      • High Level Production of Glycoprotein H of HSV-1 (F) Using HcNPV Vector System

        The Herpes simplex virus type 1 (HSV-1) strain F glycoprotein H (gH) gene in the pHLB-4 plasmid was recombinated into a baculovirus expression vector (lacZ-HcNPV) to construct a recombinant virus GH-HcNPV expressing gH. The sequences of gH and its expression were analyzed. The gH gene was located in the 6.41 kb BglII fragment. The open reading frame (ORF) of the gH gene was 2,517 bp and codes 838 amino acid residues. Insect cells infected with this recombinant virus synthesized a high level of the matured and gX-gH fusion protein with approximately 112 kDa. The fusion gH protein was localized on the membrane of the insect cells as seen by using immunofluorescence assay and accumulated in the cultured media by the SDS-PAGE and immunoprecipitation assays. The amino acid sequence presents additional characteristics compatible with the structure of a viral glycoprotein: signal peptide, putative glycosylation sites and a long C-terminal transmembrane sequence. Antibodies raised in mice to this recombinant protein recognized viral gH and neutralized the infectivity of HSV-1 in vitro. These results demonstrate that it is possible to produce a mature protein by gene transfer in eukaryotic cells, and indicate the utility of the HcNPV-insect cell system for producing and characterizing eukaryotic proteins. Furthermore, the neutralizing antibodies would appear to protect mice against HSV; accordingly, this particular recombinant protein may be useful in the development of a subunit vaccine.

        Trial sequential analysis: novel approach for meta-analysis

        Systematic reviews and meta-analyses rank the highest in the evidence hierarchy. However, they still have the risk of spurious results because they include too few studies and participants. The use of trial sequential analysis (TSA) has increased recently, providing more information on the precision and uncertainty of meta-analysis results. This makes it a powerful tool for clinicians to assess the conclusiveness of meta-analysis. TSA provides monitoring boundaries or futility boundaries, helping clinicians prevent unnecessary trials. The use and interpretation of TSA should be based on an understanding of the principles and assumptions behind TSA, which may provide more accurate, precise, and unbiased information to clinicians, patients, and policymakers. In this article, the history, background, principles, and assumptions behind TSA are described, which would lead to its better understanding, implementation, and interpretation.

      • An Empirical Study on the Effects of Official Mentoring System Exercise on Employees' Organization Commitments through Trust

        This research was conducted to find the importance of a mentor in restaurant businesses. 178 people working for companies which operate formal mentoring system were selected and a positive experiment was executed to verify the mutual relation between mutual trust and organization conrritrrent of errployees in rrentoring. The surrrmry follows below. It was found out that the career development functions influence the trust among organizations, inferiors, and colleagues in the connection between the perception and mutual trust about the mentoring system. The social psychological functions do among organizations, staffs, and colleague. And then, for the role model, in case of mentor do not have huge impacts on the trust among superiors and colleagues but on trust in group of constitutions. It is believed that the origins of these results are lack of recognition about mentors' capability in official mentoring. There should be some supports on mentors.

        지방자치단체 공직부패방지 정책수단 확산 연구 : 사건사 분석을 통한 신고포상금제도를 중심으로

        신고포상금제도는 정책수단으로서 중앙정부-지방정부, 지방정부-지방정부 간 정책확산이 이루어진 대표적인 사례이다. 본 연구는 지방정부의 신고포상금제도 도입·운영을 정책확산이론의 관점으로 설명할 수 있는지 살펴보고, 사건사 분석을 통해 신고포상금제도의 확산 메커니즘을 분석하였다. 특히, 동 제도의 확산을 시간적 차원, 공간적 차원, 내용적 차원으로 구분하여 확산 다이나믹스(dynamix)를 증명하고자 하였다. 시간적 차원의 Kaplan-Meyer 생존분석결과 누적생존 확률과 확산은 S-곡선을 보이며 나타났고, 공간적 차원의 분석에서도 Moran′s Index 값이 양의 관계를 보였다. 내용적 차원의 분석에서는 Cox 회귀분석결과 제도도입에서는 상위정부의 도입권고(압력효과), 인접 지자체 도입여부(인접효과), 타 신고포상제 도입경험(학습효과) 등이 통계적으로 유의미하게 나타났다. 한편 제도운영에서는 상위정부의 도입권고, 청렴도, 타 신고포상제 도입경험이 영향을 미치는 것으로 분석되었다. Report Bounty System(RBS) is a representative case of the policy diffusion in the government. This study examined whether the introduction and operation of the RBS of local governments can be explained from the perspective of policy diffusion theory, and analyzed the mechanism of spreading the RBS through event history analysis. In particular, the diffusion of the system was divided into time, spatial, content dimensions to prove the diffusion dynamics. The Kapplan-Meyer survival analysis of the time dimension showed that the cumulative survival probability and diffusion showed a S-curve, and the Moran′ s Index value showed a positive relation in the spatial dimension analysis. In the analysis of the context level, the Cox regression analysis showed that the introduction recommendation(pressure effect), the introduction of neighboring local governments(neighbor effect), and the introduction of other RBS(learning effect) were statistically significant in the introduction of the system. On the other hand, in the system operation, the recommendation of the upper government, the integrity index, and the experience of introducing other RBS were influenced.

        소파 방정환의 일본 유학 시기 활동

        방정환은 1920년 9월부터 1923년 9월까지 만 3년간 일본에서 활동하였다. 지금까지 방정환의 일본 활동은 도요대학의 유학 생활로 알려졌지만 이는 방정환의 3년간의 일본 생활의 일부분이었다. 방정환은 기본적으로 개벽사 동경특파원으로 활동하였다. 그는 3년간의 일본 생활을 통해 본명과 필명으로 21회에 걸쳐 11편의 글을 『개벽』에 발표하였다. 천도교청년회 창립의 중심 멤버였던 방정환은 동경에 도착한 직후부터 천도교 유학생을 모아 천도교청년회 동경지회를 조직하고 초대 지회장을 맡았다. 그는 자신의 하숙집인 계림사에서 천도교청년회 동경지회의 준비모임과 종교행사를 거행하며 1921년 2월부터 동경지회 활동을 전개하였으며 천도교의 최고기념일인 4월 5일의 천일기념일에 발회식을 가졌다. 동경지회장으로 활동하던 방정환은 1923년 어린이 운동에 전념하기 위해 지회장에서 물러났다. 방정환은 1921년 동경연수영어학교를 다니면서 정식으로 일본의 대학에 입학하려 하였지만 민원식 살해사건으로 20여 일간 구금되어 조사를 받았다. 이 여파로 방정환은 도요대학 문화학과의 청강생으로 입학하여 조선인 유학생들과 친분을 맺고, 이들과 함께하기 방학을 맞아 국내에 순회 강연대를 조직하여 유학생으로서 학문에만 전념하는 것이 아니라 사회계몽에도 적극적으로 앞장섰다. 방정환은 1922년 3월 30일 도요대학을 자퇴함에 따라서 그의 유학 생활은 1년이었다. 그의 자퇴 이유 역시 어린이 운동에 전념하기 위한 것이었다. 방정환은 일본에서 아동문학에 관해 관심을 갖고 당시 일본의 아동문학과 세계의 동화를 공부하였다. 그 결과 1921년 말에 세계의 동화를 선정해 번안한 『사랑의 선물』을 탈고하였고, 1922년 7월 국내에서 발행하였다. 『사랑의 선물』 출판은 방정환이 아동문학가로 인식되는 계기가 되었다. 방정환은 1921년 5월 1일 천도교청년회에서 분립한 천도교소년회를 중심으로 김기전과 함께 어린이운동에 매진하였다. 또 방정환은 일본에 있으면서 색동회를 조직해 어린이 운동을 뒷받침할 수 있는 인재를 양성하였다. 방정환은 어린이라는 용어를 대중화하고 어린이날을 제정해 어린이 운동에 앞장섰다. 그는 1923년에 들어와서는 『어린이』의 발간과 색동회 활동에 전념했다. 방정환은 1923년 9월 1일 관동대지진을 계기로 국내에 정착하였다. 그는 3년간 일본에서 활동했지만 1/3인 12개월 가량은 국내에서 활동하였다. 3년간 방정환은 국내와 일본을 오가며 개벽사 특파원으로, 천도교동경지회의 지회장으로, 천도교소년회의 핵심 지도자로 어린이날과 『어린이』를 창간하고, 색동회 회원으로 활동하였다. 방정환은 일본 활동 시기를 통해 자신의 활동 방향을 어린이 운동으로 정립하였다는 점이 가장 큰 업적이라고 할 수 있다. Bang Jung-Hwan was active in Japan for three years from September 1920 to September 1923. Until now, Bang Jung-Hwan’s activities in Japan were known as studying abroad at Toyo University, but this was a part of Bang Jung-Hwan’s three-year life in Japan. Bang Jung-Hwan basically worked as a Tokyo correspondent for Gaebyeoksa. Through his three years of living in Japan, he published 11 articles in ‘Gaebyeok’, 21 times under his real name and pseudonym. Bang Jung-Hwan, who was a central member of the founding of the Chondogyo Youth Association, gathered Chondoist international students right after arriving in Tokyo to organize the Tokyo Branch of the Chondogyo Youth Association and served as the first branch president. He held preparatory meetings and religious events for the Tokyo Branch of the Chondogyo Youth Association at his boarding house, Gyerimsa, and conducted the activities of the Tokyo Branch from February 1921. Bang Jung-Hwan resigned from the branch president in 1923 to focus on the children’s movement. In the second half of 1920, while attending the Donggyeong Language Training School, Bang Jung-Hwan tried to formally enter a Japanese university, but was rescued for 20 days due to Min Won-Sik’s murder case and was investigated. In the aftermath of this, Bang Jung-Hwan entered the Department of Culture at Toyo University as an audit student and became close with Korean international students. took the lead As Bang Jung-Hwan dropped out of Toyo University on March 30, 1920, his study abroad life was one year. His reason for dropping out was to devote himself to the children’s movement. Bang jung-hwan was interested in children’s literature in Japan and studied Japanese children’s literature and world fairy tales at the time. As a result, at the end of 1921, he published ‘The Gift of Love’, which was adapted by selecting fairy tales from around the world, and was published in Korea in June 1922. The publication of ‘The Gift of Love’ made Bang Jung-Hwan recognized as a children’s writer. Bang Jung-Hwan devoted himself to the children’s movement together with Kim Ki-Jeon, centering on the Chondogyo Youth Association, which was spun off from the Chondogyo Youth Association on May 1, 1921. In addition, while in Japan, Bang Jung-Hwan organized the Saekdong Association to foster talents who could support the children’s movement. Bang Jung-Hwan popularized the term children and established Children’s Day, leading the children’s movement. In 1923, he devoted himself to the publication of ‘Children’ and the activities of the Saekdonghoe. Bang Jung-Hwan settled in Korea in the wake of the Great Kwandong Earthquake on September 1, 1923. He was active in Japan for three years, but he was active in Korea for about one-third (12 months). For three years, Bang Jung-Hwan traveled back and forth between Korea and Japan, serving as a correspondent for Gaebyeoksa, as the branch president of the Chondogyo Tokyo branch, and as the core of the Chondogyo Youth Association, he founded Children’s Day and ‘Children’ and worked as a member of the Saekdonghoe. It can be said that Bang Jung-Hwan’s greatest achievement is that he established the direction of his activities as a children’s movement through his Japanese career.

      • Development of biofilm on the fractured rock under oligotrophic environment : a experimental study using in-situ reactor

        Development of biofilm in the subsurface environment occurs as a result of complex interaction between indigenous microbes and various physicochemical characteristics of surrounding media. Furthermore, growth and decay of the biofilm can also alter the hydrological properties of both fractured and porous media. Many recent studies focused on biofilm formation under static conditions with sufficient supply of nutrients. However, development of the biofilm in deep subsurface environment could indicate different characteristics and morphology because of the lack of energy and nutrient sources. Therefore, oligotrophic, and continuous flow conditions should be considered to simulate the biofilm growth at deep subsurface and investigate the contributing factors. In this study, we conducted a long-term in-situ experiment using reactor to reproduce the biofilm development in deep fractured rock media. The morphology and microbial community of the biofilm will be compared along with the types of rock and different geochemical conditions. Two replicates of biofilm reactors were constructed using fractured gneiss and sandstone cores. Each reactor was placed in two adjacent groundwater wells with different depth: 1) bedrock groundwater (depth= 100 m) in anaerobic condition (dissolved oxygen (DO) < 0.6 mg/L), and 2) alluvial groundwater (depth= 30 m) in aerobic condition (DO= ~5.6 mg/L). Both groundwater wells indicated neutral pH (6.7 and 6.3, respectively). The first samples were taken after 11 months from the installation. Visualization of the biofilm using confocal laser scanning microscopy (CLSM) revealed distinct morphology of biofilm along rock types and oxygen levels. Biofilm on the surface of gneiss showed a thin layered structure of 10-20 μm thick, compared to sandstone which indicated randomly distributed pattern. In both gneiss and sandstone, the proportion of dead cells consisting of biofilm was increased at the samples from the bedrock layer. Subsamples were taken at the fracture surface to analyze microbial community using Illumina MisSeq sequencing of 16S rRNA genes. With these results, we will discuss the microbial communities preferentially attaching to different rocks, and the geochemical factors affecting the biofilm formation and community distribution.

        Prosthetic management of a growing patient with Russell-Silver syndrome: a clinical report

        Russell-Silver syndrome (RSS) is a congenital disease characterized by short stature due to growth hormone deficiency, physical asymmetry, inverted triangular face, micrognathia, prominent forehead, and hypodontia. This case report presents a prosthetic management of a 6-year-old patient with Russell-Silver syndrome treated with overdentures on the maxilla and the mandible using the remaining primary teeth. Subsequent and comprehensive dental management considering the growth and development of a young patient will be necessary.
