        Human lipocortin-I (LCI) is a calcium ion-dependent and phospholipid-binding protein which exhibits an anti-inflammatory activity by inhibiting phospholipase A₂ activity. In this study, the LCI gene containing its own terminator region was joined to GAL10 promoter-ppL (prepro-leader sequence of mating factor a). An ATG start codon of LCI gene was placed at downstream with KR endoprotease recognition site (Lys-Arg) of ppL. Recombinant S. cerevisiae harboring the LCI expression/secretion vector, pYGLPT5, was aerobically grown on a liquid YPDG medium at 30℃ for 72hrs. The whole cell and culture supernatant were separated after centrifugation, and the expressed LCI was analyzed by SDS-PAGE and western blotting methods. A majority fraction of the expressed LCI was found to be accumulated in the intracellular fraction, resulting in very low secretion efficiency of about 7.4%. About 500mg/ℓof LCI was extracellularly produced by the fed-batch culture employing the controlled-feeding of glucose and galactose. The secreted LCI was purified by ultrafiltration and hydroxylapatite column chromatography, and a purity of more than 99% was obtained.

      • 실리콘의 TMAH 식각을 사용한 Suspended-type 마이크로 가스센서의 제조

        In recent years, different types of microfabricated 'suspended-type' structure have been used as freestanding membranes for a variety of micro gas sensors. We examined the silicon etching characteristics of tetramethyl ammonium hydroxide(TMAH). A suspended type micro gas sensor with platinum heater, sensing electrode and temperature sensor using TMAH etching of silicon was fabricated on silicon nitride(1,500Å)/ silicon dioxide (3,000Å)/ silicon nitride(1,500Å) dielectric membrane. It has low thermal conductivity and balanced stress with silicon substrate for the purpose of improving the thermal isolation between heater and silicon substrate. To avoid any chemical or physical damages to gas sensing material during the photolithography or shadow mask process, the tin dioxide film was deposited onto whole surface of device by using maskless electron beam(e-beam) evaporation technology. The power consumption of the fabricated device was about 40mW at the operating temperature of 250°C. In case of adding Pt(2wt.%) or Pd(2wt.%) catalyst, the sensitivity to CO was higher and the sensitivity to C_(4)H_(10) was higher than those to other gases, respectively.

        In this paper the compact microstrip array antenna with CP(circularly polarized) radiation for GPS(Global Positioning System) is designed. the compact operation of microstrip antenna is obtained by inserting suitable slits in the radiating patch, and CP radiation by giving the unequal length of two pairs of slits, which splits the resonant mode of interest into two orthogonal near degerate modes. Feed network of array antenna is made by using a two-layer structure and via connection probe. By numerical and experimental results, the patch size of compact CP microstrip antenna is reduced about 40% than conventional one.

      • 음운환경을 고려한 음성DB용 단어세트의 설계

        Speech database is a indispensable part of speech research. This paper introduces the PBW(Phonetically Balanced Words) set for Korean speech database. To collect a speech database, all possible phonological phenomenon should be included. In addition, PBW set has the properties that it has minimal nember of words and even probability distribution between phonological events. In this paper, we investigate some considerations to construct word set for Korean speech database. We extract PBW set composed of 452 words from 5,000 high-frequency words and it's properties are presented.

      • 농촌지역 중학생들의 체력과 정신건강에 관한 연구

        This study was performed to investigate the relationship between the physical fitness and mental health state of the rural middle school students. Two-hundred thirty-four middle school students were placed into two gender groups(boys and girls) and/or three age groups(14, 15, and 16 yrs). All subjects completed the physical fitness tests and the mental health questionnaire. After those tests, the data was collected and analyzed by MANOVA and ANOVA, and Pearson's correlation and multiple regression. The conclusions were as follows. First, in consequence of the physical fitness comparison by gender and age, while aging, boys did not show a significant difference in power, balance, ability, and flexibility, having significant improvements in muscle strength, muscular endurance, and cardiovascular endurance. Second, in consequence of the analysis of mental health state by gender, results showed that boys were better than girls in the mental health state, especially boys were better in Somatization, Anxiety, Photic Anxiety, and Psychotism. Third, in consequence of the analysis of mental health status by age, results showed a significant difference in Obsession and Interpersonal Sensitivity among nine symptom check-up lists. The older the students were, the more they were suffering from Obession, and Interpersonal Sensitivity. Finally, in consequence of the analysis of the relationship between physical fitness and mental health status by gender, the physical fitness factors of boy students had no meaningful relationship with mental health factors. On the other hand, girl students had a meaningful relationship between muscle strength and mental health factors-Anxiety, Interpersonal Sensitivity, Hostility, Phobic Anxiety, Paranoid Adaption, and Psycotism. Also girl students had a significant relationship between flexibility and mental health factors-Obsessive, Somatization, Depression, Hostility, Phobic Anxiety, Paranoid Ideation and Psychotism.

      • 도시공학 핵심과목의 심화교육 프로그램 개발에 관한 연구

        본 연구는 핵심과목 요목의 작성과 난이도별 문제은행의 지도과정, 핵심과목에 대한 자율학습 문제은행을 데이타베이스화하여 상시 활용화 추구, 교수와 학생의 상호 협동시스템에 의한 학습방법론 도출을 시도하였다. 시론적 성격에 한정되어 있긴 하나 본 연구는 협동시스템을 기반으로한 자율학습모형을 구축함으로써 학습과 교육효과 제고, 협동심과 선의의 경쟁을 매개로 하는 생활구현, 취업대비 등의 목표에 기여하는데 의의가 있다. This paper examines that drawing up a syllabus of core courses and an item pool classified by the degree of difficulty, and that inquiring practical use an databased item pool, and that proposing study method by cooperative system between students and professors. The result of this study is expected to contribute to studying, life, and taking up an occupation by constructing autonomous study model founded the cooperative system.

      • 溫泉觀光地의 目的地 選擇要因에 관한 硏究

        The purpose of this study was to examine factors of destination choice for hot spring resorts in Korea. Data were collected by questionnaire survey in January 1994, at three typical hot spring resorts : Suanbo, Paikam, and Chuksan. A total of 331 questionnaries were completed by vistiors of three survey sites. The findings of this study were summarized as follows : The seven factors of destnation choice was extracted by factor analysis : 1) the quality of hot spring water, 2) appendant facilites, 3) crowding and cleanness, 4) vicinity tourist area, 5) accessibility and cost, 6) recommendation from friends, 7) popularity. In conclusion, by evaluating the factors, first, endogenous character of resort(e.g. the quality of hot spring water) is the most prefered factor. Second, the factors of destination choice were closely correlated to the incomes of household. Third, the medical benefit of the hot water should be considered when making up hot spring resort.

      • 취업 및 국가기술자격 취득 증진 방안 연구

        본 연구는 도시공학 전공 학생의 국가기술자격 취득 및 취업, 진학 등에 필수적으로 요구되는 도시공학 관련 기초지식을 연마하고 전공지식을 심화 교육함으로서 산·학·연 연계체계에 기초하여 도시공학 전공 학생의 취업증진을 도모하기 위한 종합적이고 구체적인 방안을 모색하고자 하는 것이다. 이를 위하여 본 연구에서는 사회수요에 부응하기 위한 기술교육의 강화, 국가기술자격 취득 제고, 현장실습제도 등 3가지 측면에서 취업증진을 위한 추진실적 및 문제점을 점검하고 이에 대한 향후 개선방안을 모색하고자 하였다. The purpose of this study is to search for the comprehensive and concrete methods of increasing the graduates' employment based on both fundamental and advanced knowledge concerned urban engineering major. For this , existing condition, problems and improvement of students employment are searched in 3 view points such as strengthening of technical education to follow the social demands, enhancement of acquisition of the national qualifying examination and making the best of the field training system.

      • 대학 입학 학력고사의 생물 문항 분석 연구

        Not only in science but also in all subjects, the management of high school curriculum in Korea is received the influence of National Testing for the University Entrance Examination. And this influence has more negative aspects than positive ones. In this view point, this study is to analyze the domains of content, behavior, and context, find out the controversial problems, and make some suggestions for the improvement of the National Testing for the University Entrance Examination. The materials of this study are consist of 372 items in the National Testing for the University Entrance Examination from 1969 to 1992. The tool of this analysis is the "three-dimensional analysis tool of science education" which was set up for this study by means of correcting the fifth NAEP Assessment Frame(NAEP, 1984). To keep the validity of study, "the objectivity of the analysits" was measured before the analysis, and the measured objectivity index of analysits is 90.8%. According to the result of this analysis, in the domain of content many items have been set on the area of "development and genetics" and "structures and functions of organs" and the area of "ecology" is increasing its importance. In the domain of behavior, the items about the knowledge and comprehension covered 70.7% and the items about the science process took only 8.3% in spit of the emphasis in the science curriculum. The domain of value and responsibility of biological science hasn't been treated in the items. This analysis shows the fact that the National Testing for the University Entrance Examination have an influence on the way of the normal management of the curriculum and the achievement of educational objective. In the domain of context, the items of National Testing for the University Entrance Examination pay no attention to the area of context such as technological, social, and individual context. This will lead the science education in high school to ignore important educational function of the problem-solving learning and decision-making and this will inevitably result in the danger of cultivating scientifically illiterate people. On the basis of these analysis, when the National Testing for the and University Entrance Examination is taken into consideration on the aspect of its influence on the high-school curriculum, it should take the following into consideration. First, in the domain of content, it should focus on both the application of the area of bioecological aspect such as 'environmental biology', 'conservation biology', 'behavioral biology' and the area of technoogical aspect such as 'cell biology', 'molecular biology', 'microbiology'. Second, in the domain of behavior, scientific process, the value of science, and the application of science in the technological and social aspects should be considered. Third, in the domain of context, the items about the technological, social, and individual context should be treated. Finally, besides all these suggestions, the introduction of the system for previous problem analysis, the consideration study on the validity, difficulty, and discrimination should be conducted.

      • 灰鑄鐵의 低周波爐 熔解에서 鑄鐵 加工屑 添加에 依한 黑鉛組織 改善

        It is clear that while the use of electric melting and holding makes possible very high levels of casting quality and consistency, there are a number of operational factors to be controlled. A prolonged holding of molten iron at a higher temperature is likely to cause an increase in chilling tendency, tensile strength and hardness with the formation of undercooled structures. The post inoculation of materials such as Ca-Si, Fe-Si or graphite can produce the opposite effect but it is limited in its possible quantity, resulted in drop of the temperature of tapped melt. In contrast the furnace addition of iron chips prior to tapping makes castings of satisfied quality with a high level of uncleation a low chilling tendency and a typed graphites graph in the electric furnace melt by suppling active nuclei materials.
