The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor triggered by xenobiotics such as 3-methylcholanthrene (3MC). The hypoxia-inducible factor-1a1pha (HIF-1a1pha) is a conditionally regulated transcription factor activated by low oxygen conditions. Since both transcription factors share their common partner, AhR nuclear translocator (ARNT) to form heterodimeric complex for AhR- and hypoxia-mediated signaling, activation of the HIF-1alpha may lead to interference with 3MC induced AhR signaling by sequestering ARNT. To test this hypothesis, wild type zebrafish embryos and transgenic zebrafish embryos that express GFP in response to AhR ligands were exposed to 3MC (60 µM) at 72 h after fertilization for 3 h and then transferred to hypoxic condition caused by cobalt chloride (10-4 M). Semiquantative RT-PCR PCR analysis showed that hypoxic conditions by cobalt chloride decreased 3MC–induced cyp1a1 mRNA expression in wild type zebrafish embryos. GFP expression of 3MC-induced transgenic zebrafish embryos were decreased in hypoxic conditions. Our results demonstrated that hypoxic conditions inhibited 3MC-induced AhR signaling. But it is not clear whether this is mediated through competition for ARNT or through other mechanism.
Cadmium-Induced transient expression of a reporter gene in zebrafish (Danio rerio)

Cadmium (Cd) is a serious environmental pollutant and can cause a variety of adverse effects in humans and animals. Heat shock proteins (HSPs) play a major role in a self-protective mechanism after exposure to heat shock, toxic chemicals, and other environmental stresses. Recently, our laboratory developed a heat shock responsive transgenic zebrafish that expresses green fluorescence protein (GFP) after exposure to copper sulfate. In this study, we showed Cd toxicity on zebrafish larvae and tested responsiveness of our transient zebrafish system to Cd exposure. Cd treated larvae showed morphologic abnormalities such as spinal lordosis and kyphosis. Cd induced enhanced green fluorescence protein (EGFP) reporter gene expression in pHhsp70-EGFP transfected CHSE 214 cells, with EGFP expressed in a dose-responsive manner. Embryos injected with pHhsp-EGFP were exposed to DMSO (0.01%) or Cd (low dose: 2 µM; high dose: 20 µM) 72 h after fertilization for 3 h. Cd induced EGFP expression in skin epithelium, olfactory epithelium, retina, liver, pronephric ducts, and myotubes. These EGFP expressing tissues are related to absorption and detoxification of Cd. Our transient zebrafish biosensor could be utilized as a biomarker, indicating the presence of environmental toxicants, in an early warning system.

<P><B>Abstract</B></P><P>Skin ulcers, scoliosis, and dropsy-like scale edema were observed in laboratory-maintained zebrafish. Affected fish had multifocal granulomas not only in internal organs such as the liver, intestine, genital organs, kidney, muscle, and spleen but also in the fin, epithelium, gills, and sclera of the eyes. Large numbers of acid-fast-rod-shaped bacteria were observed within the necrotic centers of well-demarcated, multifocal granulomas with Gram's stain and Ziehl–Neelson's stain. The size of the <I>Mycobacterium</I> spp. was 1–2μm×2–3μm with a double-layered cell wall, based upon electron-microscopical features. Definitive diagnosis of these outbreaks was obtained by culture on selective media followed by PCR-restriction fragment length polymorphism analysis (PRA) of the <I>rpoB</I> gene for species identification. The amplified 360-bp products of the <I>rpoB</I> gene of mycobacteria isolated from zebrafish were digested with <I>Msp</I>I restriction enzyme, which revealed unique band patterns matching those of <I>Mycobacterium abscessus</I> and <I>Mycobacterium chelonae</I> which are responsible for skin and soft tissue infection caused by rapidly growing mycobacteria in humans. This is the first documentation of the precise identification of zoonotic non-tuberculous mycobacteria isolated from laboratory-maintained zebrafish by the PRA of the <I>rpoB</I> gene; this study thus provides a great deal of useful epidemiological information and reduces the likelihood that epizootics will occur.</P>

<P><B>Abstract</B></P><P><B>Aim: </B> We investigated the accuracy of liver stiffness measurement (LSM) in chronic hepatitis C (CHC) in a multicenter, prospective study in South Korea.</P><P><B>Methods: </B> Between June 2005 and July 2009, 91 CHC patients without a previous history of antiviral treatment, clinical evidences of cirrhosis, coinfection with other viruses, and heavy alcohol consumption and with alanine aminotransferase (ALT) ≤ 5x upper limit of normal, total bilirubin ≤ 1.5 mg/dL, sufficient liver biopsy quality (≥ 15 mm and more than six portal tracts), interquartile range to median liver stiffness (LS) value ratio ≤ 0.21, and more than 10 valid measurements, were recruited. The Batts and Ludwig scoring system was used for histologic assessment. Age–platelet index (API), aspartate aminotransferase (AST)–to–platelet ratio index (APRI), and age–spleen–platelet ratio index (ASPRI) were calculated. Area under the receiver operating characteristic curve (AUROC) was used to evaluate the performance of LSM and other noninvasive models.</P><P><B>Results: </B> The mean age was 47.9 years, and the mean LS value was 7.7 kPa (44 men and 47 women). LS value was highly correlated to the fibrosis stages (<I>r</I> = 0.835, <I>P</I> < 0.001). The AUROCs of LSM were 0.909 for ≥ F2, 0.993 for ≥ F3, and 0.970 for F = 4 and were superior to those of API (0.72, 0.858, and 0.948, respectively), APRI (0.780, 0.887, and 0.904, respectively), and ASPRI (0.713, 0.862, and 0.957, respectively). The optimal cutoff LS values were 6.2 kPa for ≥ F2, 7.7 kPa for ≥ F3, and 11.0 kPa for F = 4.</P><P><B>Conclusions: </B> Our data suggest that LSM can accurately assess liver fibrosis in patients with CHC and be applied in South Korea.</P>
Tripartin, a Histone Demethylase Inhibitor from a Bacterium Associated with a Dung Beetle Larva

<P>Tripartin (<B>1</B>), a new dichlorinated indanone, was isolated from the culture broth of the <I>Streptomyces</I> sp. associated with a larva of the dung beetle <I>Copris tripartitus</I> Waterhouse. The planar structure of tripartin (<B>1</B>) was identified by the spectroscopic analyses of NMR, mass, UV, and IR data. The structure was confirmed, and the absolute configuration of <B>1</B> was determined by X-ray crystallography. 원발성 난관암은 빈도가 드문 암의 하나로 여성 생식계 암의 약 1% 정도를 차지한다. 난관암은 조직학적 소견과 임상적인 성향이 난소암과 유사하며, 그 5년 생존율은 약 30-50%로 보고되고 있다. 임상 증상이 비특이적이며, 빈도가 낮아 수술전 진단이 어렵다. 또한 아직까지 질환 특유의 치료가 정립 되지 않았으나 병기 설정과 치료방법이 난소암과 유사하게 이루어지고 있다. 본 저자들은 부속기 종괴와 질출 혈로 외래 방문한 폐경기여성에서 각각 자궁내막종과 자궁내막염을 의심하였으나 시험개복술에서 조직학적으로 확진된 원발성 난관암 2 증례를 경험하였기에 간단한 문헌 고찰과 함께 이를 보고하는 바이다.
Cryptosporidium Infection in Animal Facilities in Korea

We investigated the presence of Cryptosporidium in four specific pathogen free (SPF) and four conventional facilities in Korea. From November 2000 to March 2005, 329 mice were histopathologically examined for Cryptosporidium infection, and infection was detected in 18.52% (5 of 27) and 3.68% (7 of 190) animals in 2000 and 2004, respectively. The detected Cryptosporidium was classified based on oocyst size and the parasitized organ. Oocysts were predominantly round to oval (measured by light microscopy, 5~12×5~10 ㎛) and parasitized the stomach. To confirm Cryptosporidium infection, PCR was performed to detect genes of Cryptosporidium parvum and Cryptosporidium oocyst wall protein. Our results suggest that Cryptosporidium had infected conventional mice, but that it had not infected SPF mice in Korea. The detected Cryptosporidium was presumed to be C. muris based on oocyst size and parasitized organ.
