        Endothelin Receptor Overexpression Alters Diastolic Function in Cultured Rat Ventricular Myocytes

        The endothelin (ET) signaling pathway controls many physiological processes in myocardium and often becomes upregulated in heart diseases. The aim of the present study was to investigate the effects of ET receptor upregulation on the contractile function of adult ventricular myocytes. Primary cultured adult rat ventricular myocytes were used as a model system of ET receptor overexpression in the heart. Endothelin receptor type A (ETA) or type B (ETB) was overexpressed by Adenoviral infection, and the twitch responses of infected ventricular myocytes were measured after ET-1 stimulation. Overexpression of ETA exaggerated positive inotropic effect (PIE) and diastolic shortening of ET-1, and induced a new twitch response including twitch broadening. On the contrary, overexpression of ETB increased PIE of ET-1, but did not affect other two twitch responses. Control myocytes expressing endogenous receptors showed a parallel increase in twitch amplitude and systolic Ca2+ in response to ET-1. However, intracellular Ca2+ did not change in proportion to the changes in contractility in myocytes overexpressing ETA. Overexpression of ETA enhanced both systolic and diastolic contractility without parallel changes in Ca2+. Differential regulation of this nature indicates that upregulation of ETA may contribute to diastolic myocardial dysfunction by selectively targeting myofi lament proteins that regulate resting cell length, twitch duration and responsiveness to prevailing Ca2+.

        Forest therapy program reduces academic and job-seeking stress among college students

        Background and objective: Recreation or activities in forest are regarded as therapy. Many forest therapy programs have been developed and assessed in the domestic. This study was conducted to investigate the effect of the forest therapy program on academic and job-seeking stress in college students. Methods: Thirty five subjects were selected as the experimental group and 25 as the control group, and 29 subjects in the experimental group and 11 in the control group participated in the follow-up test to verify the persistence of stress reduction effects. The forest therapy program was carried out once a week for 2 hours each from September 4 to December 4, 2018, adding up to total eight sessions. Results: The experimental group showed statistically significant reduction in both academic stress and job-seeking stress, whereas the control group did not. For the persistence of the forest therapy program, the experimental group did not show a statistically significant difference between the posttest and the follow-up test, and thus the stress reduction effect was maintained. Conclusion: This study proved the reduction of academic and job-seeking stress in forest therapy programs and the persistence of the stress reduction effect of the forest therapy program. The result is consistent with the Stress Recovery Theory (SRT) that shows the stress reduction effect of nature. In addition, it has significance in that it has verified that the program using the forest on campus can reduce stress of most college students.

        Effects of dietary supplementation with probiotic CS-A on performance in broiler chickens

        Probiotics, enzymes, organic acids, oligosaccharides, antioxidants, and other functional materials are actively being explored as alternatives to antibiotics. Probiotics include live beneficial microorganisms that colonize the intestinal tract and competitively inhibit attachment and growth of harmful microbes. Probiotics also increase feed efficiency by assisting in nutrient absorption and digestion. The current study was conducted in order to evaluate the effect of a new probiotic, CS-A, as a dietary supplement of a fermented product on growth performance, feed intake, and feed conversion efficiency in broiler chickens, and to evaluate its value as an alternative for antibiotics used as a feed additive. Antibacterial and anti-inflammatory effects of CS-A were investigated in vitro and the in vivo effects of a constant concentration of supplemented CS-A on growth rate and feed efficiency were evaluated. In addition, the safety of CS-A was assessed by examination of common symptoms and mortality. Determination of minimal inhibitory concentration revealed an excellent antibacterial effect of CS-A. Cytotoxicity was low and anti-inflammatory effects were achieved at the effective concentration of CS-A. Supplementation with 0.1% CS-A resulted in a feed efficiency score of 1.84 in broilers, compared to 2.00 in the control group. There were no adverse clinical findings, necropsy findings, hematology, and altered serum biochemistry parameters, and no mortality. Thus, it is concluded that CS-A is safe and effective as a feed additive.

        Immunohistochemical study on the expression of calcium binding proteins (calbindin-D28k, calretinin, and parvalbumin) in the cerebral cortex and in the hippocampal region of nNOS knock-out(−/−) mice

        Nitric oxide (NO) modulates the activities of various channels and receptors to participate in the regulation of neuronal intracellular Ca2+ levels. Ca2+ binding protein (CaBP) expression may also be altered by NO. Accordingly, we examined expression changes in calbindin-D28k, calretinin, and parvalbumin in the cerebral cortex and hippocampal region of neuronal NO synthase knockout(−/−) (nNOS−/−) mice using immunohistochemistry. For the first time, we demonstrate that the expression of CaBPs is specifically altered in the cerebral cortex and hippocampal region of nNOS−/− mice and that their expression changed according to neuronal type. As changes in CaBP expression can influence temporal and spatial intracellular Ca2+ levels, it appears that NO may be involved in various functions, such as modulating neuronal Ca2+ homeostasis, regulating synaptic transmission, and neuroprotection, by influencing the expression of CaBPs. Therefore, these results suggest another mechanism by which NO participates in the regulation of neuronal Ca2+ homeostasis. However, the exact mechanisms of this regulation and its functional significance require further investigation.

      • Consumer Recognition Survey for Establishing the Reasonable Expiration Dates of Health Functional Foods

        This study investigated consumers` recognition on shelf life of health functional foods to set up their rational expiration date. For this study 107 male and female adults aged 20 to 59 years were randomly selected as major consumers of health functional foods. And their data were analyzed with SPSS 17.0. The questionnaire included questions asking a consumer`s recognition on the shelf life of health functional foods and how to treat health function food passing its shelf life. In this study, the consumers were found to pay attentions considerably to the shelf life. But most of them did not know an accurate meaning of the shelf life of health functional food and mistook it for expiry date. In addition, the consumers doubted safety of health functional foods passing the shelf life. Therefore, education for consumers about an accurate meaning of the shelf life was needed. The results of this study were considered to be helpful to set up a reasonable expiration date for health functional food.

        A comparative study of pulp vitality following vital pulpotomy using calcium hydroxide-based sealants in beagle dogs

        Vital pulpotomy is a very useful method for disarming of canine tooth, tooth fracture, periodontitis, and malocclusion in veterinary dentistry. Calcium hydroxide is the material commonly used as a liner during vital pulpotomy. This creates a mineralized barrier by stimulating osteoblastic hard tissue repair, arrests the inflammatory response, and soothes dentin. However, the powder or mix type calcium hydroxide materials have many disadvantages due to complicated procedures for use and are hard to handle when vital pulpotomy is followed under general anesthesia in animals. This study was conducted in order to compare the effect of mix and premixed paste type calcium hydroxide as a liner in vital pulpotomy. Six beagle dogs underwent hemisection on the mesial root of the mandibular first molar and vital pulpotomy on the distal root of the first molar. On the distal root of the left and right mandibular first molar, mix type (DYCAL®, Dentsply, USA) and premixed paste type calcium hydroxide (VITAPEX®, Morita, Japan) were used as liners, respectively. Radiological evaluation was performed at immediate, 4, 12, and 20 weeks after vital pulpotomy. According to the results, all teeth had well-formed dentinal bridges, and there were no periradicular lucency, lamina dura loss, or anomalies of the pulp cavity. According to these results, on vital pulpotomy in animals, premixed paste type calcium hydroxide was easy to handle and decreased the anesthesia period due to a more convenient application procedure. A further study of many clinical cases is needed for evaluation of side effects and other problems.

        Anti-osteoporotic Effects of Sericin-Calcium Conjugate in Ovariectomized Rats

        This study was conducted in order to evaluate the effect of sericin-calcium (SC) as therapy for ovariectomy-induced osteoporosis in rats. Three weeks after ovariectomy (OVX), Sprague-Dawley rats were divided randomly into three groups: sham-operated group (Sham), ovariectomized group, and SC-treatment group (OVX+SC). Rats in the OVX+SC group were given drinking water containing 0.07% SC for eight weeks. Bone breaking force, mineralization, and blood parameters related to bone metabolism were analyzed. In OVX animals, blood concentration of 17β-estradiol showed a significant decrease, while osteocalcin and type I collagen C-terminal telopeptides (CTx) showed an increase. Breaking force of femurs as well as bone mineral density (BMD), ash, calcium, and phosphorus in femurs showed a significant decrease following OVX. Treatment with SC (0.07% in drinking water) resulted not only in remarkable restoration of the decreased 17β-estradiol and increased osteocalcin and CTx concentrations, but also led to recovery of decreased femoral breaking force, BMD, ash, calcium, and phosphorus. It is suggested that SC effectively improves bone density by preventing bone turnover-mediated osteocalcin, CTx, and minerals, and that it could be a potential candidate for use in therapy or prevention of post-menopausal osteoporosis.

      • RE-4 : Corifollitropin alfa versus daily recombinant fsh treatment for controlled ovarian stimulation in poor responders

        목적: To compare the effect of long-acting recombinant FSH (rFSH), corifollitropin alfa with daily rFSH administered during 1st 7days of controlled ovarian stimulation (COS) using GnRH antagonist multiple dose protocol (MDP) in poor responders undergoing IVF/intracytoplasmic sperm injection (ICSI). 방법: In this prospective randomized study total of 90 poor responders who were scheduled for IVF/ICSI were enrolled and divided into the corifollitropin alfa group (study group) or the daily rFSH group (control group). The diagnosis of poor responder was based on the Bologna criteria of the 2011 ESHRE consensus. In all subjects, GnRH antagonist MDP was used for COS. 결과: There were no differences in patients` characteristics between the two groups. Duration of COS was comparable between the study and control groups. The numbers of oocytes retrieved and grade I or II embryos were also similar in the two groups. However, the number of mature oocytes was higher in the study group with a borderline significance (p< .07) and the ratio of mature oocytes among total oocytes retrieved was significantly higher in the study group of 78.6 ± 13.0%, compared with 61.2 ± 16.0% in control group (p< .001). There were no differences in the clinical pregnancy rate per cycle initiated, embryo implantation rate and miscarriage rate between the two groups. 결론: Corifollitropin alfa is at least as effective as daily rFSH treatment for COS using GnRH antagonist MDP, and may improve the synchrony of follicular development in poor responders undergoing IVF/ICSI. Therefore, corifollitropin alfa may be a feasible, patient-friendly alternative for poor responders.
