The effect of neuropeptides on secretion of Interleukin-8(IL-8)

본 연구는 치수조직, 치은, 치주인대로부터 배양된 조직을 SP (Substance P)로 4시간, SP, CGRP (Calcitonin Gene-related Peptide), Tumor necrosis factor-α (TNF-α)로 8시간 자극 후 RNase Protection Assay를 시행하고, IL-8의 분비량을 측정해 다음 결과를 얻었다. 1. IL-8 mRNA는 모든세포에서 발현됐다. 2. IL-8 mRNA 발현은 SP (10^(-5)M)와 SP (10^(-8)M)로 4시간 자극 시 증가되지 않았다. 3. IL-8 mRNA 발현은 SP (10^(-4)M)와 CGRP (10^(-6)M)로 8시간 자극 시 증가되지 않았다. 4. TNF-α (2 ng/㎖) 자극 시, IL-8 mRNA 발현이 증가됐다. 5. 치은 세포를 CGRP (10^(-6)M)로 8시간 자극 시, IL-8 분비량이 증가했다 (p < 0.05). 6. 치주인대 세포를 SP (10^(-4)M)로 8시간 자극 시 IL-8 분비량이 증가했다 (p < 0.05). We investigated the secretion of Interleukin-8 (IL-8) from ginviva and periodontal ligament stimulated with Substance P (SP) and Calcitonin Gene-related Peptide (CGRP). Gingiva (GF), periodontal ligament (PDLF) and pulp (PF) tissues were collected from extracted instact 3rd molars. Cultured cells were stimulated with different concentrations of SP for 4 hrs, and stimulated with SP, CGRP and Tumor Necrosis Factor-α (TNF-α) for 8 hrs. Then RNase Protection Assay was carried out. ELISA was performed using supernatants of stimulated cells for quantitative analysis of IL-8. Results were assessed using supernatants of stimulated cells for quantitative analysis of IL-8. Results were assessed using student t-test with significance of P < 0.05. According to this study, the results were as follows: 1. IL-8 mRNA was detected in all type of cells studied (PF, GF and PDLF). 2. IL-8 mRNA expression was not increased after stimulating 4 hrs with SP (10^(-5)M) and SP (10^(-8)M) compared with Mock stimulation in all type of cells studied. 3. IL-8 mRNA expression was not increased after stimulating 8 hrs with SP (10^(-4)M) and CGRP (10^(-6)M) compared with Mock stimulation in all type of cells studied. 4. TNF-α (2 ng/㎖) increased the expression of IL-8 mRNA in all kind of cells studied. 5. The secretion of IL-8 from GF was increased 8 hrs after the stimulation with CGRP (10^(-6)M) (p < 0.05). 6. The secretion of IL-8 from PDLF was increased 8 hrs after the stimulation with SP (10^(-4)M) (p < 0.05). Calcitonin Gene-related Peptide (CGRP) increased Interleukin-8 (IL-8) which plays an important role in chemotaxis of neutrophil in Calcitonin Gene-related Peptide (CGRP) gingival tissue, whereas Substance P increased the secretion of IL-8 from periodontal ligament.

연구자들은 한국인 어른 남녀 시신의 머리와 목 61 을 대상으로 깊은 층 육안해부를 시행하여 혀동맥의 형태를 조사하였다 혀동맥이 일어나는 유형은 위갑상동맥, 혀동맥, 그리고 얼굴동맥이 바깥목동액에서 따로 가지를 내어 나뉘는 경우가 563% 호 가장 않았으며, 얼굴동맥과 혀동맥이 강은 가지에서 얼어나 나뉘는 경우 (312%), 그리고 위 강상동액과 혀동맥이 같은 가지에서 나뉘는 경우 (12 5%) 등이 있었다. 혀동맥과 혀동맥의 둘째 부분은 덮는 목뿔혀근 과의 영대학적 관계는 각각 혀동맥의 첫째 부분이 목뿔혀근 뒤에서 심하게 휘어져 깊은 층으로 들어가는 유형 (41 2 %), 그리고 혀동맥의 첫째 부분이 목뿔혀근의 뒤쪽 근육모셔리 바로 뒤흘 지나 깊은 층으로 들어가는 경우가 661% 로 가장 많았다 목뿔혀근 깊은층에서 혀동맥의 둘째 부분이 달리는 양상은 급하게 위쪽으로 꺾역 달리다가 다시 앞쪽으로 꺾여 혀의 깊은 곳으로 들어가는 경우와 목뿔빼에서 혀쪽으로 완만한 굽이플 이루며 올라가는 경우가 비슷한 반도로 관찰되었다. 입안바닥에 분포하는 혀밑동맥이 혀동맥 셋째 부분에서 나오는 경우슨 전체의 590% 에서만 관창되었으며 그 외는 대부분 얼굴동맥의 턱끝 및동맥 가지가 턱옥뽕근응 옳고 입안바닥에 분포하였다. 따라서 연구자들은 한국인 업안바 닥에는 혀동액이 전체의 59%에서만 분포하며 그외는 영굴동액의 턱끝밑동맥 가지 , 그리고 기타 다른 동맥의 가지들이 분포한다는 사실을 확인하였다
A Study of the Yang-gyeong-gyu-il-ui (兩景揆日儀) in the Joseon Dynasty

The Yang-gyeong-gyu-il-ui (兩景揆日儀) is a kind of elevation sundial using three wooden plates. Sang-hyeok Lee (李尙爀, 1810~?) and Byeong-cheol Nam (南秉哲, 1817~1863) gave descriptions of this sundial and explained how to use it in their Gyu-il-go (揆日考) and Ui-gi-jip-seol (儀器輯說), respectively. According to Gyu-il-go (揆日考) there are two horizontal plates and two vertical plates that have lines of season and time. Subseasonal (節候) lines are engraved between seasonal (節氣) lines, subdividing the interval into three equal lines of Cho-hu (初候, early subseason), Jung-hu (中候, mid subseason) and Mal-hu (末候, late subseason); there are 13 seasonal lines for a year, thus resulting in 37 subseasonal lines; also, there are 12 double-hour (時辰) lines for a day engraved on these plates. The only remaining artifact of Yang-gyeong-gyu-il-ui was made in 1849 (the 15th year of Heon-jong) and is kept at the Korea University Museum. We have compared and analyzed Yanggyeong- gyu-il-ui and similar western sundials. Also, we have reviewed the scientific aspect of this artifact and built a replica. Yang-gyeong-gyu-il-ui is a new model enhanced from the miniaturization development in the early Joseon Dynasty and can be applied to the southern part of the tropic line through a structure change.

Long-Term Changes in Post-Stroke Depression, Emotional Incontinence, and Anger

Background and Purpose Long-term changes in post-stroke depression (PSD), post-stroke emotional incontinence (PSEI), and post-stroke anger (PSA) have rarely been studied. Methods This is a sub-study of EMOTION, a randomized, placebo-controlled trial, that examined the efficacy of escitalopram on PSD, PSEI, and PSA in patients with stroke. We interviewed patients at the long-term period (LTP) using predefined questionnaires: Montgomery-Åsberg depression rating scale (MADRS) for PSD, modified Kim’s criteria for PSEI, and Spielberger trait anger scale for PSA. Additionally, the ENRICHD Social Support Instrument (ESSI) for the social support state and the modified Rankin Scale (mRS) were measured. We investigated the changes in and factors behind PSD, PSEI, and PSA at LTP. Results A total of 222 patients were included, and the median follow-up duration was 59.5 months (interquartile range, 50 to 70). Compared to the data at 6 months post-stroke, the prevalence of PSEI (11.7% at 6 months, 6.3% at LTP; P=0.05) and mean anger score (21.62, 16.24; P<0.01) decreased, while the prevalence of PSD (35.6%, 44.6%; P=0.03) and mean MADRS (6.16, 8.67; P<0.01) increased at LTP. ESSI was associated with PSD and PSA, but not with PSEI. The effect of the baseline National Institutes of Health Stroke Scale score on PSD decreased over time. The effect of low social support on PSD was greater than that of mRS at LTP. Conclusions The prevalence and degree of PSD significantly increased, while those of PSEI and PSA decreased at LTP. PSD in this stage appeared to be more closely associated with a lack of social support than patients' physical disabilities.

<P><B>Abstract</B></P> <P>The effects of initial {10−12} twins on the in-plane fatigue characteristics of a rolled Mg alloy were investigated by introducing these twins through precompression along the rolling direction of the material and then performing strain-controlled low-cycle fatigue tests along the rolling direction, 45° to the rolling direction, and transverse direction. The as-rolled sample with a strong basal texture was found to exhibit the same fatigue behavior in all the loading directions, whereas the precompressed samples exhibited anisotropic in-plane fatigue behaviors because the specific crystallographic orientation of the twinned region resulted in a variation in the active deformation modes during cyclic loading in the applied loading directions. An increase in the amount of precompression was also found to enhance the anisotropy of the in-plane fatigue behavior via an increase in the area fraction of the twinned region. The variation in the dominant deformation mechanism with a change in the loading direction was investigated by analyzing the crystallographic angle relationship and activation stresses, and the resultant evolutions of the cyclic flow curve and fatigue life were discussed in detail.</P>
Robotic‑assisted unicompartmental knee arthroplasty: historical perspectives and current innovations

Robotic assisted unicompartmental knee arthroplasty (RAUKA) has emerged as a successful approach for optimizing implantpositioning accuracy, minimizing soft tissue injury, and improving patient-reported outcomes. The application of RAUKAis expected to increase because of its advantages over conventional unicompartmental knee arthroplasty. This review articleprovides an overview of RAUKA, encompassing the historical development of the procedure, the features of the robotic armand navigation systems, and the characteristics of contemporary RAUKA. The article also includes a comparison betweenconventional unicompartmental arthroplasty and RAUKA, as well as a discussion of current challenges and future advancementsin the field of RAUKA.
Hypericin-assisted photodynamic therapy against anaplastic thyroid cancer

<P><B>Abstract</B></P> <P><B>Background</B></P> <P>Hypericin (HYP) extracted from St. John’s wort (Hypericum perforatum L.) is a natural photosensitizer in clinical photodynamic therapy (PDT). PDT is one of the powerful methods for cancer treatments because of its excellent tumoritropic characteristics and photosensitizing properties. However, limited reports on the efficacy of PDT on anaplastic thyroid cancer (ATC) have been published. Especially HYP-associated PDT has not been investigated <I>in vitro</I> and <I>in vivo</I>. In this study, we evaluated the effect of HYP for PDT against FRO ATC cells.</P> <P><B>Methods</B></P> <P>The activities of HYP-assisted PDT were investigated in ATC cells. The ATC FRO cells were treated with a combination of HYP dose and laser power. The viability of FRO cells was measured by MTT assay, and Trypan blue staining was performed to monitor cell death. Detection reactive oxygen species (ROS) and mitochondrial membrane potential after HYP-assisted PDT were analyzed by confocal microscopy. For <I>in vivo</I> study, FRO cells were injected into nude mice. After intravenous injection of HYP, Laser was irradiated and nude mice were monitored in Day 4, 7, 14.</P> <P><B>Results and conclusions</B></P> <P>The rate of FRO cell death was increased by applying HYP dose and laser power dependent. Moreover, HYP and laser irradiation induced FRO cell death was mediated by the intracellular ROS generation and mitochondrial damage. Finally, the HYP-assisted PDT eliminated FRO cell tumor from the mouse <I>in vivo</I>. These data demonstrate that HYP could be an effective photosensitizer for human ATC therapy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Hypericin and laser irradiation induced anaplastic thyroid cancer cell death. </LI> <LI> Hypericin-mediated photodynamic therapy promoted ROS generation and mitochondrial damage in cells. </LI> <LI> Hypericin with laser irradiation prevented anaplastic thyroid cancer growth in the mouse. </LI> </UL> </P>
Clustering siRNA conjugates for MMP-responsive therapeutics in chronic wounds of diabetic animals

<P>The MMP-responsive breakdown of siRNA clusters was translated to site-specific gene transfection and enhanced wound healing in diabetic ulcers. MMP-2 siRNA was chemically tethered to the end of multi-armed PEG via MMP-cleavable linkers (4PEG-siRNA) and subsequently clustered into submicron particles complexed with LPEI. 4PEG-siRNA was more tightly complexed with LPEI and the associated cluster showed higher resistance against RNase attack, in comparison to naked siRNA. Because the size of the clusters increased depending on the increase in charge ratio of LPEI to siRNA, cellular uptake of the 4PEG-siRNA/LPEI cluster was significantly attenuated due to the huge size of the cluster. However, upon MMP treatment, the cluster dissociated into smaller particles and was efficiently endocytosed by cells. An in vivo fluorescence resonance energy transfer (FRET) study also revealed that the clusters were effectively dissociated in MMP-rich environments of dorsal wounds in diabetic animals. In addition, diabetic ulcers treated with the clusters showed a faster wound closure rate and the recovered tissue expressed a larger amount of cytokeratin along with a lower expression level of MMP-2 compared to the other groups.</P>

Background: Consistent uptake of colorectal cancer (CRC) screening is important to reduce the incidence and mortality from advanced-stage CRC and increase the survival rate of the patients. We conducted a longitudinal study to determine the factors affecting CRC screening compliance in Korean adults using individual-level linked data from the Korean National Health and Nutrition Examination Survey, Korean National Health Insurance Service, and Korean Health Insurance Review and Assessment Service. Methods: We selected 3,464 adults aged 50–79 years as the study population and followed them for 12 years (January 2007–December 2018). The outcome variable was the level of adherence to CRC screening, categorized as nonadherent, intermittently adherent, and consistently adherent. An ordinal logistic regression model was designed to determine the socioeconomic factors, family history of CRC, and medical conditions that could facilitate the consistent uptake of CRC screening. Results: The results showed a significant and positive association between consistent uptake of CRC screening and the 100–150% income category (odds ratio [OR], 1.710; 95% confidence interval [CI], 1.401–2.088); clerical, sales and service job category (OR, 1.962; 95% CI, 1.582–2.433); residency at medium-sized cities (OR, 1.295; 95% CI, 1.094–1.532); high-school graduation (OR, 1.440; 95% CI, 1.210–1.713); married status (OR, 2.281; 95% CI, 1.946–2.674); use of employment-based national health insurance (OR, 1.820; 95% CI, 1.261–2.626); use of private insurance (OR, 2.259; 95% CI, 1.970–2.589); no disability (OR, 1.428; 95% CI, 1.175–1.737); family history of CRC (OR, 2.027; 95% CI, 1.514–2.714); and history of colorectal neoplasm (OR, 1.216; 95% CI; 1.039–1.422). Conclusion: The lack of regular participation in CRC screening programs in the Republic of Korea was found to be an issue that requires attention. Policies on CRC screening must place increased emphasis on strengthening educational and public relations initiatives.
