      • Assessment of variation in immunosuppressive pathway genes reveals <i>TGFBR2</i> to be associated with prognosis of estrogen receptor-negative breast cancer after chemotherapy

        <P><B>Introduction</B></P><P>Tumor lymphocyte infiltration is associated with clinical response to chemotherapy in estrogen receptor (ER) negative breast cancer. To identify variants in immunosuppressive pathway genes associated with prognosis after adjuvant chemotherapy for ER-negative patients, we studied stage I-III invasive breast cancer patients of European ancestry, including 9,334 ER-positive (3,151 treated with chemotherapy) and 2,334 ER-negative patients (1,499 treated with chemotherapy).</P><P><B>Methods</B></P><P>We pooled data from sixteen studies from the Breast Cancer Association Consortium (BCAC), and employed two independent studies for replications. Overall 3,610 single nucleotide polymorphisms (SNPs) in 133 genes were genotyped as part of the Collaborative Oncological Gene-environment Study, in which phenotype and clinical data were collected and harmonized. Multivariable Cox proportional hazard regression was used to assess genetic associations with overall survival (OS) and breast cancer-specific survival (BCSS). Heterogeneity according to chemotherapy or ER status was evaluated with the log-likelihood ratio test.</P><P><B>Results</B></P><P>Three independent SNPs in <I>TGFBR2</I> and <I>IL12B</I> were associated with OS (<I>P</I> <10<SUP>−3</SUP>) solely in ER-negative patients after chemotherapy (267 events). Poorer OS associated with <I>TGFBR2</I> rs1367610 (G > C) (per allele hazard ratio (HR) 1.54 (95% confidence interval (CI) 1.22 to 1.95), <I>P</I> = 3.08 × 10<SUP>−4</SUP>) was not found in ER-negative patients without chemotherapy or ER-positive patients with chemotherapy (<I>P</I> for interaction <10<SUP>−3</SUP>). Two SNPs in <I>IL12B</I> (r<SUP>2</SUP> = 0.20) showed different associations with ER-negative disease after chemotherapy: rs2546892 (G > A) with poorer OS (HR 1.50 (95% CI 1.21 to 1.86), <I>P</I> = 1.81 × 10<SUP>−4</SUP>), and rs2853694 (A > C) with improved OS (HR 0.73 (95% CI 0.61 to 0.87), <I>P</I> = 3.67 × 10<SUP>−4</SUP>). Similar associations were observed with BCSS. Association with <I>TGFBR2</I> rs1367610 but not <I>IL12B</I> variants replicated using BCAC Asian samples and the independent Prospective Study of Outcomes in Sporadic versus Hereditary Breast Cancer Study and yielded a combined HR of 1.57 ((95% CI 1.28 to 1.94), <I>P</I> = 2.05 × 10<SUP>−5</SUP>) without study heterogeneity.</P><P><B>Conclusions</B></P><P><I>TGFBR2</I> variants may have prognostic and predictive value in ER-negative breast cancer patients treated with adjuvant chemotherapy.       • Fatigue life prediction of rear axle using time series model

        The rear axle is one of the key parts of the automobile, lots failure of rear axle resulted from fatigue failure of the spiral bevel gears. A new method is proposed to solve the problem of accurately predicting the fatigue life of spiral bevel gears in rear axle. The method uses the recurrence tracing and difference method to improve the Autoregressive Moving Average (ARMA) model prediction accuracy, which uses variables determined from on-line measurements to characterize the state of the deterioration rear axle. The experimental results show the proposed method has relatively high prediction accuracy.

        Rheological Measurements of Physical Gelation in a Crystallized Poly(3-hydroxybutyrate)/Poly(p-vinylphenol) Blends

        In our previous work, the miscibility and rheology behaviors of poly(3-hydroxybutyrate)/poly(p-vinylphenol) (PHB/PVPh) blends with homogeneous amorphous compositions had been studied. In this work, the rheological characteristics and physical gelation point have been investigated for the miscible PHB/PVPh blends with crystalline/amorphous compositions. The time sweep and frequency sweep of dynamic oscillatory experiment are discussed in details. The storage modulus G’ and loss modulus G’’ grow gradually along with the isothermal crystallization time, but the storage modulus G′ exceeds the loss modulus G″ in the end. The physical gelation phenomena can be observed according to the power law method. In the flat region of loss tangent vs. angular frequency curves, the physical gelation point (tg) of the PHB/PVPh 90/10 comes forth prior to the PHB/PVPh 80/20 during the liquid-solid transition process, which results mainly from the various isothermal crystallization rate of PHB component within PHB/PVPh blends.

      • Fault Diagnosis System Based on Smart Bearing

        According to statistics, a lot of the rotating machinery faults are caused by the bearings, so the smart bearing technique is important for ensuring safety of them. For the smart bearing, a representative definition is that sensing devices of the different use are integrated into the traditional bearing in order to realize self-diagnosis. For condition of the variable speed, variable load and heavy load, the diagnostic technology cannot satisfy requirement of the fault feature extraction at present. This paper presents a new smart bearing of the multi-parameters including of two vibration acceleration sensing devices, two speed sensing devices, and two temperature sensing devices. In addition, the heavy noise can be decreased for extraction of the weak fault signals by the embedded integrated mode of bearing and sensor.

        Rolling element-bearing feature extraction based on combined wavelets and quantum-behaved particle swarm optimization

        A new approach is proposed to quickly determine the optimal band-pass filter for the resonant demodulation of rolling element bearings. The combined wavelet filter is joined with quantum-behaved particle swarm optimization for optimal filtering. The correlated kurtosisis used as an evaluation index to identify the optimal filtered signal. The proposed approach possesses good band-pass filteringproperties and improves optimal filtering speed and effect to converge rapidly to an optimal solution. Simulation and experiment resultsshow that this approach can generate fault features that are superior to those produced with the classic algorithm. It also shortens the timespent on the same filtering effect in comparison.

      • An Empirical Analysis of Factors Influencing the Information Service Quality of Recruitment Websites

        The online recruitment industry has developed rapidly under the Internet Plus environment, but the job hunters are rarely satisfied to the overall information service quality of recruitment websites. In order to improve the information service quality of recruitment websites, and maintain the competitive advantages of the industry, we conduct an empirical analysis of the factors influencing the information service quality of recruitment websites. Based on synthesizing the service quality evaluation indicators from both domestic and foreign studies,we propose the research model and hypotheses from the perspective of job hunters. We then conduct an online survey, and perform descriptive analysis, reliability analysis, factor analysis, variance analysis, correlation analysis and regression analysis on the sample data to test the research model and hypotheses with SPSS. The results show that the information content quality, reliability, ease of use, personalization and interactivity all have significantly positive correlation with the information service quality of recruitment websites.

        Crystallization Morphology Transition of Poly(3-hydroxybutyrate) Films Depending on Nucleation Temperature under Temperature Gradient

        The growth of spherulites is an intricate process and the final morphology is dependent on many process conditions. The spherulitic morphologies of poly(3-hydroxybutyrate) (PHB) crystallized from melt film are observed under various crystallization temperature gradient (the non-isothermal crystallization) in this work. Characterization reveals that the crystal morphologies (banded or nonbanded spherulites) of PHB film are determined significantly by nucleation temperature (not growth temperature). Interestingly, banded spherulites, nucleated between 60-135 oC, are able to grow completely above growth temperature 135 oC or below 60 oC yet and non-banded shperulites nucleated at above 135 oC can also develop steadily till below growth temperature 135 oC under temperature gradient condition. Therefore, the conclusion drawn from the temperature gradient or non-isothermal process is novel and different from the result of isothermal crystallization (morphology mainly depending on the growth temperature). In addition, the growth and evolution kinetics of banded and non-banded spherulites are both investigated for some polymer thin films under the temperature gradient in detail.

        The Transition of Molecular Alignment of Poly(ethylene oxide) Film via Thermal Annealing

        The molecular alignment of polymer film has attracted a lot of attentions due to its complicated process affected by processing conditions and dominating polymer applications. Here we demonstrate that the annealing process can not only change the surface morphology of annealed poly(ethylene oxide) (PEO) film significantly but also induce the formation of highly oriented lamellar structure. It indicates that sheaf-like and cluster banding morphology can be both found obviously on the surface of pristine film. However, there is only sheaf-like morphology and sporadical cluster banding cannot be found obviously on the surface of annealed film. Then, there are both tilt and flat-on orientations of PEO crystalline for pristine film. But, single flat-on orientation of PEO crystalline is present and tilt orientation mode is absent for annealed films. Furthermore, the transition of molecular alignment of PEO film via thermal annealing cannot be affected by the thickness and temperature.

        Glia maturation factor beta deficiency protects against diabetic osteoporosis by suppressing osteoclast hyperactivity

        Excessive osteoclast activation, which depends on dramatic changes in actin dynamics, causes osteoporosis (OP). The molecular mechanism of osteoclast activation in OP related to type 1 diabetes (T1D) remains unclear. Glia maturation factor beta (GMFB) is considered a growth and differentiation factor for both glia and neurons. Here, we demonstrated that Gmfb deficiency effectively ameliorated the phenotype of T1D-OP in rats by inhibiting osteoclast hyperactivity. In vitro assays showed that GMFB participated in osteoclast activation rather than proliferation. Gmfb deficiency did not affect osteoclast sealing zone (SZ) formation but effectively decreased the SZ area by decreasing actin depolymerization. When GMFB was overexpressed in Gmfb-deficient osteoclasts, the size of the SZ area was enlarged in a dose-dependent manner. Moreover, decreased actin depolymerization led to a decrease in nuclear G-actin, which activated MKL1/SRF-dependent gene transcription. We found that pro-osteoclastogenic factors (Mmp9 and Mmp14) were downregulated, while anti-osteoclastogenic factors (Cftr and Fhl2) were upregulated in Gmfb KO osteoclasts. A GMFB inhibitor, DS-30, targeting the binding site of GMFB and Arp2/3, was obtained. Biocore analysis revealed a high affinity between DS-30 and GMFB in a dose-dependent manner. As expected, DS-30 strongly suppressed osteoclast hyperactivity in vivo and in vitro. In conclusion, our work identified a new therapeutic strategy for T1D-OP treatment. The discovery of GMFB inhibitors will contribute to translational research on T1D-OP.
