        유화조건이 홍화적색소의 물성에 미치는 영향

        상기 실험으로부터 얻어진 결과를 요약하면 다음과 같다. 1. Hunter-value를 측정한 결과 hunter L-value 즉, 백색도에 있어서는 glycerin+carthamin+locithin의 경우가 실온저장 및, 열악조건 그리고 광조사시에 있어서 모두 8 시간이 경과하면서부터는 급격하게 상승하는 모습을 보였다. 2. 실온하에서 hunter a-value 즉, 적색도에 있어서는 carthamin 첨가구가, b-value 즉, 황색도에 있어서는 paprika 첨가구가 더 강하게 나타났다. 열악조건에서는 실온저장의 경우와 다소 다른 양상을 보여주었는데. D.W+carthamin+lecithin의 경우에 있어서도 hunter L-value가 낮은 값을 나타냈으며, hunter a-value의 경우 soybean oil+paprika에서도 높은값을 보여주었다. 3. 유변학적 특성에 대한 실험에서 비교적 고점도를 가지고 있는 glycerin을 base material로하였을 때는 이에 비해 저점도라고 할 수 있는 soybean oil이나 증류수가 쓰여졌을 때보다 spindle의 rpm에 따른 viscosity나 shear rate 및 shear stress등의 같이 월등히 높은 값을 나타냈다. 그리고, 일반적으로 spindle의 rpm에 대한 shear rate 및 shear stress의 값은 비례적으로 증가하는 경향을 나타냈다. 유화제의 사용여부에 있어서는 lecithin을 사용했을 경우. 사용하지 않았을 때보다 높은 점도를 나타냈고, 동일한 조건에서 carthamin보다는 paprika가 두 배 정도 더 높은 점도를 나타냈다. 4. soybean oil을 사용한 실험구에 있어서는lecithin의 첨가유무가 점도에 아무런 영향을 미치지 않는 것으로 나타났으나, glycerin을 사용한 실험구에 있어서는 유화제인 lecithin을 병행했을 경우, 특이하게도 오히려 paprika에서보다 carthamin에서 더 높은 점도를 나타냈다. 5. Shear stress의 경우 soybean oil 첨가구와 glycerin 첨가구가 현저한 차이를 보이며 증가한 반면, shear rate에 있어서는 두 가지 경우모두 거의 같은 경향을 나타낸 점이 특이했다. This study was peformed to evaluate the food technological properties according to different emulsion state of carthamus red pigment. For making emulsion, lecithin was used as an emulsifier and polyglycerol monooleate(PGMO) and polyoxyethylene sorbitan monooleate(Tween 80) were used as an assistant, and glycerin, distilled water and soybean oil were used as base materials respectively. Paprika stock solution was used for comparing carthamin on the rheological properties. The results were described as following; 1. Hunter L-value was not drastically increased until passed by 8 hours for glycerin, carthamin, and lecithin mixed sample. 2. Hunter a-value was higher at carthamin added sample than others, and b-value was higher to paprika added sample than others. 3. The viscosity, shear rate and shear stress levels in which glycerin was used as base material were higher than soybean oil or distilled water. 4. In which soybean oil was used as base material, lecithin was not affected on the rheological properties. But, in which glycerin was used, the lecithin was higher affected on carthamin than paprika. 5. The value of shear stress was increased both carthamin and soybean oil. However, that of shear rate was shown similar trends.

      • 완두 세포질 Fructose-1,6-bisphosphatase의 광의존적 발현

        Cytoplasmic fructose-1,6-bisphosphatase from pea leaves was purified and characterized. Monomeric molecular weight of the purified protein was estimated as 37,000 by SDS polyacrylamide gel electrophoresis. The enzyme was active at neutral pH (pH 6.6). The chloroplasmic fructose-l,6-bisphosphatase, isoenzyme of this enzyme, is known to be active at alkaline pH (pH 8.8). The purified enzyme was relatively stable against heat. The activity of the enzyme was not affected by reducing agent such as dithiothreitol and the enzyme was inhibited by AMP and fructose-2,6-bisphosphate as all other gluconeogenic fructose-l,6-bisphosphatase from various species. The enzyme activity of cytoplasmic fructose-1,6-bisphosphatase was gradually increased by light illumination and decreased by dark treatment. Immunoblot analysis suggests that the expression of the enzyme is stimulated by light. Light pulse experiments with blue (420nm), and red (660 nm)/far-red (730 nm) light were performed to analyze the photoreceptors related to light signal transduction and photomorphogenesis. The cytoplasmic enzyme activity was almost insensitive to blue or red/far-red light pulse, suggesting that the cytoplasmic enzyme is not directly regulated by light signal. Thus the activity and/or expression of the cytoplasmic enzyme may be closely related to plant growth and development accompanied by continuous light illumination.

        Gastrointestinal tumors(GIST) are rare group of neoplasias of the gastrointestinal track, which originate from or differentiate into interstitial cells of Cajal (ICC). GIST traditionally have been designated as smooth muscle tumor. However, with increasingly accurate analytic tools, immunohistochemistry and ultrastructure, most investigators were able to demonstrate that ICC, a complex cellular network postulated to act as pacemaker cells of the gastrointestinal track, which exhibit both myoid and neural features, could be a candidate for tumor histogenesis. Most ICC are only c-kit-positive, however a subset are positive for both c-kit and CD34. Most GISTs are asymptomatic and are thus usually discovered incidentally during routine check, at operation of other organs. When symptoms occur, GISTs usually present with gastrointestinal bleeding due to ulceration of the overlying gastric mucosa. Bleeding may be massive, leading to hematemesis and melena. Recently, we experienced a large sized(5×5×5 cm) GIST on the upper body of the stomach, which caused massive upper gastrointestinal bleeding in a 61-year-old man and was treated by local resection with adequate safety margin about 2 cm.

      • 잇꽃 Carthamin의 초임계 추출 및 농축

        Supercritical fluids (SCF) are useful substances that are recently used in bioseparation process. Extraction of carthamin from safflower using supercritical carbon dioxide were examined at various conditions of temperature (35∼40℃), pressure (3000∼5000 psi) and CO₂ flow rate (900∼1200 m/hr). SCF was less effective than solvent methods in carthamin extraction from safflower. Concentration of carthamin was 50% more efficent at the use of alginate beads with 35 g cellulose per column loadings compared to the control (15g cellulose per column loadings).

      • 대두(Glycine max L.) phy A cDNA의 선별과 Cloning

        cDNA for phytochrome A gene was screened with a cDNA library constructed from etiolated soybean seedlings. A genomic subclone (pB41, 1.5 kb) containing chromophore bearing region of phytochrome A was used as a probe. All the positive clones analyzed show identical restriction maps suggesting that phy A gene presents as a single copy. A clone with full coding region (3.6 kb) was subcloned and sequenced. The deduced amino acid sequence analysis of the coding regions suggest that pB44SC is categorized as phy A. The pB44SC is 62~63 % identical with monocot phy A and 74-84% identical with dicot phy A. It is only 49-51% identical with phy B and phy C.

        방선균 분리주가 생산하는 Phospholipase C 저해물질인 MT2617-2B의 분리 및 특성

        방선균 분리주 MT2617-2의 배양액으로 부터 phospholipase C (PLC) 저해물질인 MT2617-2B를 n-butanol 추출 및 column chromatography 법을 이용하여 분리하였다. MT2617-2B는 IR ^13C- 및 ^1H-NMR 그리고 ESI-MS에 의한 구조분석 결과, 한 개의 hemiketal ring, polyhydroxyl 및polymethyl groups으로 구성되었으며 side chain으로 한 개씩의 malonate 및 guanidine group을 가지는 분자량 1057의 macrolide 화합물이었다. 따라서, MT2617-2B를 기존의 macrolide 항생제인 copiamycin 및 niphithricin A로 동정하였다. 한편, MT2617-2B는 methanol 용액에서 실온에서 방치하였을 때 도일한 분자량을 가진 두 개의 이성질체를 생성하였다. PLC γ1과 -β1에 대해 각각 25 및 50㎍/㎖의 IC_50 값을 가지며, Staphylococcus aureus 와 Candida albicans에 대해서는 항균활성을 나타내지만 Escherichia coli에는 나타내지 않았다. A phospholipase C (PLC) inhibitor (MT2617-2B) was isolated from the culture broth of actionmycetes isolate MT2617-2 by the extraction with n-butanol and column chromatographic techniques. The molecular weight of the inhibitor was 1057, by the spectroscopic analyses of IR ^13C- and ^1H-NMR and ESI-MS. The chemical structure of MT2617-2B was found to be a macrolide compound consisted of a hemiketal ring, polyhydroxyl and polymethyl groups, which had a malonate and guanidine group as its side chain. MT2617-2B produced its two isomers having the same molecular weight by standing in methanol solution at room temperature. Therefore, MT2617-2B was identified as copiamycin and niphithricin A, macrolide antibiotics. The values of IC_50 against PLC γ1 and PLC-β1 were 25 and 50㎍/㎖, respectively. MT2617-2B had antimicrobial activities against Staphylococcus aureus and Candida albicans, but not against Escherichia coli.

      • 급성심근경색후 발생한 심실중격결손 2례

        Ventricular septal defect remains an infrequent but devastating complication of acute MI. Approximately 1∼3% of cases of acute myocardial infarction are complicated by ventricular septal rupture in the prethrombolytic era and the incidence has decreased to 0.2% with reperfusion therapy. This mechanical complication usually occurs within the first 10 to 14 days when necrotic tissue is most abundant and the collateral coronary circulation is not well developed. This lesion is generally associated with complete coronary obstruction rather than severe stenosis. Rupture of the ventricular septum is a severe mechanical complication of acute myocardial infarction, usually resulting in death unless surgical repair is performed. The bad prognosis of this event within the first 2 weeks indicates the need for early surgical rapair. This complication is more frequent after the first acute myocardial infarction in the elderly and secondary to a transmural myocardial infarction. We report two cases of ventricular septal rupture after acute myocardial infarction.

      • 옥상정원의 이용실태 조사 및 활성화에 관한 연구(1)

        Nowadays we globally recognize the importance of the environmental preservation and the environmental energy. The developed countries have already made efforts to realize the saving of energy and the technology based on environment-friendly viewpoint. 'Roof garden' means the restoration of soil eco-system destroyed by rash urbanization and construction. It can also offer the resting place to people in the city. Now it is considered as the realistic way that can solve the serious problems like heat island, pollution and flood in the city fundamentally. In this research, we found out that the following results were obtained: 1) Roof garden is used by users in various objects. Stage, meeting, event, date etc. 2) Users want to design roof garden that have various space features: open and close space, wide and small space etc. 3) The group composed from one to five or more played roof garden. 4) The roof garden aboved have similar facilities excepted one or two. Vaitalization in roof garden remains as a matter to be discussed further.

      • 제7차 교육과정에 의한 초등학교 실과 교과서 삽화 분석

        This study was to suggest the desirable direction of illustrations in the textbook by analyzing them based an the 7th Practical Arts curriculum. There were 624 and 565 in textbooks of 5th and 6th grade. 62.9% of the illustrations were pictures in the both of 5th and 6th textbooks(respectively 65.9% and 61.8% in 5th and 6th). 89.2% of them were appropriate for learning contents. The functions of them were mostly the training guides and materials. The distribution and area of them was almost the Same in each chapter. That gradually makes students recede from the learning. There was the gender discrimination in the textbook.
