Liquid Chromatography-tandem Mass Spectrometry for Quantification of Dioscin in Rat Plasma

Dioscin is a biologically active steroidal saponin with anticancer and hepatoprotective effects. A rapid, selective, and sensitive liquid chromatographic method with electrospray ionization tandem mass spectrometry was developed for the quantification of dioscin in rat plasma. Dioscin was extracted from rat plasma using ethyl acetate at acidic pH. The analytes were separated on a Halo C18 column using gradient elution of acetonitrile and 0.1% formic acid and detected by tandem mass spectrometry in selected reaction monitoring mode. The standard curve was linear (r2 = 0.998) over the concentration range of 1.100 ng/mL. The lower limit of quantification was 1.0 ng/mL using 50 μL of plasma sample. The coefficient of variation and relative error for intra- and inter-assay at four QC levels were 1.3 to 8.0% and .5.4 to 10.0%, respectively. This method was applied successfully to the pharmacokinetic study of dioscin after oral administration of dioscin at a dose of 29.2 mg/kg in male Sprague-Dawley rats.

<P>The ZrKClO4 particle is an energetic material used as a propellants in pyrotechnic applications. When hundreds of nm sized ZrKClO4 particles are ignited with electric shock, an oxygen atom which is bonded to an oxidizer (KClO4) obtains activation energy and becomes free oxygen atom resulting in the oxidation of Zr to form ZrO2. Energy generated from this oxidation reaction activates another near by oxygen atom and by a subsequent chain reaction the ZrKClO4 particles very rapidly release energy. Because this phenomenon occurs under anaerobic conditions, it is applied to combustion of spacecraft and rocket that need to be propelled under anaerobic conditions. Due to a growing interest in the aerospace industry, it is necessary to develop a more accurate energy release model of ZrKClO4 nanoparticles considering particle size and microscopic molecular structures in order to be used in various researched applications such as energy material performance evaluation tests and aging studies. In this work, a mathematical model describing the energy release of ZrKClO4 nano particles is suggested and compared with experiments.</P>
Mathematical and Experimental Study for Mixed Energetic Materials Combustion in Closed System

Modelling the energy release performance of energetic material combustion in closed systems is of fundamental importance for aerospace and defense application. In particular, to compensate for the disadvantage of the combustion of single energetic material and maximize the benefits, a method of combusting the mixed energetic materials is used. However, since complicated heat transfer occurs when the energetic material is combusted, it is difficult to theoretically predict the combustion performance. Here, we suggest a theoretical model to estimate the energy release performance of mixed energetic material based on the model for the combustion performance of single energetic material. To confirm the effect of parameters on the model, and to gain insights into the combustion characteristics of the energetic material, we studied parameter analysis on the reaction temperature and the characteristic time scales of energy generation and loss. To validate the model, model predictions for mixed energetic materials are compared to experimental results depending on the amount and type of energetic material. The comparison showed little difference in maximum pressure and the reliability of the model was validated. Finally, we hope that the suggested model can predict the energy release performance of single or mixed energetic material for various types of materials, as well as the energetic materials used for validation.

<P><B>Abstract</B></P> <P>The profiling of fatty acids (FAs) or sterols has been applied in clinical studies, but still needs to be improved to enable their simultaneous quantification. Moreover, little progress has been made in determining the levels of FAs and sterols in human saliva in a single run. In this study, gas chromatography-tandem mass spectrometry (GC–MS/MS) using one-step <I>tert</I>-butyldimethylsilyl (TBDMS) derivatization was developed for comprehensive profiling of 18 FAs (eight saturated, five monounsaturated, and five polyunsaturated FAs) and 7 sterols (cholesterol and its precursors). The TBDMS derivatization process was also optimized in terms of reaction solvent, catalyst, temperature, and reaction time. The optimized conditions resulted in better derivatization efficiency with good chromatographic separation through a high-temperature column within 23 min. The present method provided good linearity (<I>r</I> > 0.993), precision (coefficient of variation, 2.7% to 10.4%), and accuracy (91.5% to 103.4%). The overall recovery ranged from 73.8% to 114.3% for the 18 FAs, and from 68.9% to 79.8% for the 7 sterols. The validated method was applied to characterize FAs and sterols in human saliva samples. This is the first report of a GC–MS/MS method for the simultaneous determination of various FAs and sterols from a small volume (100 μL) of saliva. This approach can be used as a primary screening tool to examine the levels of both FAs and sterols in saliva, providing detailed information about their homeostasis for diagnostic and prognostic purposes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Both fatty acids and sterols were simultaneously measured from a small volume of human saliva. </LI> <LI> One-step <I>tert</I>-butyldimethylsilylation was developed for the simultaneous quantification of fatty acids and sterols. </LI> <LI> This GC-MS/MS method is a powerful analytical tool to measure salivary fatty acids and sterols in a single run. </LI> </UL> </P>
블랙푸드가 첨가된 약콩청국장분말의 영양소 및 생리활성물질 분석

본 연구는 대두청국장분말, 약콩청국장분말, 그리고 흑미, 흑임자, 다시마와 같은 블랙푸드를 첨가한 약콩청국장분말의 일반성분과 영양소 및 항산화 물질을 분석ㆍ비교하여 향후 알츠하이머성 치매관리나 인지능력 향상을 위한 블랙푸드가 첨가된 청국장분말의 in vivo 실험을 위한 기초자료로 활용하고자 실시하였다. 그 결과 대두와 약콩청국장에 비해서 블랙푸드를 첨가한 약콩청국장에서 수분함량이 높았으며, 유리당 중 sucrose, fructose, glucose, maltose 함량이 높았다. 또한 수용성과 조단백질 함량은 P<0.05 수준에서 약콩청국장과 블랙푸드를 첨가한 약콩청국장에서 높았다. 불포화지방산과 포화지방산의 비율은 대두청국장에 비해서 약콩청국장과 블랙푸드를 첨가한 약콩청국장에서 높았다. 무기질 함량은 대두청국장이 가장 높았으나 약콩청국장에서는 Zn, Cr, Mn이, 블랙푸드를 첨가한 약콩청국장에서는 Fe의 함량이 높았다. Isoflavone 중 diadzein 함량은 약콩청국장에서 가장 높았으나, genistein과 anthocyanin 함량은 블랙푸드를 첨가한 약콩청국장에서 가장 높았다. 따라서 본 연구에서는 일반 및 영양적 특성에서 대두청국장에 비해서 블랙푸드로 만든 청국장분말이 유리당과 뇌조직의 구성과 기능에 필요한 수용성 및 단백질 함량과 불포화지방산과 포화지방산의 비율이 높을 뿐만 아니라 우수한 항산화력으로 보고되고 있는 diadzein, genistein 그리고 anthocyanin 함량이 높은 것으로 나타났다. 그러므로 향후 알츠하이머성 치매의 원인 및 치료기전으로 보고되고 있는 산화적 스트레스조절에 대한 in vivo 실험에 블랙푸드청국장분말을 활용할 수 있을 것으로 판단된다. This study is carried out to investigate the analysis of nutrition and antioxidants of soybean Chungkukjang powder (SCP), Yak-Kong Chungkukjang powder (YCP) and Yak-Kong Chungkukjang powder added black foods (YCBP) for the management of Alzheimer"s disease. The water content of YCBP was higher than that of SCP and YCP. The soluble and crude protein contents of YCP were the highest among three Chungkukjang powders. In SCP, YCP and YCBP, the ratios of unsaturated and saturated fatty acids were 5.17, 5.76, and 5.78, respectively. The mineral content of SCP was higher than that of YCP and YCBP. Antioxidants analysis showed that the content of diadzein was the highest in YCP and genistein, and anthocyanin contents were higher than the others in YCBP. In conclusion, YCP and YCBP had higher contents than SCP in soluble and crude proteins, and unsaturated fatty acids, which are needed for composition and function of the brain tissue. Also, it was found that the contents of diadzein, genistein and anthocyanin with outstanding antioxidative ability were high. Thus, this study suggests that Chungkukjang powder, which is made with Yak-Kong, black sesame, black rice and sea tangle, can be utilized for in vivo experiment for the control of oxidative stress, reported as cause and therapy for Alzheimer"s Disease.
