        Lethal (2) Essential for Life [l(2)efl] Gene in the Two-spotted Cricket, Gryllus bimaculatus (Orthoptera: Gryllidae)

        쌍별 귀뚜라미(Gryllus bimaculatus)에서 lethal (2) essential for life [l(2)efl]을 코드한 cDNA를 분리하여 GBl(2)efl이라 하였다. GBl(2)efl는 N-glycosylation 한곳과 phosphorylation site를 15곳 가진 189 aa로 구성되며6.2등전점과 21.19 kDa 분자량을 가진다. GBl(2)efl 단백질의 이차구조는 random coils (56.08%), alpha-helix (22.22%), extended strands (17.99%), beta turns (3.7%)로 이루어진다. GBl(2)efl 는 지금까지 보고된 l(2)efl들과는48-69%의 상동성을 보인다. GBl(2)efl은1일, 3일 starvation일때에 각각 dorsal longitudinal flight muscle과 Malpighian tubules에서 mRNA발현이 증가하였다. 한편, ER stress 조건에서는GBl(2)efl 발현은 fat body에서 증가하였다. 본 연구는 곤충의 생존에 기여하는 생리학적 메커니즘을 이해와 효과적인 해충 관리 통제를 수행할 수 있는 능력을 향상에 많은 힌트를 줄 수 있는 실마리를 제공할 수 있을 것이다. A cDNA encoding the protein lethal (2) essential for life [l(2)efl] was cloned from Gryllus bimaculatus and named GBl(2)efl. This protein is composed of 189 amino acids, including an N-glycosylation site and 15 phosphorylation sites. Its predicted molecular mass is 21.19 kDa, with a theoretical isoelectric point of 6.2. The secondary structure of GBl(2)efl was predicted from the identification of random coils (56.08%), alpha helices (22.22%), extended strands (17.99%), and beta turns (3.7%) through sequence analyses. A homology analysis revealed that GBl(2)efl exhibited a high similarity with other species at the amino acid level, ranging from 52% to 69%. While GBl(2)efl mRNA expression was higher in the dorsal longitudinal flight muscle following a three-day starvation and in the Malpighian tubules following a one-day starvation, no changes in expression were detected in other tissues. Furthermore, tunicamycin-induced endoplasmic reticulum (ER) stress resulted in an approximately 1.8-fold higher expression in the fat body compared with the wild type.

        Upregulation of Kruppel-like Factor 4 Gene expression by Allomyrina dichotoma Hemolymph in the INS-1 Pancreatic β-cells

        The hemolymph of Korean rhinoceros Allomyrina dichotoma consists of blood and lymph in which various kinds of proteins function physiologically. We have previously demonstrated that A. dichotoma hemolymph has the potential to treatment and prevent diabetes through activating transcription factor 3-gene (ATF3) regulation. In this study, we investigate the expression of Kruppel-like factor 4 (KLF4) in A. dichotoma hemolymph-treated INS-1 pancreatic β-cells. The new findings show that A. dichotoma hemolymph, which upregulates KLF4 gene expression in a dose-dependent and timedependent manner. In addition, hemolymph combine with mild endoplasmic reticulum (ER) stress, which also differentially regulates KLF4 gene expression. These results may provide insights to KLF4 gene-related disease therapies through KLF4 gene regulation.

        Isolation and Characterization of Six Microorganisms from the Digestive Tract of the Cricket Gryllus bimaculatus

        우리는 처음으로 쌍별귀뚜라미 소화기관에서 6종류의 미생물을 분리하고 특성을 규명하였다. 이들은 16S rDNA을 기준으로 분류한 결과 4종류(Staphylococcus, Bacillus, Citrobacter, Proteus)에 속하였다. 분리된 6종류의 미생물은 공통적으로 ampicillin에 저항성을 보이지만 kanamycin 저항성은 보이지 않았다. 이들을 Gram염색하여 미생물의 형태적 특징을 확인하였다. Gram-positive한 rod-shaped GL2와 round-shaped GL4는 다른 분리 균 보다 많은 양의 세포외분비물을 만들었다, 이들을 MALDI-TOF-MS spectral analysis결과 87-kDa collagenase, 56-kDa & 200-kDa hypothetical protein이였다. 새롭게 분리된 6종류의 미생물은 귀뚜라미의 생리에 미치는 영향과 이들의 생물공학적 혹은 해충 방제에 이용될 수 있는 연구가 기대된다. We describe the isolation and characterization of six different intestinal microorganisms from the digestive tract of the cricket Gryllus bimaculatus. Based on 16S rRNA gene sequences, we obtained six isolates belonging to four different genera: Staphylococcus, Bacillus, Citrobacter, and Proteus. All the isolates were resistant to ampicillin. Ampicillin is an irreversible inhibitor of the enzymeetranspeptidase, which is needed to make bacterial cell walls. None of the isolates were resistant to kanamycin, which binds to the 30S subunit of the bacterial ribosome and then inhibits total protein synthesis. Gram staining was conducted, in addition to morphological classification under a microscope. Four grampositive isolates and two gram-negative isolates were detected. The gram-positive isolates were GL1 (round shaped, 2 am in diameter), GL2 (rod shaped, 2.5 μm in length), GL3 (rod shaped, 2 μm in length), and GL4 (round shaped, 1.5 μm in diameter). The gram-negative isolates were GL5 (rod shaped, 2 μm in length) and GL6 (rod-shaped, 2.5 μm in length). Notably, two of the isolates, GL2 and GL4, secreted specific extracellular proteins. These were determined by MALDI-TOF-MS spectral analysis to be a 87 kDa collagenase, 56 kDa hypothetical protein, and 200 kDa hypothetical protein. The six isolates in this study could be used for various biotechnological applications and pest management, both in the field and in greenhouse systems. In addition, it would be interesting to determine the relationship between these isolates and their host.

      • Depth based Sign Language Recognition System Using SVM

        In this paper, we propose a sign language recognition system using an SVM (Support Vector Machine) and a depth camera. In particular, we focus on the Korean sign language. For the sign language system, we suggest two methods, one for the hand feature extraction stage and the other for the recognition stage. Hand features consist of the number of fingers, finger length, palm radius, and hand direction. To extract hand features, we use Distance Transform and a hand skeleton. This method is more accurate and faster than a traditional method that uses contours. To recognize hand posture, we develop a decision tree with hand features. For more accuracy, we use SVM to determine the threshold value in the decision tree. In the experiment results, we show that the suggested method is more accurate and faster when extracting hand features and recognizing hand postures.

        Regulation of Endoplasmic Reticulum Stress Response by the Immobilization Stress

        많은 종류의 세포스트레스는 unfolded protein response (UPR)관련인자의 유전자발현을 조절한다. 본연구결과 부동스트레스(immobilization stress)는 세포의 소포체스트레스(ER stress)와 관련된 유전자발현의 변화를 유도한다; Heart, spleen, thymus, kidney, testis에서는 유전자발현 변화가 없었지만 adrenal gland, liver, lung에서는 유의할만한 상승변화가 있었다. 그러나 muscle에서는 다른 것들과 대조적으로 발현이 감소되었다. 이 결과는 부동스트레스도 다른 종류의 세포스트레스와 같이 세포수준에서 UPR을 조절할 수 있다는 최초의 보고이다. Many kind of cell stresses induce gene expression of unfolded protein response (UPR)-associated factors. This study demonstrated that up- and down-regulation of gene expression of endoplasmic reticulum (ER) stress chaperones and ER stress sensors was induced by immobilization stress in the rat organs (adrenal gland, liver, lung, muscle). However, no statistically significant regulation was detected in the others (heart, spleen, thymus, kidney, testis). The results are the first to show that immobilization stress induces UPR associated gene expression, will help to explain immobilization stress-associated ER stress.

        소포체스트레스에 의한 cAMP phosphodiesterase 7A1 유전자의 발현

        갑상선 배양세포(FRTL5)에서 ER stress에 의해서 ER chaperone (Bip, ERp29, Calnexin and PDI), ER stress sensor (PERK, ATF6 and Ire1)와 cAMP phosphodiesterase 7A1 (cAMP PDE7A1) 유전자발현이 증가하는 것을 알았다. 세포배양배지에서 A23187을 제거하면 cAMP PDE7A1 유전자발현이 회복되지만, thapsigagin의 경우는 회복되지 않았다. 그리고 A23187과 TSH를 함께 처리한 경우는 아주 강하게 cAMP PDE7A1 유전자의 발현이 억제되었다. 이 같은 결과는 ER stress에 의해서 cAMP PDE7A1 유전자발현이 상승 발현된다는 첫 보고이다. This study demonstrated that upregulation of gene expression of endoplasmic reticulum (ER) stress chaperones (Bip, ERp29, calnexin, and PDI), ER stress sensors (PERK, ATF6, and Ire1), and cAMP phosphodiesterase 7A1 (cAMP PDE7A1) was induced by ER stresses in FRTL5 cells. While removing A23187 from the culture medium restored upregulation of cAMP PDE7A1 gene expression, removal of thapsigargin did not recover its expression. In addition, cAMP PDE7A1 gene expression was strongly inhibited by treatment with A23187 combined with thyroid stimulating hormone (TSH). The results are the first to show that ER stress induces cAMP PDE7A1 gene expression.

        ERp29와 ADP-ribosylation factor 5의 결합특성

        ERp29는 endoplasmic reticulum (ER) lumen에 존재하는 단백질로 protein disulfide isomerase (PDI) family에 속한다. 비록 관련 연구 결과는 조금 있지만 정확한 생물학적인 기능은 아직 분명하지 않지만, 분비단백질과정과 단백질 folding에 관여하는 것으로 알려 지고 있다. ERp29의 기능 연구를 위하여 yeast two-hybrid screening/GST pull-down assay방법을 사용하여 ERp29-결합단백질인 ADP-ribosylation factor 5 (ARF5)를 동정하였다. 이들의 결합은 정상적인 세포생리상태에서는 결합하지만 ER stress 상태에서는 떨어졌다. 이 결과는 ERp29의 기능 연구를 위하여 하나의 실마리를 제공할 것이다. ERp29 is a endoplasmic reticulum (ER) lumenal resident protein that shows sequence similarity to the protein disulfide isomerase family. Its biological function is thought to play a role in the processing of secretory proteins within the ER, possibly by participating in the folding of proteins in the ER. Although some data on ERp29 have been reported, its normal functions are still unclear. To gain insights into the function of ERp29, we identified ARF5 protein as a protein that interacts with ERp29 using yeast two-hybrid screening and GST pull-down assay. Interaction between ERp29 and ARF5 was detected under normal cell conditions but not under ER stress conditions. This result may provide a clue for understanding ERp29 biological functions.

        Elderly Sarcopenia and Vitamin B Deficiency: A Relationship?

        노인들에서 근감소증은 의료-간호비용 증가의 주요 원인 중 하나가 되고 있다. 한국에서는 근감소증 예방 대책이 일반적으로 특정 질병이 없는 노인들을 대상으로 하지만, 요양원-요양병원에서 집단 거주하는 노인들의 근감소증 대책도 필요하다. 근감소증은 운동량 감소, 단백질 및 영양분(미네랄, 비타민 포함) 섭취 감소, 테스토스테론 및 성장호르몬 변화, 염증 등의 원인으로 발생한다. 분자 생물학적인 정확한 병태생리 기전의 이해가 요구된다. 근감소증은 골다공증, 낙상으로 인한 골절, 치매, 당뇨병, 심혈관 질환 등의 증상을 연결될 수 있다. 비타민 B 패밀리(B1-3, B5-7, B9 및 B12) 결핍을 근감소증 유발의 연구 대상으로 선택한 이유는 다음과 같다. 이는 비타민 B가 에너지 및 단백질 대사에 직접 관여하여 정상적인 신경 기능유지에 필수적이다. 비타민 B 결핍은 신경-근육 질환, 신경성 질환으로 나타날 수 있으며, 노인성 근감소증과 병행하는 경우가 많다. 노인들은 적정치 이하의 비타민 B 패밀리 섭취, 흡수 장애 및 무식증 문제 등을 겪을 가능성이 높다. 초고령화 사회에서 elderly가 자립적으로 일상생활을 할 수 있는 ‘health lifetime’을 유지하는 것은 개인의 행복추구와 사회경제적 부담을 줄일수 있는 최고의 목표이다. 본 연구는 근감소증과 관련하여 노인들의 근육량 감소 및 근육 기능 저하를 조절하는 수용성 비타민 B 패밀리의 최신 정보를 제공한다. 또한, 비타민 B 패밀리를 통한 마이오카인에 의한 근감소증의 조절 가능성도 소개한다. Sarcopenia is a leading cause of increased medical and nursing care costs among the elderly. In Korea, preventive measures for sarcopenia are mostly targeted toward the general elderly population without specific diseases. However, it is also necessary to implement measures for elderly individuals living in nursing homes and hospitals, where the prevalence of sarcopenia is high. Currently, computed tomography and/or magnetic resonance imaging are considered standard diagnostic tools. However, their complexity and time-consuming nature make them unsuitable for clinical use. The exact pathophysiological mechanisms of sarcopenia are unclear, as they involve various molecular biological pathways, including decreased exercise, protein and nutrient intake, changes in testosterone and growth hormone, and inflammation. Sarcopenia symptoms can lead to several diseases, such as osteoporosis, fractures, dementia, diabetes, and cardiovascular disease. Vitamin B deficiency is a significant factor in sarcopenia induction, with B vitamins being directly involved in energy and protein metabolism and nerve function. Vitamin B deficiency can lead to neuromuscular and neurogenic disorders, which often overlap with sarcopenia. Suboptimal intake of B vitamins, malabsorption, and anorexia are common among the elderly. This study aims to provide information on the role of water-soluble B vitamins in preventing and controlling muscle mass loss and deterioration among the elderly with sarcopenia. In addition, we discuss the potential of myokines from the B vitamin family in modulating sarcopenia.

        Expression of Beta-catenin-interacting Protein 1 (CTNNBIP1) Gene Is Increased under Hypothermia but Decreased under Additional Ischemia Conditions

        It has recently been shown that hypothermia treatment improves brain ischemia injury and is being increasingly considered by many clinicians. However, the precise roles of hypothermia for brain ischemia are not yet clear. In the present study we demonstrated firstly that hypothermia induced beta-catenin-interacting protein 1 (CTNNBIP1) gene expression and its expression was dramatically decreased under ischemic conditions. It was also demonstrated that hypothermia activated endoplasmic reticulum (ER) stress sensors especially both, the phosphorylation of eIF2α, and ATF6 proteolytic cleavage. However, the factors of apoptosis and autophagy were not associated with hypothermia. These findings suggested that hypothermia controlled CTNNBIP1 gene expression under ischemia, which may provide a clue to the development of treatments and diagnostic methods for brain ischemia.

        Short-term Hypothermia Induces Beta-catenin-interacting Protein 1 Gene Expression in PC12 Cells

        The effects of hypothermic treatment (32℃) on recovery from ischemia are controversial because the precise mechanisms of hypothermia remain unclear. We demonstrated previously that hypothermia induces beta-catenin-interacting protein 1 (CTNNBIP1) gene expression in vitro. In this study, we evaluated the effects of various hypothermic conditions, including lithium chloride treatment, on CTNNBIP1 gene expression. The results show that short-term hypothermic treatment resulted in relatively higher CTNNBIP1 gene expression than that of a longer treatment. These findings indicate that hypothermia controls CTNNBIP1 gene expression, which may provide clues to develop treatments to recover from and diagnose ischemia.
