Lee, Youngjoo,Jeon, Jae Hyun,Goh, Sung-Ho,Roh, Hanseong,Yun, Ji-Young,Kwon, Nak-Jung,Choi, Jin Ho,Yang, Hee Chul,Kim, Moon Soo,Lee, Jong Mog,Lee, Geon Kook,Han, Ji-Youn Elsevier 2019 Lung cancer Vol.136 No.-

Abstract

Objectives

Accumulating evidence reveals the association between the risk of never-smoker lung cancer and family history of cancer. However, the clinicogenomic effect of family history of cancer in never-smoker lung cancer remains unknown.

Material and methods

We screened 3,241 lung cancer patients who (a) underwent curative resection at National Cancer Center (Goyang, Korea) between 2001–2014, and (b) completed a pre-designed interview about family/smoking history at the time of diagnosis and identified 604 female never smoker lung adenocarcinoma. A positive family history of cancer [categorized as pulmonary cancer (FH-PC) or non-pulmonary cancer (FH-NPC)] was defined as a self-reported history of cancer in first-degree relatives. Survival data were followed up until January 2017. Multiplexed targeted next-generation sequencing was performed for genetic profiling.

Results

Of 604 patients, 29.1% (n = 176) had a FH, including 132 (21.9%) with FH-NPC and 44 (7.3%) with FH-PC. Patients with the FH-NPC had a higher proportion of young patients (≤45 years) than those without the FH-NPC (FH-NPC, FH-PC, and no FH; 13.6%, 2.3%, and 8.2%, respectively; P = 0.032). Patients with the FH-NPC had an increased risk of recurrence (hazard ratio [HR]: 1.90; 95% confidence interval [CI]: 1.40–2.56; P<0.001) and death (HR: 1.67; 95% CI: 1.18–2.37; P=0.004). In contrast, the FH-PC had no prognostic effect on recurrence (HR: 1.23; 95% CI: 0.71–2.15; P = 0.456) and death (HR: 0.93; 95% CI: 0.45–1.91; P=0.838). Among three driver oncogene alterations, EGFR mutation was significantly associated with the FH-PC (53.8%, 84.1%, and 65.8%, respectively; P = 0.016), ALK/ROS1/RET fusions was significantly associated with the FH-NPC (13.7%, 0.0%, and 5.0%, respectively; P = 0.004), but KRAS mutation was not associated with any type of the FH (13.8% vs. 6.0% vs. 7.8%, respectively; P = 0.288).

Conclusion

The type of family history of cancer was associated with distinct clinocogenomic subtypes and prognosis of never-smoker lung adenocarcinoma.

Highlights

Family history of cancer is related to distinct subtypes of never smoker lung cancer.

ALK/ROS1/RET fusions are enriched in patients with family history of nonlung cancer.

EGFR mutations are enriched in patients with family history of lung cancer.

Family history of nonlung cancer is associated with poor prognosis after operation.

Methicillin-resistant Staphylococcus aureus (MRSA) is spreading worldwide, emphasizing the need to search for new antibiotics. The anti-MRSA activities of gallic acid-grafted-chitosans (GA-g-chitosans) were investigated against 2 MRSA standards and 10 MRSA clinical isolates by determining the minimum inhibitory concentrations (MICs). GA-g-chitosan (I), which has the highest gallic acid content, exhibited the strongest anti-MRSA activities, with MICs of 32-64 μg/mL. A time-kill investigation revealed that GA-g-chitosan (I) exhibited a bactericidal effect at twice the MIC, also demonstrating good thermal and pH stability. Investigation of cell envelope integrity showed the release of intracellular components with an increasing absorbance value at 260 nm, indicating cell envelope damage caused by the GA-g-chitosan (I), which was further confirmed by transmission electron microscopy. When GA-g-chitosans were combined with β-lactams, including ampicillin and penicillin, synergistic effects were observed on the 2 standard MRSA strains and on the 10 clinical isolates, with fractional inhibitory indices ranging from 0.125 to 0.625. In the time-kill dynamic confirmation test, synergistic bactericidal effects were observed for the combinations of GA-g-chitosans with β-lactams, and over 4.0 log CFU/mL reductions were observed after 24 h when combination treatment was used. These results may prove GA-g-chitosans to be a potent agent when combined with ampicillin and penicillin for the elimination of MRSA.

Poncirus trifoliata was evaluated for antioxidant potential by several radical-scavenging assays, including 1,1-diphenyl-2-picrylhydrazyl, superoxide, hydroxyl, and alkyl radicals, using electron spin resonance technique. P. trifoliata extracts were prepared by using water and methanol, partitioning with different organic solvents. Among the tested samples, water extract and dichloromethane fraction evidenced relatively higher scavenging activities than those of other samples on reactive radicals and have high amount of antioxidant component such as flavonoids (8.5±0.3 and 8.3±0.2 mg rutin equivalents/g dry weight basis) and proanthocyanidin (2.6±0.3 and 2.8±0.5 mg catechin equivalents/g dry weight basis), respectively. Moreover, the water extract showed significantly high enhancement of cell viability against hydrogen peroxide-induced oxidative damage as well as reduction in the hydrogen peroxide-induced intracellular reactive oxygen species on CCL 13 cell line. These results indicate that P. trifoliate exerts various antioxidant effects against several reactive radicals and hydrogen peroxide-mediated cell damage. Thus, P. trifoliate could be a potential therapeutic modality for the treatment or prevention of several diseases associated with oxidative stress.

This study was aimed at making a retrospective observation of the morbidity rate of bovine tuberculosis with tuberculin reactors through the effects on the expanded program of test and slaughter for eradication and management among dairy cattle in Korea, from 1965 to 1997. The morbidity rate of tuberculin reactors during the period varied from 14.9 to 669.3 per 100,000 dairy population, however, those tended to decreased from 1975 to 1990, but the rate of reactor were trended to increased after 1991. In conclusion, expanded program of test and slaughter reduced the incidence cases and morbidity rates of bovine tuberculosis with tuberculin reactor available for management of dairy cattle in Korea, when the tuberculin of HCSM and PPD together used after 1973. Finally, it is our hope that these information could be use for reference in further study of the field of veterinary public health and preventive medicine.

The fruits of Cornus kousa Burg. were extracted with 80% aqueous MeOH, and the concentrated extract partitioned with EtOAc, n-BuOH and H$_2$O. Six lignans were isolated from the EtOAc fraction through repeated silica gel, ODS and Sephadex LH-20 column chromatographies. From the physico-chemical data, including NMR, MS and IR, the chemical structures of the compounds were determined to be (+)-pinoresinol (1), (-)-balanophonin (2), (+)-laricresinol(3), erythro-guaiacylglycerol-${\beta}$-coniferyl aldehyde ether (4), threo-guaiacylglycerol-${\beta}$-coniferyl aldehyde ether (5) and dihydrodehydrodiconiferyl alcohol (6), which were isolated for the first time from this plant. Most of these compounds showed cytotoxicity against human colon carci-noma (HCT-116) and human hepatocellular carcinoma (HepG2) cell lines in vitro, with IC$_{50}$ values ranging from 19.1 to 71.3 ${\mu}$g/mL.

The objective of this study was to select an effective microbial strain to improve the functional qualities of Eisenia bicyclis water extract by fermentation. For this purpose, several microorganisms isolated from traditional Korean fermented foods were inoculated and cultivated in E. bicyclis water extract. Ultimately, yeast strain YM-1 was selected for further study based on its total phenolic compound (TP) content and antioxidant activity, which were enhanced by microbial fermentation. The extract fermented by YM-1 exhibited a superior TP content and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity compared to extracts fermented by other microbes. The highest TP content and DPPH radical scavenging activity were observed after one day of YM-1 fermentation. Yeast strain YM-1 was identified as Candida utilis based on an analysis of its physiological characteristics. During fermentation of the extract by C. utilis YM-1, no significant difference was observed in the proximate composition, including moisture, crude lipid, crude protein, and crude ash. Fermentation by C. utilis YM-1 resulted in enhanced biological activity, including increases in the TP content and antioxidant activity. Thus, fermentation by C. utilis YM-1 is an attractive strategy for developing value-added food ingredients.
