        Purification and Characterization of a Xylanase from Bacillus sp . KK-1

        A thermophilic Bacillus sp. strain KK-1 isolated from soil produced an extracelluar xylanase. From the culture supernatant of Bacillus sp., the xylanase was purified to homogeneity by ammonium sulfate precipitation and DEAF-Sephadex A-50 chromatography. The molecular weight of the purified xylanase was estimated to be 45 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and gel permeation chromatography. The apparent K_m values for xylanase, using oat spelt xylan and birchwood xylan as substrates, were 7.1 ㎎/㎖ and 3.2 ㎎/㎖, and V_(max) values were 27.0 μ㏖·min^(-1)·㎎^(-1) and 29.0 μ㏖·min^(-1)·㎎^(-1), respectively. The xylanase hydrolyzed oat spelt xylan to mostly xylobiose, xytotriose, and xylose. The amino acid composition indicated that the xylanase contained high amounts of amino acid residues of glutamic acid and glutamine (Glx) and aspartic acid and asparagine (Asx).

      • Effects of pyridoxine on a high‐fat diet‐induced reduction of cell proliferation and neuroblast differentiation depend on cyclic adenosine monophosphate response element binding protein in the mouse dentate gyrus

        <P><B>Abstract</B></P><P>In this study, we challenged pyridoxine to mice fed a high‐fat diet (HFD) and investigated the effects of pyridoxine on HFD‐induced phenotypes such as blood glucose, reduction of cell proliferation and neuroblast differentiation in the dentate gyrus using Ki67 and doublecortin (DCX), respectively. Mice were fed a commercially available low‐fat diet (LFD) as control diet or HFD (60% fat) for 8 weeks. After 5 weeks of LFD or HFD treatment, 350 mg/kg pyridoxine was administered for 3 weeks. The administration of pyridoxine significantly decreased body weight in the HFD‐treated group. In addition, there were no significant differences in hepatic histology and pancreatic insulin‐immunoreactive (‐ir) and glucagon‐ir cells of the HFD‐treated group after pyridoxine treatment. In the HFD‐fed group, Ki67‐positive nuclei and DCX‐ir neuroblasts were significantly decreased in the dentate gyrus compared with those in the LFD‐fed mice. However, the administration of pyridoxine significantly increased Ki67‐positive nuclei and DCX‐ir neuroblasts in the dentate gyrus in both LFD‐ and HFD‐fed mice. In addition, the administration of pyridoxine significantly increased the protein levels of glutamic acid decarboxylase 67 (GAD67) and brain‐derived neurotrophic factor (BDNF) and the immunoreactivity of phosphorylated cyclic AMP response element binding protein (pCREB) compared with the vehicle‐treated LFD‐ and HFD‐fed mice. In contrast, the administration of pyridoxine significantly decreased HFD‐induced malondialdehyde (MDA) levels in the hippocampus. These results showed that pyridoxine supplement reduced the HFD‐induced reduction of cell proliferation and neuroblast differentiation in the dentate gyrus via controlling the levels of GAD67, pCREB, BDNF, and MDA. © 2012 Wiley Periodicals, Inc.</P>

        건강한 청년의 단시간 공해노출과 일회성 운동이 폐기능과 생리반응에 미치는 영향

        This study examined whether an acute exercise during a short-term exposure to a polluted environment affected pulmonary function and physiological responses in healthy young adults. Thirteen men (mean = 22.4 yrs) were randomly exposed to a polluted air (PA) or to clean air (CA) in a chamber. During the exposure, they rested for 30 min followed by a bench stepping exercise (40 cm height, 120 bpm) for 10 min. And they recovered from the exercise for 15 min in the chamber. The pulmonary function, heart rate, blood lactate, and blood pressure were measured during the exposure. At the end of 30 min resting, peak expiratory flow rate (PEFR) was higher in PA than CA. Immediately after the exercise, forced expiratory volume at 1 sec (FEV1) was higher in PA than CA. At 15th min of recovery, forced vital capacity, FEV1, and PEFR were significantly higher in PA than CA. No differences in cardiovascular and blood lactate responses were noticed. The results suggest that an acute intensive physical work out with breathing of volatile organic compounds induced changes of pulmonary function but not physiological adjustments.

      • Cutting performance of tungsten carbide tools coated with diamond thin films after etching for various times

        <P>In this study, diamond thin films were deposited on tungsten carbide tools using surface-wave plasma-enhanced chemical vapor deposition (SWP-CVD). To eliminate the adverse effects of cobalt on the diamond deposition process, the cobalt was removed from the surface of the tools by etching with Murakami’s reagent for various times (30, 60, and 120 min). The cutting performance of the untreated and the diamond-coated WC tools was examined by performing cutting test on carbon fiber-reinforced plastic (CFRP). The results showed that all the diamond-coated tools exhibited great improvement on the durability and wear resistance compared to the uncoated one. In addition, the diamond-coated tool lift time is found to be proportional to the etching time. An increase more than twofold has been achieved when the etch time was increased from 30 min to 120 min.</P>

        Virginia Woolf's the current of consciousness is one of kinds of psychological novels. Woolf paid attention to interior consciousness escaping from exterior events. Especially its technique became popular in 1920's all over the world. It's main leading writersw were James Joyce, William Faulkner and Virginia Woolf. Particularly even though Virginia Woolf agreed with realists in describing the essentials to life and its reality, she emphasized the sub-consciousness as well as interior consciousness. Woolf was influenced by William James and Henry Bergson's philosophical thoughts, style and form of writers such as Laurence Sterne, Henry James, Marcel Proust, Dorothy Richardson and James Joyce. Moreover she borrowed the alter-ego and was debt to Freud's affect. Woolf asserted that writers ought to express an evanescent interior consciousness without adding various, exterior elements. At first she introduced the technique in Jacob;s Room and developed the skill through recollection and presupposition in Mrs Dolloway forming into an unity a lot of image and representation to express the flux of experience. She magnified ordinary trivial movement into the poetic and musical style through the inner monologue of the stream of consciousness. A s we see in James Joyce, Faulkner's novel, the representation for itself consciousness constitutes plot without Aristotle's structural elements-beginning, middle and ending. Besides we can find her queer technique-the consciousness of coexistence through an advertising aeroplane in the same place, and exquisite harmony forming a public self's soul intercourse with opening a party. Virginia Woolf devoted herself to how to sublimate into an art " a luminous halo" of life and tried to find out a solution to be united otherness with individual. The representation of sub-consciousness may be true most suitable technique, for the consciousness can't be expressed by a logical order. In short her ultimate purpose for an art is to sublimate in her art a lot of illogical impressions. Woolf introduced the skill of parethesis to shorten a long period placing the focus on the simultaneousness and symbolism. In consequence our lives can be an opulent life as we get a harmony with the outer world beyond our egoism before the creation of art as we see Mr&Mrs R믄묘's relationship. The Waves as a Bildungsroman is the work enlarged the conception of prose, the nevel is a kind of poetry and drama. Above all things we must pursue the process of Bernard's growth because whole contents is filled with the current of consciousness. Bernard has been grown up with opening disposition to be able to communicate to everybody. Moreover Bernard identified himself with the others. The different six figures were presented in poetic prose style with careful geometry balance and order compared childhood,, adolescence, manhood and old age to rising and setting sun. Percival is a main character who assembles far away friends to one place like a party of Mrs Dalloway and lighthouse in To the Lighthouse. Unlike Neville, Bernard was brought up with matruation with sentimental sorrow on Percival's death. In consequence Bernard summarized lives of all characters egoism. Bernard showed the desire of challenge against death like Percival at the same time symbolically. As " The waves broke on the shore." suggested, the repetition of life and death like waves will be continued and the defiance against death be fallen to enervation according to Providence of nature.

        Comparisons of callus formation ratios from seed explants, callus sizes, regeneration ratios from callus and regeneration efficiency for 4 orchardgrass (Dactylis glomerata L.) varieties (Three were developed in Korea and one was imported from foreign nation) are as follows; Jangbeol 102 (67.0%) has the highest callus formation ratio in 4 weeks incubated callus after bedding the seed explants, but Potomac (68.4%) has the highest ratio in 6-seek callus. Potomac (3.93cm) has the highest callus size in 4-week callus, but Jangbeol 101 (4.32cm) has the highest size in 6-week callus. Jangbeol 101 (17.7%) has the highest plant regeneration ratio in 4-week callus, but Potomac (37.4%) has the highest raion in 6-week callus. Jangbeol 102 (11.5%) has the highest plant regeneration efficiency in 4-week callus, but Potomac (25.6%) has the highest efficiency in 6-week callus.

        하악골 후방이동시 중안면부 연조직의 변화양상에 대한 임상통계학적 연구

        Prediction of the soft tissue changes following hard tissue movement is very important from the esthetic view point for patients who have orthognathic surgery. There are many cephalometric analysis of facial bone and soft tissue on the lower lip and chin region but few soft tissue analysis on the midface after mandibular setback surgery. This study was performed to obtain whether the mandibular posterior movement has influence on the midface and the predictable ratio of post-operative measurement values of the soft tissue changes following mandibular setback surgery. Fifteen patients (8 males and 7 females) who had undergone mandibular setback surgery were selected and analyzed the soft tissue movement on the upper lip and the cheek region. Post-operative changes of the soft tissue measurements after mandibular surgery were examined on pre- and post-operative cephalo-metrics and the ratio of changes were analyzed after drawing the reference line on the face with the barium sulfate solution. The reference lines were perpendicular to the intercanthal line from infraorbital foramen and lateral canthus. The results obtained were as follows : 1.There were tendancy of anterior movement of soft tissue adjacent the nose after mandibular surgery 2.There were incerased tendancy of the amount of anterior movement from the nasal crease to the cheek region. 3.The amount of anterior movement of the soft tissue was larger below the palatal plane compared with above the palatal plane in the cheek region. 4.The upper lip length was increased and moved posterior direction after mandibular setback surgery 5.The lower lip was moved posterior direction by posterior movement of the mandibular structure 6.Soft tissue of the midface around the nose moved anterior direction after mandibular setback surgery but there was no correlation between the amount of mandibular setback and the amount of the soft tissue changes

        전두부 골절 환자의 임상적 연구

        Fracture of frontal bone is infrequent, but may have serious complications because of their proximity to the brain, eyes and noses. Fractures of the frontal area range from 5% to 15% of all facial bone fracture and include supraorbital rim and frontal sinus. As frontal bone fractures most frequently occur in the multiply injured patient, a thorough clinical and radiological examination of the patient is required before diagnosis and treatment plans are established. Sometimes coorperative treatment with other department is requried. It is specially considered that incision for access to frontal region and surgical methods for open reduction, cranialization, cannulization, sinus obliteration. After surgical or conservative treatment, it may have complication. Complication of frontal bone injury vary in severity and may occur at several years after the incidents. The major types of complications are those that occur directly at the time of injury, infection and chronic problems. This is clinical study on 31 patients with frontal bone fracture, at department of oral and maxillofacial surgery in dental hospital of Wonkwang university during past ten years. The results were as follows : 1. The sex ratio of all patients is 29 (94%) male to 2 (6%) female, the average age is 33 and the prominent groups are 2nd, 3rd decade age. 2. The causative factors are mostly traffic accident 22 cases (70%) and fall dawn, industrial accidents, so on. 3. The 17 cases has shown alert mental status, but neurologic problems is in 14 (45%) cases in initial accessment. 4. Associated facial bone fractures are prominent in the maxilla (42%) and panfacial fracture (39%). 5. Involved general problems are in department of neurologic surgery problems (65%), orthopaedic problems (23%) and ophthalmologic problems (19%) in order. 6. Open reduction has done in 15 cases and 16 cases with conservative management. 7. Postoperative complications are chronic headache (42%), esthetic problems (39%) and ophthalmologic problems (35%)in order.

        인간 타액선세포 및 선암종세포주의 분화 유도시 Transglutaminase발현과 고정의존성 성장과의 관계

        Transglutaminase(TGase) catalyzes the Ca^2+ dependent acyl transfer reaction to from γ-glutamyl lysine cross-links between substrate proteins. we have examined the expression of TGase 1 and TGase 2 enzyme activity and protein expression in differentiation induced salivary gland cells(HSG), and human salivary adenocarcinoma cell lines(SGT)in reation to anchorage independent growth. The TGase1 and TGase 2 activity in SGT was higher than in HSG, and there is no significant difference between TGase 2 and TGase1 cells. HSG cells showed no increase in TGase 1 and TGase 2 upon reaching post confluence and in 1.8mM Ca^2+. Expression of the TGase 1 and TGase 2 was induced by high cell density or by high calcium in SGT. No induction of TGase 1 & 2 protein was observed in postconfiuent HSG cells, but there was notable increases in TGase 1 & 2 protein in post confiuent SGT cells. This pattern of TGase 1 & 2 protein expression correlates with the pattern of particulate activity. SGT cells showed more colony size and colony forming activity in soft agar rather than HSG cells. The ability of SGT cells, containing high levels of TGase 2 and TGase1,to form colonies in soft agar suggests a role for this enzyme in anchorage independence.
