type II enzyme in that has been proven by the facts that PBR322 DNA cleaved by Bal I endonuclease.
