Detailed morphological analysis of axolotl sperm

Keskin, Ilknur,Gurgen, Duygu Gursoy,Avinca, Didem,Ozdemir, Ekrem Musa,Keskin, Suat Utku,Karabulut, Seda The Korean Society of Veterinary Science 2021 大韓獸醫學會誌 Vol.61 No.3

The axolotl has extraordinary regeneration capacity compared to other vertebrates. This remarkable potential has been attributed to its life-long neoteny, characterized by the exhibition of embryonic characteristics at the adult stage. A recent study provided a detailed morphological analysis of the sperm morphology of the Ambystoma mexicanum using routine and detailed histological techniques. The primary purpose of the present study is to describe a simple and inexpensive method for evaluating the morphology of axolotl sperm. In this study, spermatophore structures were collected and spread on slides and air-dried. The slides were stained with periodic acid Schiff, toluidine blue, Masson's trichrome, Giemsa, Spermac, and Diff-Quik dye for a morphological examination. The slides were coated with gold/palladium for a scanning electron microscopy examination. The sperm of the axolotl consisted of an elongated head, a neck, and a flagellum covered with an undulating membrane. The lengths of the midpiece, tail, and head were 8.575 ㎛, 356.544 ㎛, and 103.661 ㎛, respectively. In the flagellum part, the wavy membrane structure, whose function has not been explained, surrounds the tail. The data obtained from this study will constitute an important step in designing future research on the reproductive and regeneration capacity of the axolotl.

The Surgical Management of Traumatic C6-C7 Spondyloptosis

Keskin, Fatih,Kalkan, Erdal,Erdi, Fatih The Korean Neurosurgical Society 2013 Journal of Korean neurosurgical society Vol.53 No.1

A case of traumatic spondyloptosis of the cervical spine at the C6-C7 level is reported. The patient was treated succesfully with a anterior-posterior combined approach and decompression. The patient had good neurological outcome after surgery. A-51-year-old female patient was transported to our hospital's emergency department after a vehicle accident. The patient was quadriparetic (Asia D, MRC power 4/5) with severe neck pain. Plain radiographs, computerize tomography and spinal magnetic resonance imaging (MRI) showed C6-7 spondyloptosis and C5, C6 posterior element fractures. Gardner-Wells skeleton traction was applied. Spinal alignment was reachived by traction and dislocation was decreased to a grade 1 spondylolisthesis. Then the patient was firstly operated by anterior approach. Anterior stabilization and fusion was firstly achieved. Seven days after first operation the patient was operated by a posterior approach. The posterior stabilization and fusion was achieved. Postoperative lateral X-rays and three-dimensional computed tomography showed the physiological realignment and the correct screw placements. The patient's quadriparesis was improved significantly. Subaxial cervical spondyloptosis is a relatively rare clinical entity. In this report we present a summary of the clinical presentation, the surgical technique and outcome of this rarely seen spinal disorder.

Comparison of the shaping ability of novel thermally treated reciprocating instruments

Keskin, Cangul,Demiral, Murat,Sariyilmaz, Evren The Korean Academy of Conservative Dentistry 2018 Restorative Dentistry & Endodontics Vol.43 No.2

Objectives: The present study aimed to evaluate the shaping ability of 2 thermally treated nickel-titanium reciprocating systems in simulated curved canals. Materials and Methods: Forty simulated canals were prepared to apical size 25 using Reciproc Blue R25 (VDW) and WaveOne Gold Primary (Dentsply Sirona) instruments. Standard pre- and post-preparation images were taken and superimposed. The removal of resin material was measured at 5 standard points: the canal orifice, halfway between the canal orifice and the beginning of the curve, the beginning of the curve, the apex of the curve, and the end-point of the simulated canal. The data were analysed using the independent sample t-test with a 5% significance threshold. Results: The canals in which Reciproc Blue R25 was used showed a significantly greater widening than those in which WaveOne Gold was used at 4 of the 5 measurement points (p < 0.05). The Reciproc Blue R25 instrument removed significantly more resin from the inner aspect of the curve at 2 of the 5 points and similar amounts at the remaining 3 points. At the 2 apical points, there was no significant difference between the Reciproc Blue R25 and WaveOne Gold Primary instruments. Conclusion: Both instruments respected the original canal anatomy; however, WaveOne Gold resulted in a more conservative shape with less transportation.

Experimental study of bearing capacity of strip footing on sand slope reinforced with tire chips

        Keskin, Mehmet Salih,Laman, Mustafa Techno-Press 2014 Geomechanics & engineering Vol.6 No.3

Tire chips and tire chips-soil mixtures can be used as alternative fill material in many civil engineering applications. In this study, the potential benefits of using tire chips as lightweight material to improve the bearing capacity and the settlement behavior of sand slope was investigated experimentally. For this aim, a series of direct shear and model loading tests were conducted. In direct shear tests, the effect of contents of the tire chips on the shear strength parameters of sand was investigated. Different mixing ratios of 0, 5, 10, 15 and 20% by volume were used and the optimum mixing ratio was obtained. Then, laboratory model tests were performed on a model strip footing on sand slope reinforced with randomly distributed tire chips. The loading tests were carried out on sand slope with relative density of 65% and the slope angle of $30^{\circ}C$. In the loading tests the percentage of tire chips to sand was taken as same as in direct shear tests. The results indicated that at the same loading level the settlement of strip footing on sand-tire chips mixture was about 30% less than in the case of pure sand. Addition of tire chips to sand increases BCR (bearing capacity ratio) from 1.17 to 1.88 with respect to tire chips content. The maximum BCR is attained at tire chips content of 10%.

Effects of Remote Ischemic Conditioning Methods on Ischemia-Reperfusion Injury in Muscle Flaps: An Experimental Study in Rats

Keskin, Durdane,Unlu, Ramazan Erkin,Orhan, Erkan,Erkilinc, Gamze,Bogdaycioglu, Nihal,Yilmaz, Fatma Meric Korean Society of Plastic and Reconstructive Surge 2017 Archives of Plastic Surgery Vol.44 No.5

Background The aim of this study was to investigate the effects of remote ischemic conditioning on ischemia-reperfusion injury in rat muscle flaps histopathologically and biochemically. Methods Thirty albino rats were divided into 5 groups. No procedure was performed in the rats in group 1, and only blood samples were taken. A gracilis muscle flap was elevated in all the other groups. Microclamps were applied to the vascular pedicle for 4 hours in order to achieve tissue ischemia. In group 2, no additional procedure was performed. In groups 3, 4, and 5, the right hind limb was used and 3 cycles of ischemia-reperfusion for 5 minutes each (total, 30 minutes) was applied with a latex tourniquet (remote ischemic conditioning). In group 3, this procedure was performed before flap elevation (remote ischemic preconditoning). In group 4, the procedure was performed 4 hours after flap ischemia (remote ischemic postconditioning). In group 5, the procedure was performed after the flap was elevated, during the muscle flap ischemia episode (remote ischemic perconditioning). Results The histopathological damage score in all remote conditioning ischemia groups was lower than in the ischemic-reperfusion group. The lowest histopathological damage score was observed in group 5 (remote ischemic perconditioning). Conclusions The nitric oxide levels were higher in the blood samples obtained from the remote ischemic perconditioning group. This study showed the effectiveness of remote ischemic conditioning procedures and compared their usefulness for preventing ischemiareperfusion injury in muscle flaps.

SOME NEW IDENTITIES CONCERNING THE HORADAM SEQUENCE AND ITS COMPANION SEQUENCE

Keskin, Refik,Siar, Zafer Korean Mathematical Society 2019 대한수학회논문집 Vol.34 No.1

Let a, b, P, and Q be real numbers with $PQ{\neq}0$ and $(a,b){\neq}(0,0)$. The Horadam sequence $\{W_n\}$ is defined by $W_0=a$, $W_1=b$ and $W_n=PW_{n-1}+QW_{n-2}$ for $n{\geq}2$. Let the sequence $\{X_n\}$ be defined by $X_n=W_{n+1}+QW_{n-1}$. In this study, we obtain some new identities between the Horadam sequence $\{W_n\}$ and the sequence $\{X_n\}$. By the help of these identities, we show that Diophantine equations such as $$x^2-Pxy-y^2={\pm}(b^2-Pab-a^2)(P^2+4),\\x^2-Pxy+y^2=-(b^2-Pab+a^2)(P^2-4),\\x^2-(P^2+4)y^2={\pm}4(b^2-Pab-a^2),$$ and $$x^2-(P^2-4)y^2=4(b^2-Pab+a^2)$$ have infinitely many integer solutions x and y, where a, b, and P are integers. Lastly, we make an application of the sequences $\{W_n\}$ and $\{X_n\}$ to trigonometric functions and get some new angle addition formulas such as $${\sin}\;r{\theta}\;{\sin}(m+n+r){\theta}={\sin}(m+r){\theta}\;{\sin}(n+r){\theta}-{\sin}\;m{\theta}\;{\sin}\;n{\theta},\\{\cos}\;r{\theta}\;{\cos}(m+n+r){\theta}={\cos}(m+r){\theta}\;{\cos}(n+r){\theta}-{\sin}\;m{\theta}\;{\sin}\;n{\theta},$$ and $${\cos}\;r{\theta}\;{\sin}(m+n){\theta}={\cos}(n+r){\theta}\;{\sin}\;m{\theta}+{\cos}(m-r){\theta}\;{\sin}\;n{\theta}$$.

Comparison of Different Mathematical Models for Describing the Complete Lactation of Akkaraman Ewes in Turkey

Keskin, Ismail,Dag, Birol Asian Australasian Association of Animal Productio 2006 Animal Bioscience Vol.19 No.11

This study was carried out to investigate the use of three different mathematical models (Wood, Quadratic and Cubic) for describing the lactation curve of Akkaraman ewes. Data were collected from 42 ewes that were three years of age and from the same flock raised in The State Farm of $G{\ddot{o}}zl{\ddot{u}}$ in Konya Province. All ewes lambed in March. They were hand milked twice daily and the first milk test was performed with in the first month after lambing (mean = 27.8 day, SD = 4.26) in an attempt to describe the peak yield. The differences between estimated total milk yields by the models were not significant. The models were adequate for describing total milk yield. The differences between peak yields were not significant. The Wood model estimated the time of peak yield earlier than the other models and observed values (p<0.01). Especially the Cubic model's peak time was very close to really peak time (34.30 vs. 35.33 days). $R^2$ values of the models ranged from 85.85% to 96.20%. The Cubic model gave the best $R^2$ value (p<0.01). Correlation coefficients between descriptive values of the models changed from 0.32 to 1.00. Total milk yields of the models were highly correlated (above 0.99) with the total milk yield calculated by the Fleischmann method (p<0.01). As a result the Cubic model showed the best fit to the data collected from Akkaraman ewes and allowed a suitable description of the shape of the lactation curve.

GENERALIZED FIBONACCI AND LUCAS NUMBERS OF THE FORM wx2 AND wx2 ∓ 1

Keskin, Refik Korean Mathematical Society 2014 대한수학회보 Vol.51 No.4

        Let $P{\geq}3$ be an integer and let ($U_n$) and ($V_n$) denote generalized Fibonacci and Lucas sequences defined by $U_0=0$, $U_1=1$; $V_0= 2$, $V_1=P$, and $U_{n+1}=PU_n-U_{n-1}$, $V_{n+1}=PV_n-V_{n-1}$ for $n{\geq}1$. In this study, when P is odd, we solve the equations $V_n=kx^2$ and $V_n=2kx^2$ with k | P and k > 1. Then, when k | P and k > 1, we solve some other equations such as $U_n=kx^2$, $U_n=2kx^2$, $U_n=3kx^2$, $V_n=kx^2{\mp}1$, $V_n=2kx^2{\mp}1$, and $U_n=kx^2{\mp}1$. Moreover, when P is odd, we solve the equations $V_n=wx^2+1$ and $V_n=wx^2-1$ for w = 2, 3, 6. After that, we solve some Diophantine equations.

Model studies of uplift capacity behavior of square plate anchors in geogrid-reinforced sand

        Keskin, Mehmet S. Techno-Press 2015 Geomechanics & engineering Vol.8 No.4

An experimental investigation into the uplift capacity of horizontal square plate anchors in sand with and without geogrid reinforcement is reported. The parameters investigated are the effect of the depth of the single layer of geogrid, vertical spacing of geogrid layers, number of geogrid layers, length of geogrid layers, the effects of embedment depth, and relative density of sand. A series of three dimensional finite element analyses model was established and confirmed to be effective in capturing the behaviour of plate anchor-reinforced sand by comparing its predictions with experimental results. The results showed that the geogrid reinforcement had a considerable effect on the uplift capacity of horizontal square plate anchors in sand. The improvement in uplift capacity was found to be strongly dependent on the embedment depth and relative density of sand. A satisfactory agreement between the experimental and numerical results on general trend of behaviour and optimum geometry of reinforcement placement is observed. Based on the model test results and the finite element analyses, optimum values of the geogrid parameters for maximum reinforcing effect are discussed and suggested.

      • Antioxidant, Anticancer and Anticholinesterase Activities of Flower, Fruit and Seed Extracts of Hypericum amblysepalum HOCHST

Keskin, Cumali Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.7

Background: Cancer is an unnatural type of tissue growth in which the cells exhibit unrestrained division, leading to a progressive increase in the number of dividing cells. It is now the second largest cause of death in the world. The present study concerned antioxidant, anticancer and anticholinesterase activities and protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in methanol extracts of flowers, fruits and seeds of Hypericum amblysepalum. Materials and Methods: Antioxidant properties including free radical scavenging activity and reducing power, and amounts of total phenolic compounds were evaluated using different tests. Protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in extracts were determined by HPLC. Cytotoxic effects were determined using the MTT test with human cervix cancer (HeLa) and rat kidney epithelium cell (NRK-52E) lines. Acetyl and butyrylcholinesterase inhibitory activities were measured by by Ellman method. Results: Total phenolic content of H. amblysepalum seeds was found to be higher than in fruit and flower extracts. DPPH free radical scavenging activity of the obtained extracts gave satisfactory results versus butylated hydroxyanisole and butylated hydroxytoluene as controls. Reducing power activity was linearly proportional to the studied concentration range: $10-500{\mu}g/mL\;LC_{50}$ values for H. amblysepalum seeds were 11.7 and 2.86 respectively for HeLa and NRK-52E cell lines. Butyryl-cholinesterase inhibitory activity was $76.9{\pm}0.41$ for seed extract and higher than with other extracts. Conclusions: The present results suggested that H. amblysepalum could be a potential candidate anti-cancer drug for the treatment of human cervical cancer, and good source of natural antioxidants.
