        Ischemia-Related Changes in Galanin Expression in the Dentate Hilar Region after Transient Forebrain Ischemia in Gerbils

        <P>Although galanin (GAL) protects hippocampal neurons from ischemic damage, no study has examined ischemia-related changes in endogenous GAL in the hippocampal dentate gyrus. We investigated the chronological changes of GAL, well-known as the potassium channel opener, expression in the dentate gyrus at various times after 5 min of transient forebrain ischemia in gerbils. A few GAL-immunoreactive (IR) neurons were found in the polymorphic layer of the sham-operated group. Three hours after ischemia-reperfusion, the pattern of GAL immunoreactivity was similar to that of the sham-operated group and the number of GAL-IR neurons and immunoreactivity were highest 12 h after ischemic insult. At this time, GAL-IR neurons in the polymorphic layer showed strong GAL immunoreactivity. Thereafter, GAL-IR neurons and immunoreactivity significantly decreased in the dentate hilar region. Four days after ischemic insult, GAL-IR neurons were not detectable. In addition, the results of a Western blot study showed a pattern of GAL expression similar to the immunohistochemical changes. GAL protein content also was highest 12 h after ischemia. In conclusion, the increased expression of endogenous GAL in the dentate gyrus after ischemia is related to response to the ischemic damage.</P>

        이상기후 발생이 가을배추 생산자 및 소비자에게 미치는 영향

        The purpose of this article is analyzing the economic impacts of abnormal climate on fall chinese cabbage farmers and consumers in Korea, with employing the equilibrium displacement model. Our results show that there were little difference in gross farm income, even though there were significant yield reductions due to abnormal climate changes. However periodic occurrences of abnormal climates caused serious damage to consumption levels which had declined by 10.6~17.1 percent with higher prices by 15.3~24.6 percent than normal climate years since 1990.

        A case of erosive vitreoretinopathy

        Hereditary vitreoretinopathies are potentially blinding inherited disorders characterized by an abnormal-appearing vitreous gel and associated retinal changes. Four of these disorders, Stickler’s syndrome, Wagner’s disease, erosive vitreoretinopathy, and Goldmann-Favre syndrome, exhibit marked syneresis of the vitreous gel. Erosive vitreoretinopathy has associated retinal pigment epithelial changes, poor night vision, visual field defects, and abnormal electroretinographic findings; symptoms not found in Stickler’s syndrome. A 36-year-old man with progressive visual loss and a visual field defect had no systemic disease. His vitreous cavity was liquefied. Vitreous strands and a cataract were found in both eyes. Pronounced RPE degeneration was found superotemporally in both eyes and a bullous rhegmatogenous retinal detachment in the left eye accompanied two retinal tears. His visual field showed a ring scotoma in both eyes and the ERG finding was abnormal. We report one case of erosive vitreoretinopathy with retinal pigment epithelial changes, rhegmatogenous retinal detachment, visual field defects, abnormal electroretinographic findings, marked vitreous syneresis and cataract. These symptoms are distinct from previously described entities.

      • Newcastle Disease Virus의 Fusion 단백 유전자의 제한효소분절형태(RELP)의 특이성

        To evaluate the specificity of restriction fragment length polymorphism (RFLP) of Newcastle disease virus (NDV) fusion (F) protein gene for various NDV strains. F protein genes of NDV CBP-1 isolate, LaSota, B1 and Kyojungwon (KJW) were amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) and RFLP were examined. In RT-PCR using F primer sets for 1.7 kb F gene, the amplified products were observed in CBP-1, LaSota and B1 except KJW. However, F2 primer set for 521 bp yielded the amplified bands with all strains. The amplified F genes (1.7 kbp) were treatd with 15 different restriction enzymes to analyze RFLP and the cleavage patterns. CBP-1 showed various cleavage patterns with 8 enzymes, LaSota, with 11 enzymes and B1, with 11 enzymes. According to the cleavage patterns, the physical maps were constructed, and it was found that CBP-1, LaSota and B1 strains had specific cleavage sites with HinfI, XbaI and XhoI. These findings suggest that RFLP are worth applying in the diagnosis and molecular epidemiological studies of Newcastle disease.

        It is known that meningoencephalitis is the disease usually characterized by a hstory of headache, high fever and disturbance of consciousness. Its cause is usually virus. In most cases, inflammatory reaction involve the brain diffusely, but occasionally involve brain locally. In those cases in whom inflammatory reaction involved brain locally, specific symptoms appear according to the location involved. Clinically, we can suspect the site of brain where inflammatory reaction involved, according to clinical symptoms. We experienced a case of meningoencephalitis with downbeat nystagmus implying that the inflammatory lesion involve pontomedullary junction. It was confirmed by MRI. So, we report it with review of literatures.

      • 도축돈 장분변으로부터 Shiga Toxin-Producing Escherichia coli의 분리와 성상

        Shiga toxin-producing Escherichia coli (STEC) causes various clinical signs in human and animals, and has been indicated as a global enteropathogen with zoonotic importance. In this study, the feces of healthy pigs were collected from the slaughtered pigs of Daejon abattoir during the period from December 2001 to October 2002. Of 326 specimens, 13 STEC were confirmed by culture, PCR and colony hybridization. The isolates were further studied for toxin types, pathogenic factors, plasmid profiles, and antimicrobial resistance to characterize the genetic and toxigenic properties. In PCR, all of 13 isolates were evident to have shiga toxin gene (stx). Of 13 isolates stx1 gene was detected in 4 and stx2 gene in 9. The genes of eaeA, hlyA and rfbE were not present in any isolates. In colony hybridization using shiga toxin common primer (STXc), 2 to 9 per 100 colonies subcultured from 13 isolates showed the positive reaction. In the examination for plasmid profiles of the isolates, one to eleven plasmids with varying sizes of 1.0 Kb to 100 Kb were detected, and the 13 STEC could be classified into four groups by the plasmid patterns. The antimicrobial resistance patterns of the isolates were comparably corresponded with the plasmid profile patterns.

      • 각막형태검사법을 이용한 각막양상과 눈의 굴절력과의 관계

        We evaluated the topography of 200 normal cornea of Korean. The mean age was 37 years and mean refractive astigmatism was 0.82±0.69D. The corneal patterns were classified by Dr. Bogan such as round, oval, symmetric bow tie, asymmetric bow tie, and irregular pattern. Its incidence was 14.0%, 11.0%, 16.0%, 30.0% and 29.0%, respectively. There was no statistically significant difference in the distribution of topographic pattern in male and female and among age groups. The symmetric bow tie type had the greatest spherical equivalent and astigmatism and there was no statistically significant difference in those two values between round and oval group. Two bow-tie groups were not also statistically significantly different from each other in spherical equivalent and astigmatism. However, two bow tie groups were significantly different from round and oval group. So, it was possible to divide corneal topographic pattern into three groups such as round and oval, two bow tie, and irregualr group. We could understand normal corneal status related to corneal pattern and refractive power through this qualitative system for classifying corneal topography.

      • Baculovirus Vector System에 의해 발현된 재조합 Pseudorabies Virus Major Capsid Protein의 면역원성

        The recombinant pseudorabies virus major capsid protein (rMCP) was produced by expression of the MCP gene in Sf-9 cell using baculovirus transfer vector system. Following evaluation of the immunochemical properties of the rMCP, the immunogenicity of the recombinant subunit protiens were investigated in guinea pig and swine to obtain the preliminary guide line for the subunit vaccine using rMCP and gP50. It was proved that ultrasonication and 30% ammonium sulfate was most efficient to concentrate and purify the protein. The rMCP was safe in mice, guinea pigs and piglets. In guinea pigs, rMCP mixed with various adjuvants induced substantial degree of serum neutralizing antibody titers, but revealed incomplete protecivity against challenge. In swine, the combination of rMCP and gP50 showed the higher serum neutralizing antibody titers and cellular immune responses than rMCP alone. However, the protectivity was lower in comparison with the commercial gI-deleted inactivated vaccine. We expect these results to contribute to characterization of MCP gene of Korean isolate of PRV and to ultilize as preliminary information for prodution and evaluation of PRV recombinant subunit vaccines.

      • 생분해성 플라스틱이 포함된 유기성 생활 쓰레기의 퇴비화

        준 생산용 퇴비화 장치를 이용하여 생분해성 플라스틱을 포함하는 유기성 도시생활쓰레기기의 퇴비화를 연구하였다. 유기성 도시생활쓰레기는 실제 도시생활쓰레기 구성비에서 무기물을 제외한 유기성 물질만의 비로 만들어 사용하였다. 유기성 도시생활쓰레기를 구성하는 각각의 물질에 대한 생분해도를 ISO법으로 측정하였고, 퇴비의 화학적 물리적 특성도 조사하였다. 구성 물질에 대한 생분해도 결과에서, 생분해성 고분자를 포함한 각 물질들은 퇴비화에 충분한 생분해도를 나타내는 것으로 밝혀졌다. 또한, 유기성 도시 생활쓰레기로부터 비독성의 퇴비를 얻을 수 있음을 알았다. The composting study of organic municipal solid waste containing biodegradable plastic was performed by using pilot scale composting apparatus. The organic municipal solid waste(OMSW) was artificially prepared for composting. The composition of artificial OMSW was the same as that of real municipal solid waste excluding inorganic waste. The biodegradability of individual component of OMSW was studied according to the ISO method. The chemical and physical characteristics were also observed. Biodegradability study revealed that every component of OMSW including biodegradable plastic had sufficient degradability for real composting. And it was found that non-toxic compost could be produced from OMSW containing biodegradable plastic.

      • 대조 추출물분획이 치은 섬유아세포의 생물학적 활성화에 미치는 영향

        Final goal of periodontal treatment is to reconstruct the destructed periodontal tissue as well as to remove the necrotic pathologic elements. The purpose of this study is to investigate on the effect of Zizyphi extract to the inhibitory ability on collagenolytic activity of P gingivalis, biologic activity of gingival fibroblasts and on the collagen and protein synthesis of gingival fibroblasts. Gingival fibroblast from giniva of first bicuspids from patient for orthodontic treatment were used and cultured. For the measurement of inhibitory ability of collagenolytic activity, crude enzyme was extracted and used on the basis of modified Ono's method. On the inhibition of collagenolytic enzyme from herbal extracts, collagenokit CLN-100 were used. The cellular activity of gingival fibroblast, were studied using MTT solution and measured optical density on 570mm by ELISA reader. To measure the effects on the ability of whole protein and collagen synthesis, cell membrane was destructed with ultrasonic grinder after culturing, centrifuged and counted by liquid scintilation counter. The inhibitory effects on producing of IL-1β by monocyte, after promotion of producing IL-1β by LPS, were compared with the mixture of herbal extracts and other drugs using thymocyte stimulation assay. About inhibitory·effects of PGE_2 by gingival fibroblasts, herbal extract was compared with the addition of the other control groups using enzyme immunoassay. On the inhibition of collagenolytic activity by P. gingivalis, benzene extracts showed the most efficient inhibitory effects among the l9㎍/㎖ of the compared extracts and 40.5% by Tetracycline. On the cellular activity promoting effects, compared extracts showed a bit of more effects than PDGF of l00㎍/㎖ concentration and lGF of 20㎍/㎖ concentration. All of the PDGF, 1GF, Zizyphi Fructus extract should increase in collagen synthesis, but especially 70% ethylalcohol extracts of Zizyphi Fructus showed comparably high effects among the compared extracts. Effects on whole protein synthesis were slightly increased on every extract but especially 70% ethylalcohol extract showed significantly effective than any other estract. On the inhibitory effects of Zizyphi Fructus IL-1β production by monocyte, compared extracts showed 70% of highly inhibitory effect than that of 60% inhibition effects on controlled group and each extracts showed no significant difference. In PGE_2 production inhibitroy effect of Zizyphi Fructus gingival fibroblasts, Herbal extracts showed 70% of inhibition comparing with tat of 90.2% of controlled group, but each extracts showed similar effects excluding the H_2O extracts. These results suggested that Zizyphi Fructus might be useful medicine for inhibition of inflammatory mediator including IL-1β and PGE_2·
