All of sealing capsules to transport a special form radioactive material should be designed and fabricated in accordance with the design criteria prescribed in IAEA standards and domestic regulations. The objective of this study is to demonstrate the safety of a shipping capsule for 192Ir special form radioisotope which produceed in the HANARO. The safety tests were carried out for the impact, percussion, bending and heat test conditions. And leakage tests were carried out before and after the each test. Also, the safety analyses were performed using computer codes in order to verify the test results. The capsule showed slight scratches and deformation, and maintained its structural and thermal integrities in all tests without any severe damage or melting. It also met the allowable limits of leakage rate after earth test. Therefore, it has been verified that the capsule was designed and fabricated to meet all requirements for the special form.

Although over 30 common genetic susceptibility loci have been identified to be independently associated with coronary artery disease (CAD) risk through genome-wide association studies (GWAS), genetic risk variants reported to date explain only a small fraction of heritability. To identify novel susceptibility variants for CAD and confirm those previously identified in European population, GWAS and a replication study were performed in the Koreans and Japanese. In the discovery stage, we genotyped 2123 cases and 3591 controls with 521 786 SNPs using the Affymetrix SNP Array 6.0 chips in Korean. In the replication, direct genotyping was performed using 3052 cases and 4976 controls from the KItaNagoya Genome study of Japan with 14 selected SNPs. To maximize the coverage of the genome, imputation was performed based on 1000 Genome JPT+CHB and 5.1 million SNPs were retained. CAD association was replicated for three GWAS-identified loci (1p13.3/SORT1 (rs599839), 9p21.3/CDKN2A/2B (rs4977574), and 11q22.3/ PDGFD (rs974819)) in Koreans. From GWAS and a replication, SNP rs3782889 showed a strong association (combined P=3.95 10(-14)), although the association of SNP rs3782889 doesn't remain statistically significant after adjusting for SNP rs11066015 (proxy SNP with BRAP (r(2)=1)). But new possible CAD-associated variant was observed for rs9508025 (FLT1), even though its statistical significance did marginally reach at the genome-wide a significance level (combined P=6.07 10(-7)). This study shows that three CAD susceptibility loci, which were previously identified in European can be directly replicated in Koreans and also provides additional evidences implicating suggestive loci as risk variants for CAD in East Asian.

In this study, mesoporous silicate was applied as a matrix for the analysis of various molecules from small molecules to medium sized peptides in laser desorption/ionization mass spectrometry. In contrast with conventional matrix assisted laser desorption/ionization mass spectrometry (MALDI-MS), the proposed approach, desorption/ionization on mesoporous silicate mass spectrometry (DIOM-MS), significantly reduces the problem of matrix interference in low mass region and can be applied to the analysis of versatile chemicals including amino acids, synthetic drugs, peptides and others. In addition, distinctive advantage of DIOM-MS showed higher salt tolerance and could be applied to identify the proteins from the analysis of tryptically digested peptides. DIOM-MS has several availabilities such as easy sample preparation, rapid analysis of small molecules without noise, peptide analysis without organic matrix, high salt tolerance, versatile coupling with other separation techniques, and high throughput manner.

Abstract

Microfluidic chemotaxis analysis requires stable and reproducible formation of chemical gradient with low batch-to-batch variation. Dimensional uniformity of a junction channel plays a key role in diffusion-based gradient generation. Conventional methods for the analysis of bacterial chemotaxis are limited in stability and reproducibility of the chemical gradient. Here, we report a microfabrication method employing plasma enhanced chemical vapor deposition for silicon oxide deposition and consecutive reactive ion etching processes to reduce the experimental errors of chemotaxis analysis occurring from dimensional inaccuracy of the junction channel. This approach produces precise dimensional junction channels with below 1% variation. Thus, we confirm the reproducibility of chemotaxis analysis and obtain the chemotaxis index of Pseudomonas aeruginosa PAO1 with bacterial chemoeffectors.

Highlights

Precise fabrication of microchannel with low dimensional errors (C.V. = 0.42%).

Spatiotemporally reliable formation of a diffusion-based chemical gradient.

Improved reproducibility of chemotaxis assay by reliable chemical gradient.

Graphical abstract

[DISPLAY OMISSION]

Background

There are many studies regarding the effects of insulin on bone metabolism and changes in bone mineral density (BMD) in the setting of diabetes. The effect of prediabetes on BMD is not known.

Methods

A total of 802 men participated in the Korea Rural Genomic Cohort Study (in Geumsan County). According to the results of an oral glucose tolerance test, subjects were classified into normal, prediabetic, and diabetic categories. One hundred twenty-four subjects diagnosed with type 2 diabetes were excluded, leaving 678 subjects for the study inclusion. BMD was estimated with a quantitative ultrasonometer.

Results

The average BMD T scores of normal and prediabetic subjects were -1.34 ± 1.42 and -1.33 ± 1.30, respectively; there was no significant difference in the BMD T scores between these groups. The BMD T score was inversely associated with age and positively correlated with body weight, body mass index, total cholesterol, low density lipoprotein cholesterol, and HbA1c. On multiple linear regression analysis, low density lipoprotein cholesterol was the only statistically significant variable for prediabetes (β = 0.007, P = 0.005). On the stepwise regression analysis, age (β = -0.026, P < 0.001), the body mass index (β = 0.079, P < 0.001), and low density lipoprotein cholesterol (β = 0.004, P = 0.016) were significant variables for prediabetes.

Conclusions

There was no significant difference in the BMD T score between the normal and prediabetic subjects. Further studies are needed regarding the association of fracture risk and changes in BMD with the development of overt diabetes.
