        심근경색 모델 흰쥐에서 스트레스가 B-type Natriuretic Peptide 발현 및 심박변이율 변화에 미치는 영향

        심박변이율의 감소는 심근 경색 환자에서 예후의 악화와 관련되어 있다. 심박변이율 중 very low frequency가 심부전환자의 예후를 판단할 수 있는 독립적 인자로서 사용이 가능하다고 보고된 바 있으며, 심실의 압력 증가에 반응해서 생성되는 B-type natriuretic peptide가 심근 경색의 예후 인자로서 사용할 수 있음 역시 보고된 바 있다. 본 연구에서는 심근경색 모델을 제작하여 일정의 회복 기간을 거친 다음 다시 일정 기간의 스트레스를 겪게 한 후 심박변이율의 변화 및 심실 조직에서의 BNP 발현 정도를 비교하여 스트레스에 의한 영향을 관찰하고자 하였다. Sprague-Dawley계 수컷 흰쥐 15마리를 대상으로 하였다. 실험동물은 인위적 스트레스 및 수술을 받지 않은 대조군(CON, n=5), 심근경색 수술을 받은 후 restraint stress를 받지 않은 군(MI+No Stress, n=5), 심근경색 수술을 받은 후 1주일 동안 restraint stress를 받은 군(MI+Stress, n=5)으로 분류하였다. 심근경색 모델 제작 직후 15분 이상 심전도의 변화를 기록하였으며, 2개월 정도 후 심근경색 수술을 받은 동물을 두 군으로 나누어 그 중 한 군에는 1주일 동안 restraint stress를 가한 후 restraint stress를 가하지 않은 동물과 심박변이율을 비교 분석하였다. 심실 조직은 좌심실 전벽 부위의 위축을 관찰할 수 있었고, 대식세포에 의하여 응고, 괴사된 심근세포의 탐식과 혈관이 풍부한 육아조직 및 섬유 변화를 볼 수 있었다. 심박변이율은 심근경색 수술을 받은 직후 심박수는 유의한 증가를 보였고, standard deviation of the normal to normal intervals (SDNN), very low frequency (VLF) 및 low frequency (LF)의 유의한 감소를 보였다. 심근경색 수술을 받았던 동물에서 두 달 정도의 회복 기간을 거친 후 스트레스를 받지 않은 군은 심박수 및 기타 심박변이율 분석에서 SDNN 값을 제외하고는 정상군과 유의한 차이를 관찰할 수 없었던 반면 1주일 동안 스트레스를 받은 군에서는 심박수가 다시 유의하게 증가했을 뿐만 아니라 SDNN, VLF 및 LF 역시 정상군과 유의한 차이를 보였다. 이상의 결과를 종합하면 심근경색 동물에 대해 1주일 동안 스트레스를 가한 결과 심박변이율이 감소하고 심실에서의 BNP 발현은 더욱 증가하여 악화함을 보여주고 있다. Diminished heart rate variability (HRV) is associated with less favorable prognosis after myocardial infarction (MI). It has been reported that very low frequency (VLF) power in HRV analysis is an independent risk predictor in patients with congestive heart failure and B-type natriuretic peptide (BNP) can be used as a prognostic factor of MI. The purpose of this study was to investigate whether stress has an effect to the changes of BNP expression and/or heart rate variability in MI model in rats. Male Sprague-Dawley rats were divided into 3 groups: control group (CON), MI group (MI+No Stress), and MI followed by stress group (MI+Stress). MI+Stress group rats were raised for a two month recovery period after the operation, followed by being exposed to restraint stress for 2 hours per day for 1 week. Electrocardiogram was recorded after the operation and the last day after 1 week of stress. The frequency components of HRV were calculated in the frequency domain such as VLF, low frequency (LF), high frequency (HF) and so on. In HRV analysis, standard deviation of the normal to normal intervals (SDNN) was significantly reduced in both groups compared to the control group. VLF and LF also were significantly reduced in MI+Stress group compared to the control groups. In addition, BNP expression in western blotting was shown the strongest bands in MI+ Stress group among experimental groups. These results suggest that BNP and HRV were aggravated by stress in MI rat model. (Korean J Str Res 2010;18:275∼285)

      • Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

        <P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression.         Kim Young-rang has been known as poet who commanded elaborated type, meter, soft tone harmony of voice. Manufacture do poetry which have self-regulating structure through conscious calculation and materialize own inside sensuously. And his interest about shi-jo and pansori is reflected in production of poetry. He creates four line poetry criticizing existent poetry style(shi-jo and pansori and so on)in poetry´s circles in the 1930s that form of free poetry catches seat already, and plays a variation four line poetry learning forms and groped new modernistic poetry form. He established peculiar sensitivity of individual into compensation of shape and create chard free verse of new form. These features that shows in poetry form that defend self from colony actual broken external order is revealed can be seen. It is because construct space of imagination and found psychological pupil of the eye in to close loss persimmon formed on inside looking away actuality that can not accomplish own dream. Therefore, that appears in his poetry sensual and the trillion brazier fortune worlds can see that is product of injustice mind to look away colony actuality. Kim Young-rang did not compromise in colony actuality and could keep own inside to be upright through writing such poetry.

      • 미치광이풀 모상근 배양에서 Tropane Alkaloid 생산성 증진을 위한 최적 생물학적 엘리시터 선발

        Scopolamine and hyoscyamine which belong to tropane alkaloids are the pharmaceutically valuable anticholinergic drugs. In order to increase the productivities, the effects of elicitation were investigated during hairy root cultures of Scopolia parviflora. Biotic elicitors originated from 3 fungi and 1 yeast were prepared as homogenate and supernatant and added to 3-week-old cultures. Both of homogenate and supernatant of Candida albicans elicitors increased the scopolamine production. The production of hyoscyamine was enhanced by homogenate of Fusarium solani and supematant of C. albicans. Most of the other fungal elicitors were also improved on the tropane alkaloid production compared to non-treatment. Among the elicitors tested, C. albicans was proved the optimal biotic elicitor on tropane alkaloids production. These results will be served mass production of tropane alkaloids by large-scale production.

      • 남성화를 보이는 여성에서 발견된 난소의 Steroid Cell Tumor 1예

        Steroid cell tumor is a rare ovarian sex cord-stromal tumor which accounts for 0.1% of all ovarian tumors. Until now, only 4 cases have been reported in domestic literatures. Steroid cell tumor often secrets testosterone and presents virilization in adult women or precocious puberty in children. Treatment is often performed by surgical removal, adjuvant chemotherapy and radiation, but completely accepted treatment was not existed. We experienced a case of steroid cell tumor, which was manifested by typical virilization in a 43-year old patient, who was previously performed hysterectomy and unilateral oophorectomy. So, we present with a brief review of the literatures.

        중환자의 욕창 예방 연구 : 욕창 예방 QI팀을 중심으로

        Background : A pressure sore was defined as any skin lesion caused by unrelieved pressure and resulting in damage to underlying tissue. The health care institutions in the United States were reported the incident rate of pressure sores ranging from 6 to 14%. United states were reported the incident rate of pressure sores in patients rate of pressure sore. Also, Annual expenditures for the care of pressure sores in patients in the United States have been estimated to be $7.5 billion; furthermore, 50 percent more nursing time is required to care for patients with pressure sore in comparison to the time needed to implement preventive measures against pressure sore formation. However, In Korea, there were little reliable reports, or researches, about incidence rates of pressure sore in health care institution including intensive care unit and about the integrated approach like CQI action team for risk assessment, prevention and treatment of pressure ulders. Therefore, this study was to develop pressure sore risk assessment tool and the protocol for prevention of pressure sore formation through CQI action team activities, to monitor incident rate of pressure sore and the length of sore formation for patients at high risk, and to approximately estimate nursing time for sore dressing during research period as the effect of CQI action team. Method : CQI action team in intensive care unit, launched since early 1996, reviewed the literature for the standardized risk assessment tool, developed the pressure sore assessment tool based on the Braden Scale, tested its validity, compared on statistics including incidence rate of pressure sore for patients at high risk. Throughout these activities, CQI action team was developed the protocol.called as St. Marys hospital Intensive Care Unit Pressure Sore Protocol, shifted the emphasis from wound treatment to wound prevention, After applied the protocol to patients at high risk, the incident rate and the period of prevention against pressure development were tested with those for patients who received care before implementation of protocol by Chi-square and Kaplan-Meier Method of Survival Analysis. Result : The CQI action team found that there was significant difference of incidence rate of pressure sores between patients at high risk( control group) who received care implementation of protocol (p<.05). 25% possibility of pressure sore formation was shown for the patients with 6th hospital day in ICU in control group. In experimental group, the patients with 10th hospital day had 10% possibility of pressure sore. Therefore, there was significant difference(p< .05) in survival rate between two groups. Also, nursing time for dressing on pressure sore in experimental group was deceased as much as 50% of it in control group. Conclusion : The collaborative team effort led to reduced incidence, increased the length of prevention against pressure sore, and declined nursing care times for sore dressing. However, there have had several suggestions for future study. The preventive care system for pressure sore should be appleed to patients at moderate, or low risk throughout continuous CQI team activities based on Bed sore Indicator Fact Sheet. Hospital-wide supports, such as incentives, would be offered to participants for keeping strong commitment to CQI team. Also, Quality Information System monitoring incidents and estimation cost of poor quality. like workload(full time equivalence) or financial loss, regularly in a hospital has to be developed first for supporting CQI team activities as well as empowering hospital-wide QI implementation. Being several limitations, this study would be one of the report cards for the CQI team activities in intensive care unit of an acute hospital and a trial of quality improvement of health care in Korea.

      • 흰쥐 뇌의 냉동손상 후 항원제공세포와 신경교원섬유산단백의 발현 양상

        Background : Traumatic brain injury is a multifaceted injury that involves direct mechanical damage, intraparenchymal and subarachnoid hemorrhage, breakdown of the blood-brain barrier, excitotoxicity, and ischemia. Even though much investigations were performed, acceptable mechanical informations were rare. The aim of this study was to reveal the migration of antigen presenting cells associated with inflammation and the expression pattern of GFAP associated with gliotic scars in cerebral cortex of rats after cryoinjury. Material & Method : A total of 18 male Sprague-Dawley rats weighing 300 g, 2 months old, were used throughout the experiments. To injure the brain, rats were anesthetized for surgery with 3.5% chloral hydrate(1 ㎖/100 g, intraperitoneally); the frontal bones were exposed by elevating the skin; and craniectomies were performed adjacent to the central suture, midway between lambda and bregma. A cryoinjury was then created by applying a cold probe (3-mm-diameter steel rod chilled in liquid nitrogen) to the left frontal cortex (ipsilateral cortex) for 1 minute. Rats were sacrificed at 6 hours, 1, 4, 7 and 14 days postsurgery (n=3, per time point), and three rats were sacrificed as normal controls. Serial brain cryosections were made by cryostat. For immunohistochemistry, brain tissue sections were allowed to react with mouse anti-rat MHC class II antibody(1:500), mouse anti-rat ED2 antibody(1:500), and mouse anti-rat GFAP antibody(1:200). Results : MHC class II positive dendritic cells were absent in normal cerebral cortex parenchyme, but were more 16 times number in injured rats at 7 days after cryoinjury. ED2 positive macrophages were rare in normal cerebral cortex parenchyme, and were more 14 times number in injured rats at 4 days after cryoinjury. GFAP positive astrocytes were not observed in normal rats, and increased suddenly at 4 days rats after cryoinjury. GFAP positive neurons were not expressed in normal rats, and in cryoinjured rats. Conclusion : In conclusion, the above results suggest that the increases of the number of the two kinds of the antigen-presenting cells affect cell-mediated immune responses and phagocytosis, and GFAP positive cells attends in the formation of gliotic scars.

      • 폐 활성 슬러지의 납 흡착에 따른 기초 요소의 영향

        본 연구에서는 하수 처리장에서 발생되는 상당량의 폐 슬러지를 전처리 없이 중금속 제거에 사용하였다. 본 연구의 목적은 폐 슬러지를 이용하여 중금속을 제거하는 것이다. 온도와 pH, 초기 농도의 영향과 보관 시간에 따른 납 흡착량의 영항을 흡착등온 모델과 흡착속도 모델에 적용시켜 활성슬러지의 납 흡착 특성을 고찰하였다. 납, 카드뮴, 크롬, 니켈 중 슬러지는 납을 가장 많이 제거하였고, pH 4.5~5.5에서 최대의 흡착량을 나타냈으며, 온도가 증가할수록 흡착량은 증가하였고, Langmuir model에 잘 적용되었으며, 초기 최대 흡착속도도 Michaelis-Menten 형태 식으로부터 구하였다. In sewage treatment plant, waste sludge was one of bothering thing to be treated. The objective of this study was to examine the removal of heavy metal by waste sludge. The effect of factors such as temperature, pH, storage time, initial concentration of lead, and initial amount of biomass on adsorption of lead was investigated. And the biosorption model, adsorption isotherm, and initial adsorption rate model were evaluated. Waste sludge was proved to have an excellent ability to remove lead than other heavy metals such as cadmium, chromium, nickel in pure and mixed solution. The optimum pH of biosorption with waste sludge was 4.5~5.5, and the amount of adsorption was increased with increasing temperature. The adsorption isotherm of lead to waste sludge followed to Langmuir model. The initial adsorption rate of lead to waste sludge followed to Michaelis-Menten type kinetic.

      • 포장재질에 따른 김치의 저장성Ⅰ

        수출용 김치에 주로 사용되고 있는 유리병을 대상으로 유통과정 중 상품성 유지와 더불어 저장성 향상을 도모하고자 포장방법에 따른 저장성 효과를 비교ㆍ실험하였다. 포장방법은 유리병을 이용하여 밀폐포장과 투과성 포장을 하였으며 내부 포장재로 사용한 PE film 두께별(0.01, 0.06, 0.08, 0.1㎜) 저장성 향상능력을 비교하였다. 포장김치는 10℃에서 28일간 저장하면서 pH 및 산도, 색도의 변화와 관능적 품질변호를 측정하였다. 저장기간에 따른 pH와 산도의 변화에서 포장방법간에 큰 차이는 없었으나 밀폐 포장의 경우 film 두께가 다소 얇은 0.06, 0.01㎜ 처리구에서, 투과성 포장의 경우 film 두께가 다소 두꺼운 0.06, 0.08㎜ 처리구에서 높은 pH와 낮은 산도를 나타내었다. 특히 0.06㎜ 처리구의 경우 포장방법에 관계없이 control이나 타처리구에 비해 완만한 pH와 산도변화를 나타내었고, 상미한계점이 연장될 뿐만 아니라 효모에 의한 연부현상이나 탄산가스에 의한 뚜껑의 개봉이나 폭발음 등의 현상이 나타나지 않았다. 색도 측정시 저장 14일을 전후로 증가하던 L, a, b값이 감소하는 경향이었으며 이는 밀폐 포장과 투과성 포장에서 동일하게 나타났다. 관능적 품질평가에서 포장방법간에 큰 차이는 없었으나 신맛과 조직김이 유의적인 관계가 있는 것으로 보였고, 상미한계점을 경과할수록 조직감이 기호도를 좌우하는 것으로 나타났다. 또한 0.06㎜ 투과성 포장시 밀폐 포장보다 신맛이 좋은 것으로 나타났다. 저장 14일 이후부터는 0.06㎜ film 처리시 관능적 품질평가 전반에 걸쳐 우수한 것으로 인정(5%의 범위)되었다. 이상의 결과로 유리병포장의 경우 기체투과성이 높은 PE film으로 내부포장하는 것이 저장성 연장효과를 기대할 수 있으며, 특히 0.06㎜ film으로 밀폐 포장 또는 투과성 포장시 높은 저장성 연장효과와 더불어 탄산가스에 의한 팽창방지를 완화시킬 것으로 기대된다. The effect of various packaging methods on kimchi quality was investigated in order to develop the packaging techniques for preventing bottled kimchi products from inflation and expolsing, due to fermentative gas evolved during storage and distribution. Kimchi was packaged in bottle(400g) under atomosphere different methods; 1) control, 2) airtight packaging, 3) permeable packaging, Kimchi packaged with polyethylene(PE) film was stored at 10℃ and film thickness were used 0.01, 0.06, 0.08, and 0.1㎜. To evaluated quality change of kimchi, pH, acidity, color, and sensory score were measured periodically. Although pH and acidity change was not significant depending on the packaging method, airtight packaging with 0.06, 0.01㎜ film and permeable packaging with 0.06, 0.08㎜ film showed higher pH value and lower acidity then those of the others. Color change at kimchi as L, a, b value increased until 14 days and the decreased in airtight and permeable packaging. Sensory evaluation were not significant among packaging method during storage, but sour taste and texture scores were significant. Also, sour tast of permeable packaging with 0.06㎜ film showed higher than airtight packaging. After 14 days of storage, total sensory evaluation was excellent of packaging with 0.06㎜ film. Therefore, 0.06㎜ was found to be the most effective packaging method, although airtight and permeable packaging were not significant for storage properties.
