      • Motivation Versus Intention of Sharing Fake News Among Social Media Users during the Pandemic - A SEM Model

        Are intentions important in deciding the outcome of an action such as sharing misinformation among social media users during the pandemic? What is their role and how far they are important for the very act of fake sharing news? The social media users' actions on the social platform are determined by what they plan to do themselves; however, their motivation has an immense role to play in the dissemination of fake news on social media. The study proposes a conceptual model for understanding how select factors affect fake news sharing motivation and intentions of social media users. The study scrutinizes the relationship between content and context, fear of missing out (FoMO), news verification and news sharing gratification on the motivation and intention of social media users of networked Asian society. Empirical Data were drawn from social media users (N = 243) from India, using an online questionnaire based on prior studies and structural equation modeling (SEM) approach was used to analyze the data collected. Results indicate that news content, news verification, and news sharing gratification have a direct and positive relationship with sharing motivation. On the other hand, news context and content, FoMO and news sharing gratification have a positive significant relationship with sharing intention. Likewise, it was discovered that news verification will decrease sharing intention of the social media users. However, news context, that is the pandemic in the case of the present study and FoMO were not identified as determinant variables for sharing motivation among social media users. The research limitations and further scope were discussed.

        Identification and Characterization of Two New S-Adenosylmethionine- Dependent Methyltransferase Encoding Genes Suggested Their Involvement in Stipe Elongation of Flammulina velutipes

        Two new SAM-dependent methyltransferase encoding genes (fvsmt1 and fvsmt2) were identified from the genome of Flammulina velutipes. In order to make a comprehensive characterization of both genes, we performed in silico analysis of both genes and used qRT-PCR to reveal their expression patterns during the development of F. velutipes. There are 4 and 6 exons with total length of 693 and 978 bp in fvsmt2 and fvsmt1, respectively. The deduced proteins, i.e., FVSMT1 and FVSMT2 contained 325 and 230 amino acids with molecular weight 36297 and 24894 Da, respectively. Both proteins contained a SAM-dependent catalytic domain with signature motifs (I, p-I, II, and III) defining the SAM fold. SAM-dependent catalytic domain is located either in the middle or at the N-terminal of FVSMT2 and FVSMT1, respectively. Alignment and phylogenic analysis showed that FVSMT1 is a homolog to a protein- arginine omega-N-methyltransferase, while FVSMT2 is of cinnamoyl CoA O-methyltransferase type and predicted subcellular locations of these proteins are mitochondria and cytoplasm, respectively. qRT-PCR showed that fvsmt1 and fvsmt2 expression was regulated in different developmental stages. The maximum expression levels of fvsmt1 and fvsmt2 were observed in stipe elongation, while no difference was found in mycelium and pileus. These results positively demonstrate that both the methyltransferase encoding genes are involved in the stipe elongation of F. velutipes.

        Property profile of nanostructured blends of amine functionalized elastomer and epoxy

        Pure polystyrene-b-poly(ethylene-ran-butylene)-b-polystyrene (SEBS) was functionalized with amine moietyfirst through nitration, followed by reduction. The resulting amine modified SEBS was blended with various amountsof epoxy via in situ reactive approach. Thin blend films were initially cured at 120 oC for 30min and post cured at180 oC for 2 h. These films were then analyzed for their mechanical, thermal and morphological profile. Optimumimprovement in tensile strength, modulus and toughness was observed with different epoxy loading in the blends. These blends were found thermally stable up to 300 oC. The morphological studies indicated ample compatibilitybetween the two components of blends.

        Aberrant Promoter Methylation at CpG Cytosines Induce the Upregulation of the E2F5 Gene in Breast Cancer

        Purpose: The promoter methylation status of cell cycle regulatory genes plays a crucial role in the regulation of the eukaryotic cell cycle. CpG cytosines are actively subjected to methylation during tumorigenesis, resulting in gain/loss of function. E2F5 gene has growth repressive activities; various studies suggest its involvement in tumorigenesis. This study aims to investigate the epigenetic regulation of E2F5 in breast cancer to better understand tumor biology. Methods: The promoter methylation status of 50 breast tumor tissues and adjacent normal control tissues was analyzed. mRNA expression was determined using SYBR® green quantitative polymerase chain reaction (PCR), and methylation- specific PCR was performed for bisulfite-modified genomic DNA using E2F5-specific primers to assess promoter methylation. Data was statistically analyzed. Results: Significant (p<0.001) upregulation was observed in E2F5 expression among tumor tissues, relative to the control group. These samples were hypo-methylated at the E2F5 promoter region in the tumor tissues, compared to the control. Change in the methylation status (Δmeth) was significantly lower (p=0.022) in the tumor samples, indicating possible involvement in tumorigenesis. Patients at the postmenopausal stage showed higher methylation (75%) than those at the premenopausal stage (23.1%). Interestingly, methylation levels gradually increased from the early to the advanced stages of the disease (p<0.001), which suggests a putative role of E2F5 methylation in disease progression that can significantly modulate tumor biology at more advanced stage and at postmenopausal age (Pearson’s r=0.99 and 0.86, respectively). Among tissues with different histological status, methylation frequency was higher in invasive lobular carcinoma (80.0%), followed by invasive ductal carcinoma (46.7%) and ductal carcinoma in situ (20.0%). Conclusion: Methylation is an important epigenetic factor that might be involved in the upregulation of E2F5 gene in tumor tissues, which can be used as a prognostic marker for breast cancer.

        Identification and Expression Patterns of fvexpl1, an Expansin-Like Protein-Encoding Gene, Suggest an Auxiliary Role in the Stipe Morphogenesis of Flammulina velutipes

        Expansins are cell wall proteins that mediate cell wall loosening and promote specific tissue and organ morphogenesis in plants and in some microorganisms. Unlike plant expansins, the biological functions of fungal expansin-like proteins have rarely been discussed. In the present study, an expansin-like protein-encoding fvexpl1 gene, was identified from Flammulina velutipes by using local BLAST. It consisted of five exons with a total length of 822 bp. The deduced protein FVEXPL1 contained 274 amino acids with a predicted molecular mass and isoelectric point of 28,589 Da and pH 4.93, respectively. The first 19 amino acids from the N terminal are the signal peptide. Phylogenetic analysis and multiple protein alignment indicated FVEXPL1 was an expansin-like protein. The expression level of fvexpl1 gene in the stipe was significantly higher than that in the mycelia, primordia, and cap. However, the expression level of fvexpl1 gene was significantly higher in the fast elongation region of the stipe as compared with the slow elongation region. Expression analysis indicated that fvexpl1 gene might have an auxiliary role in the stipe morphogenesis of F. velutipes.

        Superior dye degradation using SnO2-ZnO hybrid heterostructure catalysts

        We investigated the efficiency of oxide based hierarchical heterostructure as adsorbent for the treatment of organic dyes, Methyl orange (MO) and Methylene Blue (MB), containing solution. Nanocrystals such as ZnO nanorods (at various temperatures of 30, 60 and 75 oC) and SnO2 nanoparticles were synthesized by electrodeposition method and hydrothermal approaches, respectively. SnO2-ZnO heterostructures were formed by spin coating SnO2 nanoparticles on ZnO nanorods matrix to form a heterostructured film. The surface morphologies and structural characterization of as-prepared heterostructures were investigated by scanning electron microscopy (SEM), transmission electron microscopy (TEM) and X-ray diffraction (XRD) techniques. While, absorption spectra of all samples were examined by UV-vis diffuse reflectance spectroscopy. The photocatalytic activities of as-prepared samples for organic dyes degradation were tested under UV light as model reaction. The SnO2-ZnO heterostructured photocatalyst showed superior activities than individual ZnO and SnO2 nanocrystals. This heightened behavior was attributed to its better charge separation capability and the slow charge recombination originating due to difference in energy values of conduction band edges of SnO2 and ZnO. The SnO2-ZnO heterostructure demonstrated better stability and recyclability up to five times, which is highly desirable for potential industrial applications including dye degradation and wastewater treatment systems.

        Content Based Image Retrieval Using Combined Features of Shape, Color and Relevance Feedback

        Content based image retrieval is increasingly gaining popularity among image repository systems as images are a big source of digital communication and information sharing. Identification of image content is done through feature extraction which is the key operation for a successful content based image retrieval system. In this paper content based image retrieval system has been developed by adopting a strategy of combining multiple features of shape, color and relevance feedback. Shape is served as a primary operation to identify images whereas color and relevance feedback have been used as supporting features to make the system more efficient and accurate. Shape features are estimated through second derivative, least square polynomial and shapes coding methods. Color is estimated through max-min mean of neighborhood intensities. A new technique has been introduced for relevance feedback without bothering the user.

        Mitochondrial genome and diverse inheritance patterns in Pleurotus pulmonarius

        Pleurotus pulmonarius, a member of the Pleurotaceae family in Basidiomycota, is an edible, economically important mushroom in most Asian countries. In this study, the complete mitochondrial genomes (mtDNA) of three P. pulmonarius strains – two monokaryotic commercial (J1-13 and ZA3) and one wild (X1-15) – were sequenced and analyzed. In ZA3 and X1-15, the mtDNA molecule was found to be a single circle of 68,305 bp and 73,435 bp, respectively. Both strains contain 14 core protein-coding genes and two ribosomal RNA (rRNA) subunit genes. The ZA3 strain has 22 transfer RNA (tRNA) genes and nine introns: eight in cytochrome c oxidase subunit 1 (cox1), and one in the rRNA large subunit (rnl). Monokaryotic J1-13 and ZA3 mtDNAs were found to be similar in their structure. However, the wild strain X1-15 contains 25 tRNA genes and only seven introns in cox1. Open reading frames (ORFs) of ZA3/J1-13 and X1-15 encode LAGLIDADG, ribosomal protein S3, and DNA polymerase II. In addition, mtDNA inheritance in J1-13, ZA3, and X1-15 was also studied. Results showed that the mtDNA inheritance pattern was uniparental and closely related to dikaryotic hyphal location with respect to the parent. Results also show that mtDNA inheritance is influenced by both the parental nuclear genome and mitogenome in the zone of contact between two compatible parents. In summary, this analysis provides valuable information and a basis for further studies to improve our understanding of the inheritance of fungal mtDNA.
