      • Urea 박테리아 전극에 관한 연구

        Proteus vulgaris 를 ammonia와 carbon dioxide gas-sensor에 고정시켜 감응도와 재현성이 우수한 요소에 대한 박테리아 전극을 직접 제조하여 pH, 온도, 완충용액, 박테리아의 양, 여러 가지 아미노산 및 무기염류에 대한 영향과 전극의 수명 등에 관하여 조사하였다. 먼저 ammonia-박테리아 전극은 25℃에서 pH 7.4인 0.05M phosphate 완충용액을 사용하였을 때가 최적이었으며, 7.0 ×10 exp (-4) -3.0 ×10 exp (-2)M 내에서 직선을 나타내었고, 이때 감응도는 116.7mV/decade였다. 한편, carbon dioxide-박테리아 전극은 30℃, pH7.0, 0.1M phosphate 완충용액에서 가장 우수한 감응을 보였다. 이 경우는 7.0 ×10 exp (-4) - 5.0 ×10 exp (-2)M 내에서 45.4 - 45.7mV/decade를 나타내었다. 이들 두 전극중에서 carbon deoxide-박테리아 전극을 이용하여 요소를 정량한다면 방해물질들의 방해가 최소화되었다. 실제 임상적 응용에서 분광 광도법과 비교한 결과 실험과정이 간단하고 편리하여 신속하게 많은 양의 시료분석이 가능하다. Bio-electrodes for urea have been constructed by immobilizing the Proteus vulgaris on ammonia and carbon dioxide gas-sensors. The bacteria containing urease convert each molecule of urea into two molecules of ammonia and one molecule of carbon dioxide gas. Ammonia-bacterial electrode based on ammonia gas-sensor has linearity in the range of 7.0 ×10 exp (-4) - 3.0 ×10 exp (-2)M urea in pH 7.4, 0.05M phosphate buffer solution at 25℃. While carbon dioxide-bacterial electrode based on carbon dioxide gas-sensor has linearity in the range of 7.0 ×10 exp (-4) - 5,0 ×10 exp (-2)M urea in pH 7.0, 0.1M phosphate buffer solution at 30℃. These electrodes were investigated for the effects of pH, temperature, buffer solutions, bacterial amounts and interferences, and life time. As the clinical applications, urea in urine was determined by these devices and was compared with spectrophotometric method. As the results, those methods were by far more simple and rapid than these method.

      • Ag_2Se電極을 利用한 할로겐산이온과 할로겐화이온의 混合物의 電位差 適定

        銀 이온 選擇性 電極을 指市電極으로 使用하여 브롬산이온(BrO_3^-)과 요오드산이온 (IO_3^-)의 단독정량과 요오드산이온(IO_3^-), 요도드화이온(I^-)과 브롬화이온(Br^-)의 혼합물을 전위차 적정법으로 定量하였다. 사용한 銀 이온 選擇性 電極은 直接 製作한 Ag_2Se電極 이었다. 混合物 電位差 滴定을 할때는 理論的인 當量點과 變曲點의 誤差를 감소시키기 爲하여 뭉침제로서 에칠알콜, 질산나트륨, 덱스트린, 니트로벤젠 등을 사용하였다. 뭉침제를 사용함으로서 I^-과 IO_3, Br^-와 IO-3^-, 또한 Br^-, I^-, IO_3^-의 混合物 定量時 第一, 第二變曲點에서 誤差를 줄일 수 있었으며 비교적 좋은 結果를 얻었다. Potentiometric titration of iodide, bromide and iodate in mixtures, bromate, iodate with silver nitrate using silver ion-selective electrode, indicator electrode, has been studied. Silver ion-selective electrode was a sintered silver selenide electrode which had been prepared in this laboratory. In the titration of mixture solution, early alcohol, sodium nitrate, dextrin, and nitrobenzene as flocculant have been used to decrease the relative error of inflection point to theoretical equivalent point. The relative error of first and second inflection point in the titration of iodide and iodate, bromide and iodate, bromide, iodide, and iodate, respectively, was decreased, and attained the good results.

      • 물과 Micelle 용액에서 Benzaldehyde의 전기화학적 환원에 대한 연구

        The electrochemical reduction of benzaldehyde in 10% ethanol-water solution contained surfactants has been studied by direct current (DC) differential pulse (DP) polarography, cyclic voltammetry (CV) and controlled potential coulometry (CPC). A totally irreversible electrode reaction processes of benzaldehyde reduction proceeded by one electron transfer to form anion radical which undergoes both protonation and a second one electron reduction to give benzyl alcohol as a major product. The cathodic reduction wave was seperated by inhibition of sodium lauryl sulfate due to selectivity and effect of anionic charged micelle. The possible reduction mechanism with controlled pH range was suggested.

      • The Study on the Structure of Nickel (Ⅱ) Isonitrosoacetylacetone imine Complexes

        Nickel(II) Complexes of the types Ni(R-IAI)(IAT')have been prepared. Where IAI' represents isonitrosoacetylacetone imine and R-IAI represents its N-alkyl derivatives. The molecular weight, magnetic moment, and IR, NMR, of these Complexes have been studied. It is suggested that the isonitroso group of R-IAI coordinates through the Nitrogen and that of IAI' through the Oxygen in Ni(R-IAI)(IAI'). The amine-exchange reactions of Nickel(II) complexes are discussed and compared on the basis of their structres.

      • Studies on the Bio-Electrode for L-Histidine

        L-Histidine에 對한 電位差感應器는 암모니아氣體 感應電極에 박테리아 Pseudomonas sp.를 固定시킴으로서 만들었다. 박테리아 효소는 Histidine의 每分子로부터 두分子의 암모니아로 變換시킨다. 박테리아 電極의 感應은 ?? Histidine에서 直線을 나타내고 그 기울기는 48-53 mV/decade이다. 이 感應器의 壽命은 三週程度이고, 적당한 選擇性을 보여준다. 박테리아를 固定시키데는 살균된 filter를 使用한 利點과 感應을 改善하기 爲하여 처음의 熱處理에 對하여 論議하였다. A potentiometric sensor for L-histidine has been made by immobilizing the bacterium Pseudomonas sp. on an ammonia gas-sensing electrode. The bacterial enzymes convert each molecule of histidine into two molecules of ammonia. The response of the bacterial electode is linear between ?? histidine with a slope of 48-53mV/decade. The sensor has a lifetime of three weeks and shows moderate selectivity. The advantages of using a sterilizing filter for immobilizing of bacteria and an initial heat treatment to improve the response are discussed.

        Microbial BOD Sensor Using Hansenula anomala

        A microbial sensor for BOD (Biochemical Oxygen Demand) measurement has been developed by immobilizing Hansenula anomala in a polyacrylamide gel. The optimum pH and temperature for BOD measurement using this sensor were pH 7.0 and $30^{\circ}C$, respectively. The response time was 30 min. A linear relationship was observed between the potential and the concentration below 44 ppm BOD. The potential was reproducible within ${\pm}9%$ of the relative error when a sample solution containing 20 mg/l of glucose and 20 mg/l of glutamic acid was employed. The effect of various compounds on BOD estimation was also examined. The potential output of the sensor was almost constant for 30 days. The relative error in BOD estimation was within ${\pm}10%$.

        Ferricyanide-Sensing Electrodes

        The Ferricyanide-sensing electrodes were prepared with $Ag_2S\;and\;Ag_3Fe(CN)_6$ (mole ratio 3:1-7:1). The 5:1 $(Ag_2S:Ag_3Fe(CN)_6)$ composition is superior to others in terms of potentiometric response, rapidity of response and reproducibility. Testing was done over the concentration range $10^{-1}M{\sim}10^{-6}M\; Fe(CN)_6^{3-}$ at pH 6.8 with constant ionic strength. The concentration-potential curve was linear and coincided with the Nernstian slope (19.7 mV/decade). Interfering ions were $I^-,\;Br^-,\; SCN^-\;and\;Fe(CN)_6^{4-}$ and the life time of this electrode was 3 weeks. This electrode could be used as the indicator electrode for measuring the ferricyanide and ferrocyanide.
