        CK2 phosphorylates AP-2α and increases its transcriptional activity

        Transcription factor AP-2α involves in the process of mammalian embryonic development and tumorigenesis. Many studies have shown that AP-2α functions in association with other interacting proteins. In a two-hybrid screening, the regulatory subunit β of protein casein kinase 2 (CK2β) was identified as an interacting protein of AP-2α; we confirmed this interaction using in-vitro GST pull-down and in-vivo co-immunoprecipitation assays; in an endogenous co-immunoprecipitation experiment, we further found the catalytic subunit α of protein casein kinase 2 (CK2α) also exists in the complex. Phosphorylation analysis revealed that AP-2α was phosphorylated by CK2 kinase majorly at the site of Ser429, and such phosphorylation could be blocked by CK2 specific inhibitor 4,5,6,7-tetrabromobenzotriazole (TBB) in a dose-dependent manner. Luciferase assays demonstrated that both CK2α and CK2β enhanced the transcription activity of AP-2α; moreover, CK2β increased the stability of AP-2α. Our data suggest a novel cellular function of CK-2 as a transcriptional co-activator of AP-2α.

        Development of a Loop-Mediated Isothermal Amplification Assay for Rapid Detection of Nocardia salmonicida, the Causative Agent of Nocardiosis in Fish

        Nocardia salmonicida is one of the main pathogens of fish nocardiosis. The purpose of this study was to build a loop-mediated isothermal amplification (LAMP) method for the rapid and sensitive detection of N. salmonicida. A set of four primers were designed from the 16S-23S rRNA intergenic spacer region of N. salmonicida, and conditions for LAMP were optimized as incubating all the reagents for 60 min at 64°C. LAMP products were judged with agar gel electrophoresis as well as with the naked eye after the addition of SYBR Green I. Results showed the sensitivity of the LAMP assay was 1.68 × 103 CFU/ml (16.8 CFU per reaction) and 10-fold higher than that of PCR. The LAMP method was also effectively applied to detect N. salmonicida in diseased fish samples, and it may potentially facilitate the surveillance and early diagnosis of fish nocardiosis.

        Multifunctional Antibacterial and Hydrophobic Cotton Fabrics Treated with Cyclic Polysiloxane Quaternary Ammonium Salt

        A novel polysiloxane quaternary ammonium salt (NCTSi) was synthesized and used as antibacterial andhydrophobic agent for cotton fabric. The chemical structures were characterized by FT-IR. The surface morphology of treatedcotton fabrics was characterized by scanning electron microscopy (SEM) and energy dispersive spectrometer (EDS). Thecotton fabrics treated with NCTSi showed excellent antibacterial properties against Gram-positive bacteria S. aureus andGram-negative bacteria E. coli. The E. coli antibacterial rate was about 99.52 % and the S. aureus antibacterial rate was about98.33 %, with only a slight decrease after 20 cycles of standard washing, to 91.43 % and 93.33 %, respectively. The inhibitionzone was 2 mm and 2.5 mm against E. coli and S. aureus when NCTSi concentration was 4 %. The treated cotton fabricsprovided excellent hydrophobic properties, and the water contact angle (WCA) reached 124.14 º compared to pure cottonfabrics. The mechanical properties and whiteness were also investigated. As a result of the performed modifications,multifunctional fabrics with a considerably increased hydrophobicity and high antibacterial property were obtained.

        Isolation and Preliminary Identification of a Novel Microorganism Producing Aspartame from Soil Samples

        To develop a new method for producing aspartame (APM), a simple and efficient strategy for the isolation of certain microorganisms producing APM from soil samples was designed. A newly strain with secreting certain specific dipeptidase to directly synthesize APM from the substrates of L-aspartic acid (L-Asp) and Lphenylalanine methyl ester (L-PM) without protection for amino acid side chains was screened from soil samples. APM concentration in reaction mixture was quantified by high performance liquid chromatography (HPLC), yield reached 0.015 g/L. Examination of the general morphological characteristics and data from the sequence analysis of the rDNA-ITS gene led to identification of the isolate as a strain of fungal endophyte spp. The newly isolated strain had a high potential for application in industrial processes for APM production. In particular, this new method was low cost for synthesis of APM during the reaction due to avoiding protection for amino acid side chains and optical resolution of the mixtures. As we known it, this is first report that a newly strain with a high potential for selective synthesis of the APM was isolated from soil.

        Combined water flux enhancement of PES-based TFC membranes in ultrasonic-assisted forward osmosis processes

        For PES-based TFC FO membranes, a significant water flux surge occurred after a moderate ultrasonic irradiation. The Change in membrane transport parameters illustrated that internal concentration polarization (ICP) was weakened due to decreased structural parameter. Combined water flux enhancement via real-time ultrasonic irradiation was obtained and related to the CP effect and membrane orientation. Increasing irradiation intensity was more suitable in PRO mode than in FO mode. Low FS concentration in concentration combinations represented high ratio of ultrasonic-induced flux increment to energy input. Multiplying concentration difference was beneficial to obtain high conversion ratio of ultrasonic power to water flux.

        GPR12 Selections of the Metabolites from an Endophytic Streptomyces sp. Asociated with Cistanches deserticola

        An endophytic Streptomyces sp. (AC-2) was isolated from the root of Cistanches deserticola Y.C.Ma..Chemical investigations of the culture broth of AC-2 afforded fifteen compounds including K1115 A (1), tyrosol (2), phenylethylamine derivatives (3, 4), cyclic dipeptides (5-8), nucleosides and their aglycones (9-13), N-acetyltryptamine (14), and pyrrole-2-carboxylic acid (15). Only tyrosol can promote an increase of intracellular cAMP special on GPR12 transfected cells, such as CHO and HEK293, which means it may be a possible ligand for GPR12.

        1992년 한중 수교 이후부터 한국을 방문한 중국 관광객의 수가 지속적으로 늘어났다. 1993년의 10만 명을 이어 1998년에는 21만 명으로 급속히 증가했고, 한중교류의 해를 맞이한 2007년에 중국인 방한관광객은 100만 명을 돌파하였다. 지리적으로 가까운 나라임에도 사회주의 체제 아래에 있었던 중국은 사회, 경제적으로 ‘가깝고도 먼 나라’로만 존재하였다. 사회주의 경제체제를 내려놓은 중국은 이제 동북아시대의 거점 국가로 대두하고 있다. 2008년 북경올림픽 및 상해세계엑스포를 계기로 중국인의 삶의 질이 한층 더 높아지고 있다. 유럽에서도 중국 관광객의 씀씀이를 주목하기 시작했다. 중국 관광객을 잡기 위해 쇼핑을 하는 그들을 위해 문화체험 프로그램과 연계할 필요성이 있다. 1970년대 초기부터 인구증가를 막기 위해 중국 정부는 ‘산아제한 정책’, 즉 계획생육(計劃生育) 정책을 폈다. 중국의 ‘산아제한 정책(계획생육정책)’은 효과적으로 많은 사회문제를 줄일 수 있었으나 자녀에 대한 지나친 관심과 사랑은 ‘소황제’라 일컫는 문제점을 낳았다. 중국의 주류 사회를 이루고 있는 이들은 중국인의 국민성에도 많은 영향을 미치고 있다. 중국의 부정적 이미지와 함께 ‘소황제’의 사회시스템 일탈로 전반적인 사회문제를 야기하고 있다. 이에 중국 정부와 부모는 미래 인재로 자녀의 바른 경제관념과 직업을 선택하게 해야 할 필요성이 생겼다. 따라서 본 연구의 대상은 중국 미래의 주류 사회의 근간이 될 아동을 대상으로 한다. 이와 같은 연구 대상은 공적인 부분과 사적인 부분을 효율적으로 반영하여 교육관광콘텐츠와 연계를 통해 발굴하고자 한다. 연구 대상으로, 부모와 함께, 혹은 학교 단체 활동의 이유로 한국에 입국하는 초등학생을 주목할 필요가 있다. 한국은 중국의 부모와 정부를 대신하여 놀이를 통해 초등학생의 직업체험을 수행하여 교육적 효과를 얻게 하고, 초등학생은 한국의 직업과 세계의 다양한 직업을 경험함으로써 한국을 재방문할 수 있게 될 것이다. From one hundred thousand to 1993, to 1998, only five years short time grew to Two hundred and ten thousand, and in 2007 the "China-Korea Exchange Year" is a breakthrough in the one million. The 2008 Beijing Olympics and the Shanghai World Expo for the development of Chinese economy after the reform and opening up have a chance. Therefore, European countries have gradually begun to attract attention to Chinese tourists, because of not increasing overseas consumption of these sources and bring to China"s rapid economic development of the Chinese tourists. At the beginning of the year 1970, China in order to control the rapid growth of the population, the implementation of the policy of "family planning". China through the family planning policy to solve the social problems, but at the same time, only children brought about by the family of "Little Emperors(China one)" problem is increasingly significant. And this is also to China"s social, economic and cultural changes and development has deep influence, and become the new problems need to be solved in Chinese society. In this regard, the Chinese government and parents are aware of the necessity of a correct outlook on life and the economic concept of the child"s education. Therefore, the research on how to China"s one-child social and economic right outlook on education research, and through the research on the child education system in South Korea advanced, feasibility of development for Chinese children"s occupation experience educational tourism industry.       • Predicament, Opportunity and Path: Reflections on the Development of Tai Chi in the New Era

        The new era has entrusted the era mission of high-quality development of Tai Chi. Standing in a new historical position, this thesis uses the method of literature and logical analysis to explore the realistic difficulties, opportunities of the times and breakthrough paths in the development of Tai Chi, which is a reflection on the breakthrough of the development of it. The research believes that the development of Tai Chi in the new era has the predicament of slow domestic and international development, the gradual dissipation of martial arts functions, and the gradual weakening of cultural attributes. However, the new era has also brought a new opportunity for the Tai Chi movement to have a clearer development blueprint, a more stable social foundation, and a healthy concept deeply rooted in the hearts of the people. The development of Tai Chi in the new era should be guided by a new pattern of domestic development and international promotion.
