      • Wall stretch and thromboxane A2 activate NO synthase (eNOS) in pulmonary arterial smooth muscle cells via H2O2 and Akt-dependent phosphorylation

        <P>Pulmonary arteries (PAs) have high compliance, buffering the wide ranges of blood flow. Here, we addressed a hypothesis that PA smooth muscle cells (PASMCs) express nitric oxide synthases (NOS) that might be activated by mechanical stress and vasoactive agonists. In the myograph study of endothelium-denuded rat PAs, NOS inhibition (L-NAME) induced strong contraction (96 % of 80 mM KCl-induced contraction (80K)) in the presence of 5 nM U46619 (thromboxane A(2) (TXA(2)) analogue) with relatively high basal stretch (2.94 mN, S(+)). With lower basal stretch (0.98 mN, S(-)), however, L-NAME application following U46619 (TXA(2)/L-NAME) induced weak contraction (27 % of 80K). Inhibitors of nNOS and iNOS had no such effect in S(+) PAs. In endothelium-denuded S(+) mesenteric and renal arteries, TXA(2)/L-NAME-induced contraction was only 18 and 21 % of 80K, respectively. Expression of endothelial-type NOS (eNOS) in rat PASMCs was confirmed by RT-PCR and immunohistochemistry. Even in S(-) PAs, pretreatment with H2O2 (0.1-10 mu M) effectively increased the sensitivity to TXA(2)/L-NAME (105 % of 80K). Vice versa, NADPH oxidase inhibitors, reactive oxygen species scavengers, or an Akt inhibitor (SC-66) suppressed TXA(2)/L-NAME-induced contraction in S(+) PAs. In a human PASMC line, immunoblot analysis showed the following: (1) eNOS expression, (2) Ser(1177) phosphorylation by U46619 and H2O2, and (3) Akt activation (Ser(473) phosphorylation) by U46619. In the cell-attached patch clamp study, H2O2 facilitated membrane stretch-activated cation channels in rat PASMCs. Taken together, the muscular eNOS in PAs can be activated by TXA(2) and mechanical stress via H2O2 and Akt-mediated signaling, which may counterbalance the contractile signals from TXA(2) and mechanical stimuli.</P>

        Chromatin Kinases Act on Transcription Factors and Histone Tails in Regulation of Inducible Transcription

        <P>The inflammatory response requires coordinated activation of both transcription factors and chromatin to induce transcription for defense against pathogens and environmental insults. We sought to elucidate the connections between inflammatory signaling pathways and chromatin through genomic footprinting of kinase activity and unbiased identification of prominent histone phosphorylation events. We identified H3 serine 28 phosphorylation (H3S28ph) as the principal stimulation-dependent histone modification and observed its enrichment at induced genes in mouse macrophages stimulated with bacterial lipopolysaccharide. Using pharmacological and genetic approaches, we identified mitogen-and stress-activated protein kinases (MSKs) as primary mediators of H3S28ph in macrophages. Cell-free transcription assays demonstrated that H3S28ph directly promotes p300/CBP-dependent transcription. Further, MSKs can activate both signal-responsive transcription factors and the chromatin template with additive effects on transcription. Specific inhibition of MSKs in macrophages selectively reduced transcription of stimulation-induced genes. Our results suggest that MSKs incorporate upstream signaling inputs and control multiple downstream regulators of inducible transcription.</P>

        6면 프리즘 렌즈가 장착된 렌즈메터에서 상점의 위치로부터 시험렌즈의 편심의 위치를 알아 낼 수 있는 알고리즘을 개발하였다. 6개의 프리즘에 의해 형성되는 6개의 상점은 시험렌즈의 편심의 극좌표에 의해 변하게 되며, 비틀린 광선 추적으로 계산할 수 있다. 광학 설계 프로그램인 Code V를 사용하여 렌즈메터 광학계를 분석하고, 시험렌즈의 위치에 따른 상점의 위치 변화를 함수로 나타내서 역으로 상점의 위치로부터 시험렌즈의 편심의 위치를 알아낼 수 있는 방법을 개발하였다. We have developed an algorithm to determine the decenter position from the image points in a lensmeter with 6-sided prism lens. The positions of the image points which are formed by six prisms depends on the decenter position of a test lens and can be calculated by skew ray tracing. The optical characteristics of the lens meter was analyzed using the Code V program and the positions of the image points was expressed as a function of the decenter position of the test lens. By minimizing the expectation error, we can determine the decenter position of a test lens from the image positions.

      • TRANSPORT OF SOLAR WIND H <sup>+</sup> AND He <sup>++</sup> IONS ACROSS EARTH’S BOW SHOCK

        <P>We have investigated the dependence of mass, energy, and charge of solar wind ( SW) transport across Earth's bow shock. An examination of 111 crossings during quiet SW in both quasi-perpendicular and quasi-parallel shock regions shows that 64 crossings had various degrees of heating and thermalization of SW. We found 22 crossings where the SW speed was <400 km s(-1). The shock potential of a typical supercritical quasi-perpendicular shock estimated from deceleration of the SW and cutoff energy of electron flat top distribution is similar to 50 Volts. We find that the temperatures of H+ and He++ beams that penetrate the shock can sometimes be nearly the same in the upstream and downstream regions, indicating little or no heating had occurred crossing the bow shock. None of the models predict that the SW can cross the bow shock without heating. Our observations are important constraints for new models of collisionless shocks.</P>

      • FACT complex is required for DNA demethylation at heterochromatin during reproduction in <i>Arabidopsis</i>

        <▼1><P><B>Significance</B></P><P>The chromatin remodeling activities of the FACT (facilitates chromatin transactions) complex are required for many cellular functions, including transcription, DNA replication, and repair. Here, we demonstrate that the two FACT subunits, SSRP1 and SPT16, are also required for genome-wide DNA demethylation and regulation of gene imprinting during <I>Arabidopsis</I> reproduction. Without FACT, <I>Arabidopsis</I> seeds undergo abnormal development and exhibit aberrant DNA hypermethylation, including at imprinting control region loci. We show that FACT associates with the DEMETER (DME) DNA demethylase, facilitating DNA demethylation at over half of DME’s targets, specifically those which reside in heterochromatin. These results provide insight into upstream events in the DNA demethylation pathway and reveal the importance of chromatin remodeling for DNA demethylation during <I>Arabidopsis</I> reproduction.</P></▼1><▼2><P>The DEMETER (DME) DNA glycosylase catalyzes genome-wide DNA demethylation and is required for endosperm genomic imprinting and embryo viability. Targets of DME-mediated DNA demethylation reside in small, euchromatic, AT-rich transposons and at the boundaries of large transposons, but how DME interacts with these diverse chromatin states is unknown. The STRUCTURE SPECIFIC RECOGNITION PROTEIN 1 (SSRP1) subunit of the chromatin remodeler FACT (facilitates chromatin transactions), was previously shown to be involved in the DME-dependent regulation of genomic imprinting in <I>Arabidopsis</I> endosperm. Therefore, to investigate the interaction between DME and chromatin, we focused on the activity of the two FACT subunits, SSRP1 and SUPPRESSOR of TY16 (SPT16), during reproduction in <I>Arabidopsis</I>. We found that FACT colocalizes with nuclear DME in vivo, and that DME has two classes of target sites, the first being euchromatic and accessible to DME, but the second, representing over half of DME targets, requiring the action of FACT for DME-mediated DNA demethylation genome-wide. Our results show that the FACT-dependent DME targets are GC-rich heterochromatin domains with high nucleosome occupancy enriched with H3K9me2 and H3K27me1. Further, we demonstrate that heterochromatin-associated linker histone H1 specifically mediates the requirement for FACT at a subset of DME-target loci. Overall, our results demonstrate that FACT is required for DME targeting by facilitating its access to heterochromatin.</P></▼2>

        Intranasal Adenovirus-Vectored Vaccine for Induction of Long-Lasting Humoral Immunity-Mediated Broad Protection against Influenza in Mice

        <P>Influenza vaccines aimed at inducing antibody (Ab) responses against viral surface hemagglutinin (HA) and neuraminidase (NA) provide sterile immunity to infection with the same subtypes. Vaccines targeting viral conserved determinants shared by the influenza A viruses (IAV) offer heterosubtypic immunity (HSI), a broad protection against different subtypes. We proposed that vaccines targeting both HA and the conserved ectodomain of matrix protein 2 (M2e) would provide protection against infection with the same subtype and also HSI against other subtypes. We report here that single intranasal immunization with a recombinant adenovirus (rAd) vector encoding both HA of H5 virus and M2e (rAdH5/M2e) induced significant HA- and M2e-specific Ab responses, along with protection against heterosubtypic challenge in mice. The protection is superior compared to that induced by rAd vector encoding either HA (rAdH5), or M2e (rAdM2e). While protection against homotypic H5 virus is primarily mediated by virus-neutralizing Abs, the cross-protection is associated with Abs directed to conserved stalk HA and M2e that seem to have an additive effect. Consistently, adoptive transfer of antisera induced by rAdH5/M2e provided the best protection against heterosubtypic challenge compared to that provided by antisera derived from mice immunized with rAdH5 or rAdM2e. These results support the development of rAd-vectored vaccines encoding both H5 and M2e as universal vaccines against different IAV subtypes.</P><P><B>IMPORTANCE</B> Current licensed influenza vaccines provide protection limited to the infection with same virus strains; therefore, the composition of influenza vaccines has to be revised every year. We have developed a new universal influenza vaccine that is highly efficient in induction of long-lasting cross-protection against different influenza virus strains. The cross-protection is associated with a high level of vaccine-induced antibodies against the conserved stalk domain of influenza virus hemagglutinin and the ectodomain of matrix protein. The vaccine could be used to stimulate cross-protective antibodies for the prevention and treatment of influenza with immediate effect for individuals who fail to respond to or receive the vaccine in due time. The vaccine offers a new tool to control influenza outbreaks, including pandemics.</P>

        알코올 발효사료 급여가 한우의 육성성적 및 혈액의 생리적 변화에 미치는 영향

        본 실험은 알코올 발효사료(alcohol 함량; 3.0∼3.2%, lactate 함량; 3.0∼3.5%) 처리가 한우 반추위내 혈액성분 및 비육능력에 미치는 영향을 검토하기 위하여 한우에 비지박 알코올 발효사료(Alcohol-fermented soybean curd dreg : AFS) 및 맥주박 알코올 발효사료(alcohol fermented brewery grain's : AFB)를 급여하여 평균체중이 329㎏인 한우 거세우 13두와 평균체중이 337㎏인 한우 비거세우 20두를 공시하여 증체량, 사료섭취량, 혈액성분, 혈중 알코올 함량을 측정하여 다음과 같은 결과를 얻었다. 거세우에서 비육전기의 일당증체량 및 사료효율은 알코올 발효사료 급여구들이 대조구에 비해 개선되는 경향을 보였으며, 비육후기의 일당증체량은 처리에 따른 유의적인 차이를 나타내지 않았으며, 비육후기의 사료효율은 알코올 발효사료 급여구가 개선되는 경향을 보였다. 비거세우에서 일당증체량 및 사료효율은 알코올 발효사료 급여로 개선되는 경향을 보였으나 유의성은 나타나지 않았으며, 비육 후기에는 개선효과가 없는 것으로 나타났다. 혈중 알코올 농도는 알코올 발효사료 급여구가 시험사료 급여 2시간 후에 유의적으로 높게 나타났으나(p<0.05), 사료급여 4시간째에는 급여전의 수준으로 낮아졌다. BUN 농도는 거세우가 비거세우에 비해 높았으며(14.3 vs 10.97 ㎎/㎗, p<0.05), 혈중 creatinine 농도는 거세우가 비거세우보다 낮았다(1.39 vs 1.49㎎/㎗, p<0.05). 혈중 cholesterol 농도는 전체적으로 체중이 증가함에 따라 증가하는 것으로 나타났고, 혈중 glucose 농도는 알코올 발효사료 급여로 비거세우에서는 감소하는 경향을 보인 반면 거세우에서는 증가하는 것으로 나타났다. 혈중 IP 농도는 알코올 발효사료 급여로 거세우 및 비거세우 모두 증가되었다.(p<0.05). This study was conducted to know the feeding effects of alcohol-fermented feedstuffs (alcohol concentration: 3.0∼3.2 %, lactate: 3.0∼3.5%) on the blood metabolites and fattening ability. Alcohol-fermented soybean curd dreg (AFS) and alcohol fermented brewery grain (AFB) were fed to 13 Korean steers (average body weight, 329㎏) and 20 Korean bulls (average 337㎏ B.W), and body weight gain, feed conversion rate, blood metabolites and blood alcohol concentration were analyzed. The feed conversion rates of steers at both fattening and finishing period were improved by feeding alcohol-fermented feedstuffs. However, improvement of daily body weight gain was observed only at fattening period. In bulls, body weight gains and feed conversion rate were also improved at fattening period (but not at finishing period) by feeding of alcohol-fermented feedstuffs, although a statistical significance was not detected. Blood alcohol concentrations of AFS and AFB group were significantly increased (p<0.05) at 2 hours after feeding, but dropped to the initial level at 4 hours after feeding. Blood urea nitrogen(BUN) concentrations of steers were higher than those of bulls (14.3 vs 10.97 ㎎/㎗), but creatinine was lower in steers than in bulls (1.39 vs 1.49㎎/㎗). Blood cholesterol concentration was generally proportional to the increment of body weight. While the blood glucose content was decreased in bulls by feeding alcohol-feedstuffs, it was increased in steers. Blood inorganic phosphorus(IP) level was increased in both steers and bulls.

        Endocrine Disruptive Potentials in Surface Water Samples from Taihu Lake, Yangtze Delta

        Taihu Lake is a major water source for part of Yangtze Delta, which is one of the most urbanized and economically prosperous areas in China. In last couple of decades, some parts of the lake were highly polluted due to eutrophication. This study analyzed dioxin-like potential and mutagenic potential in surface water samples from Taihu Lake. The samples were prepared by XAD-2 resin procedure. A batch of biological assays, including dioxin-like potential microassay with the rat hepatocyte cell line H411E, and Ames test was employed in the research. Results showed that jour water samples have high content of dioxin-like biological potential, the highest activity TEQ to 2,3,7,8-TCDD was 48 pg/ι in sample 1. The mutagenic effect with reading-frame shifting mechanism was confirmed in 3 of 4 samples. The effective sewage treatment facilities and reliable monitoring surveillance system are urgently needed for this area.

        Extrusion 가공처리 계분사료의 첨가수준이 재래산양의 육성능력, 영양소 소화율 및 체조성에 미치는 영향

        본 시험은 계분의 가축 사료화의 일환으로 계분, 옥수수 및 타피오카를 50:30:20의 비율로 혼합하여 extrusion 가공한 계분사료(EPM)의 급여수준이 반추가축의 증체량, 사료요구율, 영양소 소화율 및 도체성적에 미치는 영향을 구명하기 위하여 평균체중이 10㎏ 내외의 한국재래산양 수컷 15두를 이용하여 EPM을 시험사료중에 각각 0, 10, 20, 30 및 40%씩 첨가 급여하여 6주간 사양시험을 실시하였다. Extrusion 가공처리 계분사료의 수분, 조지방 및 조섬유 함량는 extrusion 가공처리 전보다 각각 10.8, 0.2 및 0.5% 감소하였고, NFE는 1.7% 증가하였다. 사료섭취량은 대조구의 596.1g/day에 비해 EPM 20 및 30% 첨가구에서 각각 620.6 및 698.5g/day로 증가하는 경향을 보였으나, 40% 첨가구에서는 590.7g/day로 감소하였다. 일당 증체량은 대조구에서 119.8g으로 EPM 10, 20 및 30% 첨가구의 96.8, 98.3 및 108.2g/day와 거의 유사한 수준을 보였으나 EPM 40% 첨가구에서는 78.3g/day로 대조구에 비해 유의적으로 낮았다(p<0.05). 사료요구율은 대조구(5.01)에 비하여 EPM 첨가수준이 증가할수록 높아지는 경향을 보였으며, 특히 EPM 40% 첨가구에서는 대조구에 비해 현저하게 높았다(p<0.05). 건물 및 조단백질 소화율은 각 처리간에 큰 차이는 인정되지 않았으나, 조섬유 소화율은 대조구의 63.7%에 비해 EPM 10, 20, 30 및 40% 첨가구에서 각각 65.3, 67.5, 70.4 및 71.8%로 증가하는 경향을 보였다. 조지방 소화율은 EPM 40% 첨가구에서 68.5%로 대조구에 비해 유의적으로 낮았고(p<0.05). 다른 EMP 첨가구에서는 거의 유사한 수준을 나타내었다. 도체율은 대조구의 49.7%에 비해 EPM 10, 20, 30 및 40% 첨가구에서 각각 49.7, 48.3, 47.8 및 45.2로 감소하는 경향을 나타내었다. 산양고기중의 지방 함량은 대조구의 16.3%에 비해 EPM 첨가구 모두에서 유의적으로 감소하였다(p<0.05). This experiment was conducted to investigate the effects of supplementary level of extruded poultry manure, corn and tapioca mixture (EPM) on growth performance, nutrient digestibility and body composition in korean native goats. Total 15 heads of Korean native goats (10 ㎏ B.W.) were randomly assigned into five treatment groups (EPM 10, 20, 30, 40% and control.) and feeding trial was done for six weeks with ad libitum. Protein level of the extrudate with poultry manure was directly proportional to corn supplements rate. NFE and Ca content in EPM also were functional of the tapioca supplements levels. Daily body weight gain was significantly (p<0.05) reduced in EPM 40% group, but no significant differences were observed in other groups. Concentrate feed intake was high in EPM 10 to 30% groups, compared to the control. Feed conversion ratio also was relatively higher in EPM groups than in the control. As EPM level increased, dry matter digestibility was proportionally decreased, but that of crude protein was enhanced. Dressed carcass percentage was significantly (p<0.05) reduced with the increase of EPM level, showing control 49.7, EPM 10% 49.8, EPM 20% 48.3, EPM 30% 47.9 and EPM 40% 45.2, respectively.

      • Broadband omnidirectional light detection in flexible and hierarchical ZnO/Si heterojunction photodiodes

        <P>The development of flexible photodetectors has received great attention for future optoelectronic applications including flexible image sensors, biomedical imaging, and smart, wearable systems. Previously, omnidirectional photodetectors were only achievable by integration of a hemispherical microlens assembly on multiple photodetectors. Herein, a hierarchical photodiode design of ZnO nanowires (NWs) on honeycomb-structured Si (H-Si) membranes is demonstrated to exhibit excellent omnidirectional light-absorption ability and thus maintain high photocurrents over broad spectral ranges (365 to 1,100 nm) for wide incident angles (0A degrees to 70A degrees), which enabled broadband omnidirectional light detection in flexible photodetectors. Furthermore, the stress-relieving honeycomb pattern within the photodiode micromembranes provided photodetectors with excellent mechanical flexibility (10% decrease in photocurrent at a bending radius of 3 mm) and durability (minimal change in photocurrent over 10,000 bending cycles). When employed in semiconductor thin films, the hierarchical NW/honeycomb heterostructure design acts as an efficient platform for various optoelectronic devices requiring mechanical flexibility and broadband omnidirectional light detection.</P>
