      • The Synergistic Anticancer Effect of Artesunate Combined with Allicin in Osteosarcoma Cell Line in Vitro and in Vivo

        Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.

        Consumer Psychological Factors that Affect Chinese Donation Satisfaction and Continuous Donation Intention with Food Banks

        Purpose The goal of this study is to improve the continuous donation behavior of China’s ordinary people to food bank and reduce food waste. Through the survey and measurement of China’s ordinary people’s donation satisfaction and continuous donation intention to the Food Bank;it is possible to find out the consumer psychological factors that affect donation satisfaction and continuous donation intention. Design/Methodology/Approach – A questionnaire survey was conducted for Chinese with experience in food bank donations and data was collected. After excluding bad answers and invalid answers;371 responses were obtained. In order to verify the research hypothesis;this research used structural equation path analysis and hierarchical multiple regression analysis. Findings – Altruistic motivation was found to have no effect on Chinese satisfaction with donations to food banks;while egoistic motivation showed a significant negative impact. The impact of unplanned impulsivity and evaluation sensitivity impulsivity on Chinese donation satisfaction to food banks is significant;but the impact of reward sensitivity impulsivity is not significant. Perspective-taking and empathic concern in consumer empathy have a significant impact on the satisfaction of Chinese donations to food banks. Finally;Chinese satisfaction with donations to food banks was found to have a significant impact on the intention of continuing donations;and only independent self-construal was found to have a moderating effect. Research Implications – This is the first empirical study conducted in the field of food banking in China;combined with consumer psychological factors;and has important academic significance. It also provides valuable references for China’s charitable donations;food distribution industry;and provides new reference ideas for China’s “saving food”.

        Investigation on separation principle of vanadium and chromium among Fe2VO4-CaO-FeCr2O4 system: Simplify and simulate calcification roasting process of vanadium-chromium slag

        The competitive reaction of vanadium and chromium with calcium additives, along with the formationmechanism of calcium vanadate and calcium chromate are the central issue to the efficient separationand extraction of vanadium and chromium from vanadium-chromium slag (V-Cr slag) by calcificationroasting. In this study, Fe2VO4 and FeCr2O4 were synthesized to simulate the vanadium chromium spinelin V-Cr slag; then, the reaction mechanism of Fe2VO4-CaO-FeCr2O4 powder system was studied by XRD,SEM and leaching experiments. Results showed that calcium vanadate and calcium chromate are generatedafter roasting Fe2VO4-CaO system and FeCr2O4-CaO system individually. In Fe2VO4-CaO-FeCr2O4 system,as n(CaO)/n(V2O3) is 2, vanadium exists in the form of Ca2V2O7 while almost all Cr2O3 from thedecomposition of FeCr2O4 reacts with Fe2O3 to continuously form solid solution with more FeCr2O4 adding. After leaching, the addition of FeCr2O4 has no obvious effect on the leaching ratio of vanadium, andthe leaching ratio of chromium decrease with FeCr2O4 adding. Consequently, 94.03% of vanadium and0.15% of chromium are leached with n(CaO)/n(V2O3)/n(Cr2O3) of 2/1/1.03 at 900 C. When n(CaO)/n(V2O3) is 4, Fe2VO4 is oxidized and calcified gradually to form Ca2V2O7 and Ca3V2O8 with increasing roastingtemperature. A small amount of CaCrO4 is generated owing to the calcification of chromium, and mostchromium still exists in the form of solid solution. The reaction result of Cr2O3 and Fe2O3 is the superpositionof solid solution reactions with different degrees. Increasing the roasting temperature can significantlyreduce the chromium leaching ratio. All above would provide a theoretical support for thecalcification roasting process from V-Cr slag.

        Improvement in Water Resistance of Desulfurized Gypsum by Novel Modification of Silicone Oil Paraffin Composite Emulsion-based Waterproofing Agent

        In this study, dimethyl silicone oil and liquid paraffin were combined and subsequently emulsified; the resulting mixture was innovatively incorporated into desulfurized gypsum to resolve its drawback of a poor water resistance. The waterproof mechanism of the composite emulsion and liquid paraffin emulsion with mass fractions of 1%, 2%, 3%, and 4% were investigated. The effect of the desulfurized gypsum on the waterproof performance and basic mechanical properties were also investigated. The configuration of the composite waterproofing agent was characterized by FTIR and <sup>1</sup>HNMR. The results showed that, compared with the traditional liquid paraffin emulsion-based waterproofing agent, the softening coefficient of the silicone oil paraffin composite emulsion-based water-repellent agent was increased by 60% and attained a value of 0.89. Combined with the waterproof mechanism and microscope morphology analysis of gypsum hydration products, the improvement in the water resistance of water resistance was primarily attributed to the formation of a silicone hydrophobic membrane between the crystals of the gypsum block; this ensured that water could not penetrate the crystal.

      • Knowledge of Cervical Cancer Screening among Health Care Workers Providing Services Across Different Socio-economic Regions of China

        Background: China carries a heavy burden of cervical cancer (CC) and substantial disparities exist across regions within the country. In order to reduce regional disparities in CC, the government of China launched the National Cervical Cancer Screening Program in Rural Areas (NCCSPRA) in 2009. Critical to the success of the program are the health care workers who play a pivotal role in preventing and managing CC by encouraging and motivating women to use screening services and by providing identification and treatment services. This study aimed to assess cervical cancer knowledge among these health care workers at the county level in maternal and child health (MCH) hospitals across different socio-economic regions of China. Materials and Methods: A cross-sectional survey was conducted and self-administered questionnaires were sent to all health care workers (a total of 66) providing cervical cancer screening services in 6 county level MCH hospitals in Liaoning, Hubei and Shaanxi provinces, representing eastern, central and western regions of China; 64 (97.0%, 64/66) of the workers responded. ANOVA and Chi-square test were used to compare the knowledge rate and scores in subgroups. Results: The knowledge level of the respondents was generally low. The overall combined knowledge rate was 46.9%. The knowledge rates for risk factors, prevention, clinical symptoms, screening and diagnostic tests and understanding of positive results were 31.3%, 37.5%, 18.1%, 56.3% and 84.4%, respectively. Statistically significant differences in scores or rates of CC knowledge were seen across the different regions. The total and sectional scores in the less developed regions were statistically significantly lower than in the other regions. Conclusions: The majority of the health care workers who provide CC screening service in NCCSPRA at county level MCH hospitals do not have adequately equipped with knowledge about CC. Given the importance of knowledge to the program's success in reducing CC burden in rural women in China, efforts are needed to improve the knowledge of health care workers, especially in less developed regions.

      • Protocols for Privacy-Preserving DBSCAN Clustering

        Cooperative computation is one of the most important fields in computer science. In recent years, the development of networking increases the desirability of cooperative computation. But privacy concerns often prevent different parties from sharing their data. Secure multiparty computation techniques can dispel parties’ doubts about revealing privacy information in this situation. On the other hand, Data mining has been a popular research area for more than a decade. However, in many applications, the data are originally collected at different sites owned by different users. This paper considers the problem of privacy preserving DBSCAN clustering over vertically partitioned data based on some results of SMC. An efficient secure intersection protocol is first proposed. The security and complexity of the protocols are also analyzed. The results show that the protocols preserve the privacy of the data and the time complexity as well as the communication complexity is acceptable.

      • Dendritic Cells Induce Specific Cytotoxic T Lymphocytes against Prostate Cancer TRAMP-C2 Cells Loaded with Freeze-thaw Antigen and PEP-3 Peptide

        Prostate cancer is the most common cancer in men. In this study, we investigated immune responses of cytotoxic T lymphocytes (CTLs) against TRAMP-C2 prostate cancer cells after activation by dendritic cells (DCs) loaded with TRAMP-C2 freeze-thaw antigen and/or PEP-3 peptide in vitro. Bone marrow-derived DC from the bone marrow of the C57BL/6 were induced to mature by using the cytokine of rhGM-CSF and rhIL-4, and loaded with either the freeze-thaw antigen or PEP-3 peptide or both of them. Maturation of DCs was detected by flow cytometry. The killing efficiency of the CTLs on TRAMP-C2 cells were detected by flow cytometry, CCK8, colony formation, transwell migration, and wound-healing assay. The levels of the IFN-${\gamma}$, TNF-${\beta}$ and IL-12 were measured by enzyme-linked immunosorbent assay (ELISA). Compared with the unloaded DCs, the loaded DCs had significantly increased expression of several phenotypes related to DC maturation. CTLs activated by DCs loaded with freeze-thaw antigen and PEP-3 peptide had more evident cytotoxicity against TRAMP-C2 cells in vitro. The secretion levels of IFN-${\gamma}$, TNF-${\beta}$ and IL-12, secreted by DCs loaded with antigen and PEP-3 and interaction with T cells, were higher than in the other groups. Our results suggest that the CTLs activated by DCs loaded with TRAMP-C2 freeze-thaw antigen and PEP-3 peptide exert a remarkable killing efficiency against TRAMP-C2 cells in vitro.

        Triterpenoid saponins from Clinopodium chinense (Benth.) O. Kuntze and their biological activity

        Four new ursane-type triterpenoid saponins, clinopoursaponins A–D (1–4), six new oleanane-type triterpenoid saponins, clinopodiside VII–XII (5–10), as well as eight known triterpene analogues (11–18), were isolated from the aerial parts of Clinopodium chinense (Benth.) O. Kuntze. The structures of the new compounds were determined based on extensive spectral analyses, including 1D (1H and 13C) and 2D NMR experiments (COSY, NOESY, HSQC, 2D TOCSY, HSQC-TOCSY and HMBC), HR-ESI-MS and chemical methods. Compounds 1–18 were evaluated for their protective effects against anoxia/reoxygenation-induced apoptosis in H9c2 cells and cytotoxicities against murine mammary carcinoma cell line 4T1. Compounds 8, 9 and 18 exhibited significant protective effects, while compound 1 exhibited cytotoxic activity with IC50 value of 7.4 μm compared to 7.6 μm for the positive control 10-hydroxycamptothecin.

      • Up-regulation of Thy-1 Promotes Invasion and Metastasis of Hepatocarcinomas

        Increasing evidence has revealed that thy-1 was a potential stem cell marker of liver cancer, but no data have been shown on how thy-1 regulates the pathophysiology of liver cancer, such as proliferation, apoptosis, invasion and migration. We previously demonstrated that thy-1 was expressed in about 1% of hepg2 cells, thy-1+hepg2 cells, but not thy-1-, demonstrating high tumorigenesis on inoculation $0.5{\times}10^5$ cells per BACA/LA mouse after 2 months. In the present study, our results showed that higher expression of thy-1 occurs in 72% (36/50 cases) of neoplastic hepatic tissues as compared to 40% (20/50 cases) of control tissues, and the expression of thy-1 is higher in poorly differentiated liver tumors than in the well-differentiated ones. In addition, thy-1 expression was detected in 85% of blood samples from liver cancer patients, but none in normal subjects or patients with cirrhosis or hepatitis. There was a significant negative correlation between thy-1expression and E-cadherin expression (a marker of invasion and migraton), but not between thy-1 expression and AFP expression in all the liver cancer and blood samples. We further investigated the relationship between thy-1 and E-cadherin in liver cancer hepg2 cell line which was transfected with pReceiver-M29/thy-1 eukaryotic expression vector followed by aspirin treatment. Lower expression of E-cadherin but higher expressions of thy-1 were detected in hepg2 cells transfected with pReceiver-M29/thy-1. Taken together, our study suggested that thy-1 probably regulates liver cancer invasion and migration.
