To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

<P> Mepivacaine is an aminoamide-linked local anesthetic with an intermediate duration that intrinsically produces vasoconstriction both in vivo and in vitro. The aims of this in-vitro study were to examine the direct effect of mepivacaine in isolated rat aortic rings and to determine the associated cellular mechanism with a particular focus on endothelium-derived vasodilators, which modulate vascular tone. In the aortic rings with or without endothelium, cumulative mepivacaine concentration-response curves were generated in the presence or absence of the following antagonists: N<SUP>ω</SUP>-nitro-l-arginine methyl ester [l-NAME], indomethacin, fluconazole, methylene blue, 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one [ODQ], verapamil, and calcium-free Krebs solution. Mepivacaine produced vasoconstriction at low concentrations (1 × 10<SUP>−3</SUP> and 3 × 10<SUP>−3</SUP> mol/L) followed by vasodilation at a high concentration (1 × 10<SUP>−2</SUP> mol/L). The mepivacaine-induced contraction was higher in endothelium-denuded aortae than in endothelium-intact aortae. Pretreatment with l-NAME, ODQ, and methylene blue enhanced mepivacaine-induced contraction in the endothelium-intact rings, whereas fluconazole had no effect. Indomethacin slightly attenuated mepivacaine-induced contraction, whereas verapamil and calcium-free Krebs solution more strongly attenuated this contraction. The vasoconstriction induced by mepivacaine is attenuated mainly by the endothelial nitric oxide - cyclic guanosine monophosphate pathway. In addition, mepivacaine-induced contraction involves cyclooxygenase pathway activation and extracellular calcium influx via voltage-operated calcium channels. </P>

Human fibroblast-derived multi-peptide factors (MPFs) have been used during treatments with energy-delivering modalities to enhance energy-induced tissue reactions. Human fibroblast-derived MPFs, which include a range of growth factors and chemoattractive factors, activate and recruit fibroblasts and endothelial cells, as well as promote extracellular matrix deposition, all of which are crucial to wound repair. Interestingly, fibroblasts from different species or anatomical sites exhibit distinct transcriptional properties with high heterogeneity. In addition, the patterns of MPF secretion can differ under a range of experimental conditions. Therefore, the use of allogeneic fibroblasts and proper cultivation thereof are necessary to obtain MPFs that can enhance the epithelial-mesenchymal interactions during wound repair. Moreover, energy-delivering devices should be selected according to evidence demonstrating their therapeutic efficacy and safety on a pathological skin condition and the major target skin layers. This paper reviewed the histologic patterns of post-treatment tissue reactions elicited by several energy sources, including non-ablative and ablative fractional lasers, intense focused ultrasound, non-invasive and invasive radiofrequency, picosecond-domain lasers, and argon and nitrogen plasma. The possible role of the immediate application of human fibroblast-derived MPFs during wound repair was proposed.
S-15 The Association of Gallstone & Cholecystectomy with Metabolic changes or Intestinal bacteria

Objectives: Gallbladder (GB) diseases including the status of gallstone or cholecystectomy could be associated with metabolic diseases or intestinal bacteria. However, their role has not been clarified. We investigate the relationship of GB diseases with fatty liver, hypertension, diabetes, metabolic syndrome, and small intestinal bacterial overgrowth (SIBO). Methods: We prospectively evaluated the consecutive outpatients with GB diseases attending a gastrointestinal clinic, who underwent demographic investigation, blood test for metabolic diseases, abdominal ultrasonography, glucose breath test (GBT). The 26 Controls without any abdominal symptoms were also enrolled. Results: 178 patients with GB diseases (131 GB stone, 47 cholecystectomy) were finally enrolled. The mean age of the patients were 55.4 years (range: 19-80) and 37.6% were male. The prevalence of fatty liver (46.1%), dyslipidemia (41.0%), hypertension (33.7%), metabolic syndrome (29.8%), SIBO (35.4%) and serum alanine aminotransferase (ALT) were significantly higher or higher tendency in patients with GB disease than in controls. Multivariate logistic regression analysis showed that dyslipidemia (OR=3.12, 95% confidence interval (CI): 1.02-9.73), hypertension (OR=4.62, 95% confidence interval (CI): 1.24-17.26), fatty liver (OR=3.42, 95% confidence interval (CI): 1.07-10.90) and metabolic syndrome (OR=10.39, 95% confidence interval (CI): 1.28-84.06) were independent factors associated with GB diseases. In subgroups with GB stones, dyslipidemia, hypertension, metabolic syndrome, and SIBO were independent factors. Whereas in subgroups with cholecystectomy, metabolic syndrome, fatty liver, and severity fatty liver were independent factors. Conclusions: The metabolic status with dyslipidemia, hypertension, metabolic syndrome is common risk factors related with both GB stone and cholecystectomy. Cholecystectomy is associated with the progression of fatty liver; whether GB stone is independently related with risk of intestinal bacteria

This study was performed to investigate the effect of Gleditsia sinensis Lam. extract on the physicochemical properties of emulsion-type pork sausages during storage at 10°C for 4 wk. Treatments were as follows: (C, control; T1, sodium ascorbate 0.05%; T2, Gleditsia sinensis Lam. 0.05%; T3, Gleditsia sinensis Lam. 0.1%; T4, Gleditsia sinensis Lam. 0.2%; T5, Gleditsia sinensis Lam. 0.1% + sodium ascorbate 0.05%). The values of pH, moisture content, lightness, redness, and sensory attributes were all significantly decreased, while the yellowness, chroma, hue angle, and texture properties were increased during storage with increase of the Gleditsia sinensis Lam. extract added. In addition, the antioxidant activity and antimicrobial activity in the sausages displayed significant increases (p<0.05). Therefore, although it was concluded that the addition of Gleditsia sinensis Lam. extract is not effective for improvement of the physical properties compared to chemical additives in sausages, it could be applied to meat products as a natural preservatives.
