고려 엉겅퀴(Cirsium setidens Nakai) 잎 조직을 이용한 callus 배양 및 항산화 활성 검증

We described the effects of antioxidant activity on the extracts of tissue and callus formation by Cirsium setidens Nakai. The callus culture were induced from leaf tissue of Cirsium setidens in both MS and B5 medium with IAA or/and kinetin (0∼5.0 ㎎/ℓ). As medium for callus culture induced from Cirsium setidens, B5 medium showed better condition than MS media. MS medium brought to decrease callus differentiation after formation, on the other hand B5 medium induced to accelerate callus differentiation. Among phytohormones is required 0.5 ㎎/ℓ of IAA and between 2.0 ㎎/ℓ and 1.0 ㎎/ℓ of Kinetin for formation of callus. In order to extract functional material from natural leaf and callus tissue of Cirsium setidens used distilled water and 75% ethanol as solvent and those extract used to determine antioxidant activity as alpha-diphenyl-2-picryl-hydrozyl (DPPH) radicals scavenging effect and TBA(thiobarbituric acid) activity. In result, the effect of DPPH and TBA had high activity more natural leaf extract than callus tissue and 75% ethanol was solvent better than distilled water. The 75% ethanol extract of natural leaf showed an excellent antioxidant activity because that particularly had highest concentration of vitamin C and total phenol among those extracts

Human eryhropoietin (EPO) is acidic glycoprotein hormone that plays key role in hematopoiesis by facilitating differentiation of erythrocyte and formation of hemoglobin (Hb) and is used for the treatment of anemia. Human EPO is consist of 166 amino acids which is modified by three N-glycosylations (24, 38, 83) and single O-glycosylation (126). N-glycosylation is reported to be related to the cellular secretion and activity of EPO. In this study, we examined effects of mutagenesis in glycosylation site of recombinat hEPO for the cellular secretion during production from cultured CHO cell. We produced rhEpo which was cloned by PCR from human liver cDNA (TaKaRa) in cultured CHO cell. Using supernatant of the culture, ELISA assay and western analysis were performed. To estimate biological activity, 20IU of rhuEpo was subcutaneously injected into four ICR mice. After 8 days, HCT level was increased average 13 per cent, RBC was increased ca. 2106//. In disease model Rat (anemia c-kit, WSRC-WS/WS), HCT was increased ca. 12%, RBC was increased ca. 1.6106//. These results suggests that rhEpo we produced has biological activity. To remove glycosylation site by substituting 24, 38, 83, and 126th asparagine (or serine) with glutamic acid, overlapping -extension site-directed mutagenesis was performed. To add novel glycosylation sites, 69, 105th leucine was mutated to asparagine. Mutant EPO construct was transfected into CHO cell. Supernatant of the cell culture was analyzed using ELISA assay with monoclonal anti-EPO antibody (Medac, Germany). Since, several reports for mutagenesis of glycosylation sites showed case-by-case results, we examined both transient expression and stable expression. Addition of novel glycosylation sites resulted no secretion while deletion mutants had little effect except some double deletion mutants (24/83 and 38/83) and triple mutant. We suggest that not single but combination of glycosyl group affect secretion of EPO.

OBJECTIVE:: To investigate the effect of saline chase injected at 2 different rates on computed tomography (CT) angiography. MATERIALS AND METHODS:: This study was approved by our institutional animal study committee. Three injection protocols were used; contrast injection (24 mL, 0.8 mL/s) without saline chase (protocol A), contrast injection with saline chase injected at the same rate as the contrast medium (protocol B), and contrast injection with saline chase injected at half the rate (0.4 mL/s) of the contrast medium (protocol C). In the 3 dogs used in our study, each of the protocols was applied twice for every dog resulting in a total of 18 sessions of monitoring scans. CT images were acquired every second at the fixed level of the aorta and pulmonary artery (PA). The duration of plateau, plateau deviation, and peak arterial enhancement were computed and compared using the Kruskall–Wallis and Mann–Whitney U test. RESULTS:: Peak contrast enhancements were significantly more delayed with protocol B than with protocol A in both the PA (B: 48 seconds, A: 30 seconds, P = 0.024) and aorta (B: 46 seconds, A: 38 seconds, P = 0.024). The duration of enhancement plateau was longer with protocol B than with protocol A in PA (B: 14.8 seconds, A: 9.0 seconds, P = 0.002) and in aorta (B: 16.2 seconds, A: 11.6 seconds, P = 0.004). Protocol C had the longest duration of plateau in both PA (34.5 seconds, P = 0.002) and aorta (33.8 seconds, P = 0.004) with uniform plateau enhancement. The peak enhancement values of protocol C, however, were substantially lower than that of protocol A and B in both the PA (A: 262 HU, B: 239 HU, C: 191 HU, P = 0.001) and aorta (A: 263 HU, B: 268 HU, C: 210 HU, P = 0.001). CONCLUSIONS:: Saline chase prolongs the duration of plateau and delays peak enhancement of the pulmonary artery and aorta. Saline chase injected at half the rate of contrast medium injection allowed more uniform and prolonged plateau contrast enhancement than other protocols.

<P>This study examined the effect of electrolyte temperature on the surface characteristics of anodized pure titanium in DL-α-glycerophosphate (DL-α-GP) and calcium acetate (CA). The anodic oxide films contained a large proportion of anatase with some rutile. The relative intensity of the anatase peaks and the surface roughness increased with increasing electrolyte temperature. As the electrolyte temperature was increased, the pore size increased to 1–4 µm, and the apatite crystals became coarser and denser. The amount of calcium and phosphate adsorbed increased with increasing electrolyte temperature. The anodizing voltage reached the onset of constant voltage mode faster with decreasing electrolyte temperature. A higher voltage is essential for the electrolyte at lower temperature to reach the constant voltage mode during this initial period. Copyright © 2008 John Wiley & Sons, Ltd.</P>

<P>An ultimately scaled multiple-gate CMOS thin-film transistor with a polysilicon (poly-Si) nanowire demonstrates feasibility for vertical integration using multiple active layers for application in the terabit memory era. The short-channel effects are suppressed using a multiple gate to wrap around the nanowire in devices with a size of a few tenths of a nanometer. The switching and output characteristics show high device performance without a crystallization process for the poly-Si nanowire.</P>

The purpose of this research is to investigate effects of different fluid ingestion on physiological metabolism, reaction time, and breath alcohol concentration after alcohol consumption For this study, seven male college students were divided into three groups as follows; alcohol consumption only (A(, water only (AW), and ion fluid consumption after the reaction time and recovery of breath alcohol concentration in AI(p<0.05). From this research, we could concluded that drinking ion fluid after alcohol consumption might draw positive effects on reaction time and recovery of the breath alcohol concentration. On the basis of these results, it is useful to diminish the concentration of alcohol quickly in the body and keep the body`s balance with ion fluid consumption that contains carbohydrate and electrolyte.
