Sensory Characteristics and Consumer Acceptability of Various Green Teas

The green tea market is rapidly growing and identifying the driving factors of consumers’ liking for the green tea is important in the tea industries. The objectives of this study were to investigate the effects of manufacturing conditions of the green tea on its sensory characteristics, to elucidate its relationship with the consumer liking. A descriptive analysis and consumer acceptability test were conducted for various green tea samples. The samples differed with regard to the source of the tea, the amino acid content, and the processing methods including the roasting temperature. Partial least square regressions (PLS-R) were performed to establish the relationship between the descriptive data and the consumer acceptability data. The PLS-R results showed that the majority of the consumers liked a green tea which has a stronger ‘sweet taste’ and roasting-related flavors such as ‘roasted barley’ and ‘burnt leaf’. Such sensory characteristics were produced when a sample made of tea leaves mixed with the tea stem was roasted at a high temperature (250℃) in this study.

This study was focused on the effect of the altitude on the geographical variations of germination characteristics in the populations of Plantago asiatica L. distribute in Mt. Chiri. There was a significant difference among the 14 groups in the phenological pattern in relation to altitudes. When the altitude becomes higher, the thermal time which was required for 10 to 80% germination rate showed higher and wider distribution. On the other hand, the germination response of increasing temperature (IT) and secreasing temperature (DT) regime was classified into 3 group. The first group was the spering germination type. This group showed that the IT regime hadhigher germination rate than that of the DT regime, and was distributed in Macheon(300m) and Packmudong (500m). The second was the spring-fall germination type which was distributed in Hadong (900m) and Saemt대 (1100m). This group also showed higher germination rate in the IT regime, but the difference of the germination rate between IT and DT regime was less than that in the first group (the spring type). The third group was the early fall germination type which was found in the Nogodan (1507m), Changetomok (1750m) and Cheonwangbong (1915m). The germination rate of this group showed almost 100% similarity between IT and DT regime. These data suggested that the geographical varations of germination characteristics within species was an important ecological strategy for the survival from severe environmental conditions.

Alcaligenes xylosoxydans strain SMN3 capable of utilizing biphenyl grew not only on phenol, and benzoate, but also on salicylate. Catabolisms of biphenyl and salicylate appear to be interrelated since benzoate is a common metabolic intermediate of these compounds. Enzyme levels in the excatechol 2. 3-dioxygenas which is meta-cleavage enzyme of catechol, but did not induce catechol 1, 2-dioxygenase. All the oxidative enzymes of biphenyl and 2, 3,-dihydroxybiphenyl (23DHBP) were induced when the cells were grown on biphenyl and salicylate, respectively. Biphenyl and salicylate could be a good inducer in the oxidation of biphenyl and 2, 3-dihydroxybiphenyl. The two enzymes for the degradation of biphenyl and salicylate were induced after growth on either biphenyl or salicylate, suggesting the presence of a common regulatory element. However, benzoate could not induce the enzymes responsible for the oxidation of these compounds. Biphenyl and salicylate were good inducers for indigo formation due to the activity of biphenyl dioxygenase. These results suggested that indole oxidation is a property of bacterial dioxygenase that form cis-dihydrodiols from aromatic hydrocarbon including biphenyl.
Expression of Human Lactoferrin Gene in Transgenic Rice (Oryza sativa L.)

Lactoferrin is an 80-kDa iron-binding glycoprotein known to exert many biological activities, such as facilitating iron absorption and having antimicrobial and anti-inflammatory effects. Rice can be a useful target for edible food plants to introduce human lactoferrin, because it has lower allergenicity and is likely to be safer than microorganisms or transgenic animals. A cDNA fragment encoding human lactoferrin (HLF) driven by the maize polyubiquitin promoter, along with herbicide resistance gene (bar) driven by CaMV 35S promoter, was introduced into rice (Oryza sativa L. cv. Dong Jin) using the Agrobacterium -mediated transformation system. Putative transformants were initially selected on the medium containing bialaphos. The stable integration of the bar and HLF genes into transgenic rice plants was further confirmed through polymerase chain reaction (PCR) and Southern blot analyses. The expression of the full length HLF protein from various tissues such as grains and young leaves of transgenic rice was verified by Western blot analysis. Analysis of progeny also demonstrated that introduced genes were stably inherited to the next generation at the Mendelian fashion.
Isolation of a Marine Bacterium Capable of Biodegrading Poly(butylene succinate)

We developed a poly(butylene succinate) (PBS) indicator plate and isolated a marine bacterial colony capable of biodegrading PBS based on the appearance of a clear zone. Growth of the PBS-2 isolate was observed over 4 days of culture at $37^{\circ}C$ in PBS-tryptone basal liquid medium, but not in PBS-deprived control medium. The PBS-2 isolate was named Paenibacillus sp. PBS-2 based on 16S rDNA gene sequencing. The PBS-biodegrading marine bacterium isolated in this study will contribute to the effective management of PBS waste problems in marine environments.
