        Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

        High performance liquid chromatography (HPLC) was used for the determination of lignans, eleutherosides B and E, in Acanthopanax sessiliflorus fruits and their fermented wine. The lignans were quantified by a reversed-phase system using a gradient of H2O and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and 8.49 μg/mg, respectively), their fermented wine (0.45 and 1.33 μg/mg, respectively) and wine residues (no detection).

        Determination of Authentic Chiisanoside in Acanthopanax senticousus by High Performance Liquid Chromatography

        High performance liquid chromatography (HPLC) was used for the analysis of chiisanoside in each stem and root of Acanthopanax senticosus collected from South Korea, North Korea, China and Russia. A reverse-phase system using a gradient of H$_{2}$O and acetonitrile as the mobile phase was developed and detection was at 210nm. The analysis was successfully carried out within 30 min. Chiisanoside was measured in the stem and root of A. senticosus collected from various countries.

        Recently, the robustness against daily impact is important according to portablization and downsizing of mobile electronics. Especially, almost parts of cellular phone should undergo drop test when they fall 1.5 m above ground. This test simulates the case when cellular phone slips through user's fingers while he is talking on the phone. This paper studies a drop test of auto focusing (AF) actuator for camera module in cellular phone. This component is composed of voice coil motor (VCM) as an actuator and leaf spring as a guide and suspension. The leaf spring's deformation is essential for the test because its permanent distortion disables the focusing, and then, a high quality photograph cannot be obtained. Up to now the drop test has been carried out after fabricating real AF actuator. We propose a dropt test model which simulates the drop test based on finite element analysis. This model makes us enable investigate the stress acting on the clamping and curved parts of leaf spring. Stress over Von Mises criterion lets the spring deformed permanently, and then AF actuator malfunctioned, It helps us to design and modify AF actuator without manufacturing the real product. And also, it saves the time and cost for the development of new products.

        Determination of the optimum porosity for 2‑CEES adsorption by activated carbon fiber from various precursors

        In this study, we prepared ACFs with a high specific surface area from various precursors (rayon, pitch, and oxidized polyacrylonitrile-based fibers) by a steam-activation technique and investigated the effects of the micropore and mesopore fraction on 2-CEES adsorption behaviors. The activation time was precisely controlled so that the activation yield was in the range of 35–40% to ensure the mechanical properties of the ACFs. The N2 adsorption isotherm characteristics at 77K were confirmed by Brunauer–Emmett–Teller, Barrett–Joyner–Halenda and non-local density functional theory equations. The adsorption capacities of the ACF were measured by breakthrough experiments in the gas phase (750 μg/mL of 2-CEES in N2 flow). The removal efficiency of the ACFs was evaluated and compared with that of AC. From the results, specific surface areas and total pore volume of the ACF were determined to be 1380–1670 m2/g and 0.61–0.82 cm3/g, respectively. It was also observed that various pore characteristics of ACF were found to be dependent on crystallite structure of each precursor. The break through time (C/C0 = 0.10) was in the order of Oxi-Pan-H-9-2 < Saratoga AC < Rayon-H-9-3 < Pitch-H-9-4. This indicates that 2-CEES adsorption capacity could be a function not only of specific surface area or total pore volume, but also of sub-mesopore volume fraction in the range of 1.5–2.5 nm of adsorbents.

        As the semiconductor wafer is required to be thinner for better compactness and higher integrity of the electronic products, mechanical dicing saw technique using the diamond blade is facing serious problems such as low cutting speed, tool wear, side wall crack and back side chipping. Recently, UV laser dicing process has emerged as an alternative methods to the blade dicing especially for the very thin wafers below 100㎛. In this study 355 nm UV laser dicing system has been developed in order to improve the productivity and reliability of ultra thin wafer dicing process. For the quality improvement of the developed dicing system various laser parameters are investigated such as power, speed and frequency of the laser beam. Moreover various measurement techniques are employed to evaluate the dicing quality using optical microscope, SEM and focused ion beam.

        고혈압 환자에서 Nicardipine과 Esmolol의 병용 투여가 기관내삽관시 혈압과 심박수에 미치는 영향

        Background: This study was designed to determine the efficacy of a combined use of esmolol and nicardipine for blunting hypertension and tachycardia after tracheal intubation in hypertensive patients. Methods: Forty-five hypertensive patients were randomly divided into three group: group E (esmolol 0.5 mg/kg, n = 15), group N (nicardipine 30㎍/kg, n = 15), group EN (esmolol 0.25 mg/kg, nicardipine 15㎍/kg, n = 15). All patients received midazolam 0.5 mg/kg, and glycopyrrolate 0.2 mg IM for premedication. Fentanyl 1㎍/kg was injected before induction of anesthesia, and then esmolol, nicardipine, or the mixed drugs were administrated as an IV bolus and immediately followed by the induction drugs: thiopental 5 mg/kg, and succinylcholine 1 mg/kg. Endotracheal intubation was performed 90 seconds after injection of the experimental drugs. Thereafter 50% Nitrous Oxide in oxygen and 2 vol % enflurane were inhaled. BP and HR were recorded at the resting state, before fentanyl injection (base {T0}), after injection test drug (T1), after induction (T2), immediately after intubation (T3), 2 min (T4), 3 min (T5), 4 min (T6), and 5 min (T7) after intubation. Results: There was a significant attenuation in SBP, MBP, DBP after tracheal intubation on the nicardipine & mixed groups compared to the esmolol group. HR was significantly lower in the esmolol and mixed groups than in the nicardipine group after tracheal intubation. Conclusions: Combined administration of esmolol (0.25 mg/kg) and nicardipine (15㎍/kg) was effective in attenuating an increase of BP and HR during tracheal intubation in hypertensive patients.       • A novel method for olfactory bulbectomy using photochemically induced lesion

        We present the photochemically induced olfactory bulbectomy (P-bulbectomy) as a novel method to ablate the olfactory bulb thus inducing an animal model of depression. The photosensitizer Rose Bengal was injected through the tail vein and then the cool halogen light illuminated the skull region overlying of the olfactory bulb for 10 min. Two weeks after surgery, P-bulbectomy had completely removed olfactory bulb uniformly in all animals. P-bulbectomy induced typical depression-related behaviors such as hyperactivity in the open field test and an enhancement of immobility time and in the forced swimming test. Depression-related neurohistological phenomenon was also seen; reduction of choline-acetyltransferase-positive cell numbers in the medial septum and a decline in cell proliferation in the dentate gyrus of hippocampus. This study shows that P-bulbectomy may be a convenient and reproducible experimental method to produce an animal model of depression.

      • Plasma arc 광원을 이용한 과산화수소의 치아미백 효과

        The purpose of this study was to compare of the effectiveness of tooth bleaching with different hydrogen peroxide concentrations and light activated plasma arc on extracted premolar tooth The 100 specimens were stained using the CEYLON black tea. Among of them 60 specimens were selected by lightness values (*L) and were classified into five groups (n=12): Group Ⅰ, Distilled water; Group Ⅱ, 5% hydrogen peroxide gel ; GroupⅢ, 5% hydrogen peroxide+plasma arc; Group Ⅳ, 15% hydrogen peroxide; GroupⅤ, 15% hydrogen peroxide +plasma arc. Specimens were bleached until 4 times (60 min). The color change (ΔE*) of specimens was measured after stained and each bleaching time by Shade Eye-NCC. The samples were analyzed qualitatively by evaluating morphological changes in the SEM images, microhardness and quantitatively by using roughness parameters (Ra). The acquired data was analyzed by paired t-test and one-way ANOVA followed by Tukey s multiple test for variable at a 0.05 significant level. The color changes (ΔE*) significantly increased after bleaching in all groups (p<0.05). Microhardness (VHN) significant decreased after bleaching in all groups AFM and SME images exhibited that the surfaces of specimens became roughed after bleaching. Especially, Group of hydrogen peroxide with plasma arc showed more rougher than hydrogen peroxide without plasma arc in SEM. This study suggested that hydrogen peroxide with plasma arc may be better tooth whitening effects than only hydrogen peroxide.

        Toxicity of the recombinant human hyaluronidase ALT-BC4 on embryonic development

        Cumulus-oocyte complexes (COCs), which contain immature oocytes, are matured in vitro for in vitro embryo production. Oocyte and cumulus cells are then separated using hyaluronidase. To date, there have only been a few reported cases of the toxic effects of hyaluronidase on porcine oocytes. The aim of this study was to compare the effects of bovine testis-derived hyaluronidase and recombinant human hyaluronidase on oocyte denudation and quality. Porcine COCs were matured for 44 h and denuded using different hyaluronidase concentrations and exposure times. Then, oocytes were activated by electrical parthenogenesis. In experiment 1, COCs were denuded using bovine-derived, ovine-derived (Hirax), and human recombinant (ALT-BC4) hyaluronidases for 10 and 20 min. In experiment 2, bovine-derived and human recombinant (ALT-BC4 and ICSI Cumulase<sup>®</sup>) hyaluronidases were used to denude the COCs for 2 and 20 min. In both experiments the oocytes were all completely denuded, and there was no degeneration. Rate of embryo development was significantly increased in group treated ALT-BC4 for 2 min and not significantly different in other treatment groups. In general it slightly decreased with longer exposure times. These results have confirmed that different sources of hyaluronidase do not have detrimental effects on the quality of porcine oocytes and suggest that the human recombinant hyaluronidase ALT-BC4 is suitable for oocyte denudation with an increased blastocyst rate.

        인체혈장 중 에탐부톨의 HPLC 분석법의 검증 및 단일용량 투여에 의한 약물동태 연구

        An HPLC method was employed for the determination of ethambutol in human plasma. After addition of internal standard (IS, octylamine, 2 μg/mL) and alkalinization of the plasma with 5 M sodium hydroxide, the drug and IS were extracted into the mixture of chloroform and diethyl ether (40:60, v/v). Following a 15-min vortex-mixing and a 10-min centrifugation, the organic phase was spiked with 100 pL of phenylethylisocyanate (2000 μg/mL) for chemical derivatization, mixed for 5 min and evaporated to dryness under a stream of nitrogen. The residue was reconstituted with 100 μL of mobile phase and 20 pL was injected into Cl8 column with a mobile phase consisting of methanol:water (70:30, v/v). The samples were detected utilizing an ultraviolet detector at 200 nm. The method was specific and validated with a limit of 0.15 μg/mL. Infra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of this method was demonstrated by analysis of human plasma after oral administration of a single 1200-mg dose to 20 healthy subjects. From the plasma ethambutol concentration vs. time curves, the mean AUC was 9.61 ± 1.64 μg hr/mL. and Cmax of 2.68 μg/mL reached 2.73 hr after administration. The mean biological half-life of ethambutol was 3.46 ± 1.21 hr. Based on the results, this simple and validated assay could readily be used in any pharmacokinetic studies using humans.
