      • Effect of ER signal peptides (KDEL) on expression and function of anti-virus monoclonal antibody in plant

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. In this study, the expression levels of mAbP SO57 with or without ER-signal peptides (Lys-Asp-Gly-Leu;KDEL) in transgenic tobacco plants were analysed in transgenic plant. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest that the potential of mAbP SO57 for rabies immunotherapy is regardless of plant specific glycan structures.

        Plant genetic engineering led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current animal expression systems. In this study, the expression level of mAbP SO57 between with and without Lys-Asp-Glu-Leu (KDEL) was compared in transgenic plant. PCR and PT-PCR analyse showed stable gene insertion transcription of heavy and light chain genes of mAbP SO57 with or without KDEL in plant, respectively. We validated expression of mAbP SO57 by western blot. Western blot showed the significantly higher expression level of mAbP SO57 with KDEL compared to without KDEL. Flow cytometry (FACS) analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb (mAbM) evidenced similar binding activity to the FcγRI receptor (CD64). High performance liquid chromatography (HPLC) analysed, glycosylation patterns of mAbP SO57 with or without KDEL. The mAbP SO57 with KDEL had glycan profile with both oligomannose type (47.6%) and golgi type (52.4%), while the mAbP SO57 without KDEL had only golgi type (100%) glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest the potential of mAbP SO57 for rabies immunotherapy, regardless of plant specific glycan structures.

        Timing and Evolution of the Most Recent Common Ancestor of the Korean Clade HIV Subtype B Based on Nef and Vif Sequences

        Molecular phylogenetic studies of the HIV-1 isolated from Koreans have suggested the presence of the so- called “Korean clade”, which can be defined as a cluster free of foreign isolates. The Korean clade accounts for more than 60% of Korean isolates and exerts characteristic amino acid sequences. Thus, it is merited to estimate when this Korean clade first emerged in order to understand the evolutionary pattern of the Korean clade. We analyzed and reconstructed the most recent common ancestor (MRCA) sequences from nef (n=229) and vif (n=179) Korean clade sequences. Linear regression analyses of sequence divergence estimates were plotted against sampling years to infer the year in which there was zero divergence from the MRCA sequences. MRCA sequences suggested the Korean clade was first emerged around 1984, before the first detection of HIV-1 in Korea in 1985. Further studies on synonymous and nonsynonymous substitution rates suggested positive selection event for the Korean clade, while other subtype B had undergone negative to neutral evolution.

      • Mito-Tempo Can Protect Against the Allergic Airway Inflammation through Reducing Mitochondrial ROS Generation

        Oxidative stress is well known to be implicated in the development of asthma. The mitochondrial respiratory chain is a major site of intracellular reactive oxygen species (ROS) generation and, at the same time, an important target for the damaging effects of ROS. Mito-Tempo is a specific mitochondrial ROS inhibitor and it is known to be associated with opening of mi-tochondrial permeability transition pore and inhibition of cell necroptosis or apoptosis. However, there is little information on the protective effects of Mito-Tempo on the inflammatory airway disorders including bronchial asthma and its acute exacerbation. We investigate the effects of Mito-tempo on the allergic airway inflammation and hyperresponsiveness using the mice sensitized with OVA and LPS and then challenged with OVA (OVALPS-OVA mice). The OVALPS-OVA mice showed the typical features of neutrophilic asthma; increased airway inflammatory cells, the pathologic changes, the increased levels of Th2 cytokines in lungs of OVALPS-OVA mice, increased mitochondrial ROS generation, and increased bronchial hyperresponsiveness. Interestingly, we found that in OVALPS-OVA mice, Mito-Tempo, a novel mitochondrial targeting agent significantly reduced the increases in inflammatory cytokines, mitochondrial ROS generation, airway inflammation, and bron-chial hyperresponsiveness. These findings indicate that mitochondrial dysfunction including oxidative damage may be im-plicated in the pathogenesis of bronchial asthma and provide the therapeutic potential of a mitochondrial targeting agent, Mito-Tempo, for bronchial asthma.

        Complete genome sequence of Sphingobium sp. strain PAMC 28499 reveals a potential for degrading pectin with comparative genomics approach

        Background Spingobium sp. PAMC 28499 is isolated from the glaciers of Uganda. Uganda is a unique region where hot areas and glaciers coexist, with a variety of living creatures surviving, but the survey on them is very poor. The genetic character and complete genome information of Sphingobium strains help with environmental studies and the development of better to enzyme industry. Objective In this study, complete genome sequence of Spingobium sp. PAMC 28499 and comparative analysis of Spingobium species strains isolated from variety of the region. Methods Genome sequencingwas performed using PacBio sequel single-molecule real-time (SMRT) sequencing technology. The predicted gene sequences were functionally annotated and gene prediction was carried out using the program NCBI nonredundant database. And using dbCAN2 and KEGG data base were degradation pathway predicted and protein prediction about carbohydrate active enzymes (CAZymes). Results The genome sequence has 64.5% GC content, 4432 coding protein coding genes, 61 tRNAs, and 12 rRNA operons. Its genome encodes a simple set of metabolic pathways relevant to pectin and its predicted degradation protein an unusual distribution of CAZymes with extracellular esterases and pectate lyases. CAZyme annotation analyses revealed 165 genes related to carbohydrate active, and especially we have found GH1, GH2, GH3, GH38, GH35, GH51, GH51, GH53, GH106, GH146, CE12, PL1 and PL11 such as known pectin degradation genes from Sphingobium yanoikuiae. These results confrmed that this Sphingobium sp. strain PAMC 28499 have similar patterns to RG I pectin-degrading pathway. Conclusion In this study, isolated and sequenced the complete genome of Spingobium sp. PAMC 28499. Also, this strain has comparative genome analysis. Through the complete genome we can predict how this strain can store and produce energy in extreme environment. It can also provide bioengineered data by fnding new genes that degradation the pectin

      • 편도체 기저외측핵의 AP5 투여가 상승된 경악반응의 습득에 미치는 영향

        본 연구는 고전적 공포조건화에 관여하는 장소로 알려진 편도체의 기저 외측핵에 장기상승작용(Long-Term Potentiation: LTP)의 생성을 차단하는 약물인 MNDA (N-methyl-D-aspartate)길항제 AP5를 미세주입함으로써 시각적 조건자극(CS)을 사용한 경우에, 상승된 경악 반응의 습득이 차단되는지를 보고자 하였다. 동물은 AP5-AP5, AP5-식염수, 식염수-AP5, 식염수-식염수 집단으로 나누어서 CS-US 배쌍직전에 AP5나 식염수를 주입한 후 훈련시켰다고 다시 검사직전에 AP5 혹은 식염수를 주입하였다. AP5-AP5집단과 AP5-식염수집단은 상승된 경악반응을 보이지 않았고, 식염수-AP5, 식염수-식염수 집단은 상승된 경악반응을 보였다. CS-US배쌍 직전에 주입한 AP5는 상승된 경악반응을 차단하였고, 검사직전에 주입한 AP5는 상승된 경악반응을 차단하지 않은 결과는 AP5가 습득을 차단했으되 그 표현에는 영향을 미치지 않았다고 해석된다. 그리고 AP5-AP5, AP5-식염수 집단이 상승률에서 유의미한 차이를 보이지 않았으므로, 상승된 경악반응의 차단이 상태의존적 인출 실패에 기인한 것이 아니고, AP5 자체의 약효때문임을 알 수 있다. It has been known that the amygadala is the neural substrate for conditioned fear as well as unconditioned fear. Among the substructures of amygdala, the basolateral nucleus where CS and US inputs converge and LTP occurs, contains high densities of NMDA receptors and so seems to have a critical role in synaptiplasticity. NMDA antagonist, AP5 prevent induction of Long-term potentiation, but not expression of LTP. LTP is an activity dependent enhancement of synaptic efficacy and is regarded the psysiological mechanisms that might underlie learning and memory. So this experiment was done to investigate what effect AP5 injection to the basolateral amygdala on the acquisition and expression of the fear conditioning, using the fear potentiated Startle paradigm. Animals were allocated to AP5-AP5, AP5-saline, saline-AP5, saline-saline groups. AP5 or saline was injected just before conditioning and testing. The re sult is that AP5-AP5, AP5-saline group didn't show the potentiated startle, comparative to the saline-AP5, saline-saline group, and AP5-AP5 group is not significantly different from AP5-saline. So we conclude that AP5 blocked the acquisition but not expression of conditio-ned fear-potentiated startle and convince that the blocking is not due to state-dependent retrieval failure.

        중재의 준거법 선택과 당사자 자치의 제한 : 국제스포츠중재를 중심으로

        Sports disputes have specific characteristics compared to disputes that arise in the field of commerce. One particularity is the judicial system in which the CAS plays a key role as the International Supreme Court for sports-related mat ers. The CAS Code ap lies whenever the parties agre to submit a sports-related dispute to the CAS(Art. R27). Once the parties to the arbitration agre ment have decided that the CAS Code should govern their proce dings. The parties’ autonomy is, however, limited to the provisions of the CAS Code that provide for such a cor esponding autonomy. The ap lication of the mandatory rules contained in the CAS Code can ot be excluded. In CAS appeals arbitration proce dings, the Panel shal decide the dispute ac ording to the ap licable sports regulations and, subsidiarily, to the rules of law chosen by the parties(Art. R58). In international sports disputes, the uniform ap lication and interpretation of the relevant regulations are es ential. Therefore, Art. R58 should be ap lied as a mandatory rule without any changes. Regulations of the sports organizations are to be qualified as valid rules of law. CAS panels may also ap ly the so-cal ed lex sportiva to the merits before considering statutory provisions of national jurisdictions. In this way, the specificities in (international) sports disputes can be taken into ac ount without the ne d to further examine the application of national legal standards.

      • 남자대학생의 흡연태도와 행동에 관한 연구

        There have been so many researches about problems of cigarettes and relation of smoking and heath since 1950's and generally known that smoking is one of the worst factor of increasing disease and mortality. No matter what such many proofs about various problems of smoking have been researched, smoking rate of current male collage students has not been decreased. Besides, most of the researches about cigarette use and its addiction have been focused on the adolescent or adult women smokers or adult male, not male collage student. Therefore, the subject of this study is about smoking attitude and smoking behavior of male collage students by general features. Smoking attitude was analyzed by modifying 'Guidelines for the content of tobacco smoking surveys of the general population'(WHO, 1982). Two hundred of male collage students participated to the distributed questionnaire for one week in May. The smoking attitude was categorized into duration of smoking and number of cigarette per day(CPD), and nicotine dependency, investigated by Fagerstrom Tolerance Questionnaire(FTQ) (Fagerstorm & Schneider, 1989). All statistics were performed by procedure of SAS according to number, percentage, mean, standard deviation, and ANOVA. The results are as follows: 1. Smoking attitude level was mean 25.73±5.15 marks, ranging from 13 to 39 marks of the total 40 marks. 2. Duration of cigarettes, number of cigarette per day, and nicotine dependency were mean 5.14±2.84 years, 12.88±6.85 pieces, and 4.78±1.69, respectively. 3. Smoking attitude showed significance according to general features such as marital state and allowance, but the significance according to marital state is not valuable because the numerical difference is so big between married one and single that they cannot be compared with each other. 4. Duration of smoking was significant according to general features such as age and allowance. 5. Smoking amount showed significance the factors of age only. 6. ll1ere was no significance in nicotine dependency according to general features. As a result, smoking attitude of male collage student in this study shows moderate level. Smoking attitude and duration is significant according to allowance, and smoking amount is significant according to age. Nicotine dependency according to general features is not significant but comparing with the preceding study about adult male, male collage students in this study are respectively more nicotine-dependent than adult male smokers. In conclusion, the right allowance usage strategies and effective heath education focused on the young are needed. In addition, helpful programs and information spreading strategies are necessary for establishing higher level of smoking attitude. At last, systemic regulations in daily living for ""anti-nico-dependent"" and perspective reforming of male university student are needed for reducing nicotine dependency.

        Chemical Properties and Nutrient Loadings of Rainwater during Farming Season

        Recently, special attention has been given to acid rain and its problem to environment such as acid precipitation and air pollution in East Asia. In the present study, rainwater samples were collected from Apr to Nov in 2012. The samples were chemically characterized for the assessment of emission sources. Suwon and Yeoju regions, typical agricultural areas in South Korea, were chosen for study sites. Ion composition and cation-affected neutralization were determined to evaluate the contribution of cations to the acidity of rainwater. Ion and electrical conductivity between the measured and the estimated showed high correlation. The cations observed in Suwon and Yeoju were Na<SUP>+</SUP> > NH4<SUP>+</SUP> > K<SUP>+</SUP> > Ca<SUP>2+</SUP> > Mg<SUP>2+</SUP> > H<SUP>+</SUP> and Na<SUP>+</SUP> > K<SUP>+</SUP> > NH4<SUP>+</SUP> > Ca<SUP>2+</SUP> > Mg<SUP>2+</SUP> = H<SUP>+</SUP>, respectively. The anions of all sites were SO4<SUP>2-</SUP> > NO3- > Cl<SUP>-</SUP>. While the amounts of sulfate, one of the major dissolved components of rainwater, were 77.6 and 75.6 ueq L<SUP>-1</SUP> in Suwon and Yeoju, the ones of NSS-SO4<SUP>2-</SUP> (Non-Sea Salt sulfate) were 83 and 82% in Suwon and Yeoju, respectively. The comparison of observed pH values (pHobs) with the theoretical pH values (pHthe) showed that the neutralization of rain water considerably went along during the study periods. The highest amounts of rainfall throughout the year in Suwon and Yeoju were 572.3 and 484.6 mm in July, and its corresponding nitrogen loadings in Suwon and Yeoju were 5.28 and 3.50 kg ha-1, respectively. The major ion contents for crop growth with SO4<SUP>2-</SUP>, Ca<SUP>2+</SUP>, K<SUP>+</SUP> and Mg<SUP>2+</SUP> were 51.7, 5.2, 11.8 and 1.8 kg ha<SUP>-1</SUP> in Suwon and 34.2, 4.0, 4.2 and 1.1 kg ha<SUP>-1</SUP> in Yeoju.

        Despite all that is known about music theory and psychodynamics what actually occurs within a composer's mind remains poorly understand. There are so many composers, Handel, Schumann, Berlioz, all of whom had manic-depressive illness, whose lives were used to illustrate the nature and impact of manic-depressive illness, and their music was used to illustrate the essence of human moods, from joy to socrrow and exaltation to despair. We find the effects of depressive mood in Mozart's and Beethoven's work. Therefore we must understand the musical characteristics as well as the composer's psychological status, for we comprehend the composer's work. If we play any songs perfectly, we must understand the misucal characteristic of the work and the composer's psychological status, and then add our feelings.
