        In Vitro Antiviral Activity of Ribavirin Against Severe Fever with Thrombocytopenia Syndrome Virus Myung Jin LEE1, Kye-Hyung KIM1, Jongyoun YI2, SuJin CHOI1, Chung-Jong KIM1, Nak- Hyun KIM1, Kyoung-Ho SONG1, Pyoeng Gyun CHOI1, Ji-Hwan BANG1, Wan Beom PARK1, Eu Suk KIM1, Sang-Won PARK1, Hong Bin KIM1, Nam Joong KIM1, Myoung- Don OH1 Seoul National University College of Medicine, Korea1, Pusan National University School of Medicine, Korea2 Background: Severe fever with thrombocytopenia syndrome (SFTS) is an emerging infectious disease caused by a novel Bunyavirus, severe fever with thrombocytopenia syndrome virus (SFTSV). No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

        정수처리공정상 염소소독부산물형성에 미치는 오존의 영향

        The effect of ozone on the formation and the removal of disinfection byproducts(DBPs) of chlorination process was studied to elucidate the performance of water treatment process. The samples of raw water, prechlorination process, and preozonation process were analyzed quantitatively according to the Standard Methods for the Examination of drinking water. As a result, most of total trihalomethanes(THMs) which were formed in prechlorine treatment process was not removed in the preozonation process. Most of haloacetic acids(HAAs), haloacetonitriles(HANs), and chloral hydrate(CH) was removed in sedimentation and biological activated carbon(BAC) filtration processes. However, DBPs were increased more or less by postchlorine step. In particular, the formation of THMs and HAAs depends on ozone more than chlorine, but, the formation of HANs and CH depends on chlorine more than ozone. The seasonal variation of DBPs concentration for the year needs to be investigated to study the temperature effect because DBPs strongly depend on temperature among various efficient factors.

        This study is carried out to investigate the water quality changes in Busan Metropolitan streams. The results are as follows. In Busan Metropolitan streams, the loading rates of contaminants are greater in industrial waste water streams than domestic waste water streams. Some sampling points show a little high loading in autumn, but this needs more investigation to make clear the causes. The level of organic contaminants(BOD_(5) and COD_(MN)) is found to be low and does not show seasonal variation in domestic waste water streams. But, the level of organic contaminants in industrial waste water streams is relatively high and seems to be seasonally variable, which is affected by other factors. The nutrient materials, such as nitrogen(as T-N, about 20mg/L) and phosphorus(as T-P, about 2.0mg/L), are abundant than Nak Dong River and the general trends of contaminants level are similar to those of organic contaminants.

      • 정수슬러지와 폐굴껍질 분말의 혼합첨가에 의한 산성토양 개선에 관한 연구

        In this study, farm soil was tested to find out acid soil amendment by adding water purification sludge(WPS) and waste oyster shell(WOS) under artificial acid rain(pH=534). which is average pH of Busan metropolitan area. Soil acidity, CEU(cation exchange capacity) and akaline meatals, such as K, Ca and Na were tested to find out the acid neutralization capacity of soil Because WOS had large portion of alkaline metals, WOS could play a role in acid soil amendment And soil addition of WPS could be an alternative for conventional disposal method. The purpose of this study is to evaluate acid soil amendment using LWS and WOS. In case of soil and WPS, mixing ratio was 10:0, 9:1, 8:2 and 7:3 And WPS mixed WOS was also added In acid soil as the same ratio as WPS. Throughout the experiment, soil acidity and CEC were stationary state with slight increase. Alkaline metals showed differences in concentration, but all of them were tend to decrease. In conclusion, WPS mixed WOS could play a part in amendment of acid soil.

        As the fiscal scale of Korean Government expands, the size of procurement on the products for the Government is increased. This trend needs more effective and scientific management techniques that will classify the materials systematically in order to make materials management easier. In this reason, the Materials Catalog Information System (MCIS) has been put into effect since January 1, 2001. With the advent of Government Electronic Procurement System beginning on September of 2002, however, the parallel usage of materials catalog code with GZB catalog code specially made for electronic commerce in G2B system gave rise to confusion in the exchange and utilization of the materials information, by maintaining and applying the different materials classification systems respectively. This paper analyzes the existing situation and problems of MCIS and suggests several options or alternatives to improve the system and establish a more efficient MCIS.

        高麗中期 道敎의 綜合的 硏究

        This is a Synthetic study on Taoism in the middle of Ko-ryo Dynasty which is unfold around the King Ye-Jong's days. Through the work coureses, besides synthesizing existing study, this study focuses on the Characters of historical development aspects and relation between the ideas of literature and religion on the basis of cooperated work by various fields such as history, religion and literature. Therefore, I'm going to grasp characters of the thought of Taoism Ages and the aspect of concret development. First, I examined aspects of historical Progress of Taoism in Ko-ryo dynasty. This study observed progress and fact, Religion Taoism Progressed, Cons- truction Courses on the basis of Tao-Temple's pok Won Kung(福源 宮) As a result, it raised Formed Toaism from the level of Ko-ryu Taoism by Pok Won Kung and produced systematization of religious ceremonies. And It was turned out that they spread customs of Taoism religion in Ko-ryo society and did far toward creating climate of the idea of seclusion. In the religious aspect, the writer considered the passage of the three religions. such as Confucianism, Buddhism, Taoism in Ko-ryo dynasty and the position of taoism thought, The writer also examined accepted thought of Taoism and religious rites and analyzed Taoisitic Funeral Oration written by literary men. Through these, we could realize that. Ko-ryo had already accepted the edification of Taoism in depth. The writer also examined that how Chinese Naidanhak(內丹學) had been accepted at that period. Thry took a general view of the literary spirit transfering into the learning of Song Dynasty and l inked it with Taoism. And it grafted Taoism thought. The writer researched the achieve ments and literary work of Kwak-Yo(郭與) Lee Ja-Hyun(李資賢), Chukko- ch'ilhyon(竹高七賢) and literary works and made it clear the relation between these and Taoism thought. The writer examined Taoism thought reflect on works. separately such as the idea of seclusion, the idea about Taoist Hermit, preservation of health and enjoyment of drunken state and so on. Through serious of examination, in the middle of Ko-ryu swept the Ko-ryo society with considerable influence and propagation We also could confirm that I had influened deeply religiously and spiritually. When we study continuously and deeply relating to the fore and after the period carefully, will be more wide and deep gradually the Korean history of thouhts.

        노인요양시설 서비스 공간 계획에 관한 연구

        The demand of the care facilities for the elderly will be increased, because of the social changes which desire well organized medical treatment service. However, they have been illogically administrating such as preparing service at living space because of lacking places which provide living service. So the side of administration, it is necessary to have a security and improvement in that kind of needed place. The studies of elderly care facilities have been placing too much emphasis on the living space of the residential facilities and their welfare facilities compare with the service providing place which is ignored just as a part of the living space. This study focus on a situation of elderly care facilities and analyze a service space using in facilities, so it can be applied to service place plan for the future.

        This study inspected community concept, previous community activation studies and relationship between physical surrounding improvement and programs that is the most typical community activation method. Analyses of existing community activation method are as follows ; 1) Physical surrounding improvement to dress up space and expand facilities in recent situation 2) Method to use programs Physical surrounding improvement Method classifies applicable plan regard to community of exterior space in residence complex and application of space expansion, needed to communication behavior in residence complex in the present. Methods to use programs are to develope communality as leading community subject, whether the subject is anyone. It is true that the programs are taking a primary role in activating community. but programs will enlarge the role in mutual complementary relation with the surrounding improvement than independent in relation. In the end, it is propose that we can enhance not only to change better surrounding by programs and to activate programs by the surrounding improvement but also to make the best use of physical surrounding by programs application.

      • 장생도라지의 약리작용

        도라지 (길경, Platycodon grandiflorum A. DC)는 예부터 천식, 기관지염 등에 효능이 인정되어 약용 및 식용으로 널리 이용되어 왔다. 장생 도라지에 대한 연구로 식이성 고혈압 흰쥐에 도라지 분말을 각 5%와 10% 함유한 식이를 3주간 급이하였을 때 혈청과 간장의 지질 농도가 현저하게 감소하였으며, 5%의 도라지 분말 함유 식이의 급이시 도라지를 급이하지 않은 군에 비하여 혈청과 간장의 총콜레스테롤과 중성지질의 농도가 현저하게 감소하였고, alloxan minihydrate를 체중 kg당 15 mg의 농도로 3주간 복강내 주사하여 당뇨를 유발시킨 흰쥐에 6년근과 22년근 도라지 추출물의 급이시 혈당 및 혈청과 간장의 지질 개선에 효과가 있었고, 또한 항암효과에 관한 연구로는 도라지 추출물을 B16-F1 mouse melanoma 세포에 처리해 laminin의 결합력을 실험한 결과 0.125 mg/ml 이상의 농도에서는 100%저해능을 보였으며, mice에 있어서 고형암의 무게를 61.5% 감소시켰으며 sacoma-180 ascites에 대한 생존일수 연장율이 128%에 달하는 것으로 관찰되었다. 도라지의 에탄올 추출물을 극성을 달리한 용매로 분획하여 각 획분의 항균성을 실험한 결과 23년근 도라지의 부탄올획분의 M. luteus에 대한 초소저해농도는 3년근에 비하여 28배나 더 낮은 값을 보였으며, 이를 실리카겔 칼럼 그로마트그래피와 HPLC를 이용하여 에틸아세테이트획분의 항균물질을 분리·정제한 결과 데틸아세테이트획분보다 S. epidermidis에 대해 2∼3배 더 강한 항균력이 관찰되었고 23년근 도라지의 주요 항균물질을 동정한 결과 giycerol linolate임이 확인되었다. The root of Platycodon grandiflorum (p.g.) A. DC (Platicodi radix) has been prescribed frequently for cough and bronchitis as an expectorant in a traditional oriental medicine, and used as a food. Diet-induced hyperlipidenic rats were fed diets containing 5% and 10% p.g (twenty-three years). powder for 3 weeks, the p.g. feeding markedly decreased both serum and liver lipid concentrationds in hyperlipidemic rats. Especially, 5% p.g. diet significantly decreased the concentrations of total cholesteril and triglycerides in serum and liver as compared with those of thr hyperlipidemic control group. The effects of the feeding p.g. (6 or 22 years) extract on the blood glucose, lipids in the serum and liver of alloxan induced hyperglycemic rats (S.D. strain, ♂), alloxan monohydrate 15㎎/㎏ B.W./day, I.P. injection for 3 weeks. concentrations of blood glucose were significantly higher in the allozan administration groups than those in the control group. When p.g. extracts treated in vitro with B16-F1 mouse melanoma cell lines, it showed 100% laminin-binding inhibitory activities at the concentration over 0.125 mg/ml in vivo test it showed 61.5% reduction of the solid tumor weight transplanted in mice and exhibited anticancer activity of 128% ILS against sarcoma-180 ascites. The ethanol extract of p.g. was fractionated by various solvents with different polarities, and the antibacterial activities of each fraction were tested. The minimal inhibitory concentration (MIC) values of each graction of Platycodi radix (twenty-three years) ethanol extract was lower than that of Platycodi radiz (three years) ehanol extract. the MIC value of Platycodi radix(twenty-three years) butanol fraction against M. luteus was 28 times lower than that of Platycodi radiz (three years) butanol fraction. The antibacterial compound was isolated and purified from the ethylacetate fraction by silica gel column chromatography and HPLC, and exhibited 2 to 3 times more antibacterial activity against S. epidermidis than ethylacetate fraction. The major antibacterial compound of Platycodi radix (twenty-three years) was identified to be a glycerol linolate.
