        전치부 개방교합과 정상교합자의 근활성도에 관한 비교연구

        This study was undertaken to compare the activity of masticatory muscle between normal occlusion and anterior openbite. 33 subjects without the experience of orthodontic treatment, missing teeth and the symptom of T. M. disorders were selected for this study : 25 subjects were normal occlusion and 8 subjects were anterior openbite. The ten items were measured from the cephalometric headplates, and EMG recordings of the anterior temporal and masseter muscle were taken at rest position and during maximum clenching at centric occlusion. All data were analyzed and processed with the computer statistical method. The following results were obtained : 1. At rest position, the muscle activities of both temporal and masseter muscle were higher in anterior openbite than in normal occlusion. 2. During maximum clenching, the muscle activities of both temporal and masseter muscle were prominently lower in anterior openbite than in normal occlusion. 3. At rest position, the temporal muscle of anterior openbite showed the highest activity, but showed the lowest muscle activity during maximum clenching. 4. Anterior openbite showed closer interrelationship between facial morphology and the muscle activity, and the muscle activity was more influenced by the form of mandible than that of maxilla.

      • 지구성 운동이 자연발생 고혈압 흰쥐의 혈압에 미치는 영향

        The purpose of this study is to investigate the effects of indurance exercise on BP of SHR Experimental animals were SHR(180) and SD(90) and they were divided into three groups ; a experimental group1( practice group), a experiment group2(non practice group), and a comparative group (normal group). The experimental group 1 was practicing on a treadmill at 15m/min exercise intensity for 30 minutes a day four times per week for eight weeks. While raising experimental animals, We measured BP for each group once a week, and took the weight of the heart after killing a rat. and then we examined the concentration of ?? and ??, compared, and analyzed them. we have reached the following results. 1. As time passed, BP in the experimental group 1 was almost equal to BP eight weeks ago. BP in the experimental group 2 continued to rise and then showed typical hypertension. 2. The three groups were inclined to increase the weight of their hearts as time passed. However, the experimental group 2 presented the weight of hearts increasing compared with the experimental group1 after eight weeks. 3. In the experimental group 1, and experimental group 2 groups, ?? concentration was but ?? concentration increased as time passed. However, the experimental group1 showed fewer changes than the experimental group 2. 4. The weight of all three groups were inclined to increase as time passed, but it didn't present any noteworthy differences. According to the analysed result of the above experiment, endurannsce exercise becomes an important factor in preventing the raising of BP of SHR, and the hypertrophy of hearts.       • Bupivacaine과 ropivacaine이 Xenopus oocyte에 발현된 HERG 전류에 미치는 영향

        Bupivacaine is an amide-type local anesthetic widely used for regional anesthesia. Ropivacaine is developed as a less cardiotoxic alternatives to bupivacaine. In the present study, we have analyzed the effects of bupivacaine and ropivacaine on HERG currents expressed in Xenopus oocytes. Bupivacaine and ropivacaine(3∼1,000μM) blocked HERG currents in a concentration dependent manner. EC_(50) was 26.1±3.1μM(n_(R) 0.65±0.04) and 43.5±7.9μM(n_(H) 0.99±0.13) in bupivacaine and ropivacaine, respectively. Bupivacaine and ropivacaine did not affect the activation and deactivation kinetics of HERG channels. However, the drugs decreased the slope conductance measured from fully activated current-voltage relationship curves. These results suggest that bupivacaine and ropivacaine have a similarinhibitory effect on HERG channels, which could be a possible cellular mechanism of LQT or ventricular arrythmia by the drugs.

      • Endothelium-dependent Contraction of Aorta in One-kidney, One-clip Goldblatt Hypertensive Rat

        The mechanism of impaired endothelium-dependent relaxation in the aorta of one-kidney, one clip Goldblatt hypertensive (1K,1C-GBH) rats was investigated. 8 week-old Wistar-Kyoto (WKY) rats were made hypertensive by left renal artery stenosis with contralateral nephrectomy. Endothelium-dependent relaxation was significantly reduced in 1K,1C-GBH rats as compared with WKY rats. However, the relaxation by sodium nitroprusside in 1K,1C-GBH rats was not reduced as compared with WKY rats. The impairment of endothelium-dependent relaxation in 1K,1C-GBH rats was partially restored by the pretreatment of indomethacin or SQ29548. When the nitric oxide production was inhibited by L-nitroarginine methyl ester, acetylcholine (ACh) induced a endothelium-dependent contraction that was greater in 1K,1C-GBH rats than in WKY rats. Endothelium-dependent contraction by ACh was completely abolished by indomethacin or SQ29548. However, imidazole, tranylcypromine and superoxide dismutase did not affect the endothelium-dependent contraction in 1K,1C-GBH rats. These results suggest that impaired endothelium-dependent relaxation in the 1K,1C-GBH rats might be due to the simultaneous release of EDCF, and that prostaglandin B2 may be involved as a mediator of endothelium-dependent contraction.

      • Effect of Blood Pressure on the Endothelium-Dependent Contraction in Rat Aorta

        To investigate the mechanisms of increased endothelium-dependent contraction by acetylcholine in hypertensive rats, the relationship between endothelium-dependent contraction by acetylcholine and blood pressure was studied in spontaneously hypertensive rats (SHR), one-kidney, one clip Goldblatt hypertension (1K,1C-GBH) rats, and Wistar-Kyoto rats (WKY). SHR were treated orally with enalapril or nicardipine in order to prevent development of hypertension or suppress the developed hypertension. 1K,1C-GBH rats were made by renal artery stenosis with contralateral nephrectomy in 8 week-WKY. 1. Endothelium-dependent contractions by acetylcholine (10<sup>-6</sup> ~ 10<sup>-5</sup> M) in SHR were significantly greater than those in WKY. 2. Chronic treatment with enalapril or nicardipine reduced the endothelium-dependent contraction in SHR 3. The degree of reduction of endothelium-dependent contraction was greater in SHR which was prevented from developing hypertension than in SHR of which high blood pressure was suppressed. 4. In aortic rings from 1K,1C-GBH rats, endothelium-dependent contractions by acetylcholine were augmented as compared with WKY. 5. There is good relationship between the value of blood pressure and magnitude of endothelium-dependent contraction. Thus, it is suggested that increased endothelium-dependent contraction in hypertensive rats may he due to the high blood pressure and endothelium-dependent contraction may not be a cause of the initiation of hypertension in SHR.

        A standardized bamboo leaf extract inhibits monocyte adhesion to endothelial cells by modulating vascular cell adhesion protein - 1

        Bamboo leaves (Phyllostachys pubescens Mazel ex J. Houz (Poacea)) have a long history of food and medical applications in Asia, including Japan and Korea. They have been used as a traditional medicine for centuries. We investigated the mechanism of anti-inflammatory activity of a bamboo leaf extract (BLE) on tumor necrosis factor-alpha (TNF-α)-induced monocyte adhesion in human umbilical vein endothelial cells (HUVECs). Exposure of HUVECs to BLE did not inhibit cell viability or cause morphological changes at concentrations ranging from 1 μg/ml to 1 mg/ml. Treatment with 0.1 mg/ml BLE caused 63% inhibition of monocyte adhesion in TNF-α-activated HUVECs, which was associated with 38.4% suppression of vascular cell adhesion molecule-1 expression. Furthermore, TNF-α-induced reactive oxygen species generation was decreased to 47.9% in BLE treated TNF-α-activated HUVECs. BLE (0.05 mg/ml) also caused about 50% inhibition of interleukin-6 secretion from lipopolysaccharide-stimulated monocyte. The results indicate that BLE may be clinically useful as an anti-inflammatory or anti-oxidant for human cardiovascular disease including atherosclerosis.

      • 가토 신동맥 평활근에서 Barium의 수축작용

        The contractile action of barium (Ba<sup>2+</sup>) was investigated in the arterial strip of rabbit renal artery. The helical strip of isolated renal artery was immersed in the Tris-buffered Tyrode s solution equilibrated with 100% O<sup>2</sup> at 37℃ and its isometric tension was measured. Ba<sup>2+</sup>-induced contraction of arterial strip was dose-dependent and its maximal tension corresponded to 92.1±4.5% of tension by K<sup>+</sup>(100 mM). Ba<sup>2+</sup>-induced contraction did not show the tachyphylactic phenomenon in the normal Tyrode s solution. Ba<sup>+2</sup> induced the tonic contraction in the Ca<sup>2+</sup>-free tyrode s solution and that was increased by the extracellula addition of Ca<sup>2+</sup>. During the repeated exposure of the same dose of Ba<sup>2+</sup> (10 mM) in the Ca<sup>2+</sup>-free Tyrode s solution, Ba<sup>2+</sup>-induced contraction was progressively decreased. Even though the intracellular NE-and caffeine-sensitive Ca<sup>2+</sup> was depleted, Ba<sup>+2</sup> induced the tonic contraction. After the pretreatment of lanthnum or verapamil, Ba<sup>+2</sup> did not induce contraction. Ba<sup>2+</sup>-inducedcontraction was suppressed by extracellular K<sup>+</sup> in the normal Tyrode s solution and that was dependent on K<sup>+</sup> concentration. Suppressive effect of K<sup>+</sup> (14 mM) on the Ba<sup>2+</sup>-induced contraction was also dependent on the intracellular Ca<sup>2+</sup> concentration. From the above resuts, it is suggested that Ba<sup>+2</sup> activate indirectly the contractile process by promoting the mobilization of intracellular Ca<sup>2+</sup> and the influx of extracellular Ca<sup>2+</sup>. It is also suggested that action of Ba<sup>+2</sup> on the Ca<sup>2+</sup>-activated K<sup>+</sup> channel can result in the depolarization of cell membrane in the rabbit renal artery.

      • 고혈압백서 대동맥에서 Ca에 의한 수축에 미치는 Na/Ca교환기전의 영향

        We investigated the contraction by sodium-calcium(Na-Ca) exchanges in the aortic strips of spontaneously hypertensive rats(SHR) and Wistar-Kyoto (WKY) rats. Resting tension of aortic strips in SHR was decreased by the removal of extracellular Ca and those were recovered by the addition of Ca. However, resting tone of aortic strips in WKY was not changed by Ca removal or Ca addition. Ca-induced contraction after exposure to Ca-free solution was suppressed by verapamil 10^-6M to 90% or more. In the presence of verapamil 10^-5M, phentolamine 10^-6M, caffeine 5mM and atropine 10^-6M, the reduction of extracellular sodium concentration([Na]o) from normal(158 mM) to 0 mM(replaced by choline chloride) caused the contraction(Ca entry by Na-Ca exchange). The amplitude of contraction was more greater in SHR than in WKY rats. In the presence of verapamil 10^-5M, phentolamine 10^-6M, caffeine 5mM and atropine 10^-6M, aortic strips were not contracted by Naand Ca-free solution, but was contracted by the addition of Ca. In the presence of verapamil 10^-5M and phentolamine 10^-6M and ouabain 10^-5M, Ca did not induce contraction after exposure to Ca-free Tyrode ' s solution, but in the presence of verapamil 10^-5M and phentolamine 10^-6M and monensin 10^-5M a sodium ionophore, Ca induced a little contraction in the aortic strips of SHR. These observations suggest that Na-Ca exchange does not contribute the development of Ca-induced contraction after exposure to Ca-free Tyrode ' s Solution in the presence of normal extracellular Na concentration. However, Na/Ca exchange in vascular smooth muscle is increased in SHR and may be involved in the mechanism for hypertension in SHR.
