        We performed a variety of lymphocyte stimulation tests, delayed hypersensitivity skin tests, and enumeration of several lymphocyte subpopulations in 21 patients with gastric cancer who did not receive any form of anticancer therapy, and in 20 healthy controls. The gastric cancer patients had significantly decreased number and total score of positive reactions in delayed hypersensitivity skin tests compared with the healthy controls. The percent of CD_(4) positive lymphocytes in the gastric cancer patients was significantly decreased compared to the normal control subjects, but it had no correlation with the total score of delayed cutaneous hypersensitivity reactions. The ratio of helper to suppressor cells was lower in the cancer group. The uptake of 3H-thymidine was markedly depressed in cancer patients when stimulated with various mitogens. There was little correlation between any of the stimulation tests and any of the lymphocyte subpopulation proportions to delayed hypersensitivity cutaneous reactions. Optimal proliferative response was found in lymphocytes stimulated with 10 micrograms of concanavalin-A and 10 microgram of phytohemagglutinin. Advanced stage patients had greatly depressed delayed hypersensitivity skin reactions and proliferative responses to mitogens compared with localized diseases. These results suggest that gastric cancer patients have depressed cellular immune functions, which mainly result from the decreased helper cells and defects in functional proliferative response to mitogens. Interleukin-2 and interferon-gamma restored the in vitro proliferative response of lymphocyte in patients with gastric cancer.

        KAERI 부지 주변의 환경선량 측정을 위한 온라인 감마선량 감시시스템

        환경감마선량 변동을 감시하기 위해 한국원자력연구소 부지 주변에 온라인 감마선량 감시망을 구성하였다. 모니터링 포스트에 설치된 전리함 검출기에 의해 측정된 자료는 무선전송방식으로 중앙통제소에 전송되어 실시간으로 감시된다. 무선전송 방식은 자료의 전송, 처리 및 자자에 신뢰도가 높고 경제적이었다. 감시시스템은 급격한 선량증가시 조기경보체제와 연결할 수 있게 하였다. On-line gamma monitoring system around KSERI-site was set up to monitor the radiation fluctuations in environment. Data on gamma exposure rates measured by the ionication chamber in the monitoring posts are transmitted to a computer of central control station with radio telemetry transmission and reliable on handing and storing of data. This monitroing system can triger an early warning system in the event of abnormal radiation levels.

      • 간세포암의 고주파 열치료 후 천자침 경로를 통한 전이 1례

        Radiofrequency ablation(RFA), as a form of minimally invasive therapy for hepatocellular carcinoma, has become an important treatment modality. Because of limitation of surgery, RFA has become standard therapy for hepatocellular carcinoma in some situations. But there are some complications of RFA such as bleeding, infection, hematoma, adjacent organ thermal damage including intestinal perforation, needle track seeding, and so on. There are few reports in the literature that systematically evaluate the incidence of needle tract seeding and its associated risk factors. And only 2 cases of needle tract seeding of hepatocellular carcinoma after RFA were reported in Korea. We report a case of needle tract seeding of hepatocellular carcinoma after RFA.

      • 일차배양된 설치류 호흡기 상피세포로부터의 점액소 분비에 대한 수종 약물의 영향

        1. PKC activator인 PMA는 일차배양 HTSE세포로부터의 뮤신분비를 0.1μM 농도에서 30%, 1μM 농도에서 80% 가량 증가시켰다. 2. 식물 유래 성분으로, flavonoid의 일종인 TFR은 일치배양 HTSE 세포로부터의 뮤신분비를 10μM 농도에서 50%, 100μM 농도에서 80% 가량 증가시켰다. 3. 양이온성 폴리펩티드인 PLL 및 PLA는 일차 배양 HTSE 세포로부터의 뮤신분비를 0.01 - 10μM 농도에서 용량의존적으로 감소시켰다. 4. 결론적으로, 본 연구에서 얻어진 결과들은 새로운 거담제 및 점액용해제나 단백분해 효소제가 아닌 호흡기 류신의 생성/분비를 조절해 줄 수 있는 신개념의 약물을 개발함에 있어 극히 일부분이나마 단서를 제공하고 있다고 하루 수 있을 것이다. In the present study, we tried to investigate whether phorbol myristate acetate(PMA), trihydroxymethoxy-flavanone rutinoside(TFR) and cationic polypeptides significantly affect mucin release(secretion) from cultured hamster tracheal surface epithelial cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with 3H-glucosamine for 24 hr and chased for 30 min in the presence of each agent to assess the effect on 3H-mucin release. The results were as follows : (1) Both PMA and TFR significantly increased mucin release from cultured HTSE cells ; (2) Cationic polypeptides including po1y-L-lysine(PLL, mw 7,500) and poly-L-arginine(PLA, mw 10,800) significantly inhibited mucin release from cultured HTSE cells, in a dose-dependent manner. This finding suggests us that PMA and TFR be further studied for the possible use as mild expectorants and cationic polypeptides might function as a regulator for hyper-secretion of mucus, both by direct acting on airway mucin-secreting cells, during the treatment of chronic airway diseases.

        The effects of MA packaging on the low temperature storage of satsuma mandarin, overwintering satsuma and Kiyomi tangor were investigated. Thickness of film, holes in film and incorporation of antimicrobial agent were considered as parameters for MA packaging. Chitosan was applied to surface of fruits to determine the effects of coation during MA storage. Thickness of film and holes in a bag did not affect decay ratios in three fruits. Decay ratio of satsuma mandarin packaged with LDPE film with 2-5% ceramic antimicrobial agent slightly decreased. Chitosan coating lowered decay ratio of fruit during MA storage. MA packaging with LDPE film was effective in lowering weight loss and preventing delamination of fruits. MA packaging did not affect qualities of fruit during storage.

        A study on the effect of anaerobic threshold level of skier(Nordic) to the game record has been performed and coucluded as follows 1. Comparing domestic long-distance skiers to foreign superiors, a Wide difference in maximum oxgen uptake has been found to be 8㎖/㎏·min and 16.3㎖/㎏·min for male and female respectively. 2. Comparing domestic long-distance skiers to foreign superiors, anaerobic threshold level has been found to be 10∼20% and 8∼13% lower for male and female respectively. 3. Anaerobic threshold level, the index represent sporting ability with nere maximum oxygen uptake and simultaneously without accumulation of mamary acid for a long time should be promoted to get a better record.

        The differences of the liver scintigraphic features between postnecrotic and nonpostnecrotic liver cirrhosis were studied for 69 patients. Degrees of mottling and splenomegaly were no different between two groups but shrinked form was more frequent finding in postnecrotic type(23:10), and normal shape & enlarged left lobe with shrinked right lobe were relatively dominant in non-postnecrotic group(15:6, 10:2). Grade of extrahepatic uptake of ^99mTc-tin colloid was significantly higher in Child-Pugh B & C group than A group(p<0.05), but there was no difference between postnecrotic and non-postnecrotic groups. These will be important results on the dicision for the presence of cirrhosis according to the variable causes.

        Elevated extracellular calcium ions promote proliferation and migration of mesenchymal stem cells via increasing osteopontin expression

        <▼1><P>Supplementation of mesenchymal stem cells (MSCs) at sites of bone resorption is required for bone homeostasis because of the non-proliferation and short lifespan properties of the osteoblasts. Calcium ions (Ca<SUP>2+</SUP>) are released from the bone surfaces during osteoclast-mediated bone resorption. However, how elevated extracellular Ca<SUP>2+</SUP> concentrations would alter MSCs behavior in the proximal sites of bone resorption is largely unknown. In this study, we investigated the effect of extracellular Ca<SUP>2+</SUP> on MSCs phenotype depending on Ca<SUP>2+</SUP> concentrations. We found that the elevated extracellular Ca<SUP>2+</SUP> promoted cell proliferation and matrix mineralization of MSCs. In addition, MSCs induced the expression and secretion of osteopontin (OPN), which enhanced MSCs migration under the elevated extracellular Ca<SUP>2+</SUP> conditions. We developed in vitro osteoclast-mediated bone resorption conditions using mouse calvaria bone slices and demonstrated Ca<SUP>2+</SUP> is released from bone resorption surfaces. We also showed that the MSCs phenotype, including cell proliferation and migration, changed when the cells were treated with a bone resorption-conditioned medium. These findings suggest that the dynamic changes in Ca<SUP>2+</SUP> concentrations in the microenvironments of bone remodeling surfaces modulate MSCs phenotype and thereby contribute to bone regeneration.</P></▼1><▼2><P><B>Bone formation: released calcium could stimulate stem cells</B></P><P>Studies with mouse cells and bone samples indicate how calcium ions released during the normal turnover of bone promote the supplementation of stem cells needed to make new bone during routine “remodeling” and regeneration after injury. Our bones are degraded and remodeled continuously, requiring the controlled supplement of stem cells that create bone-forming cells called osteoblasts. Jeong-Tae Koh and colleagues at Chonnam National University, Gwangju, South Korea, exposed the relevant stem cells to increased concentrations of external calcium ions. This promoted cell proliferation and migration that allow the cells to support new bone formation. The research suggests this effect on stem cells may be achieved by calcium ions released directly from bone as it is naturally degraded. The findings may help researchers develop new ways to encourage bone regeneration after injury and surgery.</P></▼2>

        수 종의 상피기원 종양 세포주에서 방사선 조사와 표피성장인자 투여에 따른 세포 주기의 변화와 apoptosis 유발에 관한 연구

        Purpose : This study was aimed to evaluate the cell cycle arrest and apoptosis induction after irradiation and epidermal growth factor(EGF) treatment in three human epithelial tumor cell lines (A43l, Siha, KB). Materials and Methods : Single irradiation of 2, 5 and 10 Gy was done on three cell lines with 5.38 Gy/min dose rate using Cs-137 irradiator at room temperature. Also, EGF of 10 ng/ml was added immediately after 10 Gy irradiation. Cell growth was evaluated by counting the living cell number using a hemocytometer at 1 day, 2 days, 3 days, 4 days and 5 days after irradiation. Cell cycle arrest and apoptosis induction were assayed with the flow cytometry at 8 hours, 12 hours, 1 day, 2 days, 3 days, 4 days and 5 days after irradiation. Results : Growth of irradiated three cell lines were inhibited in proportion to radiation dose, EGF treatment after irradiation showed various results according to cell lines. On all cell lines, G2 arrest was detected after 8 hours and maximized after 12 hours or 1 day. Amount of G2 arrest was positively dose dependent. However, EGF showed no significant change on G2 arrest. G2 arrest was recovered with time at 2 Gy and 5 Gy irradiation. However, at 10 Gy irradiation, G2 arrest was continued. Apoptosis was detected at 10 Gy irradiation. On EGF treated group after irradiation, A431 and Siha cell lines showed slightly increased apoptosis but there was no statistically significant difference. KB cell line showed no marked change of apoptosis induction. Conclusion : Irradiation effects on cell cycle arrest and apoptosis induction in three human epithelial tumor cell lines, however epidermal growth factor doesn't effect on.
