        Synthesis and characterization of ester-diol based polyurethane: a potentiality check for hypopharyngeal tissue engineering application

        Hypopharyngeal tissue engineering is increasing rapidly in this developing world. Tissue damage or loss needs the replacementby another biological or synthesized membrane using tissue engineering. Tissue engineering research is emergingto provide an effective solution for damaged tissue replacement. Polyurethane in tissue engineering has successfully beenused to repair and restore the function of damaged tissues. In this context, Can polyurethane be a useful material to dealwith hypopharyngeal tissue defects? To explore this, here ester diol based polyurethane (PU) was synthesized in two steps:firstly, polyethylene glycol 400 (PEG 400) was reacted with lactic acid to prepare ester diol, and then it was polymerizedwith hexamethylene diisocyanate. The physical, mechanical, and biological testing was done to testify the characterizationof the membrane. The morphology of the synthesized membrane was investigated by using field emission scanning electronmicroscopy. Functional groups of the obtained membrane were characterized by fourier transform infrared spectroscopyspectroscopy. Several tests were performed to check the in vitro and in vivo biocompatibility of the membrane. A highlyconnected homogeneous network was obtained due to the appropriate orientation of a hard segment and soft segment in thesynthesized membrane. Mechanical property analysis indicates the membrane has a strength of 5.15 MPa and strain 124%. The membrane showed high hemocompatibility, no cytotoxicity on peripheral blood mononuclear cell, and susceptible todegradation in simulated body fluid solution. Antimicrobial activity assessment has shown promising results against clinicallysignificant bacteria. Primary hypopharyngeal cell growth on the PU membrane revealed the cytocompatibility andsubcutaneous implantation on the back of Wistar rats were given in vivo biocompatibility of the membrane. Therefore, thesynthesized material can be considered as a potential candidate for a hypopharyngeal tissue engineering application.

        Bootstrapping Regression Residuals

        The sample reuse bootstrap technique has been successful to attract both applied and theoretical statisticians since its origination. In recent years a good deal of attention has been focused on the applications of bootstrap methods in regression analysis. It is easier but more accurate computation methods heavily depend on high-speed computers and warrant tough mathematical justification for their validity. It is now evident that the presence of multiple unusual observations could make a great deal of damage to the inferential procedure. We suspect that bootstrap methods may not be free from this problem. We at first present few examples in favour of our suspicion and propose a new method diagnostic-before-bootstrap method for regression purpose. The usefulness of our newly proposed method is investigated through few well-known examples and a Monte Carlo simulation under a variety of error and leverage structures.

        Simultaneous Identification of Multiple Outliers and High Leverage Points in Linear Regression

        The identification of unusual observations such as outliers and high leverage points has drawn a great deal of attention for many years. Most of these identifications techniques are based on case deletion that focuses more on the outliers than the high leverage points. But residuals together with leverage values may cause masking and swamping for which a good number of unusual observations remain undetected in the presence of multiple outliers and multiple high leverage points. In this paper we propose a new procedure to identify outliers and high leverage points simultaneously. We suggest an additive form of the residuals and the leverages that gives almost an equal focus on outliers and leverages. We analyzed several well-referred data set and discover few outliers and high leverage points that were undetected by the existing diagnostic techniques.

        Induction of Apoptosis in Intestinal Toxicity to a Histone Deacetylase Inhibitor in a Phase I Study with Pelvic Radiotherapy

        Purpose When integrating molecularly targeted compounds in radiotherapy, synergistic effects of the systemic agent and radiation may extend the limits of patient tolerance, increasing the demand for understanding the pathophysiological mechanisms of treatment toxicity. In this Pelvic Radiation and Vorinostat (PRAVO) study, we investigated mechanisms of adverse effects in response to the histone deacetylase (HDAC) inhibitor vorinostat (suberoylanilide hydroxamic acid, SAHA) when administered as a potential radiosensitiser. Materials and Methods This phase I study for advanced gastrointestinal carcinoma was conducted in sequential patient cohorts exposed to escalating doses of vorinostat combined with standard-fractionated palliative radiotherapy to pelvic target volumes. Gene expression microarray analysis of the study patient peripheral blood mononuclear cells (PBMC) was followed by functional validation in cultured cell lines and mice treated with SAHA. Results PBMC transcriptional responses to vorinostat, including induction of apoptosis, were confined to the patient cohort reporting dose-limiting intestinal toxicities. At relevant SAHA concentrations, apoptotic features (annexin V staining and caspase 3/7 activation, but not poly-(ADP-ribose)-polymerase cleavage) were observed in cultured intestinal epithelial cells. Moreover, SAHA-treated mice displayed significant weight loss. Conclusion The PRAVO study design implemented a strategy to explore treatment toxicity caused by an HDAC inhibitor when combined with radiotherapy and enabled the identification of apoptosis as a potential mechanism responsible for the dose-limiting effects of vorinostat. To the best of our knowledge, this is the first report deciphering mechanisms of normal tissue adverse effects in response to an HDAC inhibitor within a combined-modality treatment regimen.

        Impact of altered environment and early postnatal methamphetamine exposure on serotonin levels in the rat hippocampus during adolescence

        Background: Methamphetamine (MA) is a highly abused psychostimulant across all age groups including pregnant women. Because developing brain is vulnerable by the action of drugs, or other noxious stimuli, the aim of our study was to examine the effect of early postnatal administration of MA alone or in combination with enriched environment (EE) and/or stress of separate housing, on the levels of serotonin (5HT) in the hippocampus of male rat pups at three stages of adolescence (postnatal day (PND) 28, 35 and 45). MA (5 mg/kg/ml) was administered subcutaneously (sc) to pups (direct administration), or via mothers' milk between PND1 and PND12 (indirect administration). Controls were exposed saline (SA). Pups were exposed to EE and/or to separation from the weaning till the end of the experiment. Results: On PND 28, in sc-treated series, EE significantly increased the muted 5HT in SA pups after separation and restored the pronounced inhibition of 5HT by MA. No beneficial effect of EE was present in pups exposed to combination of MA and separation. 5HT development declined over time; EE, MA and separation had different effects on 5HT relative to adolescence stage. Conclusions: Present study shows that MA along with environment or housing affect 5HT levels, depending on both the age and the method of application (direct or indirect). These findings extend the knowledge on the effects of MA alone and in combination with different housing conditions on the developing brain and highlight the increased sensitivity to MA during the first few months after birth.

        Calculation and measurement of Al prompt capture gammas above water in a pool-type reactor

        Prompt capture gammas are an important part of the fission reactor gamma field. Because some of the structural materials after neutron capture can emit photons with high energies forming the dominant component of the gamma spectrum in the high energy region, the following study of the high energy capture gamma was carried out. High energy gamma radiation may play a major role in areas of the radiation sciences as reactor dosimetry. The HPGe measurements and calculations of the high-energy aluminum capture gamma were performed at two moderator levels in the VR-1 pool-type reactor. The result comparison for nominal levels was within two sigma uncertainties for the major 7.724 MeV peak. A larger discrepancy of 60% was found for the 7.693 MeV peak. The spectra were also measured using a stilbene detector, and a good agreement between HPGe and stilbene was observed. This confirms the validity of stilbene measurements of gamma flux. Additionally, agreement of the wide peak measurement in 7e9.2 MeV by stilbene detector shows the possibility of using the organic scintillators as an independent power monitor. This fact is valid in these reactor types because power is proportional to the thermal neutron flux, which is also proportional to the production of capture gammas forming the wide peak
