This study given here presents the results of an experimental investigation of the iron contents contained in the city water which not used for a long time in the winter. The measurements were performed after turning on and for flowing water in a moment. And the quantitative analysis was studied by chemical form and the results were as follows. 1. The content of the ferrous ion was 113.8-14.4 ppm, water soluble iron was 22.5-14.4 ppm and acid soluble iron was 150-10 ppm. 2. Iron contents of the each chemical from gradually reduced but it appeared high value than standard value of city water until five minutes.

The aim of this survey is employment perception of martial arts major students in yongin university. The university education has various objects, one of the most important purpose is opportunities for employment. As time have changed, martial arts major students pointed out that their needs for employment. If universities accept their needs, it can be raise competitiveness of martial arts major departments in the academic world. This research is released its earnings report, the data will have significant meanings for the martial arts major departments sector grappling with traditional understanding. Therefore a realistic employment measure is urgently needed for martial arts major students, because it will need to be tailored to fit into the realities and by the limitations of martial arts major's departments curriculum. The research found the martial arts major students's cognitions on employment planning of individualized department education, employment education management of martial arts major departments and martial arts education system from university to society through questionnaires. From this point of view, this research was based on students who belonging to the college of martial arts(judo, taekwondo, combative martial arts, oriental martial arts, security service) in yongin university.
Genome wide DNA methylation analysis of chromo methylase CMT3 and E3 sumo ligase AtSIZ1 mutants.

Gene expression is regulated by DNA and histone methylation by DNA and histone methyltransferases, respectively. In animal system, DNA methyltransferase with CG methylation activity is modified by SUMO conjugation and then its activity was increased, which means that the activity of DNA methyltransferase is modulated by posttranslational modification. so Chromatin remodeling is a new concept for expression of controlling of gene function. We thus analyzed the effect of E3 SUMO ligase AtSIZ1 in CMT3 (chromometnylase 3)-mediated genome methylation by next-generation sequencing (NGS), methyl binding domain MeDIP-sequencing and gene analysis using siz1-2 and cmt3 mutants. we carried out CG-enrich analysis by MeDIP sequencing revealed that the methylation level of the genome including transposons was significantly low in siz1-2 mutants compared to wild-type. Result showed the genes regulated by methylation, that genes related of embryo and root development, cellulose metabolism, and post-translational modifications. All of our data indicate that the methyltransfearse activity of CMT3 may be able to be regulated by AtSIZ1 and thereby CMT3-mediated gene expression and plant development also can be controlled by E3 SUMO ligase activity. Besides, our data also suggest that ammonium (NH4+) can stimulate AtSIZ1- and CMT3- mediated DNA methylation.

식용 및 약용으로 이용되는 산초(Zanthoxylum schinifolium)의 줄기로부터 항암활성 성분을 분리하기 위하여, 산초 줄기를 유기용매로 추출하고 각 추출물의 인체 급성백혈병 암세포에 대한 독성 및 세포자살 유도 활성을 조사하였다. Methanol (SS-7), methylene chloride (SS-8), ethyl acetate (SS-9), n-butanol (SS-10)로 추출한 각 시료와 유기용매 추출 후 잔여분획 (SL-14)의 세포 독성을 인체 급성백혈병 Jurkat T 세포주를 대상으로 조사한 결과, 암세포에 대한 세포독성이 주로 methylene chloride 추출분획인 (SS-8)에서 확인되었다. Methylene chloride 추출물(SS-8)의 Jurkat T 세포주에 대한 세포독성의 기전은 mitochondria로부터cytochrome c 방출, caspase-9 및 caspase-3의 활성화, PARP 분해, internucleosomal DNA fragmentation 등의 일련의 생화학적 반응을 수반하며, 항 세포자살 단백질인 Bcl-xL단백질의 과발현에 의해 억제되는 세포자살 기전임을 확인하였다. FADD가 disruption된 Jurkat T cell clone I2.1 (FADD<SUP>-/</SUP>-) 및 caspase-8가 결핍된 Jurkat T cell clone I9.2 (caspase-8<SUP>-/-</SUP>)와 함께 the wild-type Jurkat T cell clone A3에 미치는 SS-8의 세포독성작용을 비교 분석한 결과, wild-type Jurkat A3, FADD-deficient Jurkat clone I2.1및 caspase-8-deficient Jurkat clone I9.2 모두는 SS-8의 세포독성에 대해 유사한 정도의 감수성을 나타내었다. 이는 SS-8에 의해 유도되는 apoptosis에 있어서, Fas/FasL system이 관계되지 않음을 시사한다. 한편, SS-8를 GC-MS 분석하여, 9,12-octadecanoic acid (18.62%), 2,4-dihydro-5-methyl-4-(1-methylethylidene)-2-(4-nitrophenyl)-3H-pyrazol-3-one (14.97%), hexadecanoic acid (14.23%), (z,z)-6,9-pentadecadien-1-ol (13.73%), 5,6-dimethoxy-2-methyl benzofuran (10.95%), 그리고 4-methoxy-2-methylcinnamic acid (5.38%) 등을 포함한 16가지의 구성성분과 그 조성비를 확인하였다. 이상의 연구결과는 산초 줄기에 함유된 항암 활성에 대한 규명과 이해를 증진시킨다. To examine antitumor activity of the edible plant Zanthoxylum schinifolium, the cytotoxic effect of various organic solvent extracts of its stems on human acute leukemia Jurkat T cells was investigated. Among these extracts such as methanol extract (SS-7), methylene chloride extract (SS-8), ethyl acetate extract (SS-9), n-butanol extract (SS-10), and residual fraction (SL-11), SS-8 exhibited the most cytotoxic activity against Jurkat T cells. The methylene chloride extract (SS-8) possessed the apoptogenic activity capable of inducing sub-G1 peak along with apoptotic DNA fragmentation in Jurkat T cells. Western blot analysis revealed that SS-8 induced apoptosis via mitochondrial cytochrome c release into cytoplasm, subsequent activation of caspase-9 and caspase-3, and cleavage of PARP, which could be blocked by overexpression of Bcl-xL. Jurkat T cell clone I2.1 (FADD<SUP>-/-</SUP>) and Jurkat T cell clone I9.2 (caspase-8<SUP>-/-</SUP>) were as sensitive as was the wild-type Jurkat T cell clone A3 to the cytotoxic effect of SS-8, suggesting no contribution of Fas/FasL system to the SS-8-mediated apoptosis. The GC-MS analysis of SS-8 showed that it was composed of 16 ingredients including 9,12-octadecanoic acid (18.62%), 2,4-dihydro-5-methyl-4- (1-methylethylidene)-2-(4-nitrophenyl)-3H- pyrazol-3-one (14.97%), hexadecanoic acid (14.23%), (z,z)-6,9-pentadecadien- 1-ol (13.73%), 5,6-dimethoxy-2-methyl benzofuran (10.95%), and 4-methoxy-2-methylcinnamic acid (5.38%). These results demonstrate that the methylene chloride extract of the stems of Z. schinifolium can induce apoptotic cell death in Jurkat T cells via intrinsic mitochondria-dependent caspase cascade regulated by Bcl-xL without involvement of the Fas/FasL system.
Flowering time is repressed by sumoylation of FLC

The transition from vegetative growth to flowering is a major developmental switch in the plant cycle and the timing of flowering is very critical for reproduction of plant species. In transition to flowering in plants, Flowering locus C (FLC) is one of the crucial factors. Here, we showed How the stability and activity of FLC are regulated by sumoylation mechanism. By pull-down assay, we showed that FLC interact with E3 SUMO ligase in vitro and vivo. And we showed that FLC is sumoylated in vitro condition with AtSUMO1 protein. In transgenic plants with overexpression of FLC and inducible expression of AtSIZ1, sumo E3 ligase led to increase of FLC protein level and delayed the post-translation degradation of FLC indicating that Arabidopsis E3 sumo ligase AtSIZ1 stabilizes FLC. Also, the plants with overexpression of mutant FLC (K154R, a mutation of the sumoylation site on FLC) flowered considerably earlier than plants with overexpression of FLC but comparable with wild type indicating that sumoylation is a important part for function of FLC. Our data indicate that the sumoylation of FLC is critical for its role in the control of flowering time.

Background: Granulocyte-macrophage colony-stimulating factor (GM-CSF) gene-transduced tumor cell vaccines induce very potent systemic anti-tumor immunity in preclinical and clinical models. Our previous phase I clinical trial in patients with metastatic renal cell carcinoma (RCC) has demonstrated both immune cell infiltration at vaccine sites and T cell-mediated delayed-type hypersensitivity (DTH) response to whole tumor cell vaccines. Methods: To investigate the immune responses to autologous genetically- modified tumor cell vaccines, tumor-specific $CD8^+$ T cell lines were generated from peripheral blood lymphocytes (PBL) of a RCC patient 1.24 by repeated in vitro stimulation with either B7.1-transduced autologous RCC tumor cells or B7.1-transduced autologous tumor cells treated with interferon gamma ($IFN{\gamma}$), and cloned by limiting dilution. Results: Among several RCC-specific cytotoxic T lymphocytes (CTLs), a $CD4^+/CD8^+$ double positive T cell clone (17/A2) appeared to recognize $IFN{\gamma}$-treated autologous RCC restricted by HLA-B39. The 17/A2 also recognized other HLA-B39 positive RCC tumor cells after $IFN{\gamma}$ treatment. Conclusion: These results demonstrate that autologous RCC vaccination successfully generates the tumor-specific CTL 17/A2, and suggest that the presentation and recognition of the tumor antigen by the 17/A2 might be upregulated by $IFN{\gamma}$.
