        Mechanical properties of Metals for biomedical Applications Using Powder Metallurgy Process: A Review

        The uses of biomaterials have been revolutionizing the biomedical field in deployment as implants for humans. During the past five decades, many implant materials made of metals have been put into practical use. Powder metallurgy techniques have been used to produce controlled porous structures, such as porous coatings applied for dental and orthopedic surgical implants, which allow bony tissue ingrowth within the implant surface, thereby improving fixation. This paper examines various important metals using powder metallurgy technology to produce elements of a total hip replacement. These alloys are 316L stainless steel alloy, Co-Cr-Mo alloy, and Ti-6Al-4V alloy. Also, this paper examines current information on the mechanical properties. Mechanical properties are discussed as a function of type of materials and process of fabrication. This article addresses the engineering aspects concerning the advantages and disadvantages of each type of material.

        Occurrence of Three Strains of Cucumber mosaic virus Affecting Tomato in Kuwait

        Three strains of Cucumber mosaic virus (CMV) have been found to cause a lethal disease, referred to as fern leaf syndromes and mild mosaic symptoms in tomato (Lycopersicon esculentum Mill.) crops grown in Kuwait. CMV strains were detected and identified based on host range, symptomatology, serology, electron microscopy, and ribonucleic acid (RNA) electrophoresis in polyacrylamide gels. A high degree of viral genomic heterogeneity was detected among CMV strains isolated in Kuwait, with no apparent correlation to symptomatology in tomato host plants. Two different virus satellites of 'CMV associated RNA 5', designated CARNA 5, were detected in two virus strains that caused both lethal disease and mild symptoms, designated CMV-D1 and CMV-S1 respectively. CARNA5 was not detected in the third CMV strain that caused fern leaf syndromes designated CMV-F. All the three isolated strains were serologically indistinguishable from each other and may belong to one serotype according to Ouchterlony gel diffusion tests. These strains transmitted via aphids (Myzus persicae Sulz) in a non-persistent manner. Physical properties of the virus strains were very similar where thermal inactivation test showed that virus withstood heating for 10 min at $70^{/circ}$, dilution end point was $10^{-4}$, and the longevity in vitro at room temperature was less than 5 days for all virus strains. CMV-D1 and CMV-F were the most devastating diseases spreading in both greenhouse and field-grown tomato where aborted flower buds failed on fruit setting due to the viral infection. This is the first report to isolate three different strains of CMV in Kuwait.

        Biological Control of n Severe Viral Strain Using a Benign Viral Satellite RNA Associated with Cucumber mosaic virus

        Two strains of Cucumber mosaic virus (CMV) isolated in Kuwait were confirmed their infectivity based on symptomatology and host range on different cultivars of tomato (Lycopersicon esculentum), tobacco(Nicotiana tabacum L.) and squash (Cucurbita pepo). The pattern of symptoms differed for the two CMV strains in tomato and tobacco, showing severe stunting and mosaic symptoms with one strain designated KU2, and almost symptomless with the other strain designated KU1. A satellite RNA 5 (sat-RNA) was found to be associated with the KU1 strain and was characterized as a benign viral satellite RNA. Using reverse transcription and polymerase chain reaction (RT-PCR) with sat-RNA specific primers, an amplified PCR product of about 160bp was determined and analyzed by gel electrophoresis. This naturally occurring benign viral satellite RNA was successfully used as a biological control agent to protect tomato plants against the severe KU2 strain. Tomato plants grown in plant-growth chambers, were preinoculated with KU1 containing the benign viral satellite and then challenge inoculated with the severe KU2 strain at different time intervals. All plants challenged three weeks after preinoculation showed nearly complete protection from subsequent infection by the severe strain. This biological control technology using plant viruses was found protective and could be successfully established sooner after the preinoculation.

        Identification of Luteovirus Nucleotide Sequences in Mild Yellow-Edge Diseased Strawberry Plants

        The availability of nucleotide sequences of the coat protein gene of Potato leafroll virus (PLRV) permitted the construction of DNA primers that were utilized for cDNA synthesis. Polymerase chain reaction (PCR) products of a 487 bp. and approximately 500 bp DNA fragments were amplified from nucleic acid extracts of PLRV-infected tissue and strawberry mild yellow-edge (SMYE) diseased strawberry tissue, respectively. The amplified DNA fragments were further differentiated by hybridization analysis with a CDNA probe for the coat protein gene of PLRV and restriction fragment length polymorphism (RFLP) analysis. These results suggest that a luteovirus is associated with the SMYE disease.

        Nitrogen Biofixing Bacteria Compensate for the Yield Loss Caused by Viral Satellite RNA Associated with Cucumber Mosaic Virus in Tomato

        To overcome the problem of the yield reduction due to the viral satellite mediated protection, a culture mix of three nitrogen-fixing bacteria species of the genus Azospirillum (A. brasilienses N040, A. brasilienses SP7, and A. lipoferum MRB16), and one strain of cyanobacteria (Anabena oryzae Fritsch) were utilized as biofertilizer mixture in both greenhouse and field experiments. When protected plants were treated with biofertilizer mixtures, the fruit yield of biofertilized plants increased by 48% and 40% in a greenhouse and field experiment, respectively, compared to untreated plants inoculated with the protective viral strain alone. Polyacrylamide gel electrophoresis (PAGE) analysis of total nucleic acid (TNA) extracts revealed that biofertilization did not affect the accumulation of the viral satellite RNA (CARNA 5) that is required for plant protection against other destructive viral strains of CMV. The yield increment was a good compensation for the yield loss caused by the use of the protective viral strain associated with CARNA 5.

      • Overlapping Region of p53/Wrap53 Transcripts: Mutational Analysis and Sequence Similarity with microRNA-4732-5p

        Background: Although the majority of investigations concerned with TP53 and its protein have focused on coding regions, recently a set of studies highlighted significant roles of regulatory elements located in p53 mRNA, especially 5'UTR. The wrap53${\alpha}$ transcript is one of those that acts as a natural antisense agent, forming RNA-RNA hybrids with p53 mRNA and protecting it from degradation. Materials and Methods: In this study, we focused on the mutation status of exon $1{\alpha}$ of the WRAP53 gene (according to exon 1 of p53) in 160 breast tumor tissue samples and conducted a bioinformatics search for probable miRNA binding site in the p53/wrap53 overlapping region. Mutations were detected, using single stranded conformation polymorphism (SSCP) and sequencing. We applied the miRBase database for prediction of miRNAs which target overlapping region of p53/wrap53 transcripts. Results: Our results showed all samples to have wild type alleles in exon 1 of TP53 gene. We could detect a novel and unreported intronic mutation (IVS1+56, G>C) outside overlapping regions of p53/wrap53 genes in breast cancer tissues and also predict the presence of a binding site for miR-4732-5p in the 5'UTR of Wrap53 mRNA. Conclusions: From our findings we propose designing further studies focused on overexpression of miRNA-4732-5p and introducing different mutations in the overlapping region of wrap53 and p53 genes in order to study their effects on p53 and its ${\Delta}N$ isoform (${\Delta}$40p53) expression. The results may provide new pieces in the p53 targeting puzzle for cancer therapy.

      • Lack of Association of Intron 3 16 bp Polymorphism of TP53 with Breast Cancer among Iranian-Azeri Patients

        Background: p53 gene is a well-known tumor suppressor gene that has several polymorphisms in both its exons and introns. It has been suggested that intron 3 16 bp duplication polymorphism may affect the gene function resulting in reduction or suppression of p53 anti tumor activity. In most case control studies a duplicated allele has been noticeably more frequent in cases rather than controls but there are also conflicting results. The aim of this study was to assess the association of intron 3 16 bp duplication polymorphism of p53 with breast cancer risk among Iranian-Azeri population. We also analyzed the clinicopathological information of patients as an epidemiological description of breast cancer in the north-west of Iran. Materials and Methods: This case-control study was performed on 221 breast cancer patients and 170 controls. Genomic DNA was extracted from peripheral blood samples and tumor tissues. p53 PIN3 genotype was determined using electrophoresis of PCR products on 8% non-denaturing polyacrylamide gels and silver staining. Results: In the control and case groups, respectively, 62.9% and 61.1% had no 16 bp insertion (A1A1 genotype), 7.1% and 7.7% had insertion in both p53 alleles (A2A2) and 30% and 31.2% were heterozygous (A1A2). There was no significant difference between genotype frequencies as well as allelic frequencies in two case and control groups. Conclusions: According to the result of the present study, the intron 3 16 bp duplication polymorphism of p53 could not be assessed as a marker of risk factor for predisposition to breast cancer in Azeri population. However, a high frequency of A2 allele (22.1%) in our population suggested that intron 3 16 bp duplication polymorphism may be a valuable marker for study in other cancers with well designed large groups.

        Phytoplasma Associated with Yellowing Disease of Washingtonia sp. in Kuwait

        Yellowing disease of palms caused by phytoplasma is spreading in the Arabian Gulf region. Surveys were conducted to determine the occurrence of the disease. Electron and fluorescence microscopy, and polymerase chain reaction (PCR) techniques were used to detect the phytoplasma associated with the yellowing disease of ornamental palm Washingtonia sp. grown in Kuwait. An accumulation of phytoplasmal DNA was observed by fluorescence microscopy in phloem tissues of diseased palms. Electron microscopy showed that phytoplasma cells were primarily confined to the phloemsieve elements of tissue samples collected from infected mature palms in the field. The pathogen was identified on the basis of molecular analysis using universal and specific nested primers in PCR amplifications. Prokaryotic 16S rDNA gene was detected in amplified PCR products. Nested PCR resulted in DNA amplification of 1.2 kbp fragment. This is the first report of a phytoplasmal rDNA gene identified from the putative causal pathogen of yellows in ornamental palms in the Arabian Gulf region.
