Proteus vulgaris K80 lipase was expressed in Escherichia coli BL21 (DE3) cells and immobilized on amine-terminated magnetic microparticles (Mag-MPs). The immobilization yield and activity retention were 84.15% and 7.87%, respectively. A homology model of lipase K80 was constructed using P. mirabilis lipase as the template. Many lysine residues were located on the protein surface, remote from active sites. The biochemical characteristics of immobilized lipase K80 were compared with the soluble free form of lipase K80. The optimum temperature of K80-Mag-MPs was 60℃, which was 20℃ higher than that of the soluble form. K80-Mag-MPs also tended to be more stable than the soluble form at elevated temperatures and a broad range of pH. K80-Mag-MP maintained its stable form at up to 40℃ and in a pH range of 5.0-10.0, whereas soluble K80 maintained its activity up to 35℃ and pH 6.0-10.0. K80-Mag-MPs had broader substrate specificity compared with that of soluble K80. K80-Mag-MPs showed about 80% residual relative activity after five recovery trials. These results indicate the potential benefit of K80-Mag-MPs as a biocatalyst in various industries.
Formation of social scientific fields in Indonesian universities

This study examines the processes of the formation of social scientific fields in Indonesian universities. In contrast to the convergence perspective that is currently dominating the research on university reform, this paper enriches the divergence perspective. In essence, this study argues that individual universities respond differently to university reform and draws on Beckert’s three interrelated social forces and Paradeise and Thoenig’s concept of divergence. It conducts in-depth interviews with 40 social scientists at four state universities across the country and presents the following conclusions. State-led university reform contributes to the creation of the plurality of academic cultures, which, in turn, influences the processes of facilitation and negotiation over resources within university governance. In addition, this plurality promotes the reactions of coupling and decoupling of circles of scientific production and distribution, which respectively bolster and impede the development of social sciences. Top and wannabe circles promote peer-reviewed scientific publications as a universal standard of academic performance, whereas the missionary circle embraces the idea of public engagement as the sole mission of a university. Re-coupling among circles has to become the new rule of the game. In this manner, the quality of individual academics, university teaching, and public engagement will be considerably improved.

The purpose of this comprehensive research is to observe the impact of scintillator crystal type on entire detection process. For this aim, MCNPX (version 2.6.0) is used for designing of a physical plastic scintillation detector available in our laboratory. The modelled detector structure is validated using previous studies in the literature. Next, different types of plastic scintillation crystals were assessed in the same geometry. Several fundamental detector properties are determined for six different plastic scintillation crystals. Additionally, the deposited energy quantities were computed using the MCNPX code. Although six scintillation crystals have comparable compositions, the findings clearly indicate that the crystal composed of PVT 80% þ PPO 20% has superior counting and detecting characteristics when compared to the other crystals investigated. Moreover, it is observed that the highest deposited energy amount, which is a result of the highest collision number in the crystal volume, corresponds to a PVT 80% þ PPO 20% crystal. Despite the fact that plastic detector crystals have similar chemical structures, this study found that performing advanced Monte Carlo simulations on the detection discrepancies within the structures can aid in the development of the most effective spectroscopy procedures by ensuring maximum efficiency prior to and during use.

Vitamin D has been known for its anti-inflammatory properties. Extracts derived from Lentinula edodes (Shiitake) edible mushroom exert an anti-inflammatory effect. These extracts contain high levels of ergosterol, which converts into ergocalciferol (vitamin D2) following exposure to ultraviolet light, followed by absorption and hydroxylation into the active form 25-hydroxyvitamin D [25(OH)D]. To determine the anti-inflammatory effect of overexpression of vitamin D in edible mushrooms, L. edodes mushrooms were exposed to ultraviolet-B light, freeze-dried, followed by measurement of vitamin D2 contents, in their dry weight. C57B1/6 mice were orally treated with vitamin D2-enriched or nonenriched mushroom extract prior and during concanavalin A-immune-mediated liver injury. Exposure to ultraviolet light increased vitamin D2 content in Shiitake edible mushrooms. Following feeding of vitamin D-enriched mushroom extracts to mice with immune-mediated hepatitis, a significant decrease in liver damage was noted. This was shown by a decrease in alanine aminotransferase and aspartate aminotransferase serum levels, a decrease in proportion of mice with severe liver injury, and by improvement in liver histology. These effects were associated with a decrease in serum interferon gamma levels. A synergistic effect was noted between the anti-inflammatory effect of the mushroom extracts and that of vitamin D. Oral administration of vitamin D-enriched L. edodes edible mushroom exerts a synergistic anti-inflammatory effect in the immunemediated hepatitis. The data support its potential use as safe immunomodulatory adjuvant for the treatment of HCV and nonalcoholic steatohepatitis.

Chikungunya fever is an arboviral disease caused by the Chikungunya virus (CHIKV). The disease has similar clinical manifestations with other acute febrile illnesses which complicates differential diagnosis in low-resource settings. We aimed to develop a rapid test for CHIKV detection based on the nucleic acid lateral flow immunoassay technology. The system consists of a primer set that recognizes the E1 region of the CHIKV genome and test strips in an enclosed cassette which are used to detect amplicons labeled with FITC/biotin. Amplification of the viral genome was done using open-source PCR, a low-cost open-source thermal cycler. Assay performance was evaluated using a panel of RNA isolated from patients’ blood with confirmed CHIKV (n = 8) and dengue virus (n = 20) infection. The open-source PCR-NALFIA platform had a limit of detection of 10 RNA copies/ml. The assay had a sensitivity and specificity of 100% (95% CI: 67.56% - 100%) and 100% (95% CI: 83.89% - 100%), respectively, compared to reference standards of any positive virus culture on C6/36 cell lines and/or qRT-PCR. Further evaluation of its performance using a larger sample size may provide important data to extend its usefulness, especially its utilization in the peripheral healthcare facilities with scarce resources and outbreak situations.
