        Protective Effect of Heme Oxygenase-1 on High Glucose-Induced Pancreatic β-Cell Injury

        <P><B>Background</B></P><P>Glucose toxicity that is caused by chronic exposure to a high glucose concentration leads to islet dysfunction and induces apoptosis in pancreatic β-cells. Heme oxygenase-1 (HO-1) has been identified as an anti-apoptotic and cytoprotective gene. The purpose of this study is to investigate whether HO-1 up-regulation when using metalloprotophyrin (cobalt protoporphyrin, CoPP) could protect pancreatic β-cells from high glucose-induced apoptosis.</P><P><B>Methods</B></P><P>Reverse transcription-polymerase chain reaction was performed to analyze the CoPP-induced mRNA expression of HO-1. Cell viability of INS-1 cells cultured in the presence of CoPP was examined by acridine orange/propidium iodide staining. The generation of intracellular reactive oxygen species (ROS) was measured using flow cytometry. Glucose stimulated insulin secretion (GSIS) was determined following incubation with CoPP in different glucose concentrations.</P><P><B>Results</B></P><P>CoPP increased HO-1 mRNA expression in both a dose- and time-dependent manner. Overexpression of HO-1 inhibited caspase-3, and the number of dead cells in the presence of CoPP was significantly decreased when exposed to high glucose conditions (HG). CoPP also decreased the generation of intracellular ROS by 50% during 72 hours of culture with HG. However, decreased GSIS was not recovered even in the presence of CoPP.</P><P><B>Conclusion</B></P><P>Our data suggest that CoPP-induced HO-1 up-regulation results in protection from high glucose-induced apoptosis in INS-1 cells; however, glucose stimulated insulin secretion is not restored.</P>

      • Curcumin protects retinal pigment epithelial cells against oxidative stress via induction of heme oxygenase-1 expression and reduction of reactive oxygen

        <P><B>Purpose</B></P><P>To determine whether curcumin induces expression of the defensive enzyme heme oxygenase-1 (HO-1) and protects cells against oxidative stress in cultured human retinal pigment epithelial cells.</P><P><B>Methods</B></P><P>Effective concentrations and toxicities of curcumin were determined after 3 h of curcumin treatment with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Confluent human retinal pigment epithelium cell lines (ARPE-19) were preincubated with curcumin and oxidatively challenged with H<SUB>2</SUB>O<SUB>2</SUB>. HO-1 expression was determined with western blot analysis. To confirm the protective role of HO-1 in oxidative stress, small interfering RNA (siRNA) against HO-1 or inhibitor of HO-1 was treated with curcumin in retinal pigment epithelium cells. Intracellular levels of reactive oxygen species (ROS) were measured with flow cytometry and confocal microscopy. Apoptosis was evaluated with Annexin V-fluoroscein isothiocyanate staining.</P><P><B>Results</B></P><P>Curcumin had little cytotoxicity at concentrations less than 30 μM, and HO-1 expression was the highest at the 15 μM concentration. At this concentration, curcumin also increased the cytoprotective effect against the oxidative stress of H<SUB>2</SUB>O<SUB>2</SUB> through the reduction of ROS levels in human retinal pigment epithelial cells. Curcumin’s effect on the reduction of ROS was mediated by the increase in HO-1 expression.</P><P><B>Conclusions</B></P><P>Curcumin upregulated the oxidative stress defense enzyme HO-1 and may protect human retinal pigment epithelial cells against oxidative stress by reducing ROS levels.</P>

The purpose of this study is development of posture evaluation and Range of Motion(ROM) system by using digital vision analysis method. The results of this study are as follows. First, Scoliosis evaluation through this research measurement system represent 3mm error in 7 cervical point and deepest lumbar point, 0.7mm error in other point. This mean this research measurement system have a reliability for scoliosis evaluation. Second, for spine line evaluation on high fat subject, we need reconstrection spine line after mesurement for fat thickness in 7 cervical point and deepest lumbar point. Third, In pedioscope error test, it present 0.01848cm in X axis and 0.01757cm in Y axis. This results mean pedioscope have a reliability foot evaluation. Forth, Posture evaluation and Range of Motion measurement system by using digital vision analysis method can fast measure in range of motion and foot evaluation and posture. therefore we can expect this system application in young people posture clinic center and hospital and so on.

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

      • Antioxidant and cytoprotective effects of morin against hydrogen peroxide-induced oxidative stress are associated with the induction of Nrf-2-mediated HO-1 expression in V79-4 Chinese hamster lung fibroblasts

        <P>Natural phytochemicals of plant origin, including flavonoids, have been found to be potent antioxidants providing beneficial effects against oxidative stress-related diseases. The present study was carried out to investigate the antioxidant properties of morin, a flavonoid originally isolated from the flowering plants of the Moraceae family. Superoxide dismutase (SOD)-like activity and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) radical scavenging activity were determined. We also investigated the cytoprotective effects of morin against hydrogen peroxide (H2O2)-induced DNA damage and apoptosis in V79-4 Chinese hamster lung fibroblasts. Our results demonstrated that morin had strong scavenging effects against ABTS' radicals with enhanced SOD activity, which varied in a dose-dependent manner. Morin was found to reduce H2O2-induced intracellular reactive oxygen species generation and nuclear DNA damage, and it recovered cell viability damaged by H2O2 via inhibition of mitochondrial dysfunction-mediated apoptosis. Notably, the treatment of V79-4 cells with morin markedly enhanced the expression of heme oxygenase-1 (HO-1) but not quinone oxidoreductase-1, which was associated with the increased expression and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the downregulation of Kelch-like ECH-associated protein 1 expression. Based on our findings, we conclude that morin effectively ameliorated oxidative stress-induced DNA damage through intrinsic free radical scavenging activity and activation of the Nrf2/HO-1 pathway.</P>

        Sarcomatoid Carcinoma Arising from Mature Cystic Teratoma

        Malignant transformation of mature cystic teratoma (MCT) is rare. Sarcomatoid carcinoma is a neoplasm comprising malignant mesenchymal cells and a conventional carcinomatous area. Here, we report on a case of sarcomatoid carcinoma arising from an MCT in the left ovary of a 45-year-old female. A unilocular cyst consistent with MCT was observed; however, a nodule within the cyst was confirmed from the resected ovary. Microscopically, the nodule showed both squamous cell carcinoma and pleomorphic sarcomatous components admixing with each other. Lining epithelial cells at the periphery of the main tumor showed squamous metaplasia. When a sarcomatous component is observed in the ovary tumor, it is important to find a squamous cell component, either benign or malignant.

        한 도시지역 저소득층 주민의 정신건강수준

        연구목적: 본 연구에서는 A시 지역사회 정신보건사업을 시행하면서, 지역사회 정신보건사업의 일차적 관리대상인 저소득층 주민들의 정신건강수준을 파악하고, 정신건강수준에 영향을 미치는 요인을 알아봄으로써 지역사회에 정신질환자들이 통합되어지기 위해서 시행해야 하는 사업계획을 수립하는데 기초자료로 활용함에 그 목적이 있다. 대상 및 방법: 본 연구의 대상은 A시에 거주하는 20-59세의 저소득층 주민들 536명이며, 조사도구로는 정신건강수준을 집단적으로 선별 검사할 수 있는 조사도구인 GHQ와 자아존중감 정도를 측정하는 SES도구를 이용하여 설문지를 통한 면접조사를 실시하였다. 대상인구의 인구. 사회학적 특성과 자아존중감 정도에 따른 정신건강수준을 알아보았고, 또한 정신건강수준을 세 가지 유형(전반적 정신건강수준, 사회생활 적응 정도, 불안·우울감 정도)으로 나누어 분석하였다. 결 과: 1) 대상인구의 인구학적 특성에 따른 정신건강수준을 살펴보면 여성보다 남성이, 연령이 높을수록, 배우자 없이 혼자 사는 계층이, 그리고 신체적 건강상태가 나쁠수록 전반적 정신건강수준과 사회생활 적응 정도가 낮았으며, 불안·우울감 정도도 높았다. 2) 대상인구의 사회. 경제적 특성에 따른 정신건강수준을 살펴보면 무직이나 단순근로직이, 교육수준이 낮을수록 월세나 임대로 사는 저소득계층에서 전반적 정신건강수준과 사회생활 적응 정도가 낮았으며, 불안·우울감 정도도 높은 것으로 조사되었다. 3) 자아존중감 정도가 높을수록 전반적 정신건강수준과 사회생활 적응 정도가 높았으며, 불안·우울감 정도는 낮았다(p<0.01). 4) 정신건강수준에 영향을 미치는 요인으로는 자아존중감과 신체적 건강상태, 연령을 들 수 있는데 이 가운데 자아존중감이 가장 많은 영향을 주었으며, 그리고 자아존중감이 높은 상태에서 신체적 건강상태가 좋을수록, 또한 연령이 증가할수록 전반적인 정신건강수준이 유의하게 증가되는 것으로 조사되었다. 5) 정신건강 위험집단의 특성에 대해 비위험집단을 비교 분석한 결과 위험집단이 비위험집단에 비해 20대와 40대가 이환율이 높은 것으로 조사되었으며, 배우자가 없는 계층이, 학력수준이 낮은 계층, 단순 근로직인 계층, 신체적 건강상태가 나쁜 계층에서 이환율이 높았다.(p<0.05). 결 론: 위의 결과로 보아 향후 지역사회 정신보건사업은 전반적으로 정신건강수준이 낮은 저소득층을 주요사업대상으로 추진해야 하고, 이환율이 높은 계층 즉, 20대와 40대이고 배우자가 없이 혼자 살며 학력수준이 낮아 단순근로직에 종사하고 신체적 건강상태가 나쁜 계층에 우선적으로 역점을 두어 정신건강 프로그램을 개발하고 교육과 홍보를 지속적으로 해 나가야 될 것으로 판단한다. Objective: This study was performed to support and enhance the community mental health for the urban poor. The purpose of the study were as follows: 1) to investigate the mental health states of the urban poor, who are the first target population, 2) to identify the factors influencing their mental health states. Methods: The subjects were community residents in low socioeconomic level, who are 20-59 years old. And the scale used was GHQ(General Health Questionnaire) which is used for screening the mental health states of groups and self-esteem. The survey subjects were directly interviewed by staffs in Public Mental Health Center. SPSS PC 6.0 was used for the statistical analysis. It investigated the mental health states, according to social and demographic attributes, and self-esteem of target population. And the mental health states were analyzed to three subdomains: overall mental health states, social dysfunction, and anxiety-depression domains. Results: First, in demographic attributes, the old men, who are of low socioeconomic level, single, bad physical health, low community life satisfaction, low educational background, and having no religions, were low in overall mental health states, social function, and high in anxiety-depression domain. Second, the residents, who had high self-esteem, were high in overall mental health states, social function, and low in anxiety-depression domain. Third, the residents, who had high self-esteem scores, were significantly high in overall mental health states, social function, and low in anxiety-domain. Fourth, Among the various factors influencing the mental health states, the self-esteem was the most important factor, and the physical health was the next important factor. And the higher self-esteem they had, the better physical health states, the order they were, the higher the mental health states became. Fifth, comparing the high risk group with non-risk group, those who were in their 20`s and 40`s, had no wives or husbands, low educational background, physical workers, and were not well healthy, were apt to have mental diseases. Conclusion: In the future, the first target population in the community mental health services will be the urban poor in low mental health states. For those who are apt to have mental diseases, the first step of community mental health works is to find, select and give basic mental health services. The staffs in community mental health center have to develop various mental health programs and inform the public of them.

      • Effects of Stressed and Unstressed Reinforcements on Prestressed Concrete Members with Unbonded Tendons

        The research purpose of this paper is to investigate the influential Parameters on the unbonded tendon stress. The parameters were the reinforcing ratio, the prestressing ratio, and the loading type. To this end. first, the influence of parameters were examined with twenty eight test results obtained from references. Then, an experimental study was carried out with nine specimens. Test variables were the reinforcing ratio and the prestressing ratio. Specimens were divided equally into three groups and each group had a different level of the reinforcing ratio. Each specimen within a group has a different level of the prestressing ratio. The investigation with previous and current tests revealed the followings; (1) the length of crack distribution zone does not have a close relation with the length of plastic hinge. (2) the prestressing ratio does not affect both the length of crack distribution and the length of plastic hinge, (3) the tendon stress variation is in reverse relation with the ratios of mild steels and tendons, (4) the loading type nay not affect significantly the length of crack distribution zone, (5) AASHTO LRFD Code equation and Moon/Lim's design equation predicted the test results well with some safety margins.

        This paper aims at providing a concrete plan to improve technical and vocational teacher education in the College of Industrial Education of Chungnam National University. It is intended to propose a practical four-year regular and short-term course. Also, the estimated required facilities is included in this study as well as their management. The main conclusions and recommendations are summarized as follows; 1) The numbers of students enrollment should be reduced to 220 students which is the approximate number of future demand. 2) In order to offer to the students a broader education and to cope with the increase/decrease in demand of technical and vocational teacher within a specific field, the 8 department existing at the present (Department of Architecture Engineering Education, Civil Eng. Educ., Metallurgical Eng. Educ., Mechanical Eng. .Educ., Electrical Eng. Educ., Electronic Eng. Educ., Industrial Chemistry Educ. and Industrial Technology Educ.) would be combined into 5 (Department of Construction Engineering Education, Mechanical Eng. Educ., Electrical Eng. Educ., Industrial Chemistry Educ. and Industrial Technology Educ.). 3) The short-term course program is recommened to provide additional training and education to those candidates who already have experience in technical fields to meet an occasional demand. 4) It is necessary to add the Teaching Profession Section in the administrative organization of the College.
