        PCR을 이용한 국내 및 수입 축산물 유래 Listeria monocytogenes의 유전학적 분석연구

        One hundred-fifty two of Listeria strains were isolated from domestic chicken carcasses, a slaughter-houses from nationwide from 1996 to 1997 and imported livestock products from a ten foreign countries including the USA, China, France and Thailand. To substitute the laborious and time consuming procedures of clinical laboratory standard diagnostic methods, the rapid and specific multiplex PCR(M-PCR), which was designed to amplify a three kinds of genes simultaneously f3r specific detection and differentiation of L. monocytogenes(LM) by one step procedure, was established. The LM strains confirmed with M-PCR were analysed to their genetic diversities by RAPD using the two primers(D87 & MMTl), and also evaluated on their discriminatory abilities(na) between two primers. our study also conducted the REP-PCR and ERIC-PCR for evaluation of the possibility of REP and ERIC elements for genetic subtyping of LM strains. According to the REP-PCR and ERIC-PCR fingerprinting patterns, LM strains were divided into 7(ERIC-type) and 4(REP-type) major clusters at the relative clone cut off value of 80%. According to the computer analysis of PCR results using the GelCompar Ⅱ software, ERIC-PCR(DI=0.955) had expressed the most high discriminatory ability and followed by REP-PCR(DI=0.952), RAPD(D87; 0.954, MMT1; 0.937), in order. The combined analysis of the present PCR results was also expressed the reliable and useful discriminatory ability(DI=0.916). This study suggested that PCR based methods could be used as an reliable, reproducible, rapid, and highly discriminatory method for the genetic differentiation of genus Listeria.

        Pendulum 장치의 상악대구치의 원심이동에 대한 효과

        Ⅱ급 치성교합을 가진 환자에서는 I급 구치부 교합관계를 형성하기 위하여 상악 대구치의 원심이동이 필요하다 본 연구에서는 pendulum장치를 이용한 상악 대구치의 원심이동시 대구치의 이동양상과 고정원의 전방이동 및 정출정도 등 치아와 골격에 미치는 영향을 평가하기 위하여 시행되었다. 상악 제1대구치를 원심이동시켜 1급 구치관계가 형성된 19명의 환자 (남: 8명. 여: 11명, 평균연령 11.68±1.52세)에서 장치 장착 전과 장치제거 직후에 얻어진 측면두부방사선사진을 비교 ·분석하여 다음과 같은 결과를 얻었다. 1.I급 교합관계를 형성하기 위해서 제 1대구치는 평균 2.94± 1.54mm 원심이동하였으며 . 1.17±0.97mm 함입되었고, 소요기간은 평균 18.13±7.95일이었다. 2.고정 원으로 사용된 치아는 1.34± 1.40mm로 유의하게 전방 이동하였으며, 0.48±0.99mm 정출되었다. 또한 절치는 유의한 전방이동 및 순측경사가 발생하였다(P<0.05) . 3.구개평면과 하악평면이 이루는 각은 유의한 증가를 보였다(P(0.05). 4.제1소구치 또는 제1유구치가 고정 원으로 이용된 경우 제2소구치 또는 제2유구치가 이용된 경우 보다 전방이동이 증가하였다(P<0.005). 5. 제2대구치의 발육단계에 따른 제1대구치 및 제2대구치의 이동양상은 어떤 항목과도 유의성이 없었다(P<0.05). 6.얻어진 총 공간 중 74%는 제1대구치의 원심이동이,26%는 소구치의 전방이동에 의하여 얻어졌다. 이상의 결과를 종합하여 볼 때 pendulum appliance는 상악 대구치의 원심이동에 매우 효과적이었으며. 고정원의 소실을 최소화하기 위해 제2유구치 또는 제 2소구치가 포함되어야 한다. 또한 제1대구치의 원심경사는 필연적으로 일어나며. 이를 최소화하기 위한 노력이 계속되어야 할 것으로 사료된다. Treatment of class Ⅱ malocclusions require distalization of maxillary molars into class I relationship. Intra-arch distal molar movement techniques have recently assumed an important role in young patients. In this study, the dental and skeletal effects of the pendulum appliance were evaluated by means of cephalometric radi-ographs. The samples were consisted of 19 patients: 11 females and 8 males, mean age 11.68± 1.52 years. Measurements were obtained from cephalometric prior to and the day of removal of the pendulum appliance. Treatment changes were analyzed. The following results were obtain : 1. The pendulum appliance produced 2.94±1.54mm distal molar movement with a mean intrusion of 1 17± 0.97mm, mean period 18.13±7.95 weeks. 2. The anchor tooth was 1.34± 1.40mm forward movement and 0.48±0.99mm extrusion, and labial tilting of incisors. 3. The angle between palatal plane and mandibular plane increased significantly 4 There was no significant difference in according to 2nd molar position. 5. Total movement was consisted of 74% distal movement of 1st molar and 26% forward movement of the an-chor tooth

        피로와 의료이용, 업무상 사고 및 질병결근

        Objectives: This study examined the relationship of fatigue to medical utilization, occupational accident and sickness absence. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project, 2002~2004). A total of 30,146 employees categorized within the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were recruited. A structured-questionnaire was used to assess the participants' sociodemo-graphics, job-related factors, health-related behaviors, personality trait, self=perceived fatigue (MFS) and the frequency of medical utilization, occupational accident and sickness absence. Data on medical utilization and occupational accident were confirmed from company health records. Chi-square test and logistic regression analysis were used to elucidate the relationship of self-perceived fatigue to medical utilization, occupational accident and sickness absence. Results: In logistic regression analyses, self-perceived fatigue was associated with medical utilization (outpatient), occupational accident and sickness absence although the strength of the associations was higher in the blue collar workers except for medical utilization. For medical utilization, the white collar worker's adjusted odds ratios of outpatient utilization for workers scoring in the third quartile and the highest quartile, compared to workers scoring in the lowest quartile were 1.50 (95% CI: 1.20-1.87) and 2.16 (95% CI: 1.69-2.75), respectively. The blue collar workers' adjusted odds ratios of outpatient utilization for workers scoring in the second (OR: 1.23, 95% CI: 1.06-1.43), third (OR: 1.42, 95% CI: 1.22-1.66) and highest quartile (OR:1.83, 95% CI: 1.55-2.17), respectively, compared to workers scoring in the lowest quartile were statistically significant. However, no associations were found between fatigue and inpatient utilization for either the white or blue collar workers. The blue collar workers' adjusted odds ratios of being injured for workers scoring in the second, third and highest quartile were 1.41 (95% CI: 1.00-2.00), 1.57 (95% CI: 1.10-2.22) and 2.41 (95% CI: 1.69-3.44), respectively, compared to those scoring in the lowest quartile, but, those of the white collar workers were not significant. There was more likely to an increased risk of sickness absence in workers scoring in the second, third and highest quartile compared to those scoring in the lowest quartile in both white and blue collar workers, but the magnitudes of risk were higher in the blue collar workers than the white collars workers. Conclusion: This result suggests that fatigue is a determinant predictor of medical utilization, occupational accident and sickness absence, and that the pattern of risks for the three outcomes differs according to the occupational type. Some limitations of this study and considerations for future study were also discussed.

        Objective: The purpose of this study is to have the basis data for indoor air quality at barracks to prevent soldier's respiratory disease in military Method: We measured the indoor air quality of barracks in concentration with five factors such as airborne microbes, carbon dioxide, fine dust, temperatures and relative humidity at every month from June to August in 2009. We measured the indoor air quality at various time and at the three buildings that were classified year built. Results: The concentration shows significant difference by the inspected time not by the year built of the buildings. The concentration of airborne microbes, carbon dioxide were highest at 11:00 pm (p<0.01), and those of fine dust were highest at night(p<0.01). Temperatures were highest at 3:00 pm (p<0.01), and relative humidity were highest at 9:00 pm (p<0.05). Conclusion: Government rules should be legitimated for engineered ventilation system and periodical estimation of indoor air quality at barracks to prevent soldier's respiratory disease.

      • Transglutaminase 2 inhibition found to induce p53 mediated apoptosis in renal cell carcinoma

        <P>Renal cell carcinoma (RCC), the predominant form of kidney cancer, is characterized by high resistance to radiation and chemotherapy. This study shows that expression of protein cross-linking enzyme transglutaminase 2 (TGase 2) is markedly increased in 7 renal cell carcinoma (RCC) cell lines in comparison to HEK293 and other cancer cell lines, such as NCI 60. However, the key role of TGase 2 in RCC was not clear. The down-regulation of TGase 2 was found to stabilize p53 expression, thereby inducing a 3- to 10-fold increase in apoptosis for 786-O, A498, CAKI-1, and ACHN cell lines by DAPI staining. MEF cells from TGase 2<SUP>−/−</SUP> mice showed stabilized p53 under apoptotic stress to compare to MEFs from wild-type mice. TGase 2 directly cross links the DNA binding domain of p53, leading to p53 depletion <I>via</I> autophagy in RCC. TGase 2 and p53 expression showed an inverse relationship in RCC cells.       • A Disease Model Pig Over-Expressing Mouse PEA15 Showed Symptoms of Diabetes Mellitus

        Phosphoprotein Enriched in Astrocytes (PEA15) is a 15kD-sized intracellular signaling protein, highly expressed in astrocytes and constitutively expressed in peripheral tissues. Recently it was found that PEA15 expression was elevated in patients suffering type 2 diabetes and suggested to be involved in the syndrome of insulin resistance. To investigate the functional role of PEA15 for the control of blood glucose level, we produced a transgenic pig over-expressing mouse PEA15 (mPEA15). As a model animal, pig has many advantages. They have a higher fecundity and a short generation time and are physiologically similar to human. Using the transgenic pig, we carried out a series of experiments to establish a link between PEA15 expression and the insulin resistance. Our results suggested that, compared with control pig, mPEA15 pig has, (1) a higher blood resistin level, (2) a lower cell membrane-embeded GLUT4 level, and (3) a lower glucose clearing ability based on oral glucose tolerance test (OGTT). When our results combined, it can be concluded that mPEA15 over-expressing pig has many symptoms of insulin resistance and these pigs will become a useful disease model to investigate diabetes mellitus in the near future.
