        A First-Time-in-Human Study of GSK2636771, a Phosphoinositide 3 Kinase Beta-Selective Inhibitor, in Patients with Advanced Solid Tumors

        <P><B>Background:</B> The PI3K/protein kinase B (AKT) pathway is commonly activated in several tumor types. Selective targeting of p110β could result in successful pathway inhibition while avoiding the on- and off-target effects of pan-PI3K inhibitors. GSK2636771 is a potent, orally bioavailable, adenosine triphosphate-competitive, selective inhibitor of PI3Kβ.</P><P><B>Methods:</B> We evaluated the safety, pharmacokinetics, pharmacodynamics and antitumor activity of GSK2636771 to define the recommended phase II dose (RP2D). During the dose-selection and dose-escalation stages (parts 1 and 2), patients with <I>PTEN</I>-deficient advanced solid tumors received escalating doses of GSK2636771 (25–500 mg once daily) using a modified 3+3 design to determine the RP2D; tumor type-specific expansion cohorts (part 3) were implemented to further assess tumor responses at the RP2D.</P><P><B>Results:</B> A total of 65 patients were enrolled; dose-limiting toxicities were hypophosphatemia and hypocalcemia. Adverse events included diarrhea (48%), nausea (40%), and vomiting (31%). Single- and repeat-dose exposure increased generally dose proportionally. GSK2636771 400 mg once daily was the RP2D. Phospho/total AKT ratio decreased with GSK2636771 in tumor and surrogate tissue. A castrate-resistant prostate cancer (CRPC) patient harboring <I>PIK3CB</I> amplification had a partial response for over a year; an additional 10 patients derived durable (≥24 weeks) clinical benefit, including two other patients with CRPC with <I>PIK3CB</I> alterations (≥34 weeks). GSK2636771 400 mg once daily orally induced sufficient exposure and target inhibition with a manageable safety profile.</P><P><B>Conclusions:</B> Genomic aberrations of <I>PIK3CB</I> may be associated with clinical benefit from GSK2636771. <I>Clin Cancer Res; 23(19); 5981–92. ©2017 AACR</I>.</P>

        Sensorless Maximum Power Control of a Stand-Alone Squirrel-Cage Induction Generator Driven by a Variable-Speed Wind Turbine

        For the cost-eff ectiveness of variable-speed wind energy conversion systems (WECSs), it is extremely important to extract the maximum available power at diff erent wind speeds within the normal operating range. Furthermore, reduction of the electric generator losses additionally contributes to the effi ciency of the WECS, whereas reduction of the number of sensors is benefi cial in terms of reliability and cost. This paper presents sensorless control of a stand-alone WECS comprising a squirrel-cage induction generator (IG) and a battery system. An indirect rotor-fi eld-oriented control (IRFOC) algorithm including stray load and iron losses and online tuning of IG’s equivalent resistances and magnetising inductance is adopted for IG control to achieve high level of agreement with the actual machine. Fuzzy-logic-based optimisations of the wind turbine (WT) and IG are implemented to maximise the IG output power. The IG speed, which is required by the IRFOC algorithm, is estimated by using a model-reference-adaptive-system. The estimated IG speed is also utilised for online WT optimisation in combination with the IG torque obtained from the IRFOC equations. The performance of the proposed control strategy has been experimentally evaluated and compared with two competing sensorless control strategies for two 1.5 kW IGs of diff erent effi ciency class.

        Small-Size Induction Machine Equivalent Circuit Including Variable Stray Load and Iron Losses

        The paper presents the equivalent circuit of an induction machine (IM) model which includes fundamental stray load and iron losses. The corresponding equivalent resistances are introduced and modeled as variable with respect to the stator frequency and flux. Their computation does not require any tests apart from those imposed by international standards, nor does it involve IM constructional details. In addition, by the convenient positioning of these resistances within the proposed equivalent circuit, the order of the conventional IM model is preserved, thus restraining the inevitable increase of the computational complexity. In this way, a compromise is achieved between the complexity of the analyzed phenomena on the one hand and the model’s practicability on the other. The proposed model has been experimentally verified using four IMs of different efficiency class and rotor cage material, all rated 1.5 kW. Besides enabling a quantitative insight into the impact of the stray load and iron losses on the operation of mains-supplied and vector-controlled IMs, the proposed model offers an opportunity to develop advanced vector control algorithms since vector control is based on the fundamental harmonic component of IM variables.

        Use of ChatGPT for Determining Clinical and Surgical Treatment of Lumbar Disc Herniation With Radiculopathy: A North American Spine Society Guideline Comparison

        Objective: Large language models like chat generative pre-trained transformer (ChatGPT) have found success in various sectors, but their application in the medical field remains limited. This study aimed to assess the feasibility of using ChatGPT to provide accurate medical information to patients, specifically evaluating how well ChatGPT versions 3.5 and 4 aligned with the 2012 North American Spine Society (NASS) guidelines for lumbar disk herniation with radiculopathy. Methods: ChatGPT's responses to questions based on the NASS guidelines were analyzed for accuracy. Three new categories—overconclusiveness, supplementary information, and incompleteness—were introduced to deepen the analysis. Overconclusiveness referred to recommendations not mentioned in the NASS guidelines, supplementary information denoted additional relevant details, and incompleteness indicated omitted crucial information from the NASS guidelines. Results: Out of 29 clinical guidelines evaluated, ChatGPT-3.5 demonstrated accuracy in 15 responses (52%), while ChatGPT-4 achieved accuracy in 17 responses (59%). ChatGPT-3.5 was overconclusive in 14 responses (48%), while ChatGPT-4 exhibited overconclusiveness in 13 responses (45%). Additionally, ChatGPT-3.5 provided supplementary information in 24 responses (83%), and ChatGPT-4 provided supplemental information in 27 responses (93%). In terms of incompleteness, ChatGPT-3.5 displayed this in 11 responses (38%), while ChatGPT-4 showed incompleteness in 8 responses (23%). Conclusion: ChatGPT shows promise for clinical decision-making, but both patients and healthcare providers should exercise caution to ensure safety and quality of care. While these results are encouraging, further research is necessary to validate the use of large language models in clinical settings.

        Hierarchical Structured Multi-agent for Distributed Databases in Location Based Services

        Location management is very important in location-based services to provide services to the mobile users like banking, city guides and many more. Ubiquitous and mobile devices are the source of data in location management and its significant operations are update and search method. Some studies to improve these were presented by using optimal sequential paging, location area scheme and hierarchical database scheme. In addition, not all location services have the same access methods on data and it lead to difficulties of providing services. A proposed location management of multi-agent architecture is presented in this study. It shows the coordination of the agents on the distributed database of location-based services. The proposal focuses on the location management of the mobile object presented in a hierarchical search and update. Also, it uses a nearest neighbor technique for efficient search method of mobile objects.

        Pig meat production in the European Union-27: current status, challenges, and future trends

        The main objective of this study was to present data on the current situation and future trends of pig meat production in the European Union-27 (EU). Pig production has played an important social and economic role for centuries in many states of the EU. In 2022, pig meat production in the EU reached 23 M tons, which represented 21% of total production worldwide. The two key reasons that justify such amount of pork produced, are the acceptance and high consumption of the meat by the local population and the high quality of the meat produced which facilitated pork export. However, current data show a reduction in pork production for the last three years, as a consequence of a series of events that include i) problems with the chain of ingredients supply, ii) uncontrolled increase in African Swine Fever (ASF) outbreaks, iii) fast recovery of pig production in China, iv) increasing concerns by the rural population on the high cost to meet future requirements of the EU legislation on farm management, environmental sustainability and animal welfare, v) increased cost of all inputs involved in pig production and vi) limited interest of the new farmer generation to work on the pig sector. Consequently, pork production is expected to decrease in the EU for the next years, although sales will be maintained at a relative high level because pork is the meat preferred by local consumers in most EU countries. In order to maintain the favourable position of the pork industry in the near future, strategies to implement include: i) maintain the quality of the meat destinated to export markets, ii) improve the control of outbreaks of ASF and other swine diseases, iii) implementation of technological innovations to improve working conditions making more attractive to work in the pork sector of the food chain to the new generation of farmers and workers.

      • Single-walled carbon nanotubes oust graphene and semiconductor saturable absorbers in<i>Q</i>-switched solid-state lasers at 2<i>µ</i>m

        <P>The superior performance of a saturable absorber (SA) based on randomly-oriented single-walled carbon nanotubes (SWCNTs) operating in the spectral region of their first fundamental transition E-11 over a graphene-SA containing several (n = 3) carbon layers and a commercial semiconductor SA (SESA) for passive Q-switching of bulk thulium solid-state lasers is demonstrated. Using SWCNT-SA in a compact diode-pumped Tm:KLuW laser, a maximum average output power of 1.36 W is achieved at 1932-1944 nm corresponding to a slope efficiency of 41% and pulse characteristics of 48 ns/5.5 mu J which are superior to that for graphene-SA (123 ns/3.1 mu J) and SESA (61 ns/1.1 mu J). SWCNT-SAs are promising for eye-safe miniature lasers at similar to 2 mu m generating nanosecond pulses in the MHz repetition frequency range.</P>

        Use of δ-Aminolevulinic Acid in Swine Diet: Effect on Growth Performance, Behavioral Characteristics and Hematological/Immune Status in Nursery Pigs

        Certain amino acids are essential precursors of a variety of important biomolecules in addition to their major function as protein building blocks. ${\delta}$-Aminolevulinic acid (ALA) is synthesized from the condensed form of succinyl-CoA with glycine after decarboxylation catalyzed by ALA synthase. The objective of the study was to determine the effects of ALA supplementation on growth performance, behavioral characteristics and hematological/immune status in nursery pigs. A total of 144 pigs weaned at 21 d of age were allotted to three dietary treatments representing (-) control (w/o antibiotics; NC), (+) control (w/carbadox at 50 mg/kg; PC), and the treatment group with ALA supplementation (0.05%; TA). Each treatment had 6 pens (replicates) with 8 pigs per pen. Pigs were fed phase 1 (21.9% CP, 1.40% Lys) and 2 (20.6% CP, 1.15% Lys) experimental diets for 3 and 2 wks, respectively. Feed intake and weight gain were measured weekly during phase 1 and at the end of phase 2. At the end of phase 2, blood samples were taken and analyzed using an automated hematology analyzer. Skin color and activity of pigs (48 h) from all pens in each treatment were measured at the second week of phase 2. Growth performance was not affected (p>0.05) by the dietary supplementation of ALA during the 5 wk nursery period. Pigs in the TA (6.46) and PC (6.68) had a higher (p<0.05) number of red blood cells ($10^6cell/{\mu}L$) than pigs in the NC (6.15). Pigs in PC (12.16) had a higher (p<0.05) hemoglobin level (g/dL) than pigs in the NC group (11.29) and the TA group (11.47). Pigs in the TA and PC had darker (p<0.05) and less (p<0.05) yellow skin color than pigs in the NC. Pigs in the PC tended (p = 0.081) to be less active than pigs in the other groups. There were no differences in behavioral characteristics between the NC and the TA. The data suggest that ALA supplementation has no adverse effects on growth performance of nursery pigs. Moreover, ALA supplementation increased red blood cell counts which may be beneficial to pigs.
