      • Simultaneous Determination of Six Compounds in Hedera helix L. Using UPLC-ESI–MS/MS

        <P>We previously reported a method for simultaneously determining flavonoids and saponins from ivy leaf extract using HPLC-DAD. Due to the absence of a chromophore in two triterpene saponins, alpha-hederin and hederacoside C, the previous method was conducted at a non-specific wavelength of 210 nm. Thus, in this study, we focused on a rapid and sensitive analysis method for determining two important triterpene saponins from ivy leaf extract using UPLC-ESI-MS/MS. The developed method was validated in terms of linearity (R (2) > 0.9997), precision (RSD < 2.95%), and accuracy (98.91-101.92%), and showed increased sensitivity with improved limit of detection when compared to the previous data. In addition, the analytical conditions of UPLC-ESI-MS/MS provided better sensitivity with a shorter analysis time when compared to the HPLC-DAD method. Using this established method, the content of the six compounds was measured in medicinal syrups containing ivy leaf extract and the feasibility of this new method was demonstrated for the use in the quality control of ivy leaf extract products.</P>

      • CuInS2/ZnS quantum dot-embedded polymer nanofibers for color conversion films

        <P>Color-conversion films (CCFs) were fabricated with CuInS2/ZnS quantum dot (QD)-embedded polymer nanofibers and applied for white color emission with blue light-emitting diodes (LEDs). Red-orange color emitting CuInS2/ZnS QDs were synthesized and a photoluminescence quantum yield (PL QY) of 75% was achieved by controlling the Cu : In ratio in the QDs as well as the ZnS shell thickness. QD-embedded polystyrene nanofibers were electrospun on conducting aluminium substrates. The QD-embedded nanofiber (QDEN) structure is believed to reduce the aggregation of the QDs compared to that in a thin-film polymer. Time-resolved photoluminescence (TRPL) analysis showed a longer PL lifetime (138 ns) with the QDEN film than with a mould-cast thin film (45 ns). The longer PL lifetime with the QDEN structure is attributed to decreased quenching due to reduced aggregation. For the white lighting device, the QDEN films were applied as CCFs on a light guide plate (4.3 inch) with a GaN blue LED source. The resulting device demonstrated increased efficiency (from 11.5 to 32.3 mu m W-1) and color rendering index values (from 71 to 91) compared to commercial white LEDs. These Cd-free QD-embedded nanofiber films are expected to provide efficient and safe solutions for large-area processing of lighting devices.</P>

      • Analysis of effect of load-based excess bandwidth reservation on performances of differentiated services in E-PON

        <P>Performances of differentiated services in the upstream transmission of Ethernet passive optical network (E-PON) are affected by bandwidth reservation algorithm of an optical network unit, since data packets can be transmitted to an optical line termination only during the reserved transmission window. The effects of time-slot reservation on differentiated services are analysed and an adaptive class-based excess bandwidth reservation algorithm, called service quality pre-engagement (SQP), which reserves a time-slot based on the backlog queue and traffic arrival pattern of class is proposed. SQP applies the prediction-based bandwidth reservation to selected forward reservation classes (FRCs) which are determined by the introduced dynamic FRC selection. The proposed short-term service work-based time-slot reservation and long-term adaptive FRC selection optimise time-slot reservation according to traffic load of class. Analytic and simulation results show that this approach improves the performance of differentiated services, especially in terms of system buffer size, light-load penalty and service fairness, while guaranteeing high link utilisation and throughput</P>

      • Activation of the thalamic parafascicular nucleus by electrical stimulation of the peripheral vestibular nerve in rats

        <P>The parafascicular nucleus (PFN) of the thalamus is a primary structure in the feedback circuit of the basal ganglia-thalamo-cortical system, as well as in the neural circuit of the vestibulo-thalamo-striatal pathway. We investigated the characteristics of the functional connectivity between the peripheral vestibular system and the PFN in rats. A single electrical stimulation was applied to the horizontal semicircular canal nerve in the peripheral vestibular end-organs. This resulted in polysynaptic local field potentials (LFPs) in the PFN, which were composed of long-lasting multiple waves. The LFPs were prominently seen contralateral to the stimulation site. The PFN LFPs were suppressed by transient chemical de-afferentation of peripheral vestibular activity using a 5% lidocaine injection into the middle ear. The spontaneous firing rate of the single units increased after electrical stimulation to the horizontal canal nerve in a frequency-dependent manner. The induction of cFos protein was more prominent in the contralateral PFN than in the ipsilateral PFN following horizontal semicircular canal nerve stimulation. The functional vestibulo-parafascicular connection is a neural substrate for the transmission of vestibular sensory information to the basal ganglia.</P>

        Gold nanoparticle-based signal augmentation of quartz crystal microbalance immunosensor measuring C-reactive protein

        A biotinylated anti-rat C-reactive protein (CRP) antibody was ingeniously prepared by the reaction of the unmodified antibody with a water-soluble sulfosuccinimidyl-6-(biotinamido)hexanoate. The molar biotin incorporation of the resulting modified antibody was found as 5.82. A flow-type indirect-competitive quartz crystal microbalance immunosensor system was constructed with the sensor chip immobilized with 2mg/mL CRP as the coating antigen. When 200μL of the modified antibody having the concentration of 0.250mg/mL was added with a streptavidin-coated gold nanoparticle (GNP) to the immunosensor system, the frequency shift obtained was 139.8+/-0.3Hz. Compared to the frequency shift of 91.1+/-1.3Hz found with the addition of the unmodified antibody only, the signal augmentation after GNP binding amounted to 53.4%, which resulted in sensitivity improvement of the current immunosensor.

      • Direct and precise length measurement of single, stretched DNA fragments by dynamic molecular combing and STED nanoscopy

        <P>A combination of DNA stretching method and super-resolution nanoscopy allows an accurate and precise measurement of the length of DNA fragments ranging widely in size from 117 to 23,130 bp. BstEII- and HindIII-treated lambda DNA fragments were stained with an intercalating dye and then linearly stretched on a coverslip by dynamic molecular combing. The image of individual DNA fragments was obtained by stimulated emission depletion nanoscopy. For DNA fragments longer than 1000 bp, the measured lengths of DNA fragments were consistently within 0.5 to 1.0 % of the reference values, raising the possibility of this method in a wide range of applications including facile detection for copy number variations and trinucleotide repeat disorder.</P>

        Overcoming immunoregulatory plasticity of mesenchymal stem cells for accelerated clinical applications

        <P>Mesenchymal stem cells (MSCs) are multipotent stromal cells with the potential to differentiate into different tissue lineages. In addition to their differentiation potential, MSCs possess immunomodulatory properties that have created growing interest in both pre-clinical and clinical research. Over the years, MSCs have been applied rapidly in the clinic in a wide variety of immune-mediated disorders; however, MSC therapy has shown contradictory results, often with poor clinical outcomes. Recently, studies on MSC-based immune modulation have provided possible explanations for the conflicting clinical reports. It is now generally recognized that the immunomodulatory properties of MSCs are not constitutive but are induced by various mediators present in the inflammatory microenvironment. Different inflammatory stimuli are able to polarize MSCs to elicit distinct immunomodulatory phenotypes. Thus, the concepts of plasticity and polarization of MSC-based immune modulation may have important therapeutic implications in the clinic. In this review, we focus on the underlying mechanisms of MSC-mediated immune regulation that contribute to their therapeutic potential. Importantly, we discuss novel strategic approaches that enhance the therapeutic potential of MSCs through a consideration of MSC plasticity in immune modulation.</P>

      • Improved heat dissipation in a crystalline silicon PV module for better performance by using a highly thermal conducting backsheet

        The temperature of a crystalline silicon photovoltaic module has a strong impact on the electrical performance of the module. The performance can be improved by reducing the temperature of the module. In this study, a highly thermal conducting backsheet was integrated to the mini module, consisting of one single crystalline silicon solar cell, to investigate the temperature and performance changes in the module. By applying a backsheet with graphite and aluminum film, it was found that the final temperature was decreased, compared to a solar cell with a reference backsheet. In addition, providing a suitable heat path for thermal dissipation in the frame of the module has an influence on the effectiveness of the highly thermal conducting backsheet.
